      • The Mathematic Model of "Pressing Complexion" Differential Coefficient Countermeasure Decision for Collision-avoidance

        In this article, we have done some analysis about the collision-avoidance specialty of "pressing complexion" and "pressing danger" in the meet process of two boats, and offered a mathematic model of differential coefficient countermeasure decision for collision-avoidance. which adapt to the right complexion. The basal idea is, in the right condition whatever do the coming boat do, and our boat will always adopt dynamic, continuous and the best countermeasure. When both the controlling capabilities of two boats have advantage and inferior position, we can working-out with the qualitative differential coefficient countermeasure.

        MRI T2-Weighted Imaging and Fat-Suppressed T2-Weighted Imaging Image Fusion Technology Improves Image Discriminability for the Evaluation of Anal Fistulas

        Objective: To explore whether MRI fusion technology (combined T2-weighted imaging [T2WI] and fat-suppressed T2WI [T2WI-FS]) improves signal differences between anal fistulas and surrounding structures. Materials and Methods: A total of 32 patients with confirmed diagnoses of anal fistula were retrospectively studied. All available T2WI and T2WI-FS images for each patient were used to generate fusion image (T2WI-Fusion) based on the addition of gray values obtained from each pixel via an MR post-processing work station. The discriminability of fistula, perianal sphincter, and perianal fat in T2WI, T2WI-FS, and T2WI-Fusion images was quantified with Fisher’s scoring algorithm. For subjective visual image assessment by researchers, five-point scale scores were determined using a modified double-stimulus continuous qualityscale test to evaluate T2WI-FS, T2WI, enhanced axial three-dimensional-volumetric interpolated breath-hold examination (3D-VIBE), and T2WI-Fusion sequence images. The differences were subsequently compared. Results: Mean Fisher scores for fistulas vs. sphincters obtained from T2WI-Fusion (FFusion-fistula = 6.56) were significantly higher than those from T2WI (FT2WI-fistula = 3.35) (p = 0.001). Mean Fisher scores for sphincters vs. fat from T2WI-Fusion (FFusion-sphincter = 10.84) were significantly higher than those from T2WI-FS (FSFS-sphincter = 2.57) (p = 0.001). In human assessment, T2WI-Fusion showed the same fistula discriminability as T2WI-FS, and better sphincter discriminability than T2WI. Overall, T2WI-Fusion showed better discriminability than T2WI, T2WI-FS, and enhanced 3D-VIBE images. Conclusion: T2WI and T2WI-FS fusion technology improves signal differences between anal fistulas and surrounding structures, and may facilitate better evaluation of anal fistulas and sphincters.

        Comparison of drought resistance of rootstocks 'M9-T337' and 'M26' grafted with 'Huashuo' apple

        Drought stress is one of the main limiting factors in apple (Malus domestica Borkh.) cultivation. Rootstock plays an important role in the drought tolerance of apple plants. ‘M.9-T337’ is a novel apple rootstock that was recently introduced and widely cultivated in China. In this study, we selected the new, widely promoted Chinese apple variety ‘Huashuo’ as the scion and grafted it onto ‘M.9-T337’ (HM9). Another combination, ‘Huashuo’/‘M.26’/Malus robusta Rehd. (HM26), served as the experimental control to analyse drought resistance in the two hybrids. We believe that this empirical approach is more representative than merely studying rootstock seedlings. After sustained drought stress for over 1 month, the leaf relative water content had decreased in both types of plants, but to a lesser extent in HM26 than in HM9. The SPAD values increased in both plants, but without significant difference. Drought stress reduced photosynthetic activity in both plants, and the net photosynthetic rate was higher in HM26 than in HM9. The observed changes in the chlorophyll fluorescence parameters indicated that drought had damaged the PSII activity centres of both plants, photosynthetic electron transfer was inhibited, and excessive excitation energy accumulated. However, compared to HM26, HM9 displayed lower maximal PSII quantum photochemical efficiency and potential PSII activity. Moreover, HM9 showed lower antioxidant enzyme activity than HM26 under drought stress. A membership function analysis confirmed that ‘M.9-T337’ was less drought resistant than ‘M.26’. Nevertheless, ‘M.9-T337’ could still recover after prolonged drought stress, indicating it also had good drought resistance.

        Kinetics of Transesterification of 1,4-Butanediol With Methyl Acetate by the Ion-exchange Resin

        The dihydric alcohol transesterification of 1,4-butanediol with methyl acetate catalyzed by the ion-exchange resin is researched. The chemical system involves two transesterification reactions in series, with 1,4-butanediol monoacetate as an intermediate product. The effects of the catalyst type, catalyst size, catalyst loading, stirrer speed, the initial reactant ratio, and the reactive temperature have been meticulously studied. The results show that this consecutive transesterification is exothermic and the experimental values of reaction enthalpy are −8.50 and −6.85 kJ/mol, which are in good agreement with the values computed from the standard formation enthalpy. Three kinetic models (PH, LH, and ER) are applied to correlate the experimental data, of which ER model gives the best result with the lowest mean relative error. The activation energies are calculated to be 38.53 and 51.06 kJ/mol by ER model, demonstrating that the overall reaction rates are controlled by the reaction on the catalyst surface.

