        Production and Characterization of Enzymes Involved in Chitin Catabolic Cascade from a Bacterial Strain Isolated from Soil

        The present study was aimed to isolate the bacteria producing chitinolytic enzymes including chitin deacetylase (CDA) from soil origin. About 24 bacteria were screened for their chitinolytic enzymes producing ability on the basis of colloidal chitin. Amongst one of the most potent isolates designated to JI-02 was further selected as the most prominent chitinolytic and CDAlike enzyme (CDA-X) producer based on the ability of deacetylation of an artificial substrate pNP-acetamilide. The maximum production of CDA-X in crude enzyme from JI-02 was observed in the presence of 1% colloidal chitin at 37°C and pH 6.4 after 120 h of incubation. Potent enzyme activity of CDA-X was determined toward pNP-acetamilide, demonstrating that optimal pH, temperature and ionic strength of the CDA-X were assessed to be 7.0, 52°C and 125 mM, respectively. Unexpectedly, Nacetylglucosamine (GlcNAc) was not fully converted by the action of CDA-X to glucosamine (GlcN) under the established conditions. Further study on enzyme activity toward chitin-oligosaccharides consisting of multi-N-acetylglucosamine (GlcNAc)n, n=2-5 may be necessary to elucidate the mode of action which requires the minimum size of (GlcNAc)n. However, our data suggest that CDA-X can convert chitin to chitosan with the maximum yield of approximately 0.08g/L during the fermentation of the strain in the presence of chitin. Furthermore, we found that crude enzyme showed the significant chitosanolytic activity. Taken together, our data suggest that the strain JI-02 may be a potential candidate as a particular strain for better understating of chitin catabolic pathway.

        This study attempted to determine the degree of asbestos release from the aging slate roof by comparing the asbestos content in the slate roof rain gutter with a colored steel plate (tin plate) over the slate roof. Four slate roof houses located in Haengmok-ri, Asan-si, Chungcheongnam-do, and one house constructed with a colored steel plate on the slate roof were selected to collect the sediment of the roof rain gutter. The asbestos fiber content was calculated by a point counting method using a polarization microscope after pretreatment with conversion treatment and hydrochloric acid treatment. The average asbestos content of the four slate roof rain gutter were 1.89%. However, asbestos was not detected in the Slate covering roof rain gutter, which were constructed on the slate roof. Asbestos fiber content was the highest at 2.89% in the slate roof rain gutter installed in 1976, followed by 2.44% in 1953. From the above results, it is necessary to minimize secondary damage as asbestos fibers released from slate roof houses to the surrounding atmosphere or leaked from slate roofs as rainwater may cause soil pollution and seriously affect residents' health. Although covering with colored steel plates (tin plates) has been shown to prevent the leakage of asbestos fibers to some extent, it is believed that a policy alternative to remove the slate roof as soon as possible is needed to solve the fundamental problem.

      • 혈액투석 환자에게서 발생한 재생불량성 빈혈 1예

        Aplastic anemia is a disease characterized by pancytopenia in the peripheral blood and hypocellular bone marrow with the fatty infiltration. The causes of the acquired aplastic anemia were usually related to drugs and immune-related diseases. A 34-years old man was admitted due to general weakness. He has been going hemodialysis for 2 years because of end stage renal disease (ESRD) due to chronic glomerulonephritis. At admission, laboratory tests showed hemoglobin 6.2 g/dL, hematocrit 18.0 %, WBC 5,710mm (neutrophil 71.8%, lymphocyte 19.1%, monocyte 5.9%), reticulocyte 1.9%, platelet 93,000/mm, Fe 12.8 ug/dL, TIBC 204 ug/dL, ferritin 941.47 ng /ml, haptoglobin 0.72 g/L, vitamin B12 508.17 pmol/L, folate 24 ng/mL, total protein 6.54 g/dL, albumin 3.76 g/dL, Alk. pohsphatase 79 IU/L, AST 30.0 IU/L, ALT 39.7 IU/L, total bilirubin 0.57 mg/dl, BUN 79.4 mg/dl and creatinine 10.18 mg/dl. Peripheral blood smear showed the normocytic normochromic anemia with anisocytosis. Anemia was diagnosed but it was irresponsive to the recombinant human erythropoietin (EPO) and blood transfusion. In the bone marrow aspiration smear were there the hypoplastic myeloid and erythroid precursors. The bone marrow biopsy section showed the hypoplasia of all components (10-20%) and the fatty infiltration. We have experienced one case of the idiopathic aplastic anemia in a patient going through hemodialysis and we just report it with documentary records. 저자들은 혈액 투석 중인 환자에서 약물, 면역계 이상등의 원인을 발견할 수 없었음에도 재생 불량성 빈혈이 발생한 1예를 경험하여 문헌 고찰과 함께 보고하는 바이다.

