      • Clinical Outcomes of Basaloid Squamous Cell Carcinoma of the Esophagus: A Retrospective Analysis of 142 Cases

        Background: Basaloid squamous cell carcinoma of the esophagus (BSCCE) is a rare and distinctive tumor with no standard treatment. This study aimed to explore treatment in relation to prognosis of the disease. Methods: A total of 142 patients with BSCCE that underwent treatment in our hospital from March 1999 to July 2010 were retrospectively analyzed. All patients received surgery, 42 postoperative radiotherapy and 28 patients chemotherapy. Results: There were 26 patients included in stage I, 60 in stage II, 53 in stage III and 3 in stage IV. The clinical symptoms and macroscopic performances of BSCCE did not differ from those of typical esophageal squamous cell carcinoma. Among 118 patients receiving endoscopic biopsy, only 12 were diagnosed with BSCCE. The median survival time (MST) of the entire group was 32 months, with 1-, 3- and 5-year overall survival (OS) of 81.4%, 46.8% and 31.0%, respectively. The 5-year OS of stage I and II patients was significantly longer than that of stages III/IV, at 60.3%, 36.1% and 10.9%, respectively (p<0.001, p=0.001). The MST and 5-year OS were 59.0 months and 47.4% in patients with tumors located in the lower thoracic esophagus, and 27.0 months and 18.1% in those with lesions in the upper/middle esophagus (p=0.002). However, the survival was not significantly improved in patients undegoing adjunctive therapy. Multivariate analysis showed TNM stage and tumor location to be independent prognostic factors. Furthermore, distant metastasis was the most frequent failure pattern, with a median recurrence time of 10 months. Conclusion: BSCCE is an aggressive disease with rapid progression and a propensity for distant metastasis. It is difficult to make a definitive diagnosis via preoperative biopsy. Multidisciplinary therapy including radical esophagectomy with extended lymphadenectomy should be recommended, while the effectiveness of radiochemotherapy requires further validation for BSCCE.

        Reduction of Erythrocyte Fluid Adaptability Due to Cell Membrane Hardening Based on Single‑Cell Analysis

        Because of the rapid development of precision medicine, single-cell analysis has attracted increasing research attention, especially for erythrocyte, whose potential role in the formation of vascular plaque (atherosclerosis) has emphasized the importance of flow characteristics of single erythrocytes in bionic microfluidics. Based on the high incidence of vascular plaques among the elderly and those who have received blood transfusions, we hypothesized that cell membrane hardening changes the fluid adaptability of individual erythrocytes. This hypothesis was verified using an in vitro microfluidic technique based on an analysis of the flow morphology and cell trajectory of individual cells. A symmetrical microchannel was fabricated with a central stenosis to simulate a blood vessel containing plaque. During flowing through this microchannel, normal erythrocyte predominantly exhibited deforming, rotating, and lifting morphologies, resulting in discontinuous contact with the channel wall and a narrower distribution. Conversely, hardened erythrocytes exhibited rolling, swinging, and tumbling morphologies, resulting in stable and continuous contact with the channel wall and a wider distribution. These results indicate that cell membrane hardening decrease cell fluid adaptability on a microscopic scale. This research can offer some new insights into vascular plaques research from a bio-tribological and mechanical perspectives.

        Cyclic Softening Properties of 30Cr2MoV Steel at Elevated Temperatures

        Low cycle fatigue tests for 30Cr2MoV steel are done at both room and elevated temperatures and their softening properties are investigated. Based on the experimental results, a quantitative relationship between hardness and remaining life is established, allowing prediction of the remaining life of metals. A modified Mason-Coffin equation including the aging effect is also derived. Both the experimental and theoretical results show that it is useful to be able to predict the remaining life of a steam turbine rotor through the measurement of hardness.

        Selection and evaluation of RT-qPCR reference genes for expression analysis in the tiny egg parasitoid wasp, Trichogramma dendrolimi matsumura (Hymenoptera: Trichogrammatidae)

