        DNA methylation and mRNA expression of COL6A3 in antler mesenchyme of female and male reindeer

        Backgroud Reindeer is the only deer species that both male and female produce antlers, which provides a particularly interesting case in studying the differences between antlers of the two sexes. Alpha 3(VI) Collagen Gene (COL6A3), forms a microfibrillar network associated with the structural integrity and biomechanical properties, has been found to be one of the differentially expressed genes in antler mesenchyme of female and male reindeer. Objective and Methods The promoter sequence of reindeer COL6A3 gene was obtained using the cloning technology and analyzed by the bioinformatics methods. Bisulfite sequencing PCR (BSP) was used to detect the methylation status of the COL6A3 promoter in reindeer antler mesenchyme. Real-time quantitative PCR was used to detect COL6A3 expression in the antler mesenchyme of female and male reindeer. Results Sequence analysis revealed that the reindeer COL6A3 partial promoter sequence was 983 bp including the possible promoter region at + 105 bp to + 155 bp. Homology and phylogenetic analysis indicated that the COL6A3 promoter of reindeer had the closest genetic distance with Bos taurus, Capra hircus and Ovis aries. BSP results indicated that the methylation level of COL6A3 promoter in the female reindeer antler mesenchyme was significantly higher than in the male. Correlating with increased methylation status, we also found that COL6A3 mRNA expression in female reindeer antler mesenchyme was significantly lower than in the male. Conclusion The higher methylation level of the COL6A3 gene in female reindeer antler mesenchyme coincides with decreased COL6A3 mRNA expression, thereby affecting the transposon silencing mechanism and possibly contributing to apparent differences of antlers in female and male reindeer.

      • Evaluation of the Geometric Accuracy of Anatomic Landmarks as Surrogates for Intrapulmonary Tumors in Image-guided Radiotherapy

        Objectives: The purpose of this study was to evaluate the geometric accuracy of thoracic anatomic landmarks as target surrogates of intrapulmonary tumors for manual rigid registration during image-guided radiotherapy (IGRT). Methods: Kilovolt cone-beam computed tomography (CBCT) images acquired during IGRT for 29 lung cancer patients with 33 tumors, including 16 central and 17 peripheral lesions, were analyzed. We selected the "vertebrae", "carina", and "large bronchi" as the candidate surrogates for central targets, and the "vertebrae", "carina", and "ribs" as the candidate surrogates for peripheral lesions. Three to six pairs of small identifiable markers were noted in the tumors for the planning CT and Day 1 CBCT. The accuracy of the candidate surrogates was evaluated by comparing the distances of the corresponding markers after manual rigid matching based on the "tumor" and a particular surrogate. Differences between the surrogates were assessed using 1-way analysis of variance and post hoc least-significant-difference tests. Results: For central targets, the residual errors increased in the following ascending order: "tumor", "bronchi", "carina", and "vertebrae"; there was a significant difference between "tumor" and "vertebrae" (p = 0.010). For peripheral diseases, the residual errors increased in the following ascending order: "tumor", "rib", "vertebrae", and "carina"; There was a significant difference between "tumor" and "carina" (p = 0.005). Conclusions: The "bronchi" and "carina" are the optimal surrogates for central lung targets, while "rib" and "vertebrae" are the optimal surrogates for peripheral lung targets for manual matching of online and planned tumors.

        Enhancement of Pyruvate Production by Torulopsis glabrata through Supplement of Oxaloacetate as Carbon Source

        The capability of utilizing a TCA cycle intermediates as the sole carbon source by the multi-vitamin auxotrophic yeast Torulopsis glabrata CCTCC M202019 was demonstrated with plate count method. It is indicated that T. glabrata could grew on a medium with one of the TCA cycle intermediates as the sole carbon source, but more colonies were observed when glucose, acetate and one of the TCA cycle intermediates coexisted in the medium. Among the intermediates of the TCA cycle examined in this study, cell growth was improved by supplementing oxaloacetate. Further investigation showed that the presence of acetate was necessary when oxaloacetate was supplemented. By supplementing with 10 g/L of oxaloacetate in pyruvate batch fermentation, dry cell weight increased from 11.8 g/L to 13.6 g/L, and pyruvate productivity was enhanced from 0.96 gL-1h-1 to 1.19 gL-1h-1 after cultivation of 56 h. The yield of pyruvate to glucose was also improved from 0.63 g/g to 0.66 g/g. These results indicate that under vitamins limitation, the productivity and yield of pyruvate could be enhanced via an increase of cell growth by the supplementation of oxaloacetate.

