        The Relationship between HIF-2α and VEGF with Radiographic Severity in the Primary Osteoarthritic Knee

        Purpose: The aim of this study was to determine the relationship of hypoxia-inducible factor-2 (HIF-2α) and vascular endothelial growth factor (VEGF) with radiographic severity in primary osteoarthritis (OA) of the knee. Expression of these two factors in cartilagesamples from OA knee joints was examined at mRNA and protein levels. Materials and Methods: Knee joints were examined using plain radiographs, and OA severity was assessed using the Kellgren and Lawrence (KL) grading system. Specimens were collected from 29 patients (31 knees) who underwent total knee replacementbecause of severe medial OA of the knee (KL grades 3 and 4), 16 patients who underwent knee arthroscopy (KL grade 2), and 5 patients with traumatic knees (KL grade 0). HIF-2α and VEGF expression was quantified by real-time polymerase chain reactionand western blotting. Results: Cartilage degeneration correlated with the radiographic severity grade. OA severity, determined using the Mankin scale, correlated positively with the KL grade (r=0.8790, p<0.01), and HIF-2α and VEGF levels with the radiographic severity of knee OA (r=0.7001, p<0.05; r=0.6647, p<0.05). Conclusion: In OA cartilage, HIF-2α and VEGF mRNA and protein levels were significantly and positively correlated. The expressionof both factors correlated positively with the KL grade. HIF-2α and VEGF, therefore, may serve as biochemical markers as well as potential therapeutic targets in knee OA.

        The Protective Effect of Sodium Hyaluronate on the Cartilage of Rabbit Osteoarthritis by Inhibiting Peroxisome Proliferator-Activated Receptor-Gamma Messenger RNA Expression

        Purpose: The purpose of this study was to study the protective effect and influence of sodium hyaluronate (Na- HA) on mRNA expression of peroxisome proliferators-activated receptor gamma (PPAR-γ) in cartilage of rabbit osteoarthritis (OA) model. Materials and Methods: Forty eight white rabbits were randomly divided into A, B, and C groups. Group A was normal control group, B and C groups underwent unilateral anterior cruciate ligament transection (ACLT). The rabbits in group B were injected normal saline after ACLT; and Group C received intraarticular1% sodium hyaluronate (HA) injection 5 weeks after surgery, 0.3 mL once a week. At 11th week after surgery, all the rabbits were sacrificed. The cartilage changes on the medial femoral condyles were graded separately. Cartilage sections were stained with safranin-O and HE, and messenger RNA (mRNA) expression of PPAR-γ was detected by using real time polymerase chain reaction (Real Time-PCR). Results: Cartilage degeneration in group B was significantly more severe than in A and C injection group. The grey value of Safranin- O of B group was higher than A and C groups. Expression of PPAR-γ mRNA in group B was higher than group A and C. Conclusion: This study shows that Na-HA has a protective effect on articular cartilage degeneration, and the inhibitory effect on the PPAR-γ mRNA expression may be one of therapeutic mechanism of Na-HA.         Dynamics Analysis of a New Simple Chaotic Attractor

        In this paper, a novel simple chaotic system is discussed. Some basic dynamical properties, such as Lyapunov exponents, Poincaré mapping, fractal dimension, bifurcation diagram, continuous spectrum and chaotic dynamical behaviors of the novel chaotic system are studied, either numerically or analytically. The obtained results show clearly that the system discussed in this Letter is a novel chaotic system and deserves a further detailed investigation.

        Seismic performance of mixed column composed of square CFST column and circular RC column in Chinese archaized buildings

        This paper presents some quasi-static tests for 4 mixed columns composed of CFST column and RC column. The seismic performance and failure mode were studied under low-cyclic revised loading. The failure mode was observed under different axial compression ratios. The hysteretic curve and skeleton curve were obtained. The effects of axial compression ratio on yield mechanism, displacement ductility, energy dissipation, stiffness and strength attenuation were analyzed. The results indicate that the failure behavior of CFST-RC mixed column with archaized style is mainly caused by bending failure and accompanied by some shear failure. The axial compression ratio performs a control function on the yielding order of the upper and lower columns. The yielding mechanism has a great influence on the ductility and energy dissipation capacity of specimens. Based on the experiment, finite element analysis was made to further research the seismic performance by ABAQUS software. The variable parameters were stiffness ratio of upper and lower columns, axial compression ratio, yielding strength of steel tube, concrete strength and rebar ratio. The simulation results show that with the increase of stiffness ratio of the upper and lower columns, the bearing capacity and ductility of specimens can correspondingly increase. As the axial compression ratio increases, the ductility of the specimen decreases gradually. The other three parameters both have positive effect on the bearing capacity but have negative effect on the ductility. The results can provide reference for the design and engineering application of mixed column consisted of CFST-RC in Chinese archaized buildings.

