        Genetic heterogeneity of liver cancer stem cells

        Cancer cell heterogeneity is a serious problem in the control of tumor progression because it can cause chemoresistance and metastasis. Heterogeneity can be generated by various mechanisms, including genetic evolution of cancer cells, cancer stem cells (CSCs), and niche heterogeneity. Because the genetic heterogeneity of CSCs has been poorly characterized, the genetic mutation status of CSCs was examined using Exome-Seq and RNA-Seq data of liver cancer. Here we show that different surface markers for liver cancer stem cells (LCSCs) showed a unique propensity for genetic mutations. Cluster of differentiation 133 (CD133)-positive cells showed frequent mutations in the IRF2, BAP1, and ERBB3 genes. However, leucine-rich repeat-containing G protein-coupled receptor 5-positive cells showed frequent mutations in the CTNNB1, RELN, and ROBO1 genes. In addition, some genetic mutations were frequently observed irrespective of the surface markers for LCSCs. BAP1 mutations was frequently observed in CD133-, CD24-, CD13-, CD90-, epithelial cell adhesion molecule-, or keratin 19-positive LCSCs. ASXL2, ERBB3, IRF2, TLX3, CPS1, and NFATC2 mutations were observed in more than three types of LCSCs, suggesting that common mechanisms for the development of these LCSCs. The present study provides genetic heterogeneity depending on the surface markers for LCSCs. The genetic heterogeneity of LCSCs should be considered in the development of LCSC-targeting therapeutics.

        Infection with Citrobacter rodentium in μMT Knockout Mice

        μMT knockout mice are genetically deficient in the transmembrane domain of mu chain of the immunoglobulin M (IgM) heavy chain, resulting in the absence of mature B cells. μMT knockout mice is an in vivo model system used to clarify the role of B cells in various diseases. Enteropathogenic Escherichia coli (EPEC) induces acute and chronic diarrheal disease, especially in children of developing countries. The formation of attaching and effacing (A/E) lesion is a prominent pathogenic factor in the intestinal epithelium of EPEC infection. The A/E lesion is modulated by genes located on the pathogenic island locus of enterocyte effacement (LEE) which encode a type III secretion system (T3SS) and A/E lesion-related effector proteins. Citrobacter rodentium is a murine pathogen utilized in studying the pathogenic mechanisms of EPEC in human infections. Citrobacter rodentium produce A/E lesion to attach to intestinal epithelium, thus providing a murine model pathogen to study EPEC. Several studies have investigated the pathogenesis of Citrobacter rodentium in the μMT knockout mice. In this review, we introduce the μMT murine model in the context of C. rodentium pathogenesis and describe in detail the role of B cells and antibodies in this disease.

      • Effects of advance care planning training for advanced practice nurse students in South Korea

        Aim(s): Effective communication between health care providers and patients and their families is one of the most important components of quality end-of-life care. However, nurses in South Korea are likely to be uncomfortable communicating advance care planning (ACP) due to the cultural taboo against talking about dying. The purpose of this study was to assess how an ACP training program affects Korean advanced practice nurse (APN) students’ ACP competencies. Method(s): Data collected from 34 APN students who participated in ACP training program conducted as part of graduate clinical practice course was analyzed. A training program was provided to APN students to improve their competencies in ACP conversations with patients. The program comprised three sessions: online lectures, face-to-face simulation, and discussions on ACP and ethical issues. After completing the program, the APN students were asked to describe their experiences with the training. A Wilcoxon matched pairs signed rank test was used to examine the changes in ACP knowledge, confidence in supporting patients’ advance directives, perceived nursing roles in end-of-life (EOL) treatment decisions, and perception of a good death before and after the training program. Result(s): APN students reported that their experience was positive and helpful. There was a statistically significant increase in participants’ ACP knowledge (p < .001), confidence in supporting patient’ advance directives (p < .018), and perception of the active role of nurse in patient EOL treatment decisions (p < .001) after the training. Conclusion(s): The results indicate the effects of training program on APN students’ competencies for EOL care. They also provide evidence about what contents and methods can be helpful in developing an EOL care training for APN students.

