        만성 간염에서 Interleukin-6의 간내발현

        N/A Rackground/Aims: Interleukin-6 (lL-6), also known as B cell stimulatory factor 2(BSF-2), induces the final maturation of B cells to antitxxiy-producing cells. IL-6 has many biologic properties including the immune and intlammatory responses. This study wos aimed to evaluate the role of local interleukin 6(IL-6) in the pathogenesis of chronic hepatitis. Methods; We examined the cellular site and grade of IL-6 staining in paraffin sections of the liver from 24 patients with liver disease, using immunohistochemistry with a polyclonal antitwdy. The patient. Were divided into two groups; Group A(n=l3) with high histologic uctivi1y consisted of CAH-type B(n=10) ond active cirrhosis(n=3), whilc Group B(n= l l) with low hi.itologic activity consisted of CPH-type B(n=4), inactive cirrhosis(n=2) and fatty liver(n=S). Results: There was no staining of IL-6 in normal liver tissue. Thv grade.I of IL-6 staining in Group A were three positive in seven cases (53.81o), two positive in five ca.ics(38.3%) and one positive in only one case(7.7%), while those in Group B were one positivc in three cases(27.3%) ancl trace in eight case.(72.7ln). IL-6 stained cells in chronic hepatitis were hepatocytcs, cspecially in the areu ot' piecemeol necrosi.I, bilc duct cel1., infiltrating inflammatory cells and endothelial cell.I. The score of histological activity index(HAJ), piecemeal necrosis and fibrasis and thc gradv. Of 1L-6 staining of Group A were ull significantly higher than those of Group B. The grade of IL-6 staining and HAI werc well correlated(r =0.74, p 0.0l), Conclusion: Locally produced IL-6 in the liver may contribute to the inflammatory process and immunological response in chronic hepatiti..       • Anti-inflammatory effect of Apo-9′-fucoxanthinone via inhibition of MAPKs and NF-kB signaling pathway in LPS-stimulated RAW 264.7 macrophages and zebrafish model

        <P><B>Abstract</B></P> <P>In this study, we confirmed the anti-inflammatory effect of Apo-9-fucoxanthinone (AF) in <I>in vitro</I> RAW 264.7 cells and <I>in vivo</I> zebrafish model. In lipopolysaccharide (LPS)-stimulated zebrafish, AF significantly decreased the production of reactive oxygen species (ROS), nitric oxide (NO) and cell death. In addition, the mRNA expression of inducible nitric oxide synthase (iNOS), suppressed cyclooxygenase-2 (COX-2) and an inflammatory cytokines; IL-1β, TNF-α were shown reduction. And AF significantly inhibited NO production and expression of iNOS in LPS-stimulated RAW 264.7 cells. Further, AF suppressed COX-2, prostaglandin E2 (PGE<SUB>2</SUB>), and pro-inflammatory cytokines such as interleukin-6 (IL-6), IL-1β, and tumor necrosis factor-α (TNF-α) at 25, 50 and 100 μg/mL, respectively. Further mechanistic studies showed that AF suppressed the nuclear factor-kB (NF-kB) pathway and phosphorylation of mitogen-activated protein kinase (MAPK) pathway molecules such as extracellular signal-regulated kinase (ERK) and c-Jun N-terminal kinase (JNK).         Effects of <i>Ecklonia cava</i> ethanolic extracts on airway hyperresponsiveness and inflammation in a murine asthma model: Role of suppressor of cytokine signaling

        <P><B>Abstract</B></P><P><I>Ecklonia cava</I> (EC) is a brown alga that evidences radical scavenging activity, bactericidal activity, tyrosinase inhibitory activity, and protease inhibitory activity. However, its anti-allergic effects remain poorly understood. In the current study, we attempted to determine whether pretreatment with EC induces a significant inhibition of asthmatic reactions in a mouse asthma model. Mice sensitized and challenged with ovalbumin (OVA) evidenced typical asthmatic reactions, as follows: an increase in the number of eosinophils in bronchoalveolar lavage fluid; a marked influx of inflammatory cells into the lung around blood vessels and airways, and airway luminal narrowing; the development of airway hyperresponsiveness; the detection of tumor necrosis factor-alpha (TNF-α) and Th2 cytokines, including IL-4 and IL-5 in the bronchoalveolar lavage (BAL) fluid; and the detection of allergen-specific immunoglobulin E (IgE) in the serum. However, the administration of EC extract prior to the final airway OVA challenge resulted in a significant inhibition of all asthmatic reactions. We also demonstrated that EC extracts treatment resulted in significant reductions on matrix metalloproteinase-9 (MMP-9) and Suppressor of cytokine signaling-3 (SOCS-3) expression and a reduction in the increased eosinophil peroxidase (EPO) activity. The treatment of animals with EC extracts resulted in a significant reduction in the concentrations of the Th2 cytokine (IL-4 and IL-5) in the airways, without any concomitant increase in the concentration of Th1 cytokines. These findings indicate that EC extracts may prove useful as an adjuvant therapy for allergic airway reactions via the inhibition of the Th2 response. Accordingly, this study may provide evidence that EC extract performs a critical function in the amelioration of the pathogenetic process of asthma in mice.</P>

