Novel reassortant 2.3.4.4B H5N6 highly pathogenic avian influenza viruses circulating among wild, domestic birds in Xinjiang, Northwest China

        Qian Zhang,Xindi Mei,Cheng Zhang,Juan Li,Nana Chang,Dilihuma Aji,Weifeng Shi,Yuhai Bi,Zhenghai Ma 대한수의학회 2021 Journal of Veterinary Science Vol.22 No.4

Background: The H5 avian influenza viruses (AIVs) of clade 2.3.4.4 circulate in wild and domestic birds worldwide. In 2017, nine strains of H5N6 AIVs were isolated from aquatic poultry in Xinjiang, Northwest China. Objectives: This study aimed to analyze the origin, reassortment, and mutations of the AIV isolates. Methods: AIVs were isolated from oropharyngeal and cloacal swabs of poultry. Identification was accomplished by inoculating isolates into embryonated chicken eggs and performing hemagglutination tests and reverse transcription polymerase chain reaction (RT-PCR). The viral genomes were amplified with RT-PCR and then sequenced. The sequence alignment, phylogenetic, and molecular characteristic analyses were performed by using bioinformatic software. Results: Nine isolates originated from the same ancestor. The viral HA gene belonged to clade 2.3.4.4B, while the NA gene had a close phylogenetic relationship with the 2.3.4.4C H5N6 highly pathogenic avian influenza viruses (HPAIVs) isolated from shoveler ducks in Ningxia in 2015. The NP gene was grouped into an independent subcluster within the 2.3.4.4B H5N8 AIVs, and the remaining six genes all had close phylogenetic relationships with the 2.3.4.4B H5N8 HPAIVs isolated from the wild birds in China, Egypt, Uganda, Cameroon, and India in 2016–2017, Multiple basic amino acid residues associated with HPAIVs were located adjacent to the cleavage site of the HA protein. The nine isolates comprised reassortant 2.3.4.4B HPAIVs originating from 2.3.4.4B H5N8 and 2.3.4.4C H5N6 viruses in wild birds. Conclusions: These results suggest that the Northern Tianshan Mountain wetlands in Xinjiang may have a key role in AIVs disseminating from Central China to the Eurasian continent and East African.

Fe/N-doped carbon framework derived from ZIF-8 on graphene oxide for efficient oxygen reduction reaction

        Yating Zhang,Pei He,Dongxian Zhuo,Jianlan Zhang,Nana Zhang,Xiaobo Wang,Gang Lin,Zhenghan Kong 대한금속·재료학회 2024 ELECTRONIC MATERIALS LETTERS Vol.20 No.2

Exploration of highly effi cient non-noble metal oxygen reduction reaction (ORR) catalysts is essential for the widespreadindustrial utilization of fuel cells. Herein, we demonstrated a hierarchical porous catalyst (denoted as Fe-NC-Gs) using asimple stirring and one-step pyrolysis method. ZIF-8-derived N-doped carbon framework loaded on graphene oxide, on whichFe 3 O 4 and FeS nanoparticles are uniformly dispersed. In the composite nanostructure, a high surface area (470.58 m 2 g −1 )and hierarchical porous structure were observed. The obtained Fe-NC-G-2 exhibits superior ORR properties. The half-wavepotential (E 1/2 ) and the limiting current density in alkaline media were up to 0.85 V and − 5.39 mA cm −2 , respectively, comparableto the commercially available Pt/C. The four-electron-dominated process was exhibited in the ORR catalysis. It alsomanifests a better methanol tolerance and electrochemical stability during the chronoamperometry measurement in 0.1 MKOH. These results suggest that the proposed strategy provides a new pathway to construct effi cient electrocatalysts for ORR.

Amino-modified carbon dots as a functional platform for drug delivery: Load-release mechanism and cytotoxicity

        Nana Shi,Kaiyue Sun,Zedi Zhang,Juan Zhao,Lina Geng,Yuhua Lei 한국공업화학회 2021 Journal of Industrial and Engineering Chemistry Vol.101 No.-

Carbon dot (CDs) were prepared for carrying DOX (Doxorubicin) or FAC (Ferric ammonium citrate), andthe loading mechanism and release kinetics of CDs-drugs were studied. CDs were prepared using peonypollen and urea, and characterized by TEM, XRD, 13C NMR, XPS, UV, FT-IR, DLS and zeta potential. Theobtained CDs were rich of –NH2 group on the surface, amorphous phase, spherical morphology withthe diameter about 5 nm, and had a blue fluorescence with the quantum yield (QY) of 12%. Iron disorderscan cause cancer or anemia. DOX is used for treating cancer, but has severe side effects. FAC is an ironsupplement agent, but had low bioavailability. CDs carried DOX (or FAC) through –N=C– (or –CONH–)bond, had high loading efficiency for DOX (37.2%) and FAC (54.0%) by optimizing mass ratios of mCDsto mdrug and reacted overnight. Cellular experiments showed that CDs-drugs had lower cytotoxicity thanfree drugs and CDs co-located with the drug. In vitro release indicated that CDs-DOX had pH-targetedproperty and CDs-FAC could be absorbed easily. CDs-drug release belonged to Weibull model, and releasemechanism was determined by the bonding way between CDs and drug. CDs had potential applicationsin delivery drugs with –C=O or –COOH group.