      • The P275A Polymorphism in the Macrophage Scavenger Receptor 1 Gene and Prostate Cancer Risk: a Meta-Analysis

        Background: Published data regarding associations between the P275A polymorphism in the macrophage scavenger receptor 1 (MSR1) gene and prostate cancer (PCa) risk are inconclusive. The aim of this study was to comprehensively evaluate the genetic risk of P275A polymorphism in MSR1 gene for PCa. Materials and Methods: A systematic literature search was carried out in Pubmed, Medline (Ovid), Embase, CBM, CNKI, Weipu, and Wanfang databases, covering all available publications (last search was performed on Apr 27, 2015). Statistical analysis was performed using Revman 5.2 and STATA 10.1 software. Results: A total of 5,017 cases and 4,869 controls in 12 case-control studies were included in this meta-analysis. When all groups were pooled, there was no evidence that the P275A polymorphism had a significant association with PCa under dominant (OR=0.93, 95%CI=0.81-1.06, and p=0.28), co-dominant (homogeneous OR=0.97, 95%CI=0.56-1.68, and p=0.92; heterogeneous OR=0.93, 95%CI=0.74-1.15, and p=0.49), recessive (OR=1.10, 95%CI=0.65-1.87, and p=0.73), over-dominant (OR=0.93, 95%CI=0.75-1.15, and p=0.50), and allelic (OR=0.95, 95%CI=0.77-1.16, and p=0.61) genetic models. For stratified analyses by ethnicity and study design, no significant associations were found in the white race, the yellow race, the black race and mixed ethnicity, and the population-based case-control (PCC) and hospital-based case-control (HCC) studies under all genetic models. Conclusions: Based on our meta-analysis, the P275A polymorphism in the MSR1 gene is unlikely to be a risk factor for PCa.

        Size-Controlled Silver Nanoparticles Confined in Ordered Mesoporous Silica and Their Enhanced Catalytic Activities

        A cooperative assembly route has been developed, by which silver nanoparticles with controlled sizes are incorporated into the channels of ordered cubic mesoporous silica (KIT-6) with different pore sizes (4.3–6.4 nm). The samples were characterized by XRD, TEM, FT-IR, UV-Vis and N2 physisorption. The pore wall of mesoporous silica can efficiently confine the growth of silver nanoparticles within the channels and their average sizes decreased with the pore size reduction of KIT-6. Catalytic activities of the resultant Ag/KIT-6-x (x stands for hydrothermal temperature) composite for reducing harmful organic dye Rhodamine B (Rh B) by sodium borohydride (NaBH4) were investigated. All the Ag/KIT-6 composite samples show great catalytic activities, among which Ag/KIT-6-80 with higher loading and smaller size of Ag nanoparticles exhibits higher catalytic activity than those of Ag/KIT-6-60 and Ag/KIT-6-100.

      • Analysis of Prethrombotic States in Patients with Malignant Tumors

        Background: This study aimed to investigate the incidence and risk factors for a prethrombotic state in patients with malignant tumors. Materials and Methods: Plasma d-dimer (D-D) in patients with malignant tumors was measured. Abnormal rates of D-D and possible risk factors like gender, age, type of tumor, and staging of tumor were analyzed. Results: Of 1,453 patients, 629 demonstrated plasma D-D abnormality (43.3%). The D-D abnormal rate of male patients (n=851, 43.5%) was not statistically significantly different from that for female patients (n=602, 43.0%) (p>0.05). D-D abnormal rate increased with age and was statistically significant among different age groups (p<0.05). Regarding staging of tumor, D-D abnormal rate in patients with phase I was 2.0%, 6.2% in phase II, 47.6% in phase III and 83.1% in phase IV, with statistically significant differences between phase III and II, as well as phase III and IV (p<0.01). Conclusions: A prethrombotic state was closely related to malignancy of tumors. The risk factors for a prethrombotic state include age and tumor stage.

      • Phase II Study on Dose Escalating Schedule of Paclitaxel Concurrent with Radiotherapy in Treating Patients with Locally Advanced Non-small Cell Lung Cancer

        Objective: To evaluate clinical efficacy of a dose escalating schedule of paclitaxel concurrent with radiotherapy in treating patients with locally advanced non-small cell lung (NSCLC). Methods: Patients with locally advanced NSCLC were treated with conventional fractionated radiotherapy or three dimensional conformal radiotherapy (3 DCRT), concurrently with a dose escalating schedule of paclitaxel. All patients were divided into three groups, A with paclitaxel $30mg/m^2$, B with paclitaxel $60mg/m^2$ and C with paclitaxel $90mg/m^2$. Paclitaxel was repeated every week for a total of 4 or 6 weeks. Results: Among 109 patients, response rates were 68.8%, 71.1% and 71.8% (p>0.05) for group A (n=32), B (n=38), and C (n=39) respectively. Accordingly, disease control rates were 81.3%, 81.6% and 82.1% (p>0.05). Progression-free survival time was $8.0{\pm}5.0$ months, $11.6{\pm}6.1$ months, and $14.8{\pm}7.9$ months (p<0.05), respectively. Overall survival time was $15.4{\pm}7.6$ months, $18.2{\pm}8.0$ months, and $22.0{\pm}7.6$ months (p<0.05), one-year survival rates were 62.5%, 73.1% and 90.0% (p>0.05) and two-year survival rates were 31.3%, 38.5% and 50.0% (p<0.05). Main side-effects were bone marrow suppression, radiation related esophagitis and gastrointestinal reaction. Conclusion: In treating patients with NSCLC, concurrent chemoradiotherapy with paclitaxel improves early response compared with conventional fractionated radiotherapy or 3 DCRT. The survival rate was improved with the addition of paclitaxel, but there was an increase in adverse reactions when the dose of paclitaxel was increased.