        Safety design and performance analysis of humanoid rehabilitation robot with compliant joint

        Human safety represents the key requirement in physical human-robot interaction (pHRI). However, the majority of current research works focus on the safety evaluation. There is no definite way to design a robot that not only meets the performance but also has inherent safety. A humanoid robot for human joint movement rehabilitation is developed, to fulfill the requirement of both safety and its performance. A nonlinear model of human-robot collision with effective mass and stiffness of robot’s end-effecter (EE) is proposed. An important parameter involved in the model is the joint stiffness of the robot, which has an inherent direct effect on the safety. The influence of joint compliance on the modal frequencies is analyzed, and the kinematic performance of the robot is estimated roughly by the lowest order modal frequency. The design method of compliant joint stiffness is put forward, which can balance the safety and kinematics performance requirements. When the compliance is utilized intentionally in joints to improve safety, the question whether or not rigid performance of compliant joint can be achieved or approached by control is arose naturally. Such a challenge is addressed by the cascade control, where outer position loop with link-side position feedback is constructed. Here, restoring torque in the elastic transmission is taken as virtual control that is, in turn, set as the reference command of the inner torque loop. Intuitively, once the virtual control is reproduced as rapidly as possible in the inner torque loop, quasi-rigid performance is exhibited in the outer position loop as if there were no series elasticity in compliant joint. The stability criterion is derived, and the virtual stiffness and dynamic performance are examined. Finally, experiments are performed to validate effectiveness of the suggested control scheme.

      • Relation between Ki-67, ER, PR, Her2/neu, p21, EGFR, and TOP II-α Expression in Invasive Ductal Breast Cancer Patients and Correlations with Prognosis

        The aim of the present study was to investigate the expression of the transcription factor Ki-67, ER, PR, Her2/neu, p21, EGFR, and TOP II-${\alpha}$ in the tumor tissue of patients with invasive ductal carcinoma(IDC); in addition, we examined correlations between these markers. Two hundred and sixteen IDC patients, who were not previously been treated with chemo- or radiotherapy, were included in the study. All tumors were grade I-III. Expression of molecular markers was determined by immunohistochemical analysis on paraffin-embedded tissue sections. Follow-up data were collected for 3 months to 10 years and analyzed for tumor recurrence, survival time, and prognostic risk factors. We determined Ki-67 expression correlates with the expression of ER, PR, HER-2, EGFR, and TOP-${\alpha}$, as well as lymph node involvement, high tumor grade, lymphovascular invasion, high tumor stage, and high TNM stage in IDC. Positive Ki-67 expression was a risk factor for rapid tumor recurrence and may help tumor progression, leading to poor prognosis in IDC. Ki-67 was directly correlated with EGFR, TOP II-${\alpha}$, lymph node involvement, high tumor grade, lymphovascular invasion, high tumor stage, and high TNM stage in the hormone receptor subtypes of breast cancer. In triple negative breast cancer, Ki-67 correlated with TOP II-${\alpha}$. Expression of Ki-67 correlated with that of ER, PR, HER-2, EGFR, TOP II-${\alpha}$, and p21. In addition, the biomarker Ki-67 has a role as a prognostic factor and indicates a poor prognosis in IDC.

