Needs Analysis of English Learning for Adolescents from Multicultural Families

The purpose of this study was to investigate the needs of English learning for adolescents from multicultural families in a formal classroom environment in a public alternative high school. As a case study, there were four participants: two multicultural students, their school English teacher, and a staff member from an organization for providing an initial adaptation program for multicultural students. The first and second interviews with the school English teacher and the two multicultural students were conducted sequentially to compare the thoughts in terms of the purpose of English learning, benefits, and difficulties of multicultural students in English learning in a school setting. Between the first and second interviews, learning materials used in the school context were analyzed with an evaluation checklist. The results of the learning materials analysis were utilized for the development of the second interview questions with the two multicultural students and their English teacher. The interview with the staff member from an organization for helping multicultural students was performed after the learning materials analysis and before the second interviews with the English teacher and the two multicultural students. With regard to the purpose of English learning for students from multicultural families, whereas the two participating students commented their future careers, their English teacher and the staff member from the organization expected that learning English could be a means for multicultural student in order to develop confidence and to adjust to Korean society. Cultural and linguistic benefits and difficulties regarding advantages and challenges of multicultural students in learning English were revealed in a classroom setting. Regarding cultural difficulties and advantages of multicultural students in English learning, their past experiences of Korean culture helped multicultural students become familiar with Korean culture and could be a cultural advantage in a formal learning environment. However, culture-related topics that students from multicultural families had no experience in their home countries and comments with cultural bias on learning materials may give a negative influence on multicultural students’ English learning. In terms of linguistic benefits, their English teacher in this formal context lets multicultural students use their first languages due to the teacher’s awareness of characteristics of bi-/multilinguals. Nonetheless, a lack of vocabulary knowledge in Korean and time constraints on English practices were related to linguistic difficulties of multicultural students. The irrelevant learning materials and insufficient support programs were challenges in English learning of multicultural students. Lastly, the fact that students from multicultural families learned English in their L2 or L3 in a formal classroom environment was also one of the linguistic disadvantages. This study has pedagogical implications for policy makers, schools, and English teachers in terms of practical support for multicultural students in English learning.

A simple and reliable analytical method using high-performance liquid chromatography–ultraviolet detection was established for the analysis of Matricaria chamomilla (MC) flower extract. Apigenin-7-O-glucoside (AP7G) was chosen as a target analyte considering its useful medicinal properties and high content in the MC extract. Chromatographic separation and identification of AP7G was achieved by using a Capcell Pak MGII C18 (250 × 4.6 mm, 5 um) with a mobile phase of 20% acetonitrile in an aqueous 0.05% (v/v) trifluoroacetic acid solution at a flow rate of 1.0 mL/min. The detection wavelength and column temperature were 330 nm and 30°C, respectively. The method was validated according to the guidelines of the International Conference on Harmonization (ICH) and the Association of Official Analytical Chemists (AOAC) and was found to meet the requirements thereof, that is: linearity (R2 = 0.99936 for 6.25–18.75 mg/mL), precision (≤ 2% relative standard deviation), and accuracy (98.29%–100.18%). The limits of detection and quantification were 0.57 and 1.73 mg of the extract/mL, respectively. Thus, it is concluded that the developed method is applicable for the quantitative analysis of AP7G in the extract of MC.

<P>RNA-guided engineered nucleases (RGENs) derived from the prokaryotic adaptive immune system known as CRISPR (clustered, regularly interspaced, short palindromic repeat)/Cas (CRISPR-associated) enable genome editing in human cell lines, animals, and plants, but are limited by off-target effects and unwanted integration of DNA segments derived from plasmids encoding Cas9 and guide RNA at both on-target and off-target sites in the genome. Here, we deliver purified recombinant Cas9 protein and guide RNA into cultured human cells including hard-to-transfect fibroblasts and pluripotent stem cells. RGEN ribonucleoproteins (RNPs) induce site-specific mutations at frequencies of up to 79%, while reducing off-target mutations associated with plasmid transfection at off-target sites that differ by one or two nucleotides from on-target sites. RGEN RNPs cleave chromosomal DNA almost immediately after delivery and are degraded rapidly in cells, reducing off-target effects. Furthermore, RNP delivery is less stressful to human embryonic stem cells, producing at least twofold more colonies than does plasmid transfection.</P>
