      • Expression and Prognostic Implications of FOXO3a and Ki67 in Lung Adenocarcinomas

        Liu, Hong-Bin,Gao, Xiang-Xiang,Zhang, Qing,Liu, Jian,Cui, Yuan,Zhu, Yan,Liu, Yi-Fei Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.4

To investigate the significance of FOXO3a and Ki67 in human lung adenocarcinomas. Envision immunohistochemical staining and Western blotting were used to examine the protein expression of FOXO3a in 127 cases of human lung adenocarcinoma specimens. The positive rate in lung adenocarcinoma (55.9%) was lower than that in normal tissues (80%). We found that the expression of FOXO3a was closely related with the degree of differentiation, TNM staging, lymph node metastasis and survival. In addition, significant differences in the different pathological types of lung adenocarcinoma cases (P<0.01). The FOXO3a positive rate of the acini as the main type (APA) (86.7%) and the lepidic as the main type (LPA) (82.4%) was higher than the solid as the main type (SPA) (50.0%), the papilla as the main type (PPA) (42.9%) and the micropapilla as the main type (MPA) (9.4%). Moreover, the expression of FOXO3a was negatively related with Ki67 expression. Our results suggested that the expression of FOXO3a is closely correlated with the aggressiveness of lung adenocarcinoma. It was indicated that disregulation of FOXO3a might play key roles in the occurrence and development of lung a denocarcinoma and joint detection of the two markers might play an important role in diagnosing tumors.

RhoGDI2 induced malignant phenotypes of pancreatic cancer cells via regulating Snail expression

        Yi Bin,Hu You,Zhu Dongming,Yao Jun,Zhou Jian,Zhang Yi,He Zhilong,Zhang Lifeng,Zhang Zixiang,Yang Jian,Tang Yuchen,Huang Yujie,Li Dechun,Liu Qiuhua 한국유전학회 2022 Genes & Genomics Vol.44 No.5

Background: Rho GDP dissociation inhibitor 2 (RhoGDI2) has been shown to contribute to the aggressive phenotypes of human cancers, such as tumor metastasis and chemoresistance. Objective: This study aimed to assess the effects of RhoGDI2 on tumor progression and chemoresistance in pancreatic cancer cells. Methods: The expression of RhoGDI2 in pancreatic cancer cells was detected by Western blot analysis. Gain-of-function and loss-of-function approaches were done to examine the malignant phenotypes of the RhoGDI2-expressing or RhoGDI2-depleting cells. The correlation between RhoGDI2 and Snail was also analyzed. Results: Differential expression of RhoGDI2 protein in pancreatic cancer cell lines was identified. Gain-of-function and loss-of-function experiments showed that RhoGDI2 induced the malignant phenotypes of pancreatic cancer cells, including proliferation, migration, invasion, and gemcitabine (GEM) chemoresistance. The upregulation of RhoGDI2 stimulated the expression of Snail, resulting in the altered expression of epithelial marker E-cadherin and mesenchymal marker Vimentin, which were characteristics of the tumorigenic activity of epithelial-mesenchymal transition. The expression of RhoGDI2 and Snail was upregulated in clinical tumor samples, and higher expression of RhoGDI2 or Snail was significantly associated with poor patient survival in pancreatic ductal adenocarcinoma (PDAC). Conclusion: The findings indicated that RhoGDI2 promoted GEM resistance and tumor progression in pancreatic cancer and that RhoGDI2 might be a potential therapeutic target in patients with PDAC.

      • Divergent global precipitation changes induced by natural versus anthropogenic forcing

        Liu, Jian,Wang, Bin,Cane, Mark A.,Yim, So-Young,Lee, June-Yi Nature Publishing Group, a division of Macmillan P 2013 Nature Vol.493 No.7434