        The egg parasitoid, Trichogramma spp., is an important biological control agent used against a broad range of Lepidopteran pests in agriculture and forestry. The biology of Trichogramma has been studied in details. Further studies should focus on the molecular mechanisms of Trichogramma by qualifying the expression of related genes It is critical to select appropriate reference genes for normalizing RT-qPCR results and establishing a robust method for quantifying target gene expression. This study aimed to identify and validate appropriate reference genes for use in RT-qPCR analysis of Trichogramma dendrolimi. Ten candidate housekeeping genes, namely betaactin (ACTIN), forkhead box O (FOXO), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), heat shock pro tein 90 (HSP90), ribosomal protein L10a (RPL10a), L18 (RPL18), L28 (RPL28), S13 (RPS13), S15 (RPS15), and superoxide dismutase (SOD), were tested for their suitability as reference genes for developmental stage (3rd, 4th, 5th, 6th, 7th, 8th, 9th, and 10th day after parasitization), tissue (head, thorax, and abdomen of adults), sex of adults (male and female), and temperature (17℃, 25℃, and 32℃). According to the GeNorm analysis, a robust analysis should involve using an appropriate combination of reference genes, namely, at least three genes for different development stages, two genes for different tissues, two genes for different sex, and two genes for different temperatures, respectively. According to the RelFinder method by the integrated results of GeNorm, NormFinder, BestKeeper, and the ΔCt method, we identified the developmental stage-specific reference genes SOD, GAPDH, and ACTIN; tissue-specific reference genes RPL18 and RPS15; sex-specific reference genes RPL18 and SOD; and temperature-specific reference genes RPL18 and RPL10a. This study provides a standardized procedure for the quantification of gene expression in T. dendrolimi and will be helpful for future biological control programs using Trichogramma wasps.

        ER membrane protein complex subunit 6 (EMC6) is a novel tumor suppressor in gastric cancer

        The endoplasmic reticulum (ER) membrane protein complex subunit 6 (EMC6) is a novel human autophagy-related molecule. Here, using tissue microarray and immunohistochemistry, we report that EMC6 protein is lost or reduced in glandular cells of patients with gastric adenocarcinoma, compared to normal stomach mucosa. Overexpression of EMC6 in gastric cancer cells inhibited cell growth, migration, invasion, and induced apoptosis and cell cycle arrest at S-phase. Further investigation suggested that EMC6 overexpression in BGC823 human adenocarcinoma gastric cancer cells reduced tumorigenicity in a xenograft model, demonstrating that EMC6 has the characteristics of a tumor suppressor. This is the first study to show that EMC6 induces cell death in gastric cancer cells. The molecular mechanism of how EMC6 functions as a tumor suppressor needs to be further explored.         haracterization of a Recombinant Thermostable Xylanase from Hot Spring Thermophilic Geobacillus sp. TC-W7

        A xylanase-producing thermophilic strain, Geobacillus sp. TC-W7, was isolated from a hot spring in Yongtai (Fuzhou, China). Subsequently, the xylanase gene that encoded 407 amino acids was cloned and expressed. The recombinant xylanase was purified by GST affinity chromatography and exhibited maximum activity at 75℃ and a pH of 8.2. The enzyme was active up to 95℃ and showed activity over a wide pH range of 5.2 to 10.2. Additionally, the recombinant xylanase showed high thermostability and pH stability. More than 85% of the enzyme`s activity was retained after incubation at 70℃ for 90 min at a pH of 8.2. The activity of the recombinant xylanase was enhanced by treatment with 10 mM enzyme inhibitors (DDT, Tween-20, 2-Me, or TritonX-100) and was inhibited by EDTA or PMSF. Its functionality was stable in the presence of Li+, Na+, and K+, but inhibited by Hg2+, Ni2+, Co2+, Cu2+, Zn2+, Pb2+, Fe3+, and Al3+. The functionality of the crude xylanase had similar properties to the recombinant xylanase except for when it was treated with Al2+ or Fe2+. The enzyme might be a promising candidate for various industrial applications such as the biofuel, food, and paper and pulp industries.

        Mechanism of the simultaneous removal of nitrate and Ni(II) by Enterobacter sp. CC76 through mixotrophic denitrification processes

        We studied the mechanism for the simultaneous removal of nitrate and Ni(II) by Enterobacter sp. CC76. Response surface methodology results showed that the maximum removal ratios of nitrate and Ni(II) were 95.02% and 75.99% under the following conditions: pH 7.37, 54.31mg·L1 Fe(II), and 10.00mg·L1 Ni(II). The mechanism of Ni(II) removal involved Fe-oxide adsorption and the increase of pH. In addition, meteorological chromatography analysis indicated that Ni(II) affected gas composition during denitrification. Scanning electron microscopy and X-ray photoelectron spectroscopy confirmed that Fe-oxide adsorption was the main contributor to Ni(II) removal. This study shows that Enterobacter sp. CC76 can enhance the adsorption of Ni(II) onto Fe-oxides while removing nitrate.

        Xanthones from Garcinia paucinervis with in vitro anti-proliferative activity against HL-60 cells

        Three new xanthones, paucinervins H–J (1–3), as well as eleven known compounds (4–14), were isolated from the leaves of Garcinia paucinervis. The structures of the new compounds (1–3) were elucidated by 1D, 2D NMR spectra and HR ESIMS. In vitro antiproliferative activity against human promyelocytic leukemia HL-60 cells was tested, among which, compounds 2, 5, 6 and 7 exhibited strong growth inhibitory effects with GI50 values ranging from 1.30 to 9.08 lM, respectively. Preliminary SARs were also discussed.
