        실험적 급성 허혈성 신부전에서 Verapamil 이 혈중 Polyamine 동태에 미치는 영향

        N/A It has been proposd that calcium entry from an external medium increases intracellular free calcium to toxic levels during ischemic acute renal failure, and verapamil (ARF) has been suggested as the agentblocking calicium entry into renal cells and protecting renal function during ischemic injury. Polyamines, anorganic cations that play various roles in normal cellular proliferation and differentiation, accumulate in renal failare. Alsa it has been suggested tht the synthesis and metabolism of polyamine are influence by acute changes of the general condition, such as acute renal failure, and are mediated by a calcium influx into the cells. The study was designed to study the protective effects of systemic verapamil pretreatment on renal function and the influence on polyamine metabolism in experimental ischemic ARF in cats. For these purpose, the experimental animals were divided into 2 groups. While Group I (n=5) was an ischemic ARF model by renal artery clamping for 60 minutes, Group II (n=5) was ischemic ARF with systemic verapamil (5 ml/min/kg) pretreatment. The creatinine clearance and plasma and urinary polyamine were measured in each group before and after the renal artery clamp. The results were as follows: 1) Creatinine clearance before and after the renal artery clamp were 10.64±7.18 ml/min/kg and 2.09±1. 70 ml/min/kg in Group I, 4.47±3.38 ml min/kg and 0.60±0.79ml/min/kg in Group II, respectively, So creatinine clearance decreased more significantly in Group II campared with Group I. 2) Plasma polyamine increased after ischemia in Group I. In group I, plasma levels of putrescine, spermidine, and spermine before ischemia were 4.75±0.40 nmol/ml, 0.69±0.09 nmol/ml, and 0.83±0.63 nmol/ml, were elevated to 7.17±2.91 nmol/ml, 9.83±1.46 nmol/ ml, and 2.64±1.14nmol/ml after ischemia. But in Group II, the plasma level of polyamine was not changed, and especially, spermine decreased significantly from 0.83±0.27 before ischemia to 0.49±0.23 nmol/ml after ischenmine (p=0.033). 3) Urine polyamine excretion decreased after ischemia in Group I and Group II. In Group II, urinary excretion of spermidine and spermine before ischemia, 0.13±0.10 nmol/min and 0.17±0.13nmol/min, decreased after ischemia to 0.01±0.01nmol/min (p=0.019) and 0.032±0.26 nmol/min (p=0.0257). 4) In renal tissue, spermine content vas highest. In Group II, preischemic spermine were 397.20 nmol/g and increased to 646.66nmol/g after ischema, But there were no significant changes in the polyamine contents in Group II. From these data, it was suggested that systemic verapamil pretreatment exerts no protective effect on ischemic ARF. Plasma polyamines are elevated in ischemic ARF, and verapamil may protect these elevations.

        The incidence of aseptic meningitis infection is ensuing and threatening the health of children. Enteroviruses are the major agents of aseptic meningitis and identification of virus has been a clue to diagnosis and epidemiology. The outbreak of aseptic meningitis occurred in Pusan, 1998. Patients were concentrated from April through November. Children were more susceptible than adults. Among 306 cases of specimens from stool, throat swab tested, only 7.2% were positive on virus isolation, 12 cases from stool and 10 from throat, respectively. All isolated 7 serotypes of viruses represented cytopathic effect on cultured cells. Three types of echovirus 6, 25, 30 and coxsackievirus B2, B3, B4, B6 were identified by neutralizing anti body test. Isolated coxsackievirus and echovirus were observed by an electron microscope with negative staining.

