Interferon (IFN)-λ plays an essential role in mucosal cells which exhibit strong antiviral activity. Lactobacillus plantarum (L. plantarum) has substantial application potential in the food and medical industries because of its probiotic properties. Alphacoronaviruses, especially porcine epidemic diarrhea virus (PEDV) and transmissible gastroenteritis virus (TGEV), cause high morbidity and mortality in piglets resulting in economic loss. Co-infection by these two viruses is becoming increasingly frequent. Therefore, it is particularly important to develop a new drug to prevent diarrhea infected with mixed viruses in piglets. In this study, we first constructed an anchored expression vector with CWA (C-terminal cell wall anchor) on L. plantarum. Second, we constructed two recombinant L. plantarum strains that anchored IFN-λ3 via pgsA (N-terminal transmembrane anchor) and CWA. Third, we demonstrated that both recombinant strains possess strong antiviral effects against coronavirus infection in the intestinal porcine epithelial cell line J2 (IPEC-J2). However, recombinant L. plantarum with the CWA anchor exhibited a more powerful antiviral effect than recombinant L. plantarum with pgsA. Consistent with this finding, Lb.plantarum-pSIP-409-IFN- λ3-CWA enhanced the expression levels of IFN-stimulated genes (ISGs) (ISG15, OASL, and Mx1) in IPEC-J2 cells more than did recombinant Lb.plantarum-pSIP-409-pgsA'-IFN-λ3. Our study verifies that recombinant L. plantarum inhibits PEDV and TGEV infection in IPEC-J2 cells, which may offer great potential for use as a novel oral antiviral agent in therapeutic applications for combating porcine epidemic diarrhea and transmissible gastroenteritis. This study is the first to show that recombinant L. plantarum suppresses PEDV and TGEV infection of IPEC-J2 cells.

Hereditary non-polyposis Colorectal Cancer (HNPCC) is an autosomal dominant inheritance syndrome. HNPCC is the most common hereditary variant of colorectal cancer (CRC), which accounts for 2-5% CRCs, mainly due to hMLH1 and hMSH2 mutations that impair DNA repair functions. Our study aimed to identify the patterns of hMSH2 and hMLH1 mutations in Chinese HNPCC patients. Ninety-eight unrelated families from China meeting Amsterdam or Bethesda criteria were included in our study. Germline mutations in MLH1 and MSH2 genes, located in the exons and the splice-site junctions, were screened in the 98 probands by direct sequencing. Eleven mutations were found in ten patients (11%), with six in MLH1 (54.5%) and five in MSH2 (45.5%) genes. One patient had mutations in both MLH1 and MSH2 genes. Three novel mutations in MLH1 gene (c.157_160delGAGG, c.2157dupT and c.-64G>T) were found for the first time, and one suspected hotspot in MSH2 (c.1168C>T) was revealed. A Cluster-Based Countermeasure against MAC Layer Attacks in IEEE 802.11 MANETs

<P>MANETs become a considerable interest nowadays. However, these types of networks are vulnerable to a large group of attacks, e.g., MAC layer attacks. Most of the previous researches related to security issues focus on detection of misbehaviors after it occurred. We propose a new way of thinking to evade occurrence of misbehaviors. A novel clustering algorithm is presented using analytic hierarchy process to elect clusterhead. Afterwards, the clusterhead is required to assign backoff values to originators in its transmission range, which are also the members in the corresponding cluster, rather than permitting the originators to choose backoff values. Meanwhile, the elected clusterhead also monitors whether originators obey the backoff values. With the proposed scheme, the misuse of the backoff in MAC layer in 802.11 DCF can be prevented.</P>
A Lightweight Detection Mechanism against Sybil Attack in Wireless Sensor Network

Sybil attack is a special kind of attack which is difficult to be detected in Wireless Sensor Network (WSN). So a lightweight detection mechanism based on LEACH-RSSI-ID (LRD) is proposed in this paper. Due to the characteristic of Low-Energy Adaptive Clustering Hierarchy (LEACH) protocol, none of nodes can be the cluster head forever. Meanwhile, in order to consume less energy, both factors which are called the remaining energy of nodes and relative density of nodes are taken into account. Therefore, Sybil attack can be found by analyzing the RSSI-ID tables. Different from the previous detection methods, even though Sybil attack occurs in the initialization phase, the malicious nodes can be detected by sink node. What`s more, when each malicious node frequently changes identification, it will be detected in a short time. Through the simulations, it is revealed that the LRD mechanism can detect the Sybil attack with high detection rate and accuracy.

