        Nrf2-Heme oxygenase-1 modulates autophagy and inhibits apoptosis triggered by elevated glucose levels in renal tubule cells

        Background: Autophagy is a highly balanced process in which lysosomes remove aged and damaged organelles and cellular proteins. Autophagy is essential to maintain homeostasis in the kidneys. Methods: Using human renal tubule cells HK-2, we assessed the impact of high glucose (HG) on autophagy. We also evaluated the capability of sulforaphane (SFN) to protect the HK-2 cells from HG-induced apoptosis by modulating autophagy. Results: SFN modulated autophagy and decreased apoptosis in the HK-2 cells that were cultured in 250 mM glucose medium for two days. The reactive oxygen species (ROS) levels increased, as expected, in the cells cultured in the 250 mM glucose medium. However, the SFN decreased the ROS levels in the HK-2 cells. The overexpression of heme oxygenase-1 (HO-1) by SFN decreased the expression of LC3 and beclin-1. LC3 and beclin-1 were involved in the downregulation of caspase-3 that was observed in the HG-induced cells. Conclusion: The activation of nuclear factor E2-related factor 2 (Nrf2)-HO-1 inhibited ROS expression and subsequently attenuated autophagy and cell apoptosis after HG injury was decreased. HG injury led to the activation of autophagy and HO-1 in order to combat oxidative stress and protect against cell apoptosis. Therefore, HO-1 activation can prevent ROS development and oxidative stress during HG injury, which considerably decreases autophagy and apoptosis.

        2014년 10월 2일부터 4일까지 강원도 양양군 하조대 일대를 한국거미연구회 회원(김주필, 예용택, 신정애, 예성훈, 성민규, 이정준, 채준호)들이 조사 연구한 결과 11과 18속 20종이 채집되었기에 이에 보고하는 바이다. The authors investigated the spiders of the Hajodae beach (Gangwon-Do, Yangyang-Gun). In the present paper 20 species of 18 genera belong to 11 families were identified. These specimens are deposited at the Joo-Pil Spider Museum, Namyangju-Si, Gyeonggi-Do.

        백서 척수후근절로부터 슈반세포의 효과적인 체외 배양법

        Schwann cells(SCs), an important component of the peripheral nervous system, intract with nerous to mutually support growth and replication for the peripheral nerve regentation. Recently, ading SCs to the lumen of guidance channel is widely tried to improve regeneration or to make regeneration possible over otherwise irreparable gaps. however, it is not easy to isolate and multiplicate SCs as much as enough to help the axonal regeneration. For the allogeneic SCs source for tubular nerve guidance, we developed a little bit improved technique of harvesting and multiplicating SCs. by culturing dispersed dorsal root ganglia in specially designed medium with growth factors and serial processing, we repeatedlly generate relatively homogenous SC cultures. Our technique was compared with other methods of literature using immunostaining methods such as GFAP, S100, BDNF and the total SC count assessment at different time interval after primary culture.

        한국인 직무 스트레스 측정도구의 개발 및 표준화

        Background and Purposes: Over the past three decades, numerous studies performed in Korea have reported that job stress is a determinant risk factor for chronic diseases and work disability. Every society has its own culture and occupational climate particular to their organizations, and hence experiences different occupational stress. An occupational stress measurement tool therefore needs to be developed to estimate it objectively. The purpose of this study is to develop and standardize the Korean Occupational Stress Scale (KOSS) which is considered to be unique and specific occupational stressors in Korean employees. Subjects and Methods: Data were obtained from the National Study for Development and Standardization of Occupational Stress (NSDSOS Project: 2002-2004). A total of 12,631 employees from a nationwide sample proportional to the Korean Standard Industrial Classification and the Korean Standard Occupational Classification were administered. The KOSS was developed for 2 years (2002-2004). In the first year, we collected 255 items from the most popular job stress measurement tools such as JCQ, ERI, NIOSH and OSI, and 44 items derived from the a qualitative study (depth interview). Forty-three items of KOSS, in the second year, were retained for use in the final version of the KOSS by using Delphi and factor analysis. Items were scored using conventional 1-2-3-4 Likert scores for the response categories. Results: We developed eight subscales by using factor analysis and validation process: physical environment (3 items), job demand (8 items), insufficient job control (5 items), interpersonal conflict (4 items), job insecurity (6 items), organizational system (7 items), lack of reward (6 items), and occupational climate (4 items). Together they explained 50.0% of total variance. Internal consistency alpha scores were ranged from 0.51 to 0.82. Twenty-four items of the short form of the KOSS (KOSS-SF) were also developed to estimate job stress in the work setting. Because the levels of the subscales of occupational stress were gender dependent, gender-specific standard norms for both the 43-item full version and the 24-item short form using a quartile for the subscales of KOSS were presented. Conclusion: The results of this study suggest that KOSS might be an appropriate measurement scale to estimate occupational stress of Korean employees. Further and more detailed study needs to be conducted to improve the validity of this scale.

