      • Robust MR assessment of cerebral blood volume and mean vessel size using SPION-enhanced ultrashort echo acquisition

        <P><B>Abstract</B></P> <P>Intravascular superparamagnetic iron oxide nanoparticles (SPION)-enhanced MR transverse relaxation rates (∆<I>R<SUB>2</SUB> <SUP>⁎</SUP> </I> and ∆<I>R<SUB>2</SUB> </I>) are widely used to investigate <I>in vivo</I> vascular parameters, such as the cerebral blood volume (CBV), microvascular volume (MVV), and mean vessel size index (mVSI, ∆<I>R<SUB>2</SUB> <SUP>⁎</SUP>/∆R<SUB>2</SUB> </I>). Although highly efficient, regional comparison of vascular parameters acquired using gradient-echo based <I>∆R<SUB>2</SUB> <SUP>⁎</SUP> </I> is hampered by its high sensitivity to magnetic field perturbations arising from air–tissue interfaces and large vessels. To minimize such demerits, we took advantage of the dual contrast property of SPION and both theoretically and experimentally verified the direct benefit of replacing gradient-echo based <I>∆R<SUB>2</SUB> <SUP>⁎</SUP> </I> measurement with ultra-short echo time (UTE)-based <I>∆R<SUB>1</SUB> </I> contrast to generate the robust CBV and mVSI maps. The UTE acquisition minimized the local measurement errors from susceptibility perturbations and enabled dose-independent CBV measurement using the vessel/tissue <I>∆R<SUB>1</SUB> </I> ratio, while independent spin-echo acquisition enabled simultaneous <I>∆R<SUB>2</SUB> </I> measurement and mVSI calculation of the cortex, cerebellum, and olfactory bulb, which are animal brain regions typified by significant susceptibility-associated measurement errors.</P> <P><B>Highlights</B></P> <P> <UL> <LI> <I>ΔR<SUB>2</SUB> <SUP>⁎</SUP> </I>-CBV is vessel orientation dependent and vulnerable to susceptibility artifacts. </LI> <LI> UTE acquisition minimizes susceptibility artifacts and enables dose-independent CBV. </LI> <LI> UTE-based CBV can theoretically and experimentally replace <I>ΔR<SUB>2</SUB> <SUP>⁎</SUP> </I> for vessel size. </LI> <LI> UTE-based <I>ΔR<SUB>1</SUB> </I> contrasts enable consistent CBV and mVSI maps in mouse brain. </LI> <LI> <I>ΔR<SUB>1</SUB> </I> and <I>ΔR<SUB>2</SUB> <SUP>⁎</SUP> </I> contrasts can be synergistically combined to enhance SPION contrast. </LI> </UL> </P>

      • Lipase-catalyzed enantioselective synthesis of (R,R)-lactide from alkyl lactate to produce PDLA (poly D-lactic acid) and stereocomplex PLA (poly lactic acid)

        R-lactide, a pivotal monomer for the production of poly (D-lactic acid) (PDLA) or stereocomplex poly (lactic acid) (PLA) was synthesized from alkyl ®-lactate through a lipase-catalyzed reaction without racemization. From among several types of lipase, only lipase B from Candida antarctica (Novozym 435; CAL-B) was effective in the reaction that synthesized (R,R)-lactide. Enantiopure (R,R)-lactide, which consisted of over 99% enantiomeric excess, was synthesized from methyl ®-lactate through CAL-B catalysis. Removal of the methanol by-product was critical to obtain a high level of lactide conversion. The (R,R)-lactide yield was 56% in a reaction containing 100mg of Novozym 435, 10mM methyl ®-lactate and 1500mg of molecular sieve 5A in methyl tert-butyl ether (MTBE). The important monomer (R,R)-lactide that is required for the production of the widely recognized bio-plastic PDLA and the PLA stereocomplex can be obtained using this novel synthetic method.

