      • Skin reflectance법에 의한 신생아 bilirubin측정의 실험연구

        Spectral reflectance measurements on the newborn skin provide a noninvasive screening method for neonatal jaundice and may also be useful in assessing the efficacy of phototherapy and the potential for kernicterus. In this study we briefly discussed about the bilirubin metabolism and determination of serum bilirubin by spectral reflectance of the skin. A correlation study was conducted accross Minolta jaundice meter with AO bilirubinometer as the reference assay. The correlation statistics obtained for 43 newborns were: Minolta jaundice meter (chest site) =0.810 AO bilirubinometer+7.137, correlation coefficient+0.41, Minolta jaundice meter(forehead site) = 0.887 AO bilirubinometer+8.026, correlation coefficient = 0.38. Studies from linear relationship statistics showed some evidence for the difference between two methods. It had been suggested that a noninvasive method for serum bilirubin by skin reflectance was not compatible with the AO bilirubinometer.

        Nephelometry법에 의한 IgG Subclass 참고치 설정

        We developed nephelometric assays on the Beckman Array 360 Nephelometry analyzer that allow a quantification of IgG subclass concentrations in 100 samples quickly and reproducibly. We have established new reference ranges the human IgG subclass in sera of adults. These reference ranges are the central 95th percentile of results from a study of 100 apparently healthy individuals(44 females, 56 males). The reference ranges of IgG subclasses in sera are IgG1 447-1260 mg/dL, IgG2 246-900 mg/dL, IgG3 15-147.6 mg/dL, and IgG4 12-104.4 mg/dL respectively.

        BCG 의 Sarcoma 180 cell 增植柳制(증식류제) 效果(효과)에 관한 硏究(연구)

        미생물 기원 免疫增强物質인 BCG에 의한 Sarcoma-180 tumor cell에 대한 항종양 效果를 알아보기 위해 JCL-ICR mouse 복강에 S-180 종양세포를 접종하여 대조군으로 하고 BCG를 60㎍/㎏, ip로 -1, 1, 5, 9일에 접정한 군을비교했을 때BCG접종군의 약 2배의 수명연장 效果가 있음이 관찰되었다. BCG 접종군에서는 B cell 이 시일 경과에 따라 현저히 증가하였으며 면역글로부린의 증가를 형청 단백질 던기영동과 면역글로부린 전기영동에서 확이 하였다. 한편 51Cr-release assay에 의한 細胞毒性 시험에서 NK cell의 활성은 볼 수 없었으며 혈청을 비동화하고 complement를 넣어 주었을 때%specific51 Cr-release의 증가를 보이므로 BCG에 의한 S-180 cell 증식 억제 효과는 antibody-complement-mediated cytotoxicity에 의 한 것으로 생각된다. The study reported here was designed to determine the efficacy of Bacilli Calmette-Guerin (BCG) treatment against sarcoma 180 tumor cells suppression in JCL- ICR mice. The results were as follows : 1. A group of mice previously injected with BCG 60 ,ug/kg mouse on day-1 (injected intraperitoneally on day -1, 1, 5 and 9) is the most effective and its mean survival period have two folds longer than that of non- injected control. 2. 240 ,ug/kg mouse of BCG previously injected group on day -1, 5, 9 and 13 is most effective and mean survial peried have two folds longer. 3. The amount of B lymphocyte were markedly increased in BCG sensitized mice. 4. The observation of apparent changes from serum protein electrophoresis and immunoelectrophoresis as an increase of immunoglobulins. 5. NK cell activity was measured by a standarded 4hrs 51 Cr-release assay with Sarcoma 180 used as targets. NK cell were not or, if anything ; were far less cytotoxic against the target cells. 6. By the heat- inactivated antiserum with complement cell assay, it was shown that the percent specific 51 Cr-release was increased. The results suggest that the efficacy of BCG against S- 180 cells suppression were antibodycomplement- mediated cytotoxicity.

        1-2 diglyceride를 기질로 한 Enzymatic rate법과 1,2-O-dilaurylrac- glycero-3-glutaric acid-(6-methylresorufin) ester를 기질로 한 Enzymatic Colorimetric 법에 의한 Lipase 측정 비교

        Lipases are glycoproteins with a molecular weight of 47000 daltons and are defined as triglyceride hydrolases. They catalyze the cleavage of diglycerides with the subsequent formation of monoglycerides and fatty acids. We evaluated serum lipase CX-7 Delta(1-2 diglyceride substrate) and Integra 700(1, 2-O-dilauryl-rac-glycero-3-glutaric acid-(6-methylresorufin) ester substrate). Within-run CVs of two level serums were 4.1% and 3.0%. The between-run CVs were 3.5%, 3.9%, respectively. The comparison between Integra700(y) and CX-7(x) resulted in a linear regression analysis described by the equation: y=3.002x+36.26 with a correlation coefficient r=0.9884.