        Dynamic analysis for gene expression profiles of endothelial colony forming cells under hypoxia

        Previous studies have shown that endothelial colony forming cells (ECFCs) play an important role in the neovascularization of tumors. Hypoxia is emphasized as an important promoter of angiogenesis. However, little is known about genome-wide transcriptional regulation of ECFCs under hypoxic conditions. In this study, gene expression profiles in ECFCswere evaluated under hypoxic conditions for 3, 6, 12, 24,and 48 h, using Affymetrix U133 plus 2.0 chip microarray. 1,103 hypoxia-regulated genes were filtered, with 379(0.693 %) genes up-regulated and 724 (1.32 %) genes downregulated. Most of the up-regulated genes were involved in apoptosis, cell proliferation, or metabolic processes, while most of the down-regulated genes were involved in cell adherence,cell cycle,DNAandmRNAmetabolic processes,multi-cellular organism development, protein metabolic processes, response to stress, signal transduction, or transport. This expression profile is ECFC-specific, because it is significantly different from those of endothelial cells and smooth muscle cells under hypoxic conditions. Moreover, hypoxia-regulated apoptosis in ECFCs is mainly related with the mitochondrial pathway (p53-BAX-Caspase-9) and the death receptor pathway (FASCaspase-8-Caspase-3). MAPK pathway is activated in ECFCs under hypoxic conditions. The differentially expressed genes of ECFCs were identified under hypoxic conditions, and related with cell apoptosis, cell cycle and MAPK pathways, shedding light on the mechanism of angiogenesis.

        Molecular Cloning and Expression of cDNA Encoding the Cysteine Proteinase Inhibitor from Upland Cotton

        A cDNA encoding a novel cysteine proteinase inhibitor (CPI) was isolated from a gland mutant Xiangmian-18 of upland cotton during the pigments gland forming stage. The cDNA comprises 378 bp and encodes 125 amino acid residues with molecular mass of 13.8 kDa. It contains the conserved motif of cysteine protease inhibitors and belongs to the cystatin superfamily (Gln- Val-Val-Ala-Gly). The deduced amino acid sequences of the domains are highly similar to the normal upland cotton (96.8%). SDS-PAGE and western hybridization analysis showed that the expressed recombinant protein was recombinant CPI. The inhibitory activity of recombinant CPI was 46 u/μg which was measured by inhibiting the protease activity of papain. RT-PCR results indicated that the expression level of developing gland stage was higher than that of undeveloped gland stage.

        Identification of high-affinity VEGFR3-binding peptides through a phage-displayed random peptide library

        Objective: Vascular endothelial growth factor (VEGF) interaction with its receptor, VEGFR-3/Flt-4, regulates lymphangiogenesis. VEGFR-3/Flt-4 expression in cancer cells has been correlated with clinical stage, lymph node metastasis, and lymphatic invasion. The objective of this study is to identify a VEGFR-3/Flt-4-interacting peptide that could be used to inhibit VEGFR-3 for ovarian cancer therapy. Methods: The extracellular fragment of recombinant human VEGFR-3/Flt-4 (rhVEGFR-3/Flt-4) fused with coat protein pIII was screened against a phage-displayed random peptide library. Using affinity enrichment and enzyme-linked immunosorbent assay (ELISA) screening, positive clones of phages were amplified. Three phage clones were selected after four rounds of biopanning, and the specific binding of the peptides to rhVEGFR-3 was detected by ELISA and compared with that of VEGF-D. Immunohistochemistry and immunofluorescence analyses of ovarian cancer tissue sections was undertaken to demonstrate the specificity of the peptides. Results: After four rounds of biopanning, ELISA confirmed the specificity of the enriched bound phage clones for rhVEGFR-3. Sequencing and translation identified three different peptides. Non-competitive ELISA revealed that peptides I, II, and III had binding affinities for VEGFR-3 with Kaff (affinity constant) of 16.4±8.6 μg/mL (n=3), 9.2±2.1 μg/mL (n=3), and 174.8±31.1 μg/ mL (n=3), respectively. In ovarian carcinoma tissue sections, peptide III (WHWLPNLRHYAS), which had the greatest binding affinity, also co-localized with VEGFR-3 in endothelial cells lining lymphatic vessels; its labeling of ovarian tumors in vivo was also confirmed. Conclusion: These finding showed that peptide III has high specificity and activity and, therefore, may represent a potential therapeutic approach to target VEGF-VEGFR-3 signaling for the treatment or diagnosis of ovarian cancer.

