      • Ascorbate Peroxidase 유전자의 도입에 의한 식물의 형질전환

        This study was conducted to obtain the transformed tobacco (Nicotiana tubacum) plants with cytosolic ascorbate peroxidase gene(ApxSC7) using Agrobacterium tumefaciens LBA4404. A cDNA encoding the cytosolic ascorbate peroxidase of strawberry, ApxSC7, was introduced into tobacco plants via Agrobacterium-mediated gene transfer system. The expression vector, pIG-AP8, harboring ApxSC7 gene was used for production of transgenic tobacco plants. A large number of transgenic plants were regenerated on a medium containing hygromycin. Integration of ApxSC7 gene was confirmed by PCR and Southern blot analyses with genomic DNA. Northern blot analyses revealed that the pIGap8 gene was constitutively expressed.

        요리활동을 통한 발달지체 유아의 주의집중 및 변별력 변화

        본 연구는 발달지체 유아들에게서 공통적으로 낮게 발견되는 특성 중의 하나인 주의집중 및 변별력의 향상을 위하여 시도되었다. 이를 위하여 무엇보다도 유아들이 흥미를 가지는 매체를 선정하는 것이 중요하였으며, 선행연구의 검토를 통하여 이것은 요리활동으로 선정되었다. 즉, 본 연구의 목적은 요리활동을 통하여 발달지체유아들의 주의집중 및 변별력의 긍정적 변화를 확인하는 것이다. 연구대상으로 PEP에 의하여 발달지체를 나타내는 유아 4명을 선정하였고, 독립 및 종속변인(주의집중)의 기능적 관계를 확인하기 위하여 중다기초선설계를 이용하였으며, 또 다른 종속변인(변별력)의 변화는 프로그램 적용의 전후검사를 이용하여 확인하였다. 연구결과, 4명의 연구대상은 프로그램의 적용에 따라 급격한 주의집중 행동변화를 보임으로써 요리활동과 주의집중 행동의 기능적 관계를 확인할 수 있었다. 변별력은 4명 모두 상승된 점수를 나타내었는데 그중 유아 B가 가장 큰 상승을 나타내었다. 본 연구를 통하여 요리활동이 발달지체 유아들의 주의집중 행동을 긍정적으로 변화시킬 수 있음이 확인되었다. 그러나 향상된 주의집중 행동의 일반화 여부는 확인되지 않았으므로 이에 대한 후속 중재 및 연구가 필요하리라고 본다. 또한 변별력은 전후검사를 통하여 확인된 것이므로 미세한 행동의 변화를 측정할 수 없었음이 고려되어 해석되어야 할 것이다. The purpose of this study was to explore the effects of a cooking program on the sustained attention and the visual discrimination of children with developmental delays. The subjects for this study were 4 children with developmental delays enrolled in special kindergarten G, who scored below 50 developmental quotient on the Psycho-educational Test. Multiple Baseline Design Across Subjects (MBDAS) was used to determine the effects of a cooking program on the sustained attention and discrimination of children with developmental delays. In this study, MBDAS was performed on sustained attention while the children were playing house. The 8 types of cooking intervention program was conducted a total of 15 sessions. It was included while the children were playing house for duration of 30 to 40 minutes everyday. The data collection for sustained attention was completed every 10 seconds intervals for 10 minutes. The results of this experiment indicate that the cooking intervention program was very effective in increasing sustained attention. Before and after the intervention program, a visual discrimination test was performed to explore the effects of the cooking activity on discrimination, for children with developmental delays. The contents of the discrimination test sheet were extracted from an existing test sheet and reorganized by the researcher. The results of this test demonstrate that the cooking program positively influenced children with developmental delays in regards to discrimination and development of concepts. In summary, the cooking program proved to be effective in increasing sustained attention in children with developmental delays. The cooking program also positively influenced discrimination, specifically color, shape, and classification discrimination. The results of this study suggest a need for developing an interesting, specific, well-structured curriculum for children with developmental delays, which emphasized a variety of learning programs integrated within a cooking course.

