        Effect of six sugars on the longevity, oviposition performance and nutrition accumulation in an endoparasitoid, Meteorus pulchricornis (Hymenoptera: Braconidae)

        The effects of six sugar resources (fructose, glucose, sucrose, trehalose, raffinose and honey) on the longevity, oviposition performance and nutrition levels of Meteorus pulchricornis, a thelytokous larval endoparasitoid of the common cutworm Spodoptera litura were examined under laboratory conditions. Female adults of M. pulchricornis fed 1 M fructose, glucose, trehalose or sucrose solutions survived longer than those fed on other sugar solutions or water. When provided with honey or sucrose solutions, the female parasitoids laid more offspring than those fed other sugar diets or the control. The body size of offspring driven from honey-, fructose-, sucrose-, and glucose-fed females, along with water-fed group, were larger than the trehalose- and raffinose-fed females. However, the emergence rates of all offspring generated from different sugars- and water-fed females were similar. When separately given honey, sucrose or fructose, M. pulchricornis females accumulated fructose at a higher level than the other groups. Parasitoid wasps fed trehalose solution accumulated the highest level of total sugar. Glycogen levels and lipid content were highest at emergence and then decreased across all diets. In addition, females fed on trehalose had the highest level of glycogen compared to other sugar diets and water control regardless of emergency level. Females fed trehalsoe, fructose, and glucose solutions had a higher level of lipid than those fed other sugar solutions and water at life end. The outcome of this study can benefit both laboratory rearing and management interventions that improve sugar sources for the parasitoid in the field.

        ELECTROCHEMICAL Co 3 O 4 NANOPOROUS THIN FILMS SENSOR FOR HYDROGEN PEROXIDE DETECTION

        The nanoporous Co 3 O 4 thin ¯lms were prepared on indium tin oxide (ITO) glasses by anelectrodeposition method. The surface morphology and composition of the nanoporous Co 3 O 4¯lms were characterized by scanning electron microscopy (SEM), energy-dispersive X-rayanalysis (EDS) and X-ray photoelectron spectroscopy (XPS). The results show that the as-deposited nanoporous Co 3 O 4 ¯lm is constructed by many interconnected nano°akes withthickness of about 40 nm. The cyclic voltammetry (CV) measurement indicates that thenanoporous Co 3 O 4 ¯lms exhibit remarkable electrocatalytic activities for the hydrogen peroxide(H 2 O 2 ) reduction which shows that it is a good candidate to be employed as electrode materialsfor electrochemical sensing of H 2 O 2 . Further analysis indicated that the detection sensitivityof the sensor was 1.357 mA mM? 1 cm? 2 and the detection limit was estimated to be about0.2 mM.

        The Effect of Agomelatine in Behavioral and Psychological Symptoms of Dementia

        Objective: Moderate and severe behavioral and psychological symptoms of dementia (BPSD) often need medical treatment to improve symptoms. Agomelatine is a selective melatonergic (MT1/MT2) agonist that has normalizing effects on disturbed circadian rhythms and disrupted sleep−wake cycles. Its activity of 5HT-2C receptor antagonism is associated with lessening depression and anxiety and increasing slow-wave sleep. Based on past clinical records and current findings it suggests that agomelatine can improve BPSD for patients. This retrospective cohort study was designed to compare the BPSD before and after using agomelatine. Methods: Records of dementia cases who had ever received agomelatine treatment for BPSD in a general hospital setting during the past 2.5 years were identified and reviewed. Scores from before and after 3 months of treatment with agomelatine were collected for Neuropsychiatric Inventory (NPI), Brief Psychiatric Rating Scale (BPRS), and Clinical Global Impression (CGI) to compare and analyze the difference of psychological and behavioral symptoms pre- and post-agomelatine used. Results: Records of 144 cases of dementia with BPSD who had ever used agomelatine from January 2015 to June 2017 were collected. All of the 112 cases had BPRS and CGI scores, of which 75 cases had additional NPI scores. Among these 112 cases, the BPRS and CGI scores were significantly improved in all types of dementia. NPI scores indicated that the use of agomelatine alleviated obvious symptoms and decreased overall distress, especially in the depression/poor mood, anxiety, and sleep/night behavior. Conclusion: It is consistent with an effective result of agomelatine in improving BPSD.