As a result of global warming, precipitation is likely to increase in high latitudes and the tropics and to decrease in already dry subtropical regions. The absolute magnitude and regional details of such changes, however, remain intensely debated. As is well known from El Niño studies, sea-surface-temperature gradients across the tropical Pacific Ocean can strongly influence global rainfall. Palaeoproxy evidence indicates that the difference between the warm west Pacific and the colder east Pacific increased in past periods when the Earth warmed as a result of increased solar radiation. In contrast, in most model projections of future greenhouse warming this gradient weakens. It has not been clear how to reconcile these two findings. Here we show in climate model simulations that the tropical Pacific sea-surface-temperature gradient increases when the warming is due to increased solar radiation and decreases when it is due to increased greenhouse-gas forcing. For the same global surface temperature increase the latter pattern produces less rainfall, notably over tropical land, which explains why in the model the late twentieth century is warmer than in the Medieval Warm Period (around ad 1000–1250) but precipitation is less. This difference is consistent with the global tropospheric energy budget, which requires a balance between the latent heat released in precipitation and radiative cooling. The tropospheric cooling is less for increased greenhouse gases, which add radiative absorbers to the troposphere, than for increased solar heating, which is concentrated at the Earth’s surface. Thus warming due to increased greenhouse gases produces a climate signature different from that of warming due to solar radiation changes.

Thermal decomposition kinetics of 5-fluorouracil from thermogravimetric analysis

        Yi-ling Bei,Qing-yang Liu,Gui-bin Qi,Yuan-jun Ding 한국화학공학회 2008 Korean Journal of Chemical Engineering Vol.25 No.5

The thermal decomposition kinetics of 5-fluorouracil was studied by thermogravimetric analysis methodology. The decomposition activation energy was calculated by using Ozawa method by means of TGA in nitrogen atmosphere. Moreover, the decomposition mechanism and pre-factor were obtained by Coats-Redfern and Achar methods, respectively. It is found that the decomposition activation energy of fluorouracil is 105 kJ·mol−1. The decomposition mechanisms obtained by Coats-Redfern and Achar methods are G( α)=[−ln(1− α)]2/3 and f( α)=1.5(1− α)[−ln(1−α)]1/3 respectively, lnA is 21.40 min−1.

Modeling of low viscosity oil-water annular flow in horizontal and slightly inclined pipes: Experiments and CFD simulations

        Yi-Xin Pan,Hong-Bing Zhang,Rong-Hua Xie,Xing-Bin Liu,Min Wang 한국화학공학회 2016 Korean Journal of Chemical Engineering Vol.33 No.10

To characterize the effect of pipe inclination, low viscosity, flow rate and inlet water cut on annular flow pattern, a low viscosity oil-water two-phase annular flow in horizontal and slightly inclined (+1o, +3o and +5o) pipes with diameter of 20 mm has been experimentally investigated. A modified VOF model based on the CFD software package FLUENT was used to predict the in-situ oil fraction and pressure drop. The experimental data indicate that annular flow appears at a medium-high water cut. The slip ratio increases with flow rate increase but decreases with increasing water cut. The changes are more significant as the degree of inclination increases. Pressure drop is strongly dependent on flow rate, as it increases rapidly as inlet flow rate increase. Good agreement between the experimental data and calculated results of slip ratio and pressure drop was obtained.

( Liu Bin ),( Ning Ning Zhang ),( Chao Zhao ),( Bai Xue Lin ),( Lian Hui Xie ),( Yi Fan Huang ) 한국미생물 · 생명공학회 2012 Journal of microbiology and biotechnology Vol.22 No.10

A xylanase-producing thermophilic strain, Geobacillus sp. TC-W7, was isolated from a hot spring in Yongtai (Fuzhou, China). Subsequently, the xylanase gene that encoded 407 amino acids was cloned and expressed. The recombinant xylanase was purified by GST affinity chromatography and exhibited maximum activity at 75℃ and a pH of 8.2. The enzyme was active up to 95℃ and showed activity over a wide pH range of 5.2 to 10.2. Additionally, the recombinant xylanase showed high thermostability and pH stability. More than 85% of the enzyme`s activity was retained after incubation at 70℃ for 90 min at a pH of 8.2. The activity of the recombinant xylanase was enhanced by treatment with 10 mM enzyme inhibitors (DDT, Tween-20, 2-Me, or TritonX-100) and was inhibited by EDTA or PMSF. Its functionality was stable in the presence of Li+, Na+, and K+, but inhibited by Hg2+, Ni2+, Co2+, Cu2+, Zn2+, Pb2+, Fe3+, and Al3+. The functionality of the crude xylanase had similar properties to the recombinant xylanase except for when it was treated with Al2+ or Fe2+. The enzyme might be a promising candidate for various industrial applications such as the biofuel, food, and paper and pulp industries.