      • 식사관리와 영양지식에 관한 연구

        The purpose of this study was to investigate meal management and nutrition knowledge of the housewives according to their region, age, educational level, income and occupation. The subjects consisted of 2082 housewives residing in big city, middle or small city and agricultural and fishing villages. The portion of housewives living in the middle or small city was 44.1% and the major portion of age distribution was 30's(34.2%). More than half of the housewives(58.5%) had a job. Meal management included in this sutdy was their attitude for meal, nutritional balance in their dishes and harmonizing nutrition knowledge in the life. The meal attitude of the housewives who had no job was the lowest score. Dishes arrangement and harmonizing nutrition knowledge in the life were shown the highest score at the age of 60's. They had experience of nutritional education(63.8% ), The source of nutritional information were in order of TV., radio, relatives & neighbors, newspapers and magazines. The average score of nutrition knowledge was 7.07 out of possible 10 point. Nutrition knowledge had significant correlation with region, age, educational level and income. With increasing educational level, income and decreasing age, the score of nutrition knowledge was getting higher. Also, the housewives of urban was the higher in the nutrition knowledge score than those of rural. The frequency of food intake appeared at the higher in fruits, milk & products than meats & fishes, fishes of eatable bone, yellow green vegetables, and frying and parching food etc.

      • 심전도에서 조기재분극을 보인 특발성 심실세동

        Early repolarization is a common electrocardiographic (ECG) feature found in young adults, men and athletes, and has been considered to be a benign feature for the last several decades. But recent studies suggest that early repolarization may be related to idiopathic ventricular fibrillation and sudden cardiac death. We report a young man, 35 years old, who had life threatening ventricular fibrillation and sudden cardiac arrest. He was evaluated for cardiac causes of ventricular fibrillation. There was no explanation other than that his ECG showed an early repolarization pattern so we treated him with implantable cardioverter defibrillator. Thus, we suggest that early repolarization may be related with life threatening ventricular arrhythmia.

      • Agrobacterium tumefaciens를 이용한 bialaphos 저항성 형질전환 벼의 개발

        비선택성 제초제인 bialaphos는 고등식물에 있어서 glutamine 합성을 억제하여 식물체를 고사시킨다. Acetyltransferase에 의해 encoding된 bialaphos 저항성 유전자는 세균 Streptomyces hygroscopicus SF1239로부터 cloning된 것을 사용하였다. Bialaphose 저항성 유전자를 Agrobacterium 감염법을 이용하여 국내에서 재배되는 벼(동진)에 도입한 결과 약 30%정도의 형질전환 식물체를 얻을 수 있었다. T_1세대의 17개체는 hygromycin과 bialaphos에 대한 저항성 유전형질이 3 : 1로 분리되었다. 또한 Southern 분석을 실시한 결과 wild type의 식물체에서는 Bar 유전자의 검출을 볼 수 없었으나 형질전환 식물체의 경우 Bar 유전자의 검출이 가능하였다. T_3 세대의 형질전환 식물체와 wild type의 식물체를 포장상태에서 비선택성 제초제인 바스타를 살포하고 3주 후에 관찰한 결과 형질전환 식물체는 외형적으로 아무런 피해를 받지 않고 정상적으로 생장하였으나, wild type의 식물체와 잡초는 모두 고사하였다. 이상의 결과를 종합해보면 hygromycin과 bialaphos 저항성 유전자는 Agrobacterium 감염법을 이용하여 단자엽 식물인 벼에 도입할 수 있다는 것을 보여준 것이며, 또한 bialaphos 저항성 유전자가 식물에 도입됨으로써 비선택성 제초제에 대한 저항성 식물을 개발 할 수 있다는 것을 보여 주었다. The bialaphos is a potent inhibitor of glutamine synthease in higher plants and is used as a non-selective herbicide. We have used the bialaphos resistant gene (Bar) encoding for an acetyltransferase isloated from Streptomyces hygroscopicus SF1293. Callus derived from mature seeds of rice (Oryza sativa L. cv. Dong Jin) were co-cultivated with Agrobacterium tumefacieus EHA101 carring a plasmid pGPTV-HB containing genes for hygromycin resistance (HygR) and Bar. Transgenic plants showing in vitro resistance to 50 mg/L hygromycin and 10 mg/L bialaphos were obtained by using a two-step selection/regeneration procedure. Transformation efficiency of rice was about 30% which was as high as reported in other dicotyledons. Progenies (T_1 generation) derived from primary transformant of 17 lines were segreagated with a 3 resistant:1 sensitive ratio in medium containing hygromycin and bialaphos. Stable integration of Bar gene into chromosomal DNA was proven by Southern blot analysis of genomic DNA isolated from T_2 progenies. Transgenic plants (T_3) grown in the field were resistant to bialaphos (Basta) at a dosage lethal to wild type plants.