        Type 2 당뇨병 환자의 혈당 조절 안자로1,5Anhydroglucitol과 HbAIC의 비교

        1,5Anhydroglucitol (1,5AG) is one of the major polyols in human body. It provides useful information on current glycemic control and is superior to HbA1C. To evaluate the use of plasma 1,5Ahydroglucitol (1,5AG) levels in screening for type II diabetes mellitus, the relationship between the plasma concentration of 1,5AG and HbA1C was examined in 127 patients with type II diabetes mellitus Patients. Subjects were separated into five groups based on diabetes control and complication trial (DCCT) criteria: normal group (NG), normal DM patient (ND), near normal glycemia (NN), good control (GC), action suggested (AS) groups. Fasting plasma glucose (F=70.99, P> 0.0001, 1,5AG (F=28.2, P> 0.0ω1) and HbA1C (F=15.14, P>0.0001 concentrations showed significant differences between each groups. These fmdings suggest that the plasma 1,5AG concentration can be a useful index of type II diabetes me1litus patients.

      • Studies on the Detection of mecA Gene and Distribution of Enterotoxin Genes in Pathogenic Staphylococci Isolated from Korea Using Molecular Biologic Technique

        본 연구에서는 입상 검체에서 발견되는 병원성 포도상구균(S. aureus)과 methicillin 내성 포도상구균(MRSA)에서 각각 nurc과 mecA 유전자, 장독소인 enterotoxin A에서 enterotoxin E를 encoding하는 유전자들과 toxic shock syndrome toxin(tst) 유전자를 빠르개 탐지하고 독소형의 국내·외의 역학적인 분포를 조사하기 위하여 multiplex PCR을 수행하였다. 210개의 MRSA에서 두 개의 균주가 mecA 유전자 음성을 보인 것을 제외하고 모두 nuc유전자와 mecA유전자 양성을 나타냈다. 이 중 10개(80.5%)가 한 게 흑은 그 이상의 장독소에 대한 유전자를 지니고 있었으며 다른 41개 균주(19.5%)는 장독소 유전자에 대해 양성을 나타내지 않았다. 장독소A 유전자(sea)는 분리균 중 50.5%(n=105)에서 유전자가 중폭되었고 seb는 14.료쏭(n=30), sec는 23.3% (n=49), sed는 10.0%(n=21) 그리고 tst는 12.4%(n=26)가 증폭되었다. 58개의 균주들(27.68)은 두 개 이상의 장내독소 유전자가 증폭되었다. 가장 많이 나타난 독소 조합군은 sea와 seb로 26개 균주(12.4%)에서 발견되었으며 다음으로 빈번한 조합은 sec와 tst로 14균주(6.7%)로 나타났다. 210개 중 109개의 균주는 서울 지역에서 분리된 것이었고 101개는 부산 지역에서 분리된 균이었다. 부산지역에서 장내독소 A 유전자(sea)의 발생율은 서울에서보다 두 배 더 높게 나타났다(57.1% 대 23.6%). 그러나 장내독소 c유전자(sec)와 d유전자(sed)는 서울 지역에서 각각 23.6%와 17.9%로 비슷하게 나타난 반면에 부산 지역에서는 각각 2.0% 미만으로 거의 무시할 수 있을 정도로 낮게 나타났다. 한국의 장독소 형별 검출을 외국과 비교하면 장독소A의 검출빈도가 가장 높은 것은 같으나 차순으로 빈도가 높은 것이 외국의 경우 b유전자인데 반하여 한국에서는 c유전자로 실험결과 나타났다. 또한 한국에서의 d유전자 검출빈도가 b유전자 보다 높아 주된 유행형인 a유전자를 제외한 기타 독소형의 분포는 국가간 또는 지역간에도 차가 있는 것으로 결론지을 수 있다. In this study we attempted multiplex PCR for rapid detection of genes for nuc, mecA. enterotoxin A to E and TSST-l(tst) in bacterial lysates extracted from human strains of S aureus. In 210 isolates of methicillin-resistant Staphylococcus aureus (MRSA), all strains showed both nuc gene and mecA gene positive except two strains that showed only nuc gene positive. One hundred sixty nine(80.5%) of 210 MRSA isolates tested produced one or more toxins. Other 41(19.5%) isolates did not produce any of the toxins. Enterotoxin A gene(sea) was amplified in 50.5%(n=105) of the isolates, enterotoxin B(seb) in 14.3%(n=30), enterotoxin C(sec) in 23.3%(n=49), enterotoxin D(sed) in 10.0%(n=21) and TSST-l(tst) in 12.4%(n=26). Fifty eight strains(27.6%) amplified more than two enteroxin genes. The most common combination of toxin gene was sea and seb, which was found in 26(12.4%) strains, and the next frequent combination was sec and tst in 14(6.7%) strains from 210 isolates among which 109 MRSA strains tested were isolated in Seoul area and 101 in Pusan area. The frequency rate of enterotoxin A in Pusan was twice higher than that of Seoul with 57.1% to 23.6%. But the frequency rates of enterotoxins C and D genes in Seoul area are analogous with relatively narrow margin while those rates in Pusan area were almost negligible with less than 2% each. Apparent difference of enterotoxin type distribution was also found between the countries. Beside enterotoxin A which was most predominantely found in both European countries and Korea, enterotoxin C was the subsequently prevalent type of toxin in Korea instead of enterotoxin b which was found as the second most prevalent enterotoxin in some European countries