        Realgar transforming solution-induced differentiation of NB4 cell by the degradation of PML/RARa partially through the ubiquitin–proteasome pathway

        PML/retinoic acid receptor alpha (RARa), as ahallmark of acute promyeloid leukemia (APL), is directlyrelated to the outcome of clinical APL remedy. It isreported that arsenicals can effectively degrade PML/RARa, such as arsenic trioxide and realgar. However, thehigh toxicity or insolubility have hampered their clinicalapplications. Realgar transforming solution (RTS) wasproduced from realgar by bioleaching process in our lab. Previous studies demonstrated that RTS had a significantanti-cancer ability on chronic myeloid leukemia throughoncoprotein degradation. The capacity of RTS on treatingAPL is what is focused on in this study. The results showedthat RTS had a noticeable sensitivity in NB4 cell, and RTSremarkably down-regulated PML/RARa expression andinduced cell differentiation. Further, RTS could accumulatePML/RARa into the nuclear bodies and then executedegradation, which could be reversed by proteasomeinhibitor MG132. The results also exhibited that thereduction of RTS-induced PML/RARa expression accompaniedby the elevation of ubiquitin and SUMO-1 proteinexpression. Finally, PML and SUMO-1 had been demonstratedto be co-localized after RTS treatment byimmunofluorescence co-localization assay and immunoprecipitationassay. In conclusion, these results suggestedthat RTS-induced cell differentiation may attribute to thePML/RARa degradation partially through the ubiquitin–proteasome pathway.

        S100A4 Gene is Crucial for Methionine-Choline-Deficient Diet-Induced Non-Alcoholic Fatty Liver Disease in Mice

        Purpose: To explore the influence of S100 calcium binding protein A4 (S100A4) knockout (KO) on methionine-choline-deficient(MCD) diet-induced non-alcoholic fatty liver disease (NAFLD) in mice. Materials and Methods: S100A4 KO mice (n=20) and their wild-type (WT) counterparts (n=20) were randomly divided into KO/MCD, Ko/methionine-choline-sufficient (MCS), WT/MCD, and WT/MCS groups. After 8 weeks of feeding, blood lipid and liverfunction-related indexes were measured. HE, Oil Red O, and Masson stainings were used to observe the changes of liver histopathology. Additionally, expressions of S100A4 and proinflammatory and profibrogenic cytokines were detected by qRT-PCR andWestern blot, while hepatocyte apoptosis was revealed by TUNEL staining. Results: Serum levels of aminotransferase, aspartate aminotransferase, triglyceride, and total cholesterol in mice were increasedafter 8-week MCD feeding, and hepatocytes performed varying balloon-like changes with increased inflammatory cell infiltrationand collagen fibers; however, these effects were improved in mice of KO/MCD group. Meanwhile, total NAFLD activity scoresand fibrosis were lower compared to WT+MCD group. Compared to WT/MCS group, S100A4 expression in liver tissue of WT/MCD group was enhanced. The expression of proinflammatory (TNF-α, IL-1β, IL-6) and profibrogenic cytokines (TGF-β1, COL1A1,α-SMA) in MCD-induced NAFLD mice were increased, as well as apoptotic index (AI). For MCD group, the expressions ofproinflammatory and profibrogenic cytokines and AI in KO mice were lower than those of WT mice. Conclusion: S100A4 was detected to be upregulated in NAFLD, while S100A4 KO alleviated liver fibrosis and inflammation, inaddition to inhibiting hepatocyte apoptosis.

      • Research Paper : Effects of phosphorus and light intensity on the growth and competition of the two weed species Veronica persica and Chorispora tenella