      • Structure–function studies of a plant tyrosyl-DNA phosphodiesterase provide novel insights into DNA repair mechanisms of <i>Arabidopsis thaliana</i>

        <P>TDP1 (tyrosyl-DNA phosphodiesterase 1), a member of the PLD (phospholipase D) superfamily, catalyses the hydrolysis of a phosphodiester bond between a tyrosine residue and the 3′-phosphate of DNA. We have previously identified and characterized the <I>AtTDP</I> gene in <I>Arabidopsis thaliana</I>, an orthologue of yeast and human <I>TDP1</I> genes. Sequence alignment of TDP1 orthologues revealed that AtTDP has both a conserved C-terminal TDP domain and, uniquely, an N-terminal SMAD/FHA (forkhead-associated) domain. To help understand the function of this novel enzyme, we analysed the substrate saturation kinetics of full-length AtTDP compared with a truncated AtTDP mutant lacking the N-terminal FHA domain. The recombinant AtTDP protein hydrolysed a single-stranded DNA substrate with <I>K</I><SUB>m</SUB> and <I>k</I><SUB>cat</SUB>/<I>K</I><SUB>m</SUB> values of 703±137 nM and (1.5±0.04)×10<SUP>9</SUP>M<SUP>−1</SUP>·min<SUP>−1</SUP> respectively. The AtTDP-(Δ1–122) protein (TDP domain) showed kinetic parameters that were equivalent to those of the full-length AtTDP protein. A basic amino acid sequence (RKKVKP) within the AtTDP-(Δ123–605) protein (FHA domain) was necessary for nuclear localization of AtTDP. Analysis of active-site mutations showed that a histidine and a lysine residue in each of the HKD motifs were critical for enzyme activity. Vanadates, inhibitors of phosphoryl transfer reactions, inhibited AtTDP enzymatic activity and retarded the growth of an <I>Arabidopsis tdp</I> mutant. Finally, we showed that expression of the <I>AtTDP</I> gene could complement a yeast <I>tdp1</I>Δ<I>rad1</I>Δ mutant, rescuing the growth inhibitory effects of vanadate analogues and CPT (camptothecin). Taken together, the results of the present study demonstrate the structure-based function of AtTDP through which AtTDP can repair DNA strand breaks in plants.</P>

        Forty eight strains of yeast were cultured in potato dextorse(PD) broth at 30˚C for 24 hr and centrifuged with 12,000 rpm for 20 min. After concentrated the cultures, antihypertensive angiotensin I-converting enzyme(ACE) inhibitory activities of its concentrates were investigated. Among them, the concentrates from Saccharomyces cerevisiae Y183-3 showed the highest ACE inhibitory activity of 71.8%. The ACE inhibitor from Saccharomyces cerevisiae Y183-3 was maximally produced when Saccharomyces cerevisiae Y183-3 cultured in PD broth at 30˚C for 36 hr.

        Effect of sous-vide cooking conditions on the physicochemical, microbiological and microstructural properties of duck breast meat

        Objective: Sous-vide cooking offers several advantages for poultry meat, including enhanced tenderness, reduced cooking loss, and improved product yield. However, in duck meat, there are challenges associated with using the sous-vide method. The prolonged cooking time at low temperatures can lead to unstable microbial and oxidative stabilities. Thus, we aimed to assess how varying sous-vide cooking temperatures and durations affect the physicochemical and microbial characteristics of duck breast meat, with the goal of identifying an optimal cooking condition. Methods: Duck breast meat (Anas platyrhynchos) aged 42 days and with an average weight of 1,400±50 g, underwent cooking under various conditions (ranging from 50℃ to 80℃) for either 60 or 180 min. Then, physicochemical, microbial, and microstructural properties of the cooked duck breast meat were assessed. Results: Different cooking conditions affected the quality attributes of the meat. The cooking loss, lightness, yellowness, Hue angle, whiteness, and thiobarbituric acid reactive substance (TBARS) values of the duck breast meat increased with the increase in cooking temperature and time. In contrast, the redness and chroma values decreased with the increase in cooking temperature and time. Cooking of samples higher than 60℃ increased the volatile basic nitrogen contents and TBARS. Microbial analysis revealed the presence of Escherichia coli and Coliform only in the samples cooked at 50℃ and raw meat. Cooking at lower temperature and shorter time increased the tenderness of the meat. Microstructure analysis showed that the contraction of myofibrils and meat density increased upon increasing the cooking temperature and time. Conclusion: Our data indicate that the optimal sous-vide method for duck breast meat was cooking at 60℃ for 60 min. This temperature and time conditions showed good texture properties and microbial stability, and low level of TBARS of the duck breast meat.