      • Sporadic Early-Onset Diffuse Gastric Cancers Have High Frequency of Somatic CDH1 Alterations, but Low Frequency of Somatic RHOA Mutations Compared With Late-Onset Cancers

        <P>BACKGROUND & AIMS: Early-onset gastric cancer, which develops in patients younger than most gastric cancers, is usually detected at advanced stages, has diffuse histologic features, and occurs more frequently in women. We investigated somatic genomic alterations associated with the unique characteristics of sporadic diffuse gastric cancers (DGCs) from younger patients. METHODS: We conducted whole exome and RNA sequence analyses of 80 resected DGC samples from patients 45 years old or younger in Korea. Patients with pathogenic germline mutations in CDH1, TP53, and ATM were excluded from the onset of this analysis, given our focus on somatic alterations. We used MutSig2CV to evaluate the significance of mutated genes. We recruited 29 additional early-onset Korean DGC samples and performed SNP6.0 array and targeted sequencing analyses of these 109 early-onset DGC samples (54.1% female, median age, 38 years). We compared the SNP6.0 array and targeted sequencing data of the 109 early-onset DGC samples with those from diffuse-type stomach tumor samples collected from 115 patients in Korea who were 46 years or older (late onset) at the time of diagnosis (controls; 29.6% female, median age, 67 years). We compared patient survival times among tumors from different subgroups and with different somatic mutations. We performed gene silencing of RHOA or CDH1 in DGC cells with small interfering RNAs for cell-based assays. RESULTS: We identified somatic mutations in the following genes in a significant number of early-onset DGCs: the cadherin 1 gene (CDH1), TP53, ARID1A, KRAS, PIK3CA, ERBB3, TGFBR1, FBXW7, RHOA, and MAP2K1. None of 109 early-onset DGC cases had pathogenic germline CDH1 mutations. A higher proportion of early-onset DGCs had mutations in CDH1 (42.2%) or TGFBR1 (7.3%) compared with control DGCs (17.4% and 0.9%, respectively) (P<.001 and P=.014 for CDH1 and TGFBR1, respectively). In contrast, a smaller proportion of early-onset DGCs contained mutations in RHOA (9.2%) than control DGCs (19.1%) (P=.033). Late-onset DGCs in The Cancer Genome Atlas also contained less frequent mutations in CDH1 and TGFBR1 and more frequent RHOA mutations, compared with early-onset DGCs. Early-onset DGCs from women contained significantly more mutations in CDH1 or TGFBR1 than early-onset DGCs from men. CDH1 alterations, but not RHOA mutations, were associated with shorter survival times in patients with early-onset DGCs (hazard ratio, 3.4; 95% confidence interval, 1.5 - 7.7). RHOA activity was reduced by an R5W substitution-the RHOA mutation most frequently detected in early-onset DGCs. Silencing of CDH1, but not RHOA, increased migratory activity of DGC cells. CONCLUSIONS: In an integrative genomic analysis, we found higher proportions of early-onset DGCs to contain somatic mutations in CDH1 or TGFBR1 compared with late-onset DGCs. However, a smaller proportion of early-onset DGCs contained somatic mutations in RHOA than late-onset DGCs. CDH1 alterations, but not RHOA mutations, were associated with shorter survival times of patients, which might account for the aggressive clinical course of early-onset gastric cancer. Female predominance in early-onset gastric cancer may be related to relatively high rates of somatic CDH1 and TGFBR1 mutations in this population.</P>

        N/A Malnutrition is known to be an independent risk factor of morbidity and mortality in hemodialysis patients. The causes of malnutrition were obsucure. Several catabolic cytokines such as interleukin-1β(IL-1β), interleukin-6(IL-6), and tumar necrosis fac-tor-α(TNF-α) were reported to be elevated in hemo-dialysis patients with malnutrition, To evaluate the nutritrional status and relationship among cytokines IL-1β, TNF-α, and various parameters, we performed a cross-sectional study in maintenance hemodialysis patients who have undergone for six months at least. Patients who were taking steroid. showed any sign of infection, or had residual renal function were excluded from the study. Nutritional status was classified into well nourished(A), mild to moderate malnutrition(B), and severe malnutrition(C) by using subjective global assessment(SGA). Patients were divided into two groups (I: A, II: B and C) based on SGA. The level of IL-10, TNF-a were estimated by ELISA. Total patients were 23(M/F:15/8 mean age 52.0±1.56yrs). There were significant differences in weight changes for 6 months, CRP, and SGA between two groups based on the level of serum albumin(using 4.0g/dL as a cutoff value). Moreover, based on SGA, there were significant differences in age, BMI, CRP, and albumin between the group I and IL Serum albumin levels was directly correlated with transferrin(r=0.302, p=0.04), and was inversly correlated with age(r=-0.329, p=0.007). TNF- u was inversly correlated with Kt/Vueea(r=-0.291, p= 0.047), and directly correlated with the duration of maintenance hemodialysis(r=0,346, p=0.036), BMI(r-0.453, p<0.05). These data support the fact that albumin and CRP can be a good marker of nutritional status in maintenance hemodialysis patient. However cytokines and transferrin might not be related to nutritional status.

