      • Galectin-9 Acts as a Prognostic Factor with Antimetastatic Potential in Hepatocellular Carcinoma

        Considerable research has been conducted concerning galectin-9 and carcinomas, but little information is available about any relation with the hepatocellular carcinoma. In this study, we employed a small interfering RNA (siRNA) targeting galectin-9 to down-regulate the expression in HepG2 cells. As a result, after galectin-9 expression was reduced, cell aggregation was suppressed, while other behaviour such as the proliferation, adhesion and invasion to ECM, cell-endothelial adhesion and transendothelial invasion of the cells were markedly enhanced. When tumors of 200 patients with hepatocellular carcinoma were tested for galectin-9 expression by immunohistochemistry, binding levels demonstrated intimate correlations with the histopathologic grade, lymph node metastasis, vascular invasion and intrahepatic metastasis (P<0.05). Moreover, survival analysis indicated that patients with galectin-9 expression had much longer survival time than those with negative lesions, and the Log-rank test indicated that this difference was statistical significant (P<0.0001). The Cox proportional hazards model suggested that negative galectin-9 expression in hepatocellular carcinoma represented a significant risk factor for patient survival. We propose that galectin-9 might be a new prognostic factor with antimetastatic potential in patients with hepatocellular carcinoma.

        A taxonomic study of genus Teleogryllus from East Asia (Insecta: Orthoptera: Gryllidae)

        Crickets belonging to Teleogryllus spp. are widely distributed in East Asia and they are difficult to distinguish. In this study, a phylogenetic tree of Teleogryllus is reconstructed by COI gene, and all species can be distinguished well. The possible evolutionary histories of male genitalia, songs and life history are discussed. The result supports establishing subgenera Macroteleogryllus and Brachyteleogryllus of Teleogryllus spp. from East Asia. T. boninensis is moved to Brachyteleogryllus. The calling song of four species includes both long and short chirps, and the rest just have single chirps. The species inhabit sympatrically trend to have different type of calling song. Egg overwinter type evolved several times independently.

        Heterologous overexpression of the Arabidopsis SnRK2.8 gene enhances drought and salt tolerance in Populus × euramericana cv ‘Nanlin895’

        The SnRK (sucrose non-fermenting-1 related protein kinase) families respond to salt tolerance, nutritional stress, disease stress, and carbon metabolic regulation. In this study, the SnRK2.8 gene was cloned from Arabidopsis thaliana and nominated AtSnRK2.8. Bioinformatics characterization of AtSnRK2.8 showed that it contained an ATP binding site and a serine/ threonine protein kinase binding site at the N-terminal catalytic domain. In addition, the AtSnRK2.8 protein contained special domain I at the C-terminal catalytic domain. Real-time polymerase chain reaction (PCR) results showed that the AtSnRK2.8 gene was expressed in all tested tissues and was highly expressed in seeds. Our analyses proved that the AtSnRK2.8 gene expression was upregulated by salt and drought stresses. The recombinant AtSnRK2.8 protein was expressed using a pET-28a system and purified with HisTrap HP affinity columns. The recombinant AtSnRK2.8 protein has the properties of a protein kinase. The results of overexpressing AtSnRK2.8 in Nanlin895 poplar showed that the transgenic poplar plants had a better ability to resist drought and salt stress compared to the wild type, and adapted well to drought and a high-salt environment.

        Bovine Viral Diarrhea Virus Infection Induces Autophagy in MDBK Cells

        Bovine viral diarrhea virus (BVDV) is an enveloped, positive-sense, single-stranded RNA virus that belongs to the genus Pestivirus (Flaviviridae). The signaling pathways and levels of signaling molecules are altered in Madin-Darby Bovine Kidney (MDBK) cells infected with BVDV. Autophagy is a conservative biological degradation pathway that mainly eliminates and degrades damaged or superfluous organelles and macromolecular complexes for intracellular recycling in eukaryotic cells. Autophagy can also be induced as an effective response to maintain cellular homeostasis in response to different stresses, such as nutrient or growth factor deprivation, hypoxia, reactive oxygen species exposure and pathogen infection. However, the effects of BVDV infection on autophagy inMDBK cells remain unclear. Therefore, we performed an analysis of autophagic activity after BVDV NADL infection using real-time PCR, electron microscopy, laser confocal microscopy, and Western blotting analysis. The results demonstrated that BVDV NADL infection increased autophagic activity and significantly elevated the expression levels of the autophagy-related genes Beclin1 and ATG14 inMDBK cells. However, the knockdown of Beclin1 and ATG14 by RNA interference (RNAi) did not affect BVDV NADL infection-related autophagic activity. These findings provided a novel perspective to elaborate the effects of viral infection on the host cells.

        Tunnel Lining Stability and Structural Damage Threshold under Seismic Loading

        To analyze the damage distribution characteristics and ultimate bearing capacity of tunnel linings under seismic action, damage variables were introduced into the Mohr–Coulomb constitutive model. Thus, an incremental elastic–plastic damage constitutive model for concretewas constructed. Uniaxial tensile and compressive tests were performed to verify model reliability, and the damage thresholds for different strength classes of concrete under the test conditions were numerically obtained when destruction occurred. These damage thresholds were considered as the criteria for material failure to solve the ultimate bearing capacity and failure form of the tunnel structure. Calculation results demonstrate that as the peak ground acceleration increases, the shallowly buried tunnel structure presents the three following stages: elastic bearing, local instability, and overall instability. If the compression or tensile damage values of an element exceed the threshold value, the element is regarded as destroyed. The failure of any element of a structure can be used as a criterion for local instability. If the volume of the destroyed elements exceeds 10% of the overall volume of elements, and the proportion of destroyed elements in the lining shoulders and side walls begins to abruptly increase, the structure can generally be regarded as unstable.