        다양한 적송잎 추출물의 항생제 내성 황색포도상구균에 대한 항균활성 검증

        병원 내 감염의 주된 원인균으로 알려진 황색포도상구균(5. aureus)과 항생제 내성 황색포도상구균(PRSA, MRSA)에 대해 항균활성이 우수한 천연 항균성 물질을 검색하기 위해 적송잎을 대상으로 열수-헥산(HWH), 열수-에탄올(HWE), 헥산, 에탄올 및 열수 등으로 획득된 추출물들로 항균활성을 조사하였다. 헥산, 에탄올, 열수-헥산, 열수-에탄올 추출물들은 그람양성 대표세균인 고초균(B. subfilis), 황색포도상구균 표준균주, 항생제 내성균주인 PRSA 및 MRSA에 대해서 항균활성을 나타내었으며, 특히 MRSA (ATCC 33591) 균주에 대해 다른 균주들 보다 높은 항균활성을 나타냈다. 열수 추출물은 황색포도상구균 표준균주 및 MRSA에서만 항균활성을 나타냈지만, 항균활성은 가장 높아 50 ㎎/㎖의 농도에서 33 ㎜의 생육저해환을 나타냈다. 각 균주들에 대한 생장의 최소저해 농도(MIC)는 HWH 및 HWE 추출물이 모두 0.05 ㎎/㎖의 농도로 가장 낮았고, 열수추출물이 0.5 ㎎/㎖의 농도였으며, 에탄올 추출물은 5 ㎎/㎖의 가장 높은 농도를 나타냈다. 모든 추출물들은 121℃에서 20분간의 열처리 후에도 항균활성이 유지되었으므로 열에 대해 안정한 물질임을 알 수 있었다. 본 연구결과로 식품산업에서 솔잎 추출물을 천연 항균제로 사용할 수 있을 것으로 기대할 수 있다. investigated antimicrobial activities of various pine (Pinus densiflora) needle extracts against antibiotic resistant strains of Staphylococcus aureus. Hot water extract showed the highest antimicrobial activity against normal and methicillin resistant Staphylococcus aureus (MRSA), however, it exhibited no antimicrobial activity against penicillin resistant S. aureus (PRSA). Hot water-hexane (HWH), hot water-ethanol (HWE), hexane, and ethanol extracts showed antimicrobial activity against S. aureus, PRSA and MRSA. Minimum inhibitory concentrations (MIC) of HWH, HWE, hexane, and ethanol extracts were 0.05, 0.05, 0.5 and 5 ㎎/㎖ respectively, and HWH and HWE extracts showed the strongest antimicrobial activity among these extracts. Antimicrobial activities of pine needle extracts were stable after heating at 121℃ for 20 min. These results suggested that pine needle extracts can be used as an effective natural antimicrobial agent for food and medical industries.