        경사지에서 고추 정식시기에 따른 토양유실과 유출수에 대한 식생피복 효과

        As sloped farmland is subject to runoff and soil erosion and consequently require appropriate vegetative coverage to conserve soil and water, a field study was carried out to evaluate the impact of crop canopy coverage on soil loss and runoff from the experimental plot with three different textural types (clay loam, loam, and sandy loam). The runoff and soil loss were examined at lysimeters with 15% slope, 5 m in length, and 2 m in width for five months from May to September 2009 in Suwon (37° 16" 42.67" N, 126° 59" 0.11" E). Red pepper (Capsicum annum L. cv. Daechon) seedlings were transplanted on three different dates, May 4 (RP1), 15 (RP2), and 25 (RP3) to check vegetation coverage. During the experimental period, the vegetation coverage and plant height were measured at 7 day-intervals and then the "canopy cover subfactor" (an inverse of vegetation cover) was subsequently calculated. After each rainfall ceased, the amounts of soil loss and runoff were measured from each plot. Under rainfall events >100 mm, both soil loss and runoff ratio increased with increasing canopy cover subfactor (R²=0.35, p<0.01, R²=0.09, p<0.1), indicating that as vegetation cover increases, the amount of soil loss and runoff reduces. However, the soil loss and runoff were depending on the soil texture and rainfall intensity (i. e., EI30). The red pepper canopy cover subfactor was more highly correlated with soil loss in clay loam (R²=0.83, p<0.001) than in sandy loam (R²=0.48, p<0.05) and loam (R²=0.43, p<0.1) plots. However, the runoff ratio was effectively mitigated by the canopy coverage under the rainfall only with EI30<1000 MJ mm ha<SUP>-1</SUP> hr<SUP>-1</SUP> (R²=0.34, p<0.05). Therefore, this result suggested that soil loss from the red pepper field could be reduced by adjusting seedling transplanting dates, but it was also affected by the various soil textures and EI30.

      • <i>CYP2A6</i> and <i>ERCC1</i> polymorphisms correlate with efficacy of S-1 plus cisplatin in metastatic gastric cancer patients

        <P><B>Background:</B></P><P>We evaluated the association between polymorphisms of cytochrome P450 2A6 (<I>CYP2A6</I>)/excision repair cross-complementation group 1 (<I>ERCC1</I>)/X-ray repair cross-complementing group 1(<I>XRCC1</I>) and treatment outcomes of metastatic gastric cancer (MGC) patients treated with S-1/cisplatin.</P><P><B>Methods:</B></P><P>Among MGC patients (<I>n</I>=108), who received S-1 (40 mg m<SUP>−2</SUP> b.i.d., days 1–14) and cisplatin (60 mg m<SUP>−2</SUP>, day 1) every 3 weeks, we analysed the wild-type allele (<I>W</I>) and variants (<I>V</I>) of <I>CYP2A6</I> (<I>*4</I>, <I>*7, *9, *10</I>), and the polymorphisms of <I>ERCC1</I> (rs11615, rs3212986) and <I>XRCC1</I> (rs25487).</P><P><B>Results:</B></P><P>Patients having fewer <I>CYP2A6</I> variants had better response rates (<I>W</I>/<I>W vs W</I>/<I>V</I> other than <I>*1/*4 vs V</I>/<I>V</I> or <I>*1/*4</I>=66.7 <I>vs</I> 58.3 <I>vs</I> 32.3% <I>P</I>=0.008), time to progression (TTP) (7.2 <I>vs</I> 6.1 <I>vs</I> 3.5 months, <I>P</I>=0.021), and overall survival (23.2 <I>vs</I> 15.4 <I>vs</I> 12.0 months, <I>P</I>=0.004). <I>ERCC1 19442C</I>><I>A</I> (rs3212986) was also associated with response rate (<I>C/C</I>, 46.7% <I>vs C/A</I>, 55.3% <I>vs A/A</I>, 87.5%) (<I>P</I>=0.048) and TTP (4.4 <I>vs</I> 7.6 <I>vs</I> 7.9 months) (<I>P</I>=0.012). Patients carrying both risk genotypes of <I>CYP2A6</I> (<I>V</I>/<I>V</I> or <I>1/*4</I>) and <I>ERCC1 19442C</I>><I>A</I> (<I>C/C</I>) <I>vs</I> those carrying none showed an adjusted odds ratio of 0.113 (<I>P</I>=0.004) for response, and adjusted hazard ratios of 3.748 (<I>P</I>=0.0001) for TTP and 2.961 (<I>P</I>=0.006) for death.</P><P><B>Conclusion:</B></P><P>Polymorphisms of <I>CYP2A6</I> and <I>ERCC1 19442C</I>><I>A</I> correlated with the efficacy of S-1/cisplatin.</P>