      • 鹿茸 藥針 製劑가 힌쥐 腎臟 組織의 抗酸化 作用에 미치는 影響

        Cervus elaphus for herb-acupuncture solution (CEHAS) was tested for the effects of free radical generating enzyme and lipid peroxidation in rat's kidney. In vitro, levels of lipid peroxide in tissues of kidney were proportionally decreased to concentration of extracts prepared from CEHAS. They were much more decreased, when lipid perocidation was inducesd with ferrous iron(Fe II). Also, enzyme activities of xanthine oxidase were decreased. The ratio of type conversion of zanthine oxidase was lowered, too. But, it was not seen changes on enzyme activities of aldehyde oxidase. These results suggest that CEHAS decrease the activities of free radical generating enzymes such as xanthine oxidase which form lipid peroxide.

      • 枇杷葉의 抗糖尿病藥理活性과 IL-1β誘發糖尿病 마우스의 膵臟 燐酸化酵素에 미치는 效果에 關한 硏究

        淸凉生津, 解渴上嘔의 효능이 있으며, 한국등에서 항당뇨병성 한약으로서 사용되어 온 비파엽의 열수출물의 당뇨병치료효과를 검증하기 위하여 alloxan으로 유발된 당뇨병흰쥐의 혈장포도당농도 및 인슈린농도를 측정하였다. 비파엽처리군의 혈장 포도당농도는 현저히 증가하였으며 혈장인슈린농도는 크게 감소하지 않았다. 한편, 당뇨병유발의 내재적인 요소로 알려진 사이토카안인 IL-1β로 당뇨병을 유발한 뒤 그 효과를 검정했다. 즉, 실험동물을 interleukin-1β로 처리하여 실험적으로 당뇨병을 유발한 후 glucose의 燐酸化에 관여하는 glutokinase와 hexokinase의 활성을 살펴보았다. 그 결과, 비파엽엑기스는 interleukin-1β 과잉투여 당뇨병으로 인하여 증가된 혈액내 glucose농도를 감소시키고 insulin 과잉분비를 정상으로 회복시키는데 효과적이었다. 동시에 lL-1β에 의해 유도된 당뇨마우스에 대한 枇杷葉엑기스 投與가 췌장 glucokinase와 hexokinase의 활성을 촉진하는 것으로 나타나 lL-1β유발 당뇨병에 효과가 있는 것으로 사료되었다. Studies were conducted on anti-dibetic effect of the water extract from leaves of Eriobotryae folium which had been used in Korea as a remedy for dibetes. The extract was found to inhibit the Increase in the plasma level of sugar bu the not the decrease in the plasma level of insulin in alloxan-induced dibetic rata. Also, we investigated the in vivo effect of an aqueous extract(referred to as EF) from Eriobotryae folium on glucokinase and hexokinase activities of diabetes mellilus induced by Interteukin-1β (IL-1β). After 1 week of IL-1β injection, the levels of serum glucose concentration and insulin secretion were dramatically increased. However, the insulin secretion was decreased with administration of EF. The level of glucose concentration was decreased by EF administration. Furthermore, it was observed that EF was effective in recovering the levels of insulin secretion. Enzyme activities of the glucokinase and hexokinase, which are key enzymes of glucose phosphorylastion, were decreased by IL-1β. EF administration to the mice allowed proportional increasing by stimulation of induction of enzyme activities as high as normal group. These results suggested that EF is highly effective in treatment of diabetes mellitus induce by IL-1β.

      • Saccharomyces cerevisiae에 존재하는 GPI결합 세포벽 단백질에 대한 연구

        The cell wall of Saccharomyces cerevisiae is an elastic structure that provides osmotic and physical protection and determines the shape of the cell. The cell wall of S. cerevisiae is made up of three components: glucan, mannoproteins, and chitin. The former two are the major components of the cell wall. The glucanase-extractable mannoproteins are covalently bound to β-1,6-glucan and divided into two main classes of mannoproteins, GPI-CWPs and Pir-CWPs. In yeast, threre are two groups of GPI proteins: plasma membrane-resident proteins, such as Gaslp or Yap3p, and cell wall-targeted proteins, such as Tirlp or a-agglutinon are exist. GPI proteins have three characteristics in common: a signal sequence for secretion at the N-terminus, a serine/threonine-rich sequence which provides sites for glycosyation, and a C-terminal GPI-attachment signal. In GPI-attached proteins, there are two differen signals for determining their cellular localization. These are the amino acid residues V,I, or L at the -4/5 site and Y or N at the -2 site and the basic residue(s) R and/or K in the short-minus rehion. The former residues act as a positive factor for cell wall incorporation, while the later residues act as a negative factor for incorporation, such that the proteins remain on the plasma membrane as GPI-anchored proteins. Amino acid sequence of the GPI-attachment site of yeast slightly differs from that of mammalian cells. This might be of use in the development of anti-fungal drugs and the cell wall targeting of the foreign proteins.