This paper proposes a three degree-of-freedoms (DOFs) cable-driven parallel robot (CDPR), which is actuated by three groups of parallel cables and tensioned by an elastic telescopic rod with a passive spring. Firstly, the architecture of the robot is briefly illustrated with emphasis on the three parallelogram arrangements of the cables and composition of elastic telescopic rod structure. This robot has larger workspace and greater tension than previous under-constrained CDPRs. Secondly, the kinematic and dynamic models of the robot are established. On this basis, the parameters of the spring installed in the telescopic rod are optimized and determined considering both the acceleration and cable force through using the differential evolution (DE) algorithm. Thirdly, dynamic stability analysis of the robot under impulsive disturbances is performed according to the Gauss principle of least constraint (GPLC). The results of a simulation case show that this robot has better dynamic stability in comparison with conventional under-constraint CDPR because of the presence of the elastic telescopic rod. The measurement results in the workspace are obtained through dynamic simulation. Finally, the experiments are performed based on numerical simulation. The feasibility of the CDPR is verified via the experiments and simulations.

<P>We explored a very interesting gold nanoparticle system—pegylated gold in colloidal solution—and analyzed its uptake by mice colorectal adenocarcinoma CT26 tumor cells and the impact on the cell’s response to x-ray irradiation. We found that exposure to polyethylene glycol (PEG) modified (‘pegylated’) 4.7 ± 2.6 nm gold nanoparticles synthesized by a novel synchrotron-based method enhances the response of CT26 cells to x-ray irradiation. Transmission electron microscopy (TEM) and confocal microscopy revealed that substantial amounts of such nanoparticles are taken up and absorbed by the cells and this conclusion is supported by quantitative induced coupled plasma (ICP) results. Standard tests indicated that the internalized particles are highly biocompatible but strongly enhance the cell damage induced by x-ray irradiation. Synchrotron radiation Fourier transform infrared (SR-FTIR) spectromicroscopy analyzed the chemical aspects of this phenomenon: the appearance of C = O stretching bond spectral features could be used as a marker for cell damage and confirmed the enhancement of the radiation-induced toxicity for cells.</P>

Acute graft-versus-host disease (GVHD) following liver transplantation is a rare but fatal complication. The correct diagnosis and management of GVHD after liver transplantation are still major challenges. Herein, we reported successful salvage treatment of acute GVHD by withdrawal of immunosuppression in a patient who presented with fever, skin rashes, and decreased blood cell counts after liver transplantation. This case highlights the need for awareness of drug-induced liver injury if liver function tests are elevated during treatment, especially in patients taking multiple potentially hepatotoxic drugs, such as broad-spectrum antibiotics. When occurs, an artificial liver support system is a useful tool to provide temporary support of liver function for the patient in the event of drug-induced liver injury.

Protein glycation inhibitors from Polygonatum odoratum rhizomes were investigated using a bioassay-guided procedure to characterize active compounds for preventing and treating diabetic complications. The EtOH extract and soluble fractions were evaluated using an in vivo model of renal advanced glycation end-product (AGE) accumulation in streptozotocin-induced diabetic rats and an in vitro bovine serum albumin-glucose assay. Three homoisoflavanones 3-(4’-hydroxybenzyl)-5,7-dihydroxy-6-methyl-8-methoxychroman-4-one (1), 3-(4’-hydroxybenzyl)-5,7-dihydroxy-6,8-dimethylchroman-4-one (2), and 3-(4’-methoxybenzyl)-5,7-dihydroxy-6-methyl-8-methoxychroman-4-one (3), isolated from the active CHCl3-soluble fraction of the EtOH extract, were subjected to in vitro bioassays to evaluate their inhibitory activities against AGE formation. All the isolates inhibited AGE formation more effectively than the positive control, aminoguanidine. These results indicate that pending further study these compounds could be used as novel natural product drug for mitigating diabetic complications.
Balloon dilation of epidermolysis bullosa-related esophageal strictures: A report of two cases

Epidermolysis bullosa (EB) is a very rare inherited disease featured with skin blistering resulting from minor trauma. Sometimes the esophageal mucosa could also be involved, which leads to esophageal strictures. Here we report two cases of EB-related esophageal strictures who were successfully treated with esophageal balloon dilations. The two cases with EB had severe dysphagia. Clinical examination showed signs of malnutrition, skin blisters and loss of toenails due to EB. They underwent careful fluoroscopic balloon dilation with 10- and 16-mm-sized balloon catheters, respectively. They could ingest soft and some solid foods after the procedure and maintained during the 20 months and 16 months follow-up periods.
