        Up‑regulation of miR‑27 extenuates lipopolysaccharide‑induced injury in H9c2 cells via modulating ICAM1 expression

        Background MiR-27 has been found to present an overt myocardial expression during cardiogenesis. However, whether miR-27 involves in myocarditis development and the possible molecular mechanism remain unknown. The purpose of this study was to investigate the biological characteristic of miR-27 in LPS-damaged H9c2 cells. Methods H9c2 cells were treated with lipopolysaccharide (LPS, 10 μg/ml) for 12 h to form cell injury. MiR-27 mimic and inhibitor were used to up-regulate or down-regulate miR-27 expression. MTT assay and flow cytometry analysis were conducted to test cell viability and apoptosis. The relative RNA expression level of miR-27 and intercellular adhesion molecule 1 (ICAM1) was determined by qRT-PCR. Luciferase reporter gene assay was utilized to confirm the interaction between miR-27 and ICAM1. Western blot was used to determine the protein expression levels. Results We observed that LPS treatment significantly decreased the level of miR-27 in H9c2 cells. Moreover, LPS exposure suppressed cell viability, promoted cell apoptosis and increased the relative expression of p-NF-κB p65/NF-κB p65 and p-IκBα/IκBα. Up-regulation of miR-27 increased cell proliferation and reduced cell apoptosis, while down-regulation of miR-27 suppressed cell growth and promoted cell apoptosis. ICAM1 was predicted and verified as a target of miR-27, and the expression of ICAM1 is negatively regulated by miR-27. The relative expression of p-NF-κB p65/NF-κB p65 and p-IκBα/ IκBα was dramatically decreased by miR-27 mimic and increased by miR-27 inhibitor. Conclusion Our study illustrated that up-regulation of miR-27 exhibits a protective effect on LPS-damaged H9c2 cells, which may be achieved by regulating ICAM1 and NF-κB signaling.

        A method of X-ray source spectrum estimation from transmission measurements based on compressed sensing

        A new method of X-ray source spectrum estimation based on compressed sensing is proposed in this paper. The algorithm K-SVD is applied for sparse representation. Nonnegative constraints are added by modifying the L1 reconstruction algorithm proposed by Rosset and Zhu. The estimation method is demonstrated on simulated spectra typical of mammography and CT. X-ray spectra are simulated with the Monte Carlo code Geant4. The proposed method is successfully applied to highly ill conditioned and under determined estimation problems with a good performance of suppressing noises. Results with acceptable accuracies (MSE < 5%) can be obtained with 10% Gaussian white noises added to the simulated experimental data. The biggest difference between the proposed method and the existing methods is that multiple prior knowledge of X-ray spectra can be included in one dictionary, which is meaningful for obtaining the true X-ray spectrum from the measurements.

        Effects of co-ion initial concentration ratio on removal of Pb2+ from aqueous solution by modified sugarcane bagasse

        A modified sugarcane bagasse (SCB) fixed bed column was used to remove Pb2+ from aqueous solution. To determine the optimal condition for Pb2+ separation, Ca2+ was chosen as the model interfering ion, and effects of Ca2+ and Pb2+ initial concentration ratio (C0 Ca : C0 Pb) on the adsorption of Pb2+ were investigated. Results showed that adsorption amount ratio of Ca2+ and Pb2+ (qe Ca : qe Pb) had a good linear relationship with C0 Ca : C0 Pb. Mass ratio of Pb2+ absorbed on the modified SCB was higher than 95% at C0 Ca : C0 Pb<1.95, illustrating that Pb2+ could be selectively removed from aqueous solution. To verify that, simulated waste water containing co-ions of K+, Na+, Cd2+ and Ca2+ was treated, and results showed that the equilibrium amount of Pb2+, K+, Na+, Cd2+ and Ca2+ adsorbed was 134.14, 0.083, 0.058, 1.28, and 1.28mg g−1, respectively, demonstrating that the modified SCB could be used to remove Pb2+ from aqueous solution in the investigated range.

        Isolation, Structural Characterization, and Lymphopoiesis Stimulant Activity of a Polysaccharide from the Abalone Gonad

        A novel polysaccharide (AGP-32) from the gonad of Haliotis discus hannai Ino was isolated using a protease-assisted process and successive ion-exchange and gel-filtration chromatography. The backbone of AGP-32 was determined using hydrolysis with trifluoroacetic acid. FTIR, NMR, and methylation analysis, and periodate oxidation and Smith degradation analysis revealed that the AGP-32 backbone mainly consisted of (1→6)-linked mannose, (1→3)-linked galactose, and (1→3)-linked glucose in a proportion of 2:3:1. An in vitro cell assay indicated that AGP-32 promoted mice splenic lymphocyte proliferation by 26% at a concentration of 50 μg/mL. AGP-32 had an effect on immune protection and is a candidate for consideration as a functional food.

