        Simple and green fabrication of a biomass-derived N and O self-doped hierarchical porous carbon via a self-activation route for supercapacitor application

        To meet the increased performance and cost requirements of commercial supercapacitor, a N and O self-doped hierarchical porous carbon is fabricated via a green and simple self-activation route utilizing leaves of wild hollyhock as raw materials. Comparing to commercial activated carbon, the reported material exhibits some marked merits, such as simple and green fabrication process, low cost, and superior capacitance performance. The specifc surface area of the obtained N and O co�doped hierarchical porous carbon arrives 954 m2 g−1, and the content of the self-doped nitrogen and oxygen reaches 2.64 at.% and 7.38 at.%, respectively. The specifc capacitance of the obtained material reaches 226 F g−1 while the specifc capacitance of the symmetric supercapacitor arrives 47.3 F g−1. Meanwhile, more than 90.3% of initial specifc capacitance is kept under a current density of 20 A g−1, and no arresting degradation is observed for capacitance after 5000 times cycle, perfectly demonstrating the excellent cycle and rate capability of the obtained material. The obtained N and O co-doped hierarchical porous carbon are expected to be an ideal substitution for commercial activated carbon.

        Preparation and Investigation of Reinforced PVP Blend Membranes for High Temperature Polymer Electrolyte Membranes

        Poly(vinylpyrrolidone) (PVP), as the low-cost and commercial material, exhibits superior phosphoric acid doping capability due to the presence of heterocycle and carbonyl groups in the repeat unit. However, it can’t be used as the high temperature polymer electrolyte membrane (HT-PEM) alone because of its significant hydrophilicity and poor mechanical stability. In the present work, polyethersulfone (PES), polysulfone (PSU), polyetherketone-cardo (PEK-c), polyvinylidene fluoride (PVDF) and poly(vinylidene fluoride-co-hexafluoropropylene) (PHFP), five kinds of engineering thermoplastics with excellent mechanical properties and chemical inertness, are chosen to prepare a series of PVP blend membranes by the polymer blending method in order to enhance the dimensional and mechanical stabilities of PVP based membranes. The influence of structures of enhanced polymers on properties of HT-PEMs was investigated systematically. PVP blend membranes with aromatic polymers (i.e. PES, PSU and PEK-c) exhibited decreased volume swellings, increased acid doping contents, superior conductivities and improved mechanical strengths, which determined that they are more suitable for electrolytes of fuel cell applications comparing with PVP/PVDF and PVP/PHFP membranes blended with aliphatic polymers.

        Numerical Simulation of Prestressed Precast Concrete Bridge Deck Panels Using Damage Plasticity Model

        This paper describes a three-dimensional approach to modeling the nonlinear behavior of partial-depth precast prestressed concrete bridge decks under increasing static loading. Six full-size panels were analyzed with this approach where the damage plasticity constitutive model was used to model concrete. Numerical results were compared and validated with the experimental data and showed reasonable agreement. The discrepancy between numerical and experimental values of load capacities was within six while the discrepancy of mid-span displacement was within 10 %. Parametric study was also conducted to show that higher accuracy could be achieved with lower values of the viscosity parameter but with an increase in the calculation effort.

        Using low-coverage whole genome sequencing technique to analyze the chromosomal copy number alterations in the exfoliative cells of cervical cancer

        Objectives: We analyzed the chromosomal-arm-level copy number alterations (CNAs) in the cervical exfoliative cell and tissue samples by using the low-coverage whole genomic sequencing technique. Methods: In this study, we retrospectively collected 55 archived exfoliated cervical cell suspension samples and the corresponding formalin-fixed and paraffin-embedded tissue section samples including 27 invasive cervical cancer and 28 control cases. We also collected 19 samples of the cervical exfoliative cells randomly from women to verify the new algorithm model. We analyzed the CNAs in cervical exfoliated cell and tissue samples by using the low-coverage next generation of sequencing. Results: In the model-building study, multiple chromosomal-arm-level CNAs were detected in both cervical exfoliated cell and tissue samples of all cervical cancer cases. By analyzing the consistency of CNAs between exfoliated cells and cervical tissue samples, as well as the heterogeneity in individual patient, we also established a C-score algorithm model according to the chromosomal-arm-level changes of 1q, 2q, 3p, 7q. The C-score model was then validated by the pathological diagnosis of all 74 exfoliated cell samples (including 55 cases in model-building group and 19 cases in verification group). In our result, a cutoff value of C-score >6 showed 100% sensitivity and 100% specificity in the diagnosis of cervical cancer. Conclusion: In this study, we found that CNAs of cervical exfoliated cell samples could robustly distinguish invasive cervical cancer from cancer-free tissues. And we have also developed a C-score algorithm model to process the sequencing data in a more standardized and automated way.