        Parkinson’s Disease with Fatigue: Clinical Characteristics and Potential Mechanisms Relevant to α-Synuclein Oligomer

        Background and Purpose The aim of this study was to identify the clinical characteristics and potential mechanisms relevant to pathological proteins in Parkinson’s disease (PD) patients who experience fatigue. Methods PD patients (n=102) were evaluated using a fatigue severity scale and scales for motor and nonmotor symptoms. The levels of three pathological proteins—α-synuclein oligomer, β-amyloid (Aβ)1-42, and tau—were measured in 102 cerebrospinal fluid (CSF) samples from these PD patients. Linear regression analyses were performed between fatigue score and the CSF levels of the above-listed pathological proteins in PD patients. Results The frequency of fatigue in the PD patients was 62.75%. The fatigue group had worse motor symptoms and anxiety, depression, and autonomic dysfunction. The CSF level of α-synuclein oligomer was higher and that of Aβ1-42 was lower in the fatigue group than in the non-fatigue group. In multiple linear regression analyses, fatigue severity was significantly and positively correlated with the α-synuclein oligomer level in the CSF of PD patients, after adjusting for confounders. Conclusions PD patients experience a high frequency of fatigue. PD patients with fatigue have worse motor and part nonmotor symptoms. Fatigue in PD patients is associated with an increased α-synuclein oligomer level in the CSF

        Effects of Low Level Water-soluble Pentosans, Alkaline-extractable Pentosans, and Xylanase on the Growth and Development of Broiler Chicks

        This study investigated the effects of low levels of water-soluble pentosans (WSP), alkaline-extractable pentosans (AEP), and xylanase on the growth and organ development of broiler chicks. Three hundred and fifty 1-d-old female broiler chicks were randomly allocated into seven experimental groups of five pen replicates, with ten chicks per replicate. The control group consumed a corn-soybean meal-based diet. Six dietary treatment groups consumed the basal diet supplemented with one of the following: WSP at 50 mg/kg (WSP50) or 100 mg/kg (WSP100); AEP at 50 mg/kg (AEP50) or 100 mg/kg (AEP100); or xylanase at 3 mg/kg (Xase3) or 6 mg/kg (Xase6). Data including the body weight, digestive organ weights, gut length, rectal digesta viscosity, and gut microflora and pH were collected on d 5, 10, and 15. When compared to the control group, WSP50 promoted body weight gain and organ growth throughout the study, calculated as 3-d averages (p<0.05). WSP100 increased weight gain and enhanced organ development (proventriculus, gizzard, and gut) on d 10 (p<0.05), but the 3-d averages were not different from the control group except for the weight of gizzard. Both Xase3 and Xase6 increased the 3-d average weight gain and the growth of the gizzard (p<0.05). WSP50 increased the digesta viscosity compared to Xase3 on d 10 and 15 (p<0.05). WSP50, Xase3, and Xase6 increased the concentration of Lactobacillus in the rectum when compared to the control group (p<0.05), but only Xase3 lowered the digesta pH in the ileum and cecum on d 10 and 15. AEP had minimal influence on the growth and organ development of broilers. The results showed that low levels of WSP, AEP, and xylanase had different effects and underlying mechanisms on the growth and organ development of broiler chicks. WSP50 could increase the growth performance of broilers fed a corn-soybean meal-based diet.

      • Highly Selective Colorimetric Signaling of Iron Cations Based on Fluoran Dye

        <P> A highly selective colorimetric chemosensor based on fluoran dye for iron cations, 2'-anilino-3'methyl-6'-dibuthylamino-N-(2'-(2''-hydroxybenzylideneamino)ethyl) iso-indolin-1-one-fluoran (5), was designed and synthesized. The chemical structures of all the intermediates and fluoran dye 5 were characterized by 1H-NMR, 13C-NMR, MS and elemental analysis. And its sensing behavior toward metal ions was investigated by UV-visible absorption spectroscopes. The fluoran dye 5 chemosensor showed an extreme selectivity for Fe2+ and Fe3+ in acetonitrile solution. Whereas other ions including Mg2+, Pd2+, Ni2+, Hg2+, Cd2+, Ag+, Cu2+, Zn2+ and Al3+ induced basically no spectral change, which constituted a Fe2+ highly sensitive and selective colorimetric chemosensor from colorless to black by “naked eyes.” Spectral responses at &lgr;max = 653 nm reveals that dye 5 can function as OR logic gate with Fe2+ and Fe3+ as input variables.</P>

      • Gene Silencing of β-catenin by RNAi Inhibits Proliferation of Human Esophageal Cancer Cells by Inducing G0/G1 Cell Cycle Arrest

