Development of PCR-based molecular markers by transcriptome sequencing in Platycodon grandiflorume

Platycodon grandiflorum, which is the only species in the genus Platycodon of the family Campanulaceae, is an herbaceous flowering perennial. P. grandiflorum is generally known as bellflower or balloon flower indicating its ornamental uses. It has also been traditionally used as a medicinal crop in East Asia, which is widely employed as an antiphlogistic, antitussive, and expectorant. However, marker-assisted selection and molecular breeding in P. grandiflorum has lagged behind other plants such as pepper and tomato because of the lack of genetic information and effective molecular markers. Transcriptome sequencing provides an effective way to obtain large amount of sequence data when there is no available genome sequence. In this study, we performed a transcriptome analysis in platycodons, which has not been attempted previously. We analyzed simple sequence repeats (SSRs) using RNA-seq data. Di-nucleotide motifs were the most abundant repeats (39% ~ 40%) followed by mono- (26% ~ 32%), tri- (25% ~ 31%), tetra- (1.5% ~ 1.9 %), penta- (0.3% ~ 1%) in three platycodon accessions. Based on the SNP information obtained from RNA-seq analysis, we developed 12 PCR-based markers in Platycodon. The number of alleles ranged from two to seven with the average PIC value of 0.373. These 12 markers were applied to 21 platycodon accessions and a phylogenetic tree was constructed. The markers developed in this study could be introduced in molecular breeding program of platycodons. The SSR information obtained from RNA-seq analysis could be further utilized for developing genic-SSR markers in platycodons. Since platycodon is considered as an orphan crop, which has not been actively deployed for genetic study, the sequence information obtained from this study will contribute to further genetic improvements, genomic information and gene discovery in platycodon
Development of gene-based markers for pink fruit peel color in tomatoes

Tomato fruit color, which is the most visible characteristic among the other fruit traits, is considered to have a substantial influence on consumers. The pink-colored tomatoes with high soluble solids content are considerably preferred especially in Asia compared to the other colors. Generally the pink fruit trait of tomatoes is easily determined by visual examination of intact fruit, however, it is technically determined by the characteristic of the fruit peel. The pink trait is regulated by variations of the SlMYB12(y) gene located on chromosome 1, which controls the accumulation of the naringenin chalcone, which comprises a large proportion of flavonoids. In this study, we developed a derived Cleaved Amplified Polymorphic Sequences (dCAPS) marker and a sequence characterized amplified regions (SCAR) marker in order to discriminate of pink/non-pinktomatoes in the domestic breeding lines. Quantitative RT-PCR analysis indicated that the SlMYB gene is highly expressed in non-pink fruit peel, whereas the expression is significantly lowered in the pink fruit peel. These gene based markers are expected to enhance the efficiency and accuracy of selection pink-tomatoes in tomato breeding programs.
Reduced fronto‐callosal fiber integrity in unmedicated OCD patients: A diffusion tractography study

<P><B>Abstract</B></P><P>It is widely accepted that abnormalities in the frontal area of the brain underpin the pathophysiology of obsessive‐compulsive disorder (OCD). Fundamental to this investigation is the delineation of frontal white matter tracts including dorsal and ventral frontal projections of interhemispheric connections. While previous investigations of OCD have examined the dorsal and ventral frontal regions, the corresponding callosal connections have not been investigated, despite their importance. We recruited twenty patients with OCD (15 drug‐naïve and 5 currently unmedicated) and demographically similar healthy controls, and conducted fiber tractography and <I>post hoc</I> quantitative analysis using diffusion tensor imaging. We extracted fractional anisotropy (FA) of the fronto‐callosal fibers along the entire length of the tract. Function‐specific [by the Brodmann area region‐of‐interest (ROI) approach] and region‐specific (by the length‐parameterization approach) tracts were defined. In addition, we devised a new index of dorsal‐ventral imbalance (DVII) of fiber integrity. Significant FA decreases were observed in orbitofrontal and dorsolateral prefrontal projections of the corpus callosum (<I>P</I> < 0.05, false discovery rate‐corrected) with higher function/region sensitivity than voxel‐based or ROI‐based approaches. Importantly, OCD patients also exhibited significantly higher ventral‐greater‐than‐dorsal asymmetry of FA values than normal controls (<I>P</I> < 0.05, FDR‐corrected). This study is the first to investigate fiber integrity in the dorsal/ventral frontal parts of the callosal tractography in unmedicated OCD patients. Using a more quantitative method in terms of functional and regional specificity than previous studies, we report abnormalities in interhemispheric connectivity of both dorsal and ventral networks in the pathophysiology of OCD. 산업혁명 이후 일자리를 찾기 위해 수많은 사람이 도시로 모여들어, 현재 세계 인구 50% 이상이 도시에 살고 있다. 이러한 도시 집중화는 앞으로도 급속히 전개되어 2035년에는 75% 인구가 도시에 살 것으로 전망된다. 대도시는 점점 높아지는 인구 밀도로 인하여 환경 오염, 심각한 교통 체증, 지나치게 빠른 에너지 고갈 및 자연 생태계 파괴 등 부작용이 발생하면서 지속가능성이 떨어지고 있다. 또한, 높은 범죄율과 안전사고, 불평등과 양극화로 인한 일과 삶의 불균형, 지나치게 경쟁적인 교육 등으로 대도시 시민들의 행복 지수 역시 떨어지고 있다. 이러한 문제를 해결하기 위하여 공급자, 관리자 중심이 아닌 사용자와 시민 중심으로 설계, 운영, 관리되는 IT 기술 기반 미래형 도시 모델인 스마트시티가 탄생하게 되었다. 우리나라도 국가 중점 사업으로 스마트시티의 효율적인 건설 및 운영을 통해 도시 경쟁력을 향상하고 지속 가능한 발전을 촉진하려는 시도를 적극적으로 하고 있다. 이를 뒷받침하기 위해 본 연구는 국토종합계획 및 스마트도시 종합계획, 스마트시티 전략계획 등을 기반으로 세종시의 스마트도시서비스 기반 시설 등의 기본 방향 및 추진 전략을 검토하고, 관련 계획의 연관 관계를 조사하여 세종시 스마트시티 관련 사업 검토와 스마트도시 조성을 위한 추진 체계 및 정책을 제언하고자 한다. This urban centralization is expected to develop rapidly, with 75% of the population living in the city by 2035. Large cities are becoming unsustainable due to side effects such as environmental pollution, severe traffic jams, excessive energy depletion, and destruction of the natural ecosystem. In addition, the happiness index of citizens of large cities is also falling because of high crime rates and safety accidents, the work-life imbalance caused by inequality and polarization, and overly competitive education. To solve this problem, Smart City, an IT-based future city model, was born. The Korean government is also actively attempting to improve urban competitiveness and promote sustainable development through efficient construction and operation of smart cities as a national focus project. To support the effort, we review the basic directions and strategies of Sejong City’s Smart City service infrastructure based on the comprehensive national land plan, Smart City plan, and Smart City strategy plan.

<P>The powered exoskeleton race in Cybathlon 2016 consisted of six challenging tasks that required a pilot with complete paraplegia to walk on a level floor, uphill, downhill, and on stairs; stand up and sit down; step on stones; and even pass through a tilted path. All of these tasks addressed exactly the requirements for a powered exoskeleton designed to assist with activities of daily living (ADL) for paraplegics. Every team brought unique technologies to achieve this goal.</P>