        Facile synthesis of an IRMOF-3 membrane on porous Al<sub>2</sub>O<sub>3</sub> substrate via a sonochemical route

        A densely-packed IRMOF-3 membrane (S-IRMOF-3(Mem)), ~55 μm in thickness, on an Al<SUB>2</SUB>O<SUB>3</SUB> disc support was prepared via a sonochemical synthesis route, and the optimal fabrication conditions were established. Bare IRMOF-3 particles (S-IRMOF-3(p)) were synthesized independently to analyze the contribution of the Al<SUB>2</SUB>O<SUB>3</SUB> support. The overall physicochemical properties of the IRMOF-3 products produced via the sonochemical route were superior to those prepared by microwave heating. The IRMOF-3 materials produced were characterized by XRD, SEM, EDS-mapping, and N<SUB>2</SUB> adsorption-desorption measurements. S-IRMOF-3(p) exhibited satisfactory CO<SUB>2</SUB> adsorption capacities (54 mg/g at 298 K/1 bar; 732 mg/g at 298 K/20 bar) and CO<SUB>2</SUB>/N<SUB>2</SUB> selectivity (18 at 298 K/1 bar). Both S-IRMOF-3(Mem) and S-IRMOF-3(p) were assessed as a catalyst for a liquid phase Knoevenagel condensation reaction between benzaldehyde and ethyl cyanoacetate. S-IRMOF-3(Mem) and S-IRMOF-3(p) showed comparable conversions (87-89%) with 100% selectivity after a 4 h reaction at 333 K. The heterogeneous nature of the catalyst was confirmed by a hot filtering experiment, and S-IRMOF-3(Mem) could be recovered easily after the reaction and be recycled several times with little change in product yield.

        Since the Korean Association of Quality Assurance for Clinical Laboratory was established in 1976, the numbers of the participating hospitals nowadays were increased from 40 to 450 or more hospitals all over the country. In the early day the both internal and external quality control programs were started at the same time, however, the external QC programs prevailed over the internal ones putting the cart before the horse. The accuracy and the precision of the data are not satisfying in spite of the 20 years experiences of quality control programs. All the daily data should be reported after the validity is assured by the self-evaluation not by third party evaluation. It is regret that the CVs of enzymatic tests in external QC are exceeding 10% and there is no sign of diminishing CVs. One of the major reasons is that the types of anaytical instrurments and reagents are diverisified. Moreover, most of them are subject to the standards of the respective country of manufacturer. Accordingly, we can make a suggestion that the reagents and instruments should be imported or supplied meeting the established requirements for Korean standardization.

        In European countries or Japan they made an attempt for the standardization of assays in AST, ALT, ALP, CK, LD and GGT 10 years ago. For the actualization of the trial, they managed to succeeded in development of certified reference material and enzyme reference material. And they are now forecasting the unification of test results produced differently under the respective situatons. Then, through the highly developed communication system, someone can access any test results whenever or whereever only if they want to. It means that we can expect much greater improvements in medical services through the information exchanges. In this study the authors tried to ascertain the interchangeability comparing the obtained results of the CK tests between two general hospitals. Fortunately, the data got in the internal QC programs came to fall within the normal tolerances. The reference intervals for the CK test were different accarding to the chosen reagents and the results from the pooled serum were also shown differently each other. However, from the two hospitals, we could get glucose K factors, upon which the measurements were obtained and compared. Using K factors we measured the CKs, then we multiply the CK values by correction coefficients. We could found that the two results were showing good coincidence in the comparison. The main purpose of the standardization of enzyme assays is the creation of a new unified value which can be expressed differently otherwise. Using the standardized values we can exploit the data everywhere, reduce the medical costs from repeated or duplicated tests, save the foreign exchanges preventing the diversification of impart connections.