      • 라텍스개질 콘크리트의 물-시멘트비에 따른 건조수축 특성

        Drying shrinkage cracking which may be caused by the relatively large specific suface is a matter of grave concern for latex modified concrete(LMC) overlay and rapid-setting cement latex modified concrete(RSLMC) overlay. LMC and RSLMC were studied for field applications very actively in terms of strength and durability in Korea. However, there were no considerations in drying shrinkage. Therefore, the purpose of this dissertation was to study the drying shrinkage properties of LMC and RSLMC with the main experimental variables such as cement types(ordinary portland cement, rapid setting cement), water-cement ratios and curing days at a same controlled environment of 60% of relative humidity and 20℃ of temperature. The drying shrinkage for specimens was measured with a digital dial gauge of Demec. The test results showed that the drying shrinkage of LMC and RSLMC were considerably lower with low water-cement ratio, respectively. This might be attributed to the interlocking of hydrated cement and aggregates by a film of latex particles, water retention due to hydrophobic, and colloidal properties of the latexes resulting in reduced water evaporation.

      • 일반인에서 자율신경계 기능 평가

        Background : To evaluate the autonomic nervous system functions, sympathetic skin response (SSR) and R-R interval variation (RRIV) were tested in general persons. Methods : The SSR and the RRIV were evaluated in 24 subjects with a mean age of 39.56 year (24 ~ 56 year). Bilateral SSRs were recorded on the palm and sole with stimulation of median and tibial nerves. The RRIVs were estimated at rest, during deep breathing and valsalva maneuver. Results : The SSR and the RRIV were obtained from all subjects. The mean latencies of SSR recorded from both palms and soles were 1.41 ± 0.05 sec (right palm), 1.28 ± 0.03 sec (left palm), 1.80 ± 0.09 sec (right sole) and 1.75 ± 0.11 sec (left sole). The mean amplitudes of SSR were 5.98 ± 0.67 mV (right palm), 5.70 ± 0.06 mV (left palm), 3.17 ± 0.45 mV (right sole) and 2.23 ± 0.15 mV (left sole). The mean values of RRIV at rest, during deep breathing and valsalva maneuver were 1.14 ± 0.05, 1.18 ± 0.09 and 1.27 ± 0.22. Conclusion : Functional status of autonomic nervous system is obtained easily with SSR and RRIV. Our results can be a useful basis of control values.

        The optimal medium composition for the production of carotenoid pigment from Haloarcular sp. EH-1 as a dietary for fishes were 1.0% sucrose, 1.0% yeast extract, 25% sodium chloride, 0.3% sodium citrate, 0.2% potassium chloride, 2.0% magnesium sulphate, 0.002% ferric sulphate(pH7.0). The incubation temperate, aeration rate and agitation speed were 40℃, 100ml medium/500ml vol. shaking flask, and 180 rpm/min. The carotenoid pigment production was highest after 5 days of incubation with the light.

      • 컴포넌트 기반의 건강관리 시스템 개발을 위한 UI 툴

        Component-based systems are generally known as useful for maintenance and extension. However, there are a number of difficult aspects in designing them, and skillful knowledge of component-based design is strongly requested. Against the problem, this paper presents a User Interface tool that helps to develop component-based healthcare systems more easily and effectively. This UI tool has been developed, aiming at enabling easier and faster development of such systems. Further it supports standardization and interoperability. We expect that use of the UI tool helps to add, delete, change medical contents, and maintain related systems with less effort and better performance.

      • 재활의학과적 검사 결과가 임상증상과 차이를 보인 잠수병 환자

        Decompression sickness is divided into 2 types based on clinical symptoms and severity. Type I decompression sickness involves usually the musculoskeletal system and cardiopulmonary system. Type II decompression sickness involves commonly the central nervous system. We experienced a case of type II decompression sickness involving cervical spinal cord. The patient showed hypesthesia below C5 dermatome and quadriplegia. But the whole spinal magnetic resonance image revealed high signal of the spinal cord below T2 levels and electrodiagnostic study showed thoracic cord level lesion. In decompression sickness, diagnosis of injured spinal cord levels is very delicate problem because in some cases clinical symptoms can be unmatched with magnetic resonance findings and electrodiagnostic study. In this kind of case there are little studies of diagnostic tools and prognostic factors. Therefore, focusing on clinical symptoms need to help more proper treatment and provide better prognosis to decompression sickness patients

        플라즈마 표면복합처리한 구상흑연주철의 열피로 특성

        This paper reports an investigation of the characteristics of plasma duplex treated ductile cast irons during thermal fatigue tests on a thermal cycling rig. The thermal fatigue cracking behaviors and microstructure of untreated and plasma surface treated ductile cast irons have been investigated. Both ferrite and pearlite matrix ductile cast irons were evaluated. During the tests, 60 mm long cylinders with 20 mm diameter were subjected to 1,000 high frequency induction heating and water cooling cycles, lasting 7 and 5 sec, respectively. The thermal fatigue damage was evaluated by analyzing different crack dimensions and distribution by light microscopy. The results showed that the plasma surface treatment increased thermal fatigue cracking resistance. When thermal fatigue cracking occurs, the cracks always initiate at the surface of the specimen. The major path of crack propagation is generally along the ferrite grain boundaries. The hardened layer had an effect on the thermal fatigue property. In addition, the ductile cast iron with an pearlite matrix has a better resistance to thermal fatigue than that of ferrite matrix.