        Veronica persica Poir. (Persian speedwell) is an invasive alien weed species in the upland soils throughout China. Its dominance in weed flora could be alleviated by Chorispora tenella (Pall.) (tender chorispora) in P-poor soils and in an intense light environment. Here, three experiments in growth chambers were carried out in order to explore the effect of P and light intensity on the growth and competition of V. persica and C. tenella, the pH value in their rhizospheres, and the kinetic parameters (Km and Vmax) of the H2PO4 - uptake by the two weed species. The ANOVA results showed that the light intensity and P level in the soil obviously influenced the growth of the two weed species. However, the effect of the P level in the soil was greater for V. persica and the effect of the light intensity was greater for C. tenella. The restriction of the growth of V. persica by a low P level in the soil was much more than that of C. tenella, but it was reversed in the two species in relation to a low light intensity. In addition, the pH was lower in the rhizosphere of C. tenella after the P-deficient treatment and there was a lower Km in the uptake of H2PO4 - than for V. persica, suggesting that C. tenella could adapt to a low-P environment more easily. In conclusion, C. tenella growing together with V. persica had a competitive advantage under high-light-intensity and low-P-level conditions, but it lost its advantage with an increase in P fertilization and a decrease in light intensity.

      • Anti-tumor Activity and Apoptosis-regulation Mechanisms of Bufalin in Various Cancers: New Hope for Cancer Patients

        The induction of apoptosis in target cells is a key mechanism for most anti-tumor therapies. Bufalin is a cardiotonic steroid that has the potential to induce differentiation and apoptosis of tumor cells. Research on bufalin has so far mainly involved leukemia, prostate cancer, gastric cancer and liver cancer, and has been confined to in vitro studies. The bufadienolides bufalin and cinobufagin have been shown to induce apoptosis in a wide spectrum of cancer cell. The present article reviews the anticancer effects of bufalin. It induces apoptosis of lung cancer cells via the PI3K/Akt pathway and also suppressed the proliferation of human non-small cell lung cancer A549 cell line in a time and dose dependent manner. Bufalin, bufotalin and gamabufotalin, key bufadienolides, significantly sensitize human breast cancer cells with differing ER-alpha status to apoptosis induction by the TNF-related apoptosis-inducing ligand (TRAIL). In addition, bufadienolides induce prostate cancer cell apoptosis more significantly than that in breast epithelial cell lines. Similar effects have been observed with hepatocellular carcinoma (HCC) but the detailed molecular mechanisms of inducing apoptosis in this case are still unclear. Bufalin exerts profound effects on leukemia therapy in vitro. Results of multiple studies indicate that bufalin has marked anti-tumor activities through its ability to induce apoptosis. Large-scale randomized, double-blind, placebo or positive drug parallel controlled studies are now required to confirm the efficacy and apoptosis-inducing potential of bufalin in various cancers in the cliniucal setting.

        A Study of Management Problems and Countermeasures of Home Appliance Manufacturing Enterprises Based on a Green Supply Chain during COVID-19

        Purpose There was a significant impact on the global economy in 2020 during COVID-19. Home appliance manufacturing enterprises are important to promote the rapid development of economies in Korea and China;but with rapid development;waste has caused enormous damage to the environment. Consumers take green factors into account when purchasing products. At the same time;governments promulgate environmental laws and regulations. Therefore;it is necessary and urgent for home appliance manufacturing enterprises to implement green supply chain management. Design/Methodology/Approach Based on the development of green supply chain management theory;this paper analyzes the present situation of home appliance manufacturing industry in Yancheng and the existing problems of green supply chain management. Based on a questionnaire on the restrictive factors of green supply chain management of the home appliance manufacturing industry in Yancheng using SPSS21.0;this paper studied the restraining factors and corresponding secondary factors of the green supply chain management in Yancheng home appliance manufacturing enterprises (government;enterprise;supply chain upstream;and downstream). Findings The study found that government policy was most influential;among which environmental laws and regulations had a low impact;but imperfect environmental laws and regulations and loose enforcement have a greater impact. Second was the influence of the companies themselves;among which the number and quality of green supply chain management personnel to be improved have a greater impact. Supply chain upstream and downstream were the least impact factors;among which conveying the concept of green supply chain management to suppliers had a greater impact. Research Implications Strategies and solutions are put forward to the government;enterprises;and the upstream and downstream of the supply chain to promote the improvement of green supply chain management of home appliance manufacturing enterprises in Yancheng. These findings could be applied equally to Korea.