      • RST Invariant Fragile Watermarking for 2D Vector Map Authentication

Nana Wang,Jilong Bian,Han Zhang 보안공학연구지원센터 2015 International Journal of Multimedia and Ubiquitous Vol.10 No.4

2D vector maps are very important data in geographical information systems (GISs). It is considered to be content for which verification of integrity and authentication are urgently required. Using a marking technique and a rotation, scaling and translation (RST) invariant watermark generation method, we propose a scheme that detects and locates malicious attacks with high accuracy while providing the RST invariance property. In particular, we propose setting a unique mark on each feature, calculating the authentication watermark for each spatial feature group by folding the hash results of the differences of the log-radiuses, and embedding the watermark using a RST invariant watermarking method. While the watermark ensures both the sensitivity to malicious manipulations and the RST invariance property, the mark of each feature provides superior accuracy of tamper localization. Moreover, this algorithm is immune to feature rearrangement and vertex reversing operations. Theoretical analysis and experimental results are provided to demonstrate the effectiveness of our method.

IRE1α protects against osteoarthritis by regulating progranulin-dependent XBP1 splicing and collagen homeostasis

        Liang Li,Zhang Fengmei,Feng Naibo,Kuang Biao,Fan Mengtian,Chen Cheng,Pan Yiming,Zhou Pengfei,Geng Nana,Li Xingyue,Xian Menglin,Deng Lin,Li Xiaoli,Kuang Liang,Luo Fengtao,Tan Qiaoyan,Xie Yangli,Guo Fen 생화학분자생물학회 2023 Experimental and molecular medicine Vol.55 No.-

Osteoarthritis (OA) is a full-joint, multifactorial, degenerative and inflammatory disease that seriously affects the quality of life of patients due to its disabling and pain-causing properties. ER stress has been reported to be closely related to the progression of OA. The inositol-requiring enzyme 1α/X-box-binding protein-1 spliced (IRE1α/XBP1s) pathway, which is highly expressed in the chondrocytes of OA patients, promotes the degradation and refolding of abnormal proteins during ER stress and maintains the stability of the ER environment of chondrocytes, but its function and the underlying mechanisms of how it contributes to the progression of OA remain unclear. This study investigates the role of IRE1α/ERN1 in OA. Specific deficiency of ERN1 in chondrocytes spontaneously resulted in OA-like cartilage destruction and accelerated OA progression in a surgically induced arthritis model. Local delivery of AdERN1 relieved degradation of the cartilage matrix and prevented OA development in an ACLT-mediated model. Mechanistically, progranulin (PGRN), an intracellular chaperone, binds to IRE1α, promoting its phosphorylation and splicing of XBP1u to generate XBP1s. XBP1s protects articular cartilage through TNF-α/ERK1/2 signaling and further maintains collagen homeostasis by regulating type II collagen expression. The chondroprotective effect of IRE1α/ERN1 is dependent on PGRN and XBP1s splicing. ERN1 deficiency accelerated cartilage degeneration in OA by reducing PGRN expression and XBP1s splicing, subsequently decreasing collagen II expression and triggering collagen structural abnormalities and an imbalance in collagen homeostasis. This study provides new insights into OA pathogenesis and the UPR and suggests that IRE1α/ERN1 may serve as a potential target for the treatment of joint degenerative diseases, including OA.

Polymorphism of Exon 2 of BMP15 Gene and Its Relationship with Litter Size of Two Chinese Goats

        Wang, Yuqin,Yuanxiao, Li,Nana, Zhang,Zhanbin, Wang,Junyan, Bai Asian Australasian Association of Animal Productio 2011 Animal Bioscience Vol.24 No.7

Polymorphisms of BMP15 gene exon 2 and its relationship with prolificacy of goats were detected by PCR-SSCP and DNA sequencing methods in Chinese two local goat breeds. The results showed that the product amplified by the primers displayed polymorphisms. Three genotypes (AA, BB and AB) were detected in Funiu white goats, and their frequency was 0.071, 0.715, 0.214, respectively. Two genotypes (AB and BB) were detected in Taihang black goats, and their frequency was 0.342 and 0.658, respectively. Sequencing revealed that four mutations (456T${\rightarrow}$G, 466C${\rightarrow}$G, 510C${\rightarrow}$T, 511T${\rightarrow}$C) occurred in genotype BB of Funiu white goat, which resulted in amino acid substitution of V155G and S171P. No mutation was detected in Taihang black goat. The Funiu white goat with genotype BB had 0.91 or 0.82 kids, more than those with AB or AA, respectively. The difference of the least squares means for litter size between BB and AB was not significant (p>0.05) in Taihang black goat. It is concluded that the BMP15 gene may be a major gene which affects the prolificacy in Funiu white goats. This study could provide basic molecular data on the reproductive characteristics of local breeds of Henan province in China, and a scientific basis for the conservation and utilization of those two goat breeds.