        Objectives: The aim of the present study was to explore mechanisms underlying the effects of down-regulating ${\beta}$-catenin expression on esophageal carcinoma (EC) cells. Methods: Cell cycle distribution and apoptosis were determined using flow cytometry and annexin V apoptosis assay, respectively. Transmission electron microscopy (TEM) was used to examine changes in ultrastructure, while expression of cyclin D1 protein and mRNA was detected by western blot and real-time PCR. Proliferating cell nuclear antigen (PCNA) and extracellular signal-regulated kinase (ERK) 1-2 were evaluated by Western blot analysis. PCNA labeling index (LI) was determined by immunocytochemistry. Results: Compared with pGen-3-con transfected and Eca-109 cells, the percentage of G0/G1-phase pGen-3-CTNNB1 transfected cells was obviously increased (P<0.05), with no significant difference among the three groups with regard to apoptosis (P>0.05). pGen-3-CTNNB1 transfected cells exhibited obvious decrease in cyclin D1 mRNA and protein expression (P<0.05) and the ultrastructure of Eca-109 cells underwent a significant change after being transfected with pGen-3-CTNNB1, suggesting that down-regulating ${\beta}$-catenin expression can promote the differentiation and maturation. The expression of PCNA and the ERKI/2 phosphorylation state were also down-regulated in pGen-3-CTNNB1 transfected cells (P<0.05). At the same time, the PCNA labeling index was decreased accordingly (P<0.05). Conclusion: Inhibition of EC Eca-109 cellproliferation by down-regulating ${\beta}$-catenin expression could improve cell ultrastructure by mediating blockade in G0/G1 through inhibiting cyclin D1, PCNA and the MAPK pathway (p-ERK1/2).

      • Human Papillomavirus (HPV) Infection in Women Participating in Cervical Cancer Screening from 2006 to 2010 in Shenzhen City, South China

        Purpose: Human papillomavirus (HPV) infection plays an important role in the development of cervical cancer, but the prevalence of HPV infection in women of Shenzhen city remains unclear. The present study was performed to describe the change of cervical HPV infection in females who participated in voluntary cervical cancer screening from 2006 to 2010 in Shenzhen city, China. Methods: A total of 4, 413 women were recruited. HPV infections were genotyped by polymerase chain reaction (PCR) and reversed dot blot hybridization in Shenzhen Maternity and Child Health Hospital. Results: The prevalence of HPV infection was 13.8%. The five most commonly found HPV types were HPV16 (3.47%), HPV58 (1.68%), HPV33 (1.38%), HPV43 (1.36%) and HPV18 (1.27%). The secular trends of major HPV type-specific were diverse. Among of them, the prevalence of HPV18 increased sharply while others increased slowly or even decreased in the period. The change of total HPV, single HPV and multiple HPV infection were similar during the five years. Conclusions: Our findings suggested that HPV infection is common with HPV16 and HPV 58 as the primary subtypes in women in Shenzhen city.The prevalence of HPV 18 infection is increasing faster than any others, which will lead it to be one of the main subtypes in this city in the future.

      • Effect of TLR4 and B7-H1 on Immune Escape of Urothelial Bladder Cancer and its Clinical Significance

        Background/Aim: Toll-like receptor 4 (TLR4) and B7-H1, both normally expressed restricted to immune cells, are found to be aberrantly expressed in a majority of human tumors and may play important roles in regulation of tumor immunity. It has been shown that urothelial bladder cancer (UBC) patients can manifest tumoral immune escape which may be a potential critical factor in tumor pathogenesis and progression. However, so far, the mechanisms of UBC-related immune escape have not been clarified. The aim of this study was to investigate the effect of TLR4 and B7-H1 on immune escape of UBC. Methods: Bladder cancer T24 cells were pre-incubated with LPS and co-cultured with tumor specific CTLs. CTL cytotoxicity and apoptosis rates were measured by MTT assay and flow cytometry, respectively. The effects of an ERK inhibitor on B7-H1 expression and CTL cytotoxicity against T24 cells were also evaluated. In addition, TLR4, B7-H1 and PD-1 protein expression was analyzed by immunohistochemistry in 60 UBC specimens and 10 normal urothelia. Results: TLR4 activation protected T24 cells from CTL killing via B7-H1 overexpression. However PD98059, an inhibitor of ERK, enhanced CTL killing of T24 cells by reducing B7-H1 expression. TLR4 expression was generally decreased in UBC specimens, while B7-H1 and PD-1 were greatly overexpressed. Moreover, expression of both B7-H1 and PD-1 was significantly associated with UICC stage and WHO grade classification. Conclusions: TLR4 and B7-H1 may contribute to immune escape of UBC. Targeting B7-H1 or the ERK pathway may offer new immunotherapy strategies for bladder cancer.