        The KIMS collaboration is an experimental group searching for the Weakly Interacting Massive Particle (WIMP) which is one of the strongest candidates of dark matter. In a WIMP search experiment, the neutron is an important background because the nuclear recoil signal of a neutron is indistinguishable from that of a WIMP. We have constructed a 20-liter liquid scintillation detector to measure the neutron background. We present the performance of the neutron detector, as well as its structure and electronics system. We have achieved a good neutron/gamma separation anda good position measurement. A position dependent energy calibration has also been performed to correct the nonlinear response of the detector for different particle incident positions.

        MiR-188-5p regulates the proliferation and differentiation of goat skeletal muscle satellite cells by targeting calcium/calmodulin dependent protein kinase II beta

        Objective: The aim of this study was to reveal the role and regulatory mechanism of miR-188-5p in the proliferation and differentiation of goat muscle satellite cells. Methods: Goat skeletal muscle satellite cells isolated in the pre-laboratory were used as the test material. First, the expression of miR-188-5p in goat muscle tissues at different developmental stages was detected by quantitative reverse transcription polymerase chain reaction (qRT-PCR). In addition, miR-188-5p was transfected into goat skeletal muscle satellite cells by constructing mimics and inhibitors of miR-188-5p, respectively. The changes of differentiation marker gene expression were detected by qPCR method. Results: It was highly expressed in adult goat latissimus dorsi and leg muscles, goat fetal skeletal muscle, and at the differentiation stage of muscle satellite cells. Overexpression and interference of miR-188-5p showed that miR-188-5p inhibited the proliferation and promoted the differentiation of goat muscle satellite cells. Target gene prediction and dual luciferase assays showed that miR-188-5p could target the 3'untranslated region of the calcium/calmodulin dependent protein kinase II beta (CAMK2B) gene and inhibit luciferase activity. Further functional studies revealed that CAMK2B promoted the proliferation and inhibited the differentiation of goat muscle satellite cells, whereas si-CAMK2B restored the function of miR-188-5p inhibitor. Conclusion: These results suggest that miR-188-5p inhibits the proliferation and promotes the differentiation of goat muscle satellite cells by targeting CAMK2B. This study will provide a theoretical reference for future studies on the molecular mechanisms of skeletal muscle development in goats.

        Real or Hyperreal: Bob Dylan’s Oral Poetry in the Digital Age

        This paper approaches oral poetry by setting Bob Dylan, an American singer-songwriter, as an example. Dylan has been widely recognized as a major figure in popular culture and the realm of oral poetry for over half a century. Being the only musical creator entitled to the Nobel Literature Prize (in 2016) for “having created new poetic expressions within the great American song tradition” (Nobelprize.org.), Dylan proves to be an artist and scholar who successfully integrate conventional literature with sound in the new media era. However, it remains unknown whether the combination of oral poetry with technoscientific tools brings literature infinitely closer to reality or merely fabricates another fictional domain with hyperreality. This study takes the musician as a postmodernist and explores the power of presentation in his lyrics. By appreciating the poetic lines and the patterns of expression, this paper primarily aims to provide a novel take on Dylan’s oral poetry as well as literature itself in the current digital era.

        Disrupting Conventional Representations of Time and Space in A Girl is a Half-formed Thing and How to Wash a Heart

        This paper explores the disruptive tendencies in contemporary literature regarding conventional representations of time and space, as witnessed in Eimear McBride’s A Girl is a Half-formed Thing and Bhanu Kapil’s How to Wash a Heart. Guided by the theoretical frameworks of Fredric Jameson and Mikhail Bakhtin, the analysis delves into the departure from linear time and static spatial backdrops. Both literary works challenge traditional notions by employing fragmented temporal structures, reversible temporality, and dynamic shifts between past and present experiences. The interplay of Jameson’s “Temporal Permanent” and Bakhtin’s “Chronotope” further enriches our understanding, highlighting enduring emotional facets and converging temporal trajectories within shared spatial contexts. The concept of the “Juxtaposition of Temporal Chronotopes” emerges as a pivotal analytical lens, revealing the interconnectedness of time and space in these works. In essence, this exploration contributes to a nuanced understanding of the intricate interplay between time, space, and narrative dynamics in contemporary literature.

        Mountain and Sea: Gender Dilemma in Park Chan-wook’s Decision to Leave (2022)

        This paper delves into the clash between post-feminism and remasculinization within contemporary Korean cinema, with a specific focus on Park Chan-wook’s romantic mystery film, Decision to Leave (2022). While previous research has mainly focused on diverse themes, such as romance, gender, ethical violence, and human existence with transcultural imagery, this study adopts an interdisciplinary approach to analyze the film’s artistic incorporation of metaphorical imagery, particularly the mountain and sea, and how it illuminates the gender dynamics within the play. The paper argues that these visual elements shed light on the gender dilemma inherent in the cross-cultural and multi-dimensional elements in of South Korean filmmaking. By examining the embodiment of individual concept, post-feminism and remasculinization, in the prototypes of Seo-rae and Hae-jun respectively, the paper uncovers the underlying causes of the failures in their interaction and the tragic fate of their relationship. Furthermore, the study demonstrates that these diverse and dynamic ideologies, including remasculinization and post-feminism, create and correspond to the metaphorical haze pictured in the film. The imagery of haze lingers the obscure cinematic landscape surrounding contemporary gender dilemma in South Korea.
