        In Vitro Antiviral Activity of Ribavirin Against Severe Fever with Thrombocytopenia Syndrome Virus Myung Jin LEE1, Kye-Hyung KIM1, Jongyoun YI2, SuJin CHOI1, Chung-Jong KIM1, Nak- Hyun KIM1, Kyoung-Ho SONG1, Pyoeng Gyun CHOI1, Ji-Hwan BANG1, Wan Beom PARK1, Eu Suk KIM1, Sang-Won PARK1, Hong Bin KIM1, Nam Joong KIM1, Myoung- Don OH1 Seoul National University College of Medicine, Korea1, Pusan National University School of Medicine, Korea2 Background: Severe fever with thrombocytopenia syndrome (SFTS) is an emerging infectious disease caused by a novel Bunyavirus, severe fever with thrombocytopenia syndrome virus (SFTSV). No effective antiviral therapy is proven yet, but clinical use of ribavirin (RBV) has been tried. We investigated the antiviral effect of RBV against SFTSV in vitro. Methods: To test for cytotoxicity of RBV, Vero cells were treated with different concentrations of RBV (3.90 to 500 μg/mL, two-fold dilution) and analyzed by cell viability MTS assay 48h post-infection. To determine antiviral activity of RBV against SFTSV, Vero cells were infected with SFTSV strain Gangwon/Korea/2012 at 100 TCID50 (50% tissue culture infective dose) per well in a 96-well plate, and RBV was added at the concentrations showing no or minimal cytotoxicity. Viral RNAs were extracted from the culture supernatants and quantifi ed using one-step real-time reverse transcription- PCR to amplify the partial large segment of SFTSV. Statistical analysis was done by one-way ANOVA with Tukey`s post hoc test. Results: Cytotoxicity due to RBV was not observed at RBV concentration =31.3 μg/ mL. Viral RNAs at 24h post-RBV treatment were reduced with increasing RBV concentrations (1-32 μg/mL), compared with those of mock-treated cells (P <0.01, Figure). Half maximal inhibitory concentration (IC50) of RBV was 3.69 μg/mL at 24h post-RBV treatment. Conclusions: Our study shows that RBV has antiviral effect against SFTSV in a dose-dependent manner. Further studies are required to evaluate the effi cacy of RBV in SFTS.

      • Immunoblot법을 이용한 Anisakis증의 혈청학적 진단

        Acute gastric anisakiasis is rather easily diagnosed by endoscopy or radiological technique. But preoperational diagnosis of acute intestinal anisakiasis is very difficult. Furthermore, the diagnosis of chronic anisakiasis is more difficult because the worm invade the stomach or intestinal wall and finally to degenerate. Serodignosis may helpful in such diagnostic difficulties. However, any of the serodiagnostic method in anisakiasis has limitations for practical use due to cross reactions. In this context, the study was performed to observe the specifically reacting antigenic bands of crude extract Anisakis larval worm antigen and their reaction to the serum antibody obtained from experimentally immunized and infected rabbit, confirmed human anisakiasis and other helminthic infection cases using SDS-PAGE and EITB. The results obtained were as follows; 1. Among 12 cases of confirmed anisakiasis, eleven showed positive reactions by ELISA. The average 0. D. value of the test was 1.035. Two from 4 D. latum infected cases and one from 4 A. lumbricoides infected cases showed positive reaction respectively. 2. Silver stained SDS-PAGE of crude Anisakis larval worm antigen revealed more than 45 antigenic protein bands. Among them, 186, 76, 72, 65, 59~62, 54, 52, 50, 49, 46, 43, 40, 33, 32, 29, 24, 23, 18, 17, and 13Kd bands were major bands. Protein bands of 12Kd relative molecular weight was observed in A. suum extract antigen, on the other hand 29 and 46Kd bands were not observed. 3. Major antigenic bands of crude Anisakis worm antigen which reacted with serum antibody of confirmed anisakiasis cases were 186, 116, 72, 52, 46, 40, 33 and 32Kd band. Most of them also reacted with the sera of D. latum, A. lumbricoides, T. trichiura infected cases except 46Kd band which showed positive resuits only with sera of anisakiasis. So, this band could be considered as diagnostic band for anisakiasis especially for the identification of cross reactions against A. lumbricoides and D. lafum infection. 4. Although, low molecular weight bands such as 33 and 32Kd were observed as species specific, the sensitivity of that fractions were obviously low. Consequently, the diagnostic use of the low molecular weight protein bands were not promising. 5. Serum of rabbit immunized with Anisakis larval worm antigen also showed positive reaction on 46Kd bands, but that of A. suum and T. canis did not. These results were in accord with the results in human cases. In conclusion, 46Kd bands of crude Anisakis larval worm antigen can be considered as a diagnostic one for the identification of cross reactions using immunoblot technique.