      • 침자가 성장기 및 성숙기 웅성 흰쥐의 혈액성분에 미치는 영향

        Acupuncture, an over 2500 years old therapeutic method stemming from traditional oriental medicine, is nowadays wide open to modern medical science. With the widespread usage of acupuncture, the number of article about biomechanical or therapeutic effects of acupuncture from animal experiments and clinical trials is largely reported. In a previous study, we have reported that Joksamlee(ST36) acupuncture increases serum protein and glucose but decrease serum lipid and cholesterol. For further study, we was examined effects of Shenshu(B23) and Samumgyo(SP6) acupuncture stimulation on differential blood components in growing and adult male rate. 24 male Sprague-Dawley rats were randomly assigned into three groups; A1 (B23 and SP6 acupuncture in adult rats) group, A2 (B23 acupuncture in growing rats) group, and control group. The results obtained were as follows; 1. Body weight was significantly decreased in A1 and A2 groups. 2. Serum TP level was not changed, but albumin level was significantly increased in A2 group. 3. Serum glucose level was significantly increased in A1 group. 4. Serum TG level was significantly decreased in A2 group: PL level was significantly decreased in both A1 and A2 groups; FFA level was not changed. 5. Serum T.CHO, HDL-C and LDL-C levels were not changed in both A1 and A2. 6. Serum amylase activity was significantly increased in A1 group.

      • IFCC인정 혈장단백표준품(CRM470)에 의한 면역단백 측정에 관한 연구

        Measuring proteins is unlike measuring ions and small molecules for which the chemical and physical properties are well understood. Assaying large proteins requires comparing the test sample with a reference materials. Quality Control surveys in recent years, in various parts of the world, have shown poor between laboratory agreement for measurments of immunoglobulins. Values for the commonly measured proteins may have varied a considerable amount among method depending on the protein measured and the standard materials by the manufacturer. The College of American Pathotogists and the Bureau Economic Communitaire de Reference of the European Economic Community have recently released Reference Preparation for Proteins in Human Serum(RPPHS). The assay for IgG and IgM using CRM470 standard were compared with conventional method. There was a good correlation among two methods. However, the mean value in IgG obtained by the CRM470 method was a slightly lower than that by the conventional method, but higher than that by the conventional method in IgM.

        Results of clinical laboratory tests are used in many medical situations. Ideally, the standards of test performance required to fulfill medical needs should be well defined, particularly standards for the important reliability performance characteristics, namely, imprecision and bias. Given that the objective of standardization is to ensure that the same result is obtained regardless of how, where, or by whom the test is done. The most stringency of the precision goals set by the other studies are those for calcium, sodium, chloride and potassium (CVs of≤5%), analytes under tight homeostatic control. From the results of this study, we conclude that matrix-induced biases may occur when commercial lyophilized control materials are used with some analytic systems, and that the amount and nature (positive or negative) of the observed biases are instrument, reagent, and method specific.