        Phenolic composition and nutritional attributes of diaphragma juglandis fructus and shell of walnut (Juglans regia L.)

        Phenolic composition and nutritional attributes of diaphragma juglandis fructus (Djf) and walnut shells (Ws) were investigated. Phenolic acids, hydroxybenzoic acid, isoflavone, and flavone were identified in the free phenolic fractions (FPFs) of both Djf and Ws. Bound phenolic fractions were less than FPFs both in content and diversity. The soluble dietary fiber contents of Djf and Ws were 25.56 g/100 g and 9.5 g/100 g, respectively. The contents of unsaturated fatty acids (1912.28 mg/kg and 9137.56 mg/kg, respectively) were significantly higher than that of saturated fatty acid both in Djf and Ws. The content of essential amino acids in Djf (9.67 mg/g) was significantly higher than that in Ws. More than eight types of monosaccharides were detected in Djf and Ws. The monosaccharide content of Djf (314.16 mg/g) was significantly higher than that of Ws (60.97 mg/g). Trehalose was the predominant component both in Djf (71.2%) and Ws (78.6%).

        Omentin-1 ameliorates oxidative stress in model of diabetic ophthalmopathy via the promotion of AMPK function

        Background Omentin-1 is a new type of adipokines. Objective To study that the effects of Omentin-1 on diabetic ophthalmopathy and its mechanism. Results Omentin-1 expression level in serum of patients with diabetic ophthalmopathy was reduced. In mice with diabetic ophthalmopathy, Omentin-1 mRNA and protein expressions in ocular tissue were suppressed. Furthermore, Omentin-1 presents oxidative stress and ophthalmopathy in the mice model of diabetes. In vitro model of high glucose-induced ophthalmopathy, Omentin-1 reduced oxidative stress and promoted its cell damage, and si-Omentin-1 promoted oxidative stress and protect cells activity. Omentin-1 induced p-AMPK protein expression and reduced ubiquitination of AMPK levels. AMPK inhibitor reversed the function of Omentin-1 on oxidative stress and cells activity in vitro model of high glucose-induced ophthalmopathy. On the other hand, AMPK agonist also reduced the function of si-Omentin-1 on oxidative stress and cells activity. Conclusion These findings indicate that Omentin-1 Ameliorates oxidative stress in the model of diabetic ophthalmopathy via the promotion of AMPK function.

        Impact of Dissection after Drug-Coated Balloon Treatment of De Novo Coronary Lesions: Angiographic and Clinical Outcomes

        Purpose: Dissection after plain balloon angioplasty is required to achieve adequate luminal area; however, it is associated with ahigh risk of vascular events. This study aimed to examine the relationship between non-flow limiting coronary dissections andsubsequent lumen loss and long-term clinical outcomes following successful drug-coated balloon (DCB) treatment of de novocoronary lesions. Materials and Methods: A total of 227 patients with good distal flow (Thrombolysis in Myocardial Infarction flow grade 3) followingDCB treatment were retrospectively enrolled and stratified according to the presence or absence of a non-flow limiting dissection. The primary endpoint was late lumen loss (LLL) at 6-month angiography, and the secondary endpoint was target vesselfailure (TVF, a composite of cardiac death, target vessel myocardial infarction, target vessel revascularization, and target vesselthrombosis). Results: The cohort consisted of 95 patients with and 132 patients without a dissection. There were no between-group differencesin LLL (90.8%) returning for angiography at 6 months (0.05±0.19 mm in non-dissection and 0.05±0.30 mm in dissection group, p=0.886) or in TVF (6.8% in non-dissection and 8.4% in dissection group, p=0.799) at a median follow-up of 3.4 years. In a multivariateanalysis, the presence of dissection and its severity were not associated with LLL or TVF. Almost dissections (93.9%) were completelyhealed, and there was no newly developed dissection at 6-month angiography. Conclusion: The presence of a dissection following successful DCB treatment of a de novo coronary lesion may not be associatedwith an increased risk of LLL or TVF (Impact of Drug-coated Balloon Treatment in de Novo Coronary Lesion; NCT04619277).

        Evaluating the synergistic effect of maple leaves extract and iodide ions on corrosion inhibition of Q235 steel in H2SO4 solution

        In recent years, many plant extracts have been used as green corrosion inhibitors to replace traditionalcorrosion inhibitors, but their inhibition performance still needs to be improved. The synergistic corrosioninhibition behavior and mechanism of maple leaves extract (MLE) and potassium iodide (KI) onQ235 steel in 0.5 M H2SO4 solution were firstly investigated in this work. The results demonstrate thatboth MLE and KI can prevent the corrosion of steel in corrosive media, and that their combined protectioneffect is superior. The corrosion inhibition efficiency (g) value reaches 93.4 % in corrosive solution withthe combination of 200 mg/L MLE and 200 mg/L KI, while the maximum of g is only 81.6 % using MLEalone. In addition, Fe-N bond and Fe-O bond were found on the steel surface through the X-ray photoelectronspectroscopy (XPS) analysis, which confirm the formation of a protective film. The adsorption behaviorof an inhibitory film composed of MLE and KI mixtures onto steel surface follows the Langmuiradsorption isotherm model, consisting of physisorption and chemisorption. Finally, molecular dynamics(MD) simulation indicates that inhibitor molecules can adsorb on the Fe substrate in a parallel way withhigh binding energy values, which supports the experiments well.