        인사부문의 전략적 참여와 고성과작업시스템의 실행-지각의 차이

        본 연구는 선행연구에서 깊게 다루지 않았던 고성과작업시스템의 실행-지각의 차이에 대해 고찰하고, 사회적자본과 권력 및 영향력 관점에서 인사부문의 전략적 참여가 인사관리강도와 고성과작업시스템의 실행-지각의 차이에 미치는 영향을 살펴보았다. 특히 인사부문의 전략적 참여와 고성과작업시스템의 실행-지각의 차이 간의 관계에서 인사관리강도의 매개효과를 검증함으로써 구체적인 메커니즘을 파악하고자 하였다. 국내 76개 조직의 474명 조직구성원들을 대상으로 실증적 연구를 실시한 결과, 인사부문의 전략적 참여는 고성과작업시스템의 실행-지각의 차이를 줄이는 것으로 나타났으며, 이 과정에서 인사관리강도가 부분매개역할을 하는 것으로 나타났다. 분석결과를 토대로 본 연구의 시사점과 한계점 및 향후 연구과제 등에 대해 논의하였다. The purpose of this study is to examine the influence of HR involvement on high performance work systems(HPWS) gap. In addition, we attempt to verify the mediating effect of HRM system strength between HR involvement and HPWS gap. To test the proposed hypotheses in our study, we analyzed the data set of 474 full-time employees from 76 Korean organizations in several industries. Hierarchical regression analysis is used to test the proposed hypotheses and sobel test is used to test the significance of the mediating effects of HRM system strength. And industry, union presence, the size, organization age were controlled in the model. The major findings are as follows. First, as predicted, HR involvement was negatively associated with HPWS gap. Second, HR involvement was posi- tively associated with HRM system strength. Third, the mediation effect of HRM system strength on the relationship between HR involvement and HPWS gap was partially supported. We also discussed some implications and future research directions.

        인사관리강도와 인사관리효과성의 관계에 관한 연구

        전략적 인적자원관리에 대한 그동안의 연구는 인사제도의 효과에 초점을 둔 내용(content)연구 중심으로 진행되어 인사관리가 어떻게 실행되고 그 효과는 어떠한가에 관한 과정(process)중심 연구가 미흡하였다. 이에 본 연구는 과정중심 접근방식에 입각하여 인사관리강도가 인사제도풍토와 인사관리효과성에 미치는 영향을 분석하였다. 국내 72개 기업, 443명의 종업원을 대상으로 실증적 연구를 실시한 결과, 인사관리강도는 인사관리효과성에 긍정적인 영향을 미쳤으며, 이 과정에서 인사제도풍토가 매개역할을 하는 것으로 나타났다. 이러한 결과를 바탕으로 본 연구의 시사점, 한계점 및 향후 연구과제 등에 대해 논의하였다. Theorists of strategic human resource management (HRM) who have been interested in clarifying the relationship and underlying processes between HRM and organizational performance tend to employ two different but related approaches: a contend-based approach and a process-based approach. First, The main concern of content-based approach is an effective HR system usually defined as a well designed set of HR practices necessary for achieving a firm’s strategy, such as high performance or high commitment work systems. Research in this area focuses on exploring the direct relationship between HR systems and firm performance or internal processes that how HRM practices can improve firm performance. Second, unlike a content-based approach, a process based approach highlights the way messages of HRM communicated to employees. The role of HRM practices is to send messages to employees that allow them to understand the desired and appropriate responses in order to achieve an organizational goal (Bowen & Ostroff, 2004). Research in this area focuses on the communication role of the HR practices that how they should be implemented in order to allow employees to clearly perceive what is needed. Given a desired content of the HRM system, the HRM system may still not elicit appropriate collective behaviors and attitudes needed for effectiveness, because individuals may interpret the HRM practices idiosyncratically, leading to variability in psychological climate perceptions (Bowen & Ostroff, 2004). Therefore, the manner in which HRM is implemented is very important. Despite the impotance of studies on the process of HRM system in the field of strategic human resource management research, however, most previous works have concentrated on the content of HRM system. Along this line of reasoning, Bowen & Ostroff (2004) proposed the concept of the strength of human resource management (HRM system strength) as comprising three features: distinctiveness, consistency, and consensus. which are required to allow employees to properly perceive what the HRM system is intended. Although the theoretical implication of Bowen & Ostroff (2004)’s HRM system strength have been supported by many researchers, there is very limited number of empirical studies on the processes and outcomes of it because of the absence of its appropriate measurement. In particular, few research on the HRM system strength has been performed in the context of Korea (Kang et al., 2011). In this regard, using process-based approach, we examined the effect of HRM system strength on HR effectiveness placing emphasis on the role of communication for improving the HR effectiveness. HR effectiveness, defined as a firm’s HR activities contribute to the achievement of a firm’s strategic goals, is an important antecedent of firm performance (Ryu & Kim, 2007; Wright et al., 2001). Despite its importance, a study on communication role of HR department to allow HRM to be implemented effectively have not been tried. This study fills this void. In addition, based on the work of Bowen and Ostroff (2004), we examined if HR climate, defined as employees’ shared perceptions of the HR system, has a mediating effect in the relationship between HRM system strength and HR effectiveness. HR climate seems to be predicted directly by HRM system strength, but the empirical findings suffer from a lack of agreement (ex: Li, Frenkel, & Sanders, 2011; Sanders, Dorenbosch, & Reuver, 2008). On basis of a literature review, the following hypotheses were postulated: Hypothesis 1: HRM system strength is positively related to HR effectiveness. Hypothesis 2: HRM system strength is positively related to HR climate. Hypothesis 3: Climate strength has a mediating effect in the relationship HRM system strength and HR effectiveness.         앙상블 머신러닝 모델을 활용한 치매 예측