        Hearing impairement in varying degrees constitutes one of the most common disabilites affecting school aged children. Eight out of every 1,000 students under the age of twenty have significant hearing loss. Hearing disabilities range from slight to total loss of sound perception. Statistics indicate that only one out of every ten hearing impaired children will become deaf. Most motor activities can be included students may possess low levels of static and/ or dynamic balance, but the greatest majority possess normal motor and physical capabilities. All the hearing impaired youngsters should be properly assessed and given the identical testing batteries as normal children. Hearing impaired students have, essentialy, a communication handicap and suffer from inability to understand verbal instructions. All physical educators should be able to use minimal finger spelling and signing for hearing impaired students.

        According to the investigation made by analyzing cause of the detail about sports injury. We make this things an object of all the male representative players 254 out of 286 (88.81%) that took part in the whole event from April 15, 1988 to September 16, 1988 in Seoul Olympic Games. The results are as follows. 1. The representative players in Korea started in the athlete life at age 13.1 on the average. Gymnast early started at age 8.4 and yachtist was later at age 18 in the athlete life. 2. There are about 5.7 cases on the outbreak of the sports trauma per person of player a year. This came into existence near the ankle at the high rate of damage formed 11.90%. 3. A sort of sports injury in analysis muscular bruise broke out at the highest rate of occurrence formed 15.70%. 4. The greater part of representative players themselves the injury of trauma by massage (12.53%). Some of them undergo medical treatment to the specialist hospital once a year on the average. Most of hockey players have had experience in this kind of injury treatment three times one year to per person of players. A few of them showed 0.15 times per person a year. 5. Most of representative players have been periodically to the orthopedist formed 27.59 (732) out of all 2653 cases at the high rate of fact. Especially a sort of injury in analysis, the injury of ankle region showed the percentage 11.86 (704) out of all 5,937 cases at the highest rate. 6. The period of medical treatment of the representative players in Korea came out about 14.7 days. Besides period of the other player's treatment came out mostly from one to five days formed 44.82%(935 cases) 7. Manyof them have been roughly treated with muscular bruise in analysis of sports injury. They were taking pains with muscular bruise formed 15.65% (1003) out of all 6408 case. 8. In major cause of sports injury came into existence during the training with waste of physical stamina formed 35.89% and were beyond their power with practicing the game formed 48.09%. Also, a sort of those were brought about the cause of sports injury with desire of outcome too much during in the game formed 44.20%

        Anti-inflammatory effect of polyphenol-rich extract from the red alga Callophyllis japonica in lipopolysaccharide-induced RAW 264.7 macrophages

        Despite the extensive literature on marine algae over the past few decades, a paucity of published research and studies exists on red algae. The purpose of this study was to evaluate the potential therapeutic properties of the ethanol extract of the red alga Callophyllis japonica against lipopolysaccharide (LPS)-stimulated macrophage inflammation. The C. japonica extract (CJE) significantly inhibited the nitric oxide (NO) production and the induced dose-dependent reduction of the protein and mRNA levels of inducible nitric oxide synthase and cyclooxygenase-2. Additionally, the CJE reduced the mRNA levels of inflammatory cytokines, including tumor necrosis factor-${\alpha}$, interleukin (IL)-$1{\beta}$, and IL-6. We investigated the mechanism by which the CJE inhibits NO by examining the level of mitogen-activated protein kinases (MAPKs) activation, which is an inflammation-induced signaling pathway in macrophages. The CJE significantly suppressed the LPS-induced phosphorylation of c-Jun N-terminal kinase, extracellular signal-regulated kinase and p38 MAPK. Taken together, the results of this study demonstrate that the CJE inhibits LPS-induced inflammation by blocking the MAPK pathway in macrophages.