        Profiling Gene Expression During Gland Morphogenesis of a Glanded and a Glandless Upland Cotton

        The pigment gland is an important character of the Gossypium plant. With the aim of identifying genes involved in pigment gland morphogenesis in cotton, gene expression during pigment gland morphogenesis in Chuan 2802, which is glanded both in seed and plant, and a glandless line N5 was profiled using Affymetrix Cotton microarray. The results showed that there were 564 differentially expressed genes greater than twofold during gland morphogenesis. About 60.2% of these genes shares similarity with known genes on GenBank and about 39.8% with no functional description in the database. These described genes may play roles in defense response, response to oxidative stress, peroxidase activity, and the other metabolic pathways. The KEGG Orthology-Based Annotation System indicated that these above twofold expressed genes involved seven biochemical pathways on KEGG. These findings suggest that a complicated regulation is associated with pigment gland morphogenesis and the associated defense response including gossypol biosynthesis in cotton.

        Baicalin attenuates TNBS-induced colitis in rats by modulating the Th17/Treg paradigm

        Baicalin, a flavonoid, has a wide range ofpharmacological properties, including immunomodulation. The objective of this study was to investigate the effect ofbaicalin on the balance of T helper 17 (Th17) and regulatoryT (Treg) cells in a colitis model. The rat colitis modelwas induced by 2,4,6-trinitrobenzene sulfonic acid(TNBS). Baicalin (10 ml/kg, each) or mesalazine (positivecontrol) was then administered orally for 7 days. Inflammatoryand immunological responses were evaluated bypathology, enzyme-linked immunosorbent assay, real-timepolymerase chain reaction, western blot analysis, and flowcytometry. Our study showed that baicalin not only significantlyattenuated TNBS-induced colitis by reducing thedisease activity index as well as macroscopic and microscopicscores, but it also improved the weight loss andshortening of the colon. Baicalin treatment also induced asignificant decrease in the levels of inflammatory mediators,including the myeloperoxidase activity, the levels oftumor necrosis factor a, IL-1b, and Th1-related cytokinesIL-12 and IFN-c. Furthermore, the beneficial effects ofbaicalin seem to be associated with regulation of the Th17and Treg paradigm. We found that administration ofbaicalin significantly downregulated the number of Th17cells and the levels of Th17-related cytokines (IL-17 andIL-6) and retinoic acid receptor-related orphan receptor ct. In contrast, there was an increase in Treg cells numbers,Treg-related cytokines transforming growth factor-b andIL-10, and forkhead box P3. Our results suggest that theanti-inflammatory effect of baicalin may be linked tomodulation of the balance between Th17 and Treg cells inTNBS-induced ulcerative colitis.

      • Senescence as A Consequence of Ginsenoside Rg<sub>1</sub> Response on K562 Human Leukemia Cell Line

        Aims and Background: Traditional chemotherapy strategies for human leukemia commonly use drugs based on cytotoxicity to eradicate cancer cells. One predicament is that substantial damage to normal tissues is likely to occur in the course of standard treatments. Obviously, it is urgent to explore therapies that can effectively eliminate malignant cells without affecting normal cells. Our previous studies indicated that ginsenoside $Rg_1$ ($Rg_1$), a major active pharmacological ingredient of ginseng, could delay normal hematopoietic stem cell senescence. However, whether $Rg_1$ can induce cancer cell senescence is still unclear. Methods: In the current study, human leukemia K562 cells were subjected to $Rg_1$ exposure. The optimal drug concentration and duration with K562 cells was obtained by MTT colorimetric test. Effects of $Rg_1$ on cell cycle were analyzed using flow cytometry and by SA-${\beta}$-Gal staining. Colony-forming ability was measured by colony-assay. Telomere lengths were assessed by Southern blotting and expression of senescence-associated proteins P21, P16 and RB by Western blotting. Ultrastructural morphology changes were observed by transmission electron microscopy. Results: K562 cells demonstrated a maximum proliferation inhibition rate with an $Rg_1$ concentration of $20{\mu}\;mol{\cdot}L^{-1}$ for 48h, the cells exhibiting dramatic morphological alterations including an enlarged and flat cellular morphology, larger mitochondria and increased number of lysosomes. Senescence associated-${\beta}$-galactosidase (SA-${\beta}$-Gal) activity was increased. K562 cells also had decreased ability for colony formation, and shortened telomere length as well as reduction of proliferating potential and arrestin $G_2$/M phase after $Rg_1$ interaction. The senescence associated proteins P21, P16 and RB were significantly up-regulated. Conclusion: Ginsenoside $Rg_1$ can induce a state of senescence in human leukemia K562 cells, which is associated with p21-Rb and p16-Rb pathways.