      • 토양 미생물인 Streptomyces tubercidicus에서 분리한 GTPcyclohydrolase Ⅰ 저해제

        GTP cyclohydrolase I (EC 3.5.4.16) catalyzes the conversion of GTP to D-erythro-7,8-dihydroneopterin triphosphates and formic acid. and thus performs the first committing step in the biosynthesis of the pteridine moiety of folk acid in microorganisms and of tetrahydrobiopterin in higher animals. GTP cyclohydrolase I isolated from Streptomyces tubercidicus was inhibited by DL-6-methyl-5,6,7,8-tetrahydropterin and xanthopterin, and was not inhibited by tetrahydrofolic acid, pterin, pterin-6-carboxylic acid, biopterin, neopterin, ribofravin. These results suggest that bacterial GTP cyclohydrolase I may be regulated by its metaboilic end product.

      • 정신분열병 환자의 인지기능 상태

        목적: 정신분열병 환자의 적응 상태와 증상 형성에 영향을 주는 것으로 생각되는 인지기능 상태를 주의력, 형태 인식, 단어에 대한 구별과 종합 기능을 평가해봄으로써 알아보고자 하였다 방법: 정신분열병 환자 27명과 대조군 27명을 대상으로 하였고, 평가 도구로써 주의력은 K-WAIS의 숫자외우기 문제로, 형태 인식은 K-WAIS의 모양 맞추기 문제와 숨은 그림 찾기로, 단어 기능은 K-WAIS의 공통성 문제와 단어 차이 검사로 각각 이루어졌다. 통계 처리는 SPSS 11.5를 이용하여 Chi-Square test와 Student's t-test로 검증을 하였다. 결과: 정신분열군은 숫자외우기, 숨은 그림 찾기, 모양 맞추기에서는 대조군보다 낮은 점수를 보였다. 단어 차이 검사와 공통성 문제에서는 양군에서 차이를 보이지 않았다. 결론: 본 연구에서 정신분열군의 인지 기능은 정상인 대조군에 비해 주의력과 형태 인식 기능이 유의하게 떨어져 있었으며, 단어들을 구분하고 종합하는 능력은 차이가 없었다. 이런 결과는 임상적으로 정신분열병에서 보이는 망상이 언어적인 측면에서는 어느 정도 전달이 되는 현상과 일치하지만 주의나 형태인지와 같은 기본적인 과정에서 장애가 있다는 추정을 하게 한다. Purpose: This study aimed to investigate the cognitive function in patients with schizophrenia by assessing attention, recognition of a shape, and verbal function. Methods: The subjects were 27 patients with schizophrenia group and 27 control group. Attention was assessed by the digit span task(K-WAIS subtest). The recognition of a shape was assessed by the object assembly task(K-WAIS subtest) and hidden figure task. Verbal function was assessed by Similiarities(K-WAIS subtest) and the different word task. Analysis was performed by the Chi-Square test and Student's t-test via SPSS 11.5. Result: Schizophrenia group had statistically lower scores than control group in digital span, hidden figure and object assembly. There was no difference between two groups in dimensions of difference in words and similarity. Conclusion: This study showed that patients with schizophrenia had impairments in attention and recognition of shape, and no difference between two groups in verbal ability. These explained the partial communicative function of delusion in verbal level. The formation of delusion might be related to the more basic elements such as attention and recognition of shape.