        Anti-inflammatory potential of saponins derived from cultured wild ginseng roots in lipopolysaccharide-stimulated RAW 264.7 macrophages

        <P>Ginseng, namely the root of Panax ginseng Meyer, is a well-known traditional medicine that has been used in Asian countries for thousands of years. Ginseng saponins have been shown to exert a variety of prominent pharmacological effects in a number of diseases. The aim of the present study was to identify the anti-inflammatory effects of total saponins extracted from cultured wild ginseng roots (TSWG) on lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. An elevated production of nitric oxide (NO) was detected in the RAW 264.7 cells in response to stimulation with LPS, as shown by NO detection assay using Griess reagent. However, pretreatment with TSWG inhibited the production of NO through the suppression of inducible NO synthase gene expression. Furthermore, the LPS-induced gene expression and production of tumor necrosis factor-alpha (TNF-alpha) and interleukin-1 beta (IL-1 beta) were significantly reduced by treatment with TSWG, as shown by ELISA, and western blot analysis and RT-PCR, respectively. In the LPS-stimulated RAW 264.7 cells, nuclear factor-kappa B (NF-kappa B) was translocated from the cytosol to the nucleus, while pre-treatment with TSWG induced the sequestration of NF-kappa B in the cytosol through the inhibition of the inhibitor of kappa B degradation, as shown by immunofluorescence staining. TSWG also contributed to the down-regulation of mitogen-activated protein kinases and Akt in the LPS-stimulated RAW 264.7 cells. Additionally, in the TSWG-treated RAW 264.7 cells, we observed the activation of nuclear factor (erythroid-derived 2)-like 2 and an increase in heme oxygenase-1 expression; these effects were associated with the inhibition of the generation of reactive oxygen species. The results from the present study indicate that TSWG exerts anti-inflammatory and antioxidant effects, suggesting that TSWG may be an effective therapeutic agent for inflammatory diseases and prevent cellular damage induced by oxidative stress.</P>

      • Platelet-Rich Plasma Increases the Levels of Catabolic Molecules and Cellular Dedifferentiation in the Meniscus of a Rabbit Model

        <P>Despite the susceptibility to frequent intrinsic and extrinsic injuries, especially in the inner zone, the meniscus does not heal spontaneously owing to its poor vascularity. In this study, the effect of platelet-rich plasma (PRP), containing various growth factors, on meniscal mechanisms was examined under normal and post-traumatic inflammatory conditions. Isolated primary meniscal cells of New Zealand white (NZW) rabbits were incubated for 3, 10, 14 and 21 days with PRP(−), 10% PRP (PRP(+)), IL(+) or IL(+)PRP(+). The meniscal cells were collected and examined using reverse-transcription polymerase chain reaction (RT-PCR). Culture media were examined by immunoblot analyses for matrix metalloproteinases (MMP) catabolic molecules. PRP containing growth factors improved the cellular viability of meniscal cells in a concentration-dependent manner at Days 1, 4 and 7. However, based on RT-PCR, meniscal cells demonstrated dedifferentiation, along with an increase in type I collagen in the PRP(+) and in IL(+)PRP(+). In PRP(+), the aggrecan expression levels were lower than in the PRP(−) until Day 21. The protein levels of MMP-1 and MMP-3 were higher in each PRP group, <I>i.e.</I>, PRP(+) and IL(+)PRP(+), at each culture time. A reproducible 2-mm circular defect on the meniscus of NZW rabbit was used to implant fibrin glue (control) or PRP <I>in vivo</I>. After eight weeks, the lesions in the control and PRP groups were occupied with fibrous tissue, but not with meniscal cells. This study shows that PRP treatment of the meniscus results in an increase of catabolic molecules, especially those related to IL-1α-induced inflammation, and that PRP treatment for an <I>in vivo</I> meniscus injury accelerates fibrosis, instead of meniscal cartilage.</P>

      • Sargassum horneri inhibits psoriasis-related inflammation in HaCaT keratinocytes and ameliorates imiquimod-induced psoriasis-like skin lesions in mice

        Sargassum horneri exhibits broad bioactive properties. However the anti-psoriatic effects of its have not been reported. The present study evaluated whether ethanol extract of S. horneri (ESH) can improve the symptoms of psoriasis in vitro and in vivo and elucidated the mechanisms underlying those effects in vitro. ESH treatment significantly attenuated the protein levels of IL-6, IL-8, and TNF-α in IFN-γ/TNF-α stimulated HaCaT keratinocytes. In addition, ESH inhibited (PI3K)/Akt phosphorylation and NF-κB activation in HaCaT keratinocytes. We further demonstrated that the ESH significantly ameliorates the severity of skin lesions (erythema, scaling, and thickness) in the IMQ-induced psoriasis-like mouse model. ESH attenuated the production of inflammatory cytokines by inhibiting Akt/NF-κB signaling pathways in IFN-γ/TNF-α-induced HaCaT keratinocytes. Furthermore, ESH could ameliorate the severity of skin lesions in an IMQ-induced psoriasis-like mouse model. Thus, S. horneri has considerable potential in the treatment or prevention of inflammatory disorders, such as psoriasis.