        고령 장애인의 일(WORK) 경험과 욕구에 관한 연구 : 뇌병변 장애를 중심으로

        본 연구의 목적은 고령이면서 장애를 동시에 경험하고 있는 고령 장애인들의 일 경험과 욕구에 관한 특성을 당사자의 관점에서 이해하는 것이다. 이러한 목적을 달성하기 위해 50세 이상 뇌병변장애인 13명을 대상으로 일 경험과 욕구에 관한 개방형식의 질문으로 심층 인터뷰를 실시하여 그 결과를 분석함으로써 고령 장애인 당사자의 일 경험과 욕구에 대한 심층적 이해와 현상의 의미를 깊이 있게 파악하고자 하였다. 분석결과, 고령 뇌병변장애인들은 일을 하고자 하는 높은 욕구를 가지고 있으며 일을 통해 삶의 의미와 보람을 찾고자 하나 현실적으로는 장애와 고령, 그리고 만성질환자라는 삼중고 속에서 “일하기” 에 상당한 제약을 받는 것으로 나타났다. 이러한 연구결과는 지금까지 고용정책에서 소외되었던 고령 장애인의 ‘일’ 에 관한 정책적 개입의 필요성을 강력하게 시사해 주고 있다. The purpose of this study is to understand the characteristics of the experiences and needs for work of aged people with disabilities who experience both old age and disability from their viewpoint. To this end, this study conducted in-depth interviews with 13 people aged 50 or older with encephalopathic disabilities by open-ended questions regarding their experiences and needs for work, and analyzed the results so as to more deeply understand the experiences and needs for work of aged people with disabilities as well as the implications of the present situation. The analysis results showed that aged people with disabilities have high needs for work and try to find the meaning and worth of life through work, but in reality face considerable limitations to ‘working’ in the triple handicaps of disability, old age, and chronic disease. The results of this study strongly suggest the need for policy intervention in the ‘work’ of aged people with disabilities who have been neglected by employment policies until now.

      • 브랜딩·스타일링·마케팅을 결합한 스페이스 디자인 프로세스 연구 - 작품‘Fill the Red, Feel the Red'를 중심으로 -

        본 연구는 디자인 분야와 마케팅 분야의 전략을 종합하여 전략적 연결점을 찾아내 요소들 간 융합을 이루는 사례를 제시하여 브랜드·스페이스·마케팅에서 새로운 방향성과 아이디어를 제시하는데 목적을 둔다. 이에 본 연구는 2017년 9월~12월까지 ㈜애경과 산업 연계하여 진행된 숙명여대 환경디자인학과 ‘브 랜딩·스타일링·마케팅’ 교과목에서 도출된 ‘fill the red, feel the red’ 프로젝트를 마케팅 및 디자인 프로세스, 결과물을 도출하여 그 과정들을 밝힌다. 아울러 본 연구는 브랜드 스페이스 마케팅에 대한 이론적 고찰을 통해, 브랜드 스페이스(팝업스토어)를 개발 및 스타일링하며 ㈜애경의 화장품 브랜드 ‘LUNA’를 브 랜딩, 스타일, 마케팅적 측면에서 종합적으로 연계하여 도출되는 과정들을 제안함으로서 향후 브랜드 스페이스 디자인과 마케팅 분야에 체계적이고 창의적인 방법들로 결과물을 도출하는데 기초자료로 활용하는데 의의가 있다.

        마늘설기의 재료 배합비에 따른 관능적,텍스쳐 특성

        The purpose of this study is to determine the optimal mix ratio of Haircut-Sulgi and thereby, review its availability as health cake. For this purpose, the ratios of garlic juice and powder were varied with sugar added. As a result of quantitative descriptive analysis, it was found that the more the garlic juice and powder were added to non-glutinous rice, the Garlic taste was stronger. On the other hand, the less garlic powder was added, the cake would taste softer, and the less garlic juice was added, the cake was more chewy and moist. The t cling after swallowing was best when the ratio of garlic juice was 7%. As a consequence of surveying the tastes of Maneul-Sulgi, it was foetid that the less garlic powder was added to non-glutinous rice, the cake was more preferred. It was perceived that the ratio of garlic juice should be 7% for flavor and desirable taste. As a result of testing the mechanic characteristics, it was found that the less garlic juice was used, the cake was more hard, elastic, cohesive, viscose and chewy. The more garlic powder was used, the Adhesiveness was higher. The Overall acceptability of sensory examination for Garlic taste had positive correlation 'Hardness of sensory examination and mechanical examination for adhesiveness.