        CK2 phosphorylates AP-2α and increases its transcriptional activity

        Transcription factor AP-2α involves in the process of mammalian embryonic development and tumorigenesis. Many studies have shown that AP-2α functions in association with other interacting proteins. In a two-hybrid screening, the regulatory subunit β of protein casein kinase 2 (CK2β) was identified as an interacting protein of AP-2α; we confirmed this interaction using in-vitro GST pull-down and in-vivo co-immunoprecipitation assays; in an endogenous co-immunoprecipitation experiment, we further found the catalytic subunit α of protein casein kinase 2 (CK2α) also exists in the complex. Phosphorylation analysis revealed that AP-2α was phosphorylated by CK2 kinase majorly at the site of Ser429, and such phosphorylation could be blocked by CK2 specific inhibitor 4,5,6,7-tetrabromobenzotriazole (TBB) in a dose-dependent manner. Luciferase assays demonstrated that both CK2α and CK2β enhanced the transcription activity of AP-2α; moreover, CK2β increased the stability of AP-2α. Our data suggest a novel cellular function of CK-2 as a transcriptional co-activator of AP-2α.

        Production of Transgenic Pigs with an Introduced Missense Mutation of the Bone Morphogenetic Protein Receptor Type IB Gene Related to Prolificacy

        In the last few decades, transgenic animal technology has witnessed an increasingly wide application in animal breeding. Reproductive traits are economically important to the pig industry. It has been shown that the bone morphogenetic protein receptor type IB (BMPR1B) A746G polymorphism is responsible for the fertility in sheep. However, this causal mutation exits exclusively in sheep and goat. In this study, we attempted to create transgenic pigs by introducing this mutation with the aim to improve reproductive traits in pigs. We successfully constructed a vector containing porcine BMPR1B coding sequence (CDS) with the mutant G allele of A746G mutation. In total, we obtained 24 cloned male piglets using handmade cloning (HMC) technique, and 12 individuals survived till maturation. A set of polymerase chain reactions indicated that 11 of 12 matured boars were transgene-positive individuals, and that the transgenic vector was most likely disrupted during cloning. Of 11 positive pigs, one (No. 11) lost a part of the terminator region but had the intact promoter and the CDS regions. cDNA sequencing showed that the introduced allele (746G) was expressed in multiple tissues of transgene-positive offspring of No.11. Western blot analysis revealed that BMPR1B protein expression in multiple tissues of transgene-positive $F_1$ piglets was 0.5 to 2-fold higher than that in the transgene-negative siblings. The No. 11 boar showed normal litter size performance as normal pigs from the same breed. Transgene-positive $F_1$ boars produced by No. 11 had higher semen volume, sperm concentration and total sperm per ejaculate than the negative siblings, although the differences did not reached statistical significance. Transgene-positive $F_1$ sows had similar litter size performance to the negative siblings, and more data are needed to adequately assess the litter size performance. In conclusion, we obtained 24 cloned transgenic pigs with the modified porcine BMPR1B CDS using HMC. cDNA sequencing and western blot indicated that the exogenous BMPR1B CDS was successfully expressed in host pigs. The transgenic pigs showed normal litter size performance. However, no significant differences in litter size were found between transgene-positive and negative sows. Our study provides new insight into producing cloned transgenic livestock related to reproductive traits.

        Differentiation of cystic biliary atresia and choledochal cysts using prenatal ultrasonography

        Purpose: This study explored the performance of prenatal ultrasonography in the differential diagnosis of cystic biliary atresia (CBA) and choledochal cyst (CC). Methods: Fetuses diagnosed with hepatic hilar cyst in the second trimester were included in this study. A series of prenatal ultrasound examinations were performed in the second and third trimesters. The diameter of the gallbladder (GB) and hepatic cyst were measured, as well as the wall thickness of the GB. The GB-cyst connection, visibility of the right hepatic artery (RHA), and other concomitant abnormalities were carefully evaluated. A neonatal transabdominal ultrasound examination was performed within 1 week after birth, and clinical data were followed up to 6 months after birth. Results: Between January 1, 2016 and January 31, 2020, 53 fetuses diagnosed with hepatic hilar cyst were recruited. Eight were excluded because they were lost to follow-up. Among the 45 cases included in this study, 10 were diagnosed with CBA and 35 with CC after birth. Statistically significant differences were found in GB width, wall thickness, change in GB width, change in cyst length, GB-cyst connection, and RHA visibility between the CBA and CC groups. GB width showed the best diagnostic performance with an area under the curve (AUC) of 0.899. The combination of GB width, GB wall thickness, and GB-cyst connection yielded a comparable AUC of 0.971. Conclusion: The GB should be carefully evaluated in fetuses with hepatic hilar cyst. Prenatal ultrasound findings could provide suggestive parameters for the differential diagnosis of CBA from CC.         Backstepping method based controller design for third-order truck platoon robust to dynamic uncertainty and external disturbance

        The vehicle platoon is subject to dynamic uncertainty and external disturbance, such as air drag and parameter variation in practice, so lack of consideration for those factors can deteriorate control performance. This paper proposes a nonlinear control algorithm based on a novel adaptive backstepping method to cope with the uncertainty and disturbance. The virtual controller in the backstepping method utilizes only the local information of the individual vehicle, rather than the global state information, in the control algorithm design. A third-order vehicle dynamics model was built to incorporate the nonlinearity and the factor of actuation delay. The fuzzy logic system (FLS) is applied to estimate the nonlinearity in vehicle dynamics to facilitate the adaptive control. Five scenarios with different settings were constructed to verify the control performance in the simulation, which indicates the effectiveness and robustness of the controller to dynamic uncertainty and external disturbance.