        고령화 사회가 되어가면서 노인 질병에 관해 관심이 높아지고 있다. 그 중, 일상생활에 지장을 많이 주는 노인성 치매는 기술이 발달하여도 완치할 수 있는 치료법이 없기 때문에 초기에 발견하는 것이 가장 좋은 방법이다. 하지만 이러한 치매 예측을 위하여 수많은 사전 연구들에서는 MRI, 망막 이미지 영상 데이터 등 지속적인 촬영과 비용이 필요하다. 이러한 방법들은 모든 사람들에게 적용시킬 수 없다는 단점을 가지고 있다. 이에 본 연구에서는 치매 예측 발견의 저비용에 관한 발전을 위하여 운동 시간, 고강도 활동 시간, 신진대사량 등 여러 활동 데이터와 깊은 수면 시간, 분 당 호흡 수 등 수면의 질을 알 수 있는 수면 데이터를 활용하여 XGBsoot 와 lightGBM과 같은 딥러닝 예측 모델을 이용하여 치매 여부에 가장 영향을 많이 미친 주요 변수들을 알아내는 것을 목적으로 한다. 분석 결과, 활동 데이터와 수면 데이터를 동시에 고려했을 때 치매 예측 성능이 가장 잘 나왔고, XGBoost 모델에서 가장 중요한 변수는 ‘매 시간당 활동 유지 점수’이었고, lightGBM은 ‘분당 평균 심박동 수’ 이었다. As we become an aging society, there is a growing concern about geriatric diseases. Among them, senile dementia, which disrupts daily life, is best detected at an early stage because there is no cure, even with technological advances. However, many preliminary studies to predict dementia require continuous imaging data, such as MRI and retinal imaging data, and are expensive. These methods have the disadvantage that they cannot be applied to all people, so this study aims to develop low-cost predictive dementia detection. Therefore, the purpose of this study is to identify the main variables that most influenced the presence of dementia using deep learning prediction models such as XGBsoot and lightGBM by utilizing various activity data such as exercise duration, high-intensity activity duration, and metabolic rate, and sleep data such as deep sleep duration and breathing rate per minute to determine sleep quality. The analysis showed that the best dementia prediction performance was achieved when both activity and sleep data were considered simultaneously, and the most important variable in the XGBoost model was the “activity_score_move_every_hour” and the “sleep_hr_everage” in lightGBM.

        이타미 준의 전시공간에 나타난 비판적 지역주의에 관한 연구 - 케네스 프램톤의 비판적 지역주의를 중심으로 -

        모더니즘 건축이 추구하는 보편성은 표준화와 함께 전 세계적으로 지역의 몰개성화를 불러오며 건축의 풍토성과 장소성의 상실을 가져오게 되었다. 이에 대한 대안으로 등장했던 케네스 프램튼의 비판적 지역주의는 모더니즘의 진보적 정신을 살리며 지역적 특성들과 결합된 실천적 대안의 형태로 제시되었다. 이러한 비판적 지역주의의 특성은 전통성의 재해석을 바탕으로 한 자신의 독자적인 모더니티를 추구하는 이타미준의 전시공간의 건축에서도 찾아볼 수 있었다. 비판적 지역주의의 장소 형성하기, 테토닉한 형태, 물리적인 감각의 추구 등의 특성들은 지역의 자연환경과의 융화를 꾀하며, 지역의 건축재료와 독창적인 구축 방식을 통해 다양한 감각의 소통을 추구하는 이타미 준의 전시 건축에서도 나타나며 분석되었다. 풍토, 경관, 지역의 문맥에서 핵심적인 본질을 찾아 건축의 의미와 정신을 결합시킨 이타미 준의 건축 방법은 지역의 정체성을 회복하는 현대 건축의 대안적 방법으로서 기여할 수 있을 것으로 판단되었다. The universality in modern architecture leading to standardization of the region has resulted in the loss of architectural place. Kenneth Framton's critical regionalism, which emerged as an alternative solution was presented as a practical alternative with regional characteristics. These characteristics of critical regionalism was also found in the exhibition architecture space of Itami Jun, who pursued his own modernity based on the reinterpretation of tradition. The characteristics of forming place, the tectonic form, and the tactile sensation in critical regionalism, are analyzed in the architecture of Itami Jun, seeking to integrate with the natural environment and pursuing various senses of communication through local materials of original tectonic construction. Itami Jun's architecture constructing core meaning and essential spirit of architecture may contribute as an alternative method of modern architecture to restore local identity.