        정신분열병 환자에서 Risperidone에 의해 유발된 조증 2례

        저자들은 정 신분열병 환자에서 risperidone에 의해 유발된 조증, 경조증 각 1례를 경험하였기에 국내에서는 최초로. 문헌고찰과 함께 보고한다. 증례 1은 22세의 여자 긴장형 정신분열병 환자로 3년전 발병 당시 약물치료후 유지요법 없이 지내다가 4∼5개월 전부터 사회적 위축, 식사량 저하. 말수가 줄어드는 등의 음성증상이 심해져 외래에서 risperidone 2mg을 일 주일간 처방하였다. 환자는 약물순응도가 낮아서 모두 6회에 걸쳐 억지로 투약하여 이 기간에 총 6∼8mg복용후 2∼3일 만에 조증이 발생되었다. 입원후 risperidone을 중단하고 lithium과 고용량의 정형 항정신병약물(chlorpromazine 1200mg 또는 haloperidol 20mg)을 10일간 투여하여 일부 조증이 호전되었다. 증례 2는 29세 남자로 3년 전부터 피해망상, 독극망상을 보이는 망상형 정신분열 병으로 항정신병약물을 처음 투여한 환자였다. 38일간 2mg 사용 시에는 양성증상 및 음성증상의 호전을 보이다가 3mg으로 증량된 후 일 주일만에 뚜렷한 경조증을 보였다. Risperidone을 중단하고 chlorpromazine 300mg을 투여하자 5일 만에 경조증이 완전히 회복되었다. 두 예 모두 risperidone 단독투여에 의한 것이었고, 기분안정제 병합투여, 약물남용 정신질환의 병력이나 가족력, 동반 신체질환은 없었다. Risperidone의 세로토닌(5-HT₂) 차단효과는 도파민(D₂) 차단작용과 함께 정신분열 병에서 음성증상과 정동중상을 호전시킬 뿐 아니라 항우울효과를 나타내며. 일부 환자에서 조증 및 경조증을 유발시키는 것으로 보인다. 기분장애와 분열정동장애에서도 risperidone에 의해 조증이 유발 또는 기존 조증중상이 악화되었다는 증례보고가 있고. 강박장애 등 다른 정신질환으로도 적응증이 확대되고 있으므로, 임상의들은 드물지만 이런 정신과적 부작용의 발생 가능성에 대해 알아야 하겠고. 특히 기분장애나 분열정동장애에서 사용할 경우 기분안정제의 병합투여와 최소 치료용량으로 서서히 증량할 것을 권하는 바이다. 또한 조증유발의 위험인자가 있는 경우는 기분안정효과가 있는 clozapine의 사용이 권장된다. We report the first two cases of manic and hypomanic episodes respectively induced by risperidone treatment done to schizophrenics in Korea. One case was a 22-year-old woman with catatonic schizophrenia. Since 3 years ago, she had shown psychotic symptoms, but with was poor treatment compliance. She had mainly negative symptoms such as social withdrawal, decreased food intake, mutism, and symptoms had been worsened since last 4-5 months. Prior to closed ward admission, she was prescribed 2mg/d of risperidone for a week at OPD. Two days artier taking medicine totally 6-8mg, she revealed manic features. After hospitalization, risperidone was discontinued and then, lithium 900mg/d and high dosage of conventional antipsychotics(chlorpromazine 1200mg/d or haloperidol 20mg/d) were prescribed . About on the l0th day of hospitalization, there was limited improvement of her manic symptoms. The other case was a 29-year-old man with a 3-year history of paranoid schizophrenia. He was never exposed to antipsychotics before. His main symptoms were delusions of being poisoned and of persecution. His positive and also negative symptoms were alleviated by 38 days of risperidone 2mg/d trial. However, one week after dosage increment to 3mg/d, hypomanic symptoms appeared. Risperidone medication was discontinued and was replaced by chlorpromazine 300mg/d. The hypomanic episode was resolved over 5 days. In both of the two cases, manic episodes occurred by monotherapy of risperidone without mood stabilizer, and there were no history of substance abuse and other psychiatric disorders, family history of psychiatric disorders, and comorbid physical illnesses. It is hypothesized that the potent blockade effect on serotonin(5-HT₂) receptor of risperidone causes antidepressant effect as well as therapeutic effect for negative and affective symptoms in schizophrenia. Risperidone would induce manic or hypomanic features in schizophrenic patin of preexisting manic symptoms by risperidone treatment in mood disorder and schizoaffective disorder. Risperidone is being used more widely, even for obsessive-compulsive disorder and other psychiatric disorders. It is necessary for clinicians to recognize manic switch, one of psychiatric side effects by risperidon trial. It is recommended that the combination of mood stabilizer wish risperidone or usage of talc minimum effective dose of risperidone may bewefal especially in the patients with mood disorders or schizoaffective disorders. Clozapine which has mood-stabilizing properties is also beneficial in risk groups of risperidone-induced mania.

        Background: Healthy and sustainable community is essential component of development. But there has been no systematic reporting about healthy municipality in specific city, although economic development results in considerable progress. Method: According to the Ontario progress monitoring tool partial monitoring has been done. Survey to the stakeholder including public officers were done and analyzed. Result: Economic area were most lowly reported area. By specific item, direction of improvement were induced in each area including social environment and human health area. Green space, waste treatment, traffic problem, small health service benefit were major agenda among the stockholders. Conclusion: According to the Ontario progress reporting, we got good picture about healthiness of one municipality and concrete direction of improvement.