        전경대원에서 발생한 결핵의 임상 양상

        Background: The purpose of this study was to document the incidence and clinical characteristics of patients with tuberculosis (TB) in combat and auxiliary police, living in a group, in Korea where the incidence rate of active TB in a general population is higher than in Western countries. Materials and Methods:We retrospectively reviewed the medical records of all tuberculosis patients diagnosed at National Police Hospital from January 2002 through December 2004. Results:In 2002-2004, a total of 156 cases of tuberculosis were identified with the mean (Standard deviation) age of 20.6 (±1.0) years. Of these, 134 (85.9%) patients were registered as new cases, 11 (7.1%) as relapse, 2 (1.3%) as failure, 5 (3.1%) as treatment-after-default cases whereas 4 (2.6 %) patients were not included in any categories. Average annual new TB rate and smear-positive TB rate were 86.5/10^(5) and 17.4/10^(5) person-years, respectively. In 12 of 31 smear-positive cases, time from onset of symptoms to diagnosis was more than 30 days. Two multidrug-resistance TB cases were identified and two suspected outbreak episodes of TB had occurred during 3 years. Conclusion:There was no statistically significant difference in the incidence rate of new cases of TB between the general population aged 20 to 29 years and combat and auxiliary police in Korea. 배경 : 국내 결핵의 발생은 감소 추세에 있으나 여전히 후진국형 발생양상을 보이고 있다. 특히 집단생활을 하는 젊은이들에서 발생하는 결핵의 유행이 간헐적으로 알려지고 있어 우려를 낳고 있다. 이에 저자들은 최근 전경대원의 결핵 발병 양상에 대해 알아보고자 본 연구를 시행하였다. 재료 및 방법 : 2002년 1월부터 2004년 12월까지 3년간 경찰병원에서 결핵 진료를 받은 전경대원의 의무기록을 후향적으로 분석하여 연도별 결핵 신환발생률, 도말양성결핵 신환율, 진단 지연, 집단발병, 다제내성결핵을 조사하였다. 결과 : 연도별 결핵 신환자 수(10만명당 발생추정치)는 2002년 50명(83.6명), 2003년 42명(83.7명), 2004년 42명(93.5명)이었고 도말양성 폐결핵 신환자 수는 2002년 10명(16.7명), 2003년 11명(21.9명), 2004년 6명(13.4명)이었다. 3년간 도말양성 폐결핵 환자는 31명이 있었고 그 중 12명은 호흡기 증상 발생 30일이 지나서 진단을 받았다. 다제내성 결핵은 2명이 있었으며 결핵의 집단발병이 의심되는 사례는 2차례 있었다. 결론 : 전경대원과 20-29세 연령군 일반인의 결핵 신환발생률, 도말양성 폐결핵 발생률의 통계적으로 유의한 차이는 없었다.

        A 48-year-old male visited the emergency room of the authors’ hospital due to nausea, vomiting, and myalgia for four days. Acute hepatitis A was identified from the serologic marker of the hepatitis A virus. Mild elevation of the serum creatinine and creatinine phosphokinase(CPK) suggested rhabomyolysis, which was confirmed with the serum aldolase, myoglobin, and urine myoglobin. With supportive care, both the liver and renal func- tions were recovered gradually and fully. This case shows that rhabdomyolysis can be one of the mechanisms of renal complication in cases of acute symptomatic hepatitis A.

        We consider the problem of identifying and testing multiple outlier in weighted linear regression models. When case wieights are given because of variance heterogeneity, we can estimate efficiently regression parameters using weighted least squares estimators are sensitive to outliers like ordinary least squares estimations. Thus, in this paper, We propose some statistics for regression diagnostics in weighted least square regression. On the other hand, we introduce testing procedures for the detection of multiple outliers that appear to be less sensitive to this problem. Both procedures attempt to separate the data into a set of "Clean" data points and a set of points that contain the potential outliers. The potential outliers are then tested to see how extreme they are relative to the clean subset, using an appropriately scaled version of the prediction error. As the outlier-detection methods of H.S(HadiandSimonoff), in this study, we would like to suggest that we uses σ² which definite of back and forth is trimmed instead of σ² is used on the second step of H.S method after arranging γ², in order.

      • p-Anisaldehyde가 조골세포 분화에 미치는 영향

        p-Anisaldehyde is a natural fragrance extracted from Pimpinella anisum L., and used as a preservative. This study examined the effect of p-anisaldehyde on osteoblast differentiation. First, cytotoxicity tests were carried out after examining the concentrations that did not show toxicity to the cells. The expression of osteoblast differentiation marker gene was confirmed by RT-PCR and was not effective in the treatment with p-anisaldehyde alone. However, p-anisaldehyde decreased the expression levels of inhibitor of differentiation-1 (Id1), distalless related homeobox (Dlx5), and runt-related transcription factor 2 (Runx2), which are osteogenic differentiation marker genes, which are increased in osteogenic condition. alkaline phosphatase (ALP) activity was also confirmed by ALP staining that p-anisaldehyde reduced ALP activity. These results show that p-anisaldehyde is effective in reducing osteoblast differentiation.