        정보통신 표준화 지수측정 모형 개발 연구

        The standard issue in the information and telecommunication industry is increasingly important with the rapid development of technology. This paper proposes an index model which can measure the degree of standardrzation in the Korean information and telecommunication field We first classified ICT sector into 14 sub-sectors. Then for each sub-sector, we considered a Set of important determinants to measure the level of standardization, and constructed a linear equation using this set of determinants Finally we estimated the relative degree of importance of each determinant using the AHP methodology The proposed model found that overall level of standardization In the Korean ICT industry is relatively low, and "IMT-2000 technology" and "computer network technology" among 14 sub-sectors are highly standardized sub-sectors The validity of the proposed model was also partially proved using two different methods, holistic and historical approach.

      • 정전 열 접합을 이용한 Multi-Substrate Bonding

        We performed silicon-to-glass bonding using silicon direct bonding followed by anodic bonding(SDAB). Initial bonding between glass and silicon was caused by the hydrophilic surfaces of silicon and glass ensemble using silicon direct bonding(SDB) method. We found that the bonded specimen using SDAB process had higher strength than one using anodic bonding process only. We performed multiple layer bonding by SDAB, which is glass- silicon -glass as sandwich structure. In the silicon wafer, the (1mm x 2mm x 500μm)-sized cavity was formed by the anisotropic etching of the silicon substrate in EPW(Ethylendiamin-Pyrocatechol-Water). And, the cavity was sealed with glass wafers by SDAB method.

        Metamifop, (R)-2-[4-[(6-chloro-2-benzoxazolyloxyphenoxy]-N-(2-fluorophenyl)-N-methyl propanamide, was newly developed by Dongbu Hannong Chemicals in Korea. In the present study we conducted acute oral toxicity and acute dermal toxicity test. The values of LD_(50) of metamifop was found to be over 5,000 mg/kg for ICR mice. On the other hand, metamifop showed no acute dermal toxicity to SD rats.

        In this paper, we proposes two phase chopper system with flour. separate groups of D.C motor for regenerative braking. The main circuit consists of four sets of chopping parts, eight diodes and four separate groups of D.C motor. Although the proposed chopper circuit requires more circuit elements than the conventional two phase chopper system with, combined output, it has the following advantages: (1) The region of regenerative braking becomes twice as that of the conventional two phase chopper system. (2)Current division of load becomes equalized. Therefore. it is possible to drive not only series motor but also shunt and separately excited motor (3) The harmonic component of the source current is greatly diminished.

      • 생분해성 다공질 Calcium Phosphate 세라믹스의 제조

        When porous calcium phosphate ceramics are used as a bone graft substitute, it is known that new tissue or blood vessel is grown into the porous implant due to their excellent biocompatibility. In this study, the most chemically stable β-crystalline form of calcium metaphosphate, Ca(PO₃)₂is prepared by the controlled thermolysis of monocalcium phosphate, Ca(H₂PO₄)₂. The diameter of cylindrical pores formed during cooling was controlled by a holding time at the melting point of a monocalcium phosphate and by the change of a recrystallization temperature, to obtain the most appropriate size(about 200㎛) of pores. It was observed that the increasing holding time at the melting point of monocalcium phosphate results in the decreases of pore sizes.

        To cope with corporation environments such as global economy era, increased product diversity and rapid market environmental change, the production system should be operated with capabilities to supply products in time and the lowest price that meet customer's demands. Therefore, the production system has been changed from mass production system of conveyor type to U-line system based on JIT according to market environmental change. Scheduling on the production activities is to decide proper time and methods for each activity by supplying materials within limited time. If the scheduling, however, is not favorable, it will not only fail to meet appropriate supplying time but also cause inefficiency caused by the low utilization of diverse elements for production activities. Therefore, the study is to understand characteristics of U-line and look over the theoretical background of scheduling planning. This application was developed by using Visual basic 6.0 and Active X control.
