        A Synthetic Naringenin Derivative, 5-Hydroxy-7,4′-diacetyloxyflavanone-<i>N</i>-phenyl Hydrazone (N101-43), Induces Apoptosis through Up-regulation of Fas/FasL Expression and Inhibition of PI3K/Akt Signaling Pathways in Non-Small-Cell Lung Cancer Cells

        <P>Naringenin, a well-known naturally occurring flavonone, demonstrates cytotoxicity in a variety of human cancer cell lines; its inhibitory effects on tumor growth have spurred interest in its therapeutic application. In this study, naringenin was derivatized to produce more effective small-molecule inhibitors of cancer cell proliferation, and the anticancer effects of its derivative, 5-hydroxy-7,4′-diacetyloxyflavanone-<I>N</I>-phenyl hydrazone (N101-43), in non-small-cell lung cancer (NSCLC) cell lines NCI-H460, A549, and NCI-H1299 were investigated. Naringenin itself possesses no cytotoxicity against lung cancer cells. In contrast, N101-43 inhibits proliferation of both NCI-H460 and A549 cell lines; this capacity is lost in p53-lacking NCI-H1299 cells. N101-43 induces apoptosis via sub-G<SUB>1</SUB> cell-cycle arrest in NCI-H460 and via G<SUB>0</SUB>/G<SUB>1</SUB> arrest in A549 cells. Expression of apoptosis and cell-cycle regulatory factors is altered: Cyclins A and D1 and phospho-pRb are down-regulated, but expression of CDK inhibitors such as p21, p27, and p53 is enhanced by N101-43 treatment; N101-43 also increases expression levels of the extrinsic death receptor Fas and its binding partner FasL. Furthermore, N101-43 treatment diminishes levels of cell survival factors such as PI3K and p-Akt dose-dependently, and N101-43 additionally induces cleavage of the pro-apoptotic factors caspase-3, caspase-8, and poly ADP-ribose polymerase (PARP). Cumulatively, these investigations show that the naringenin derivative N101-43 induces apoptosis via up-regulation of Fas/FasL expression, activation of caspase cascades, and inhibition of PI3K/Akt survival signaling pathways in NCI-H460 and A549 cells. In conclusion, these data indicate that N101-43 may have potential as an anticancer agent in NSCLC.</P>

      • Morphological Characteristics and Karyotypic Analysis of Aster spathulifolius According to Native Area

        The growth charateristics and karyotypes of Aster spathulifolius collected from 5 sites including coastal and island region on the Korean peninsula, were analysed. Several morphological characteristics of the plants such as leaf length, leaf width, top internode, medium internode, spike branching, flower diameter, number of petal, leaf color, leaf form, stem and leaf hair, viscosity, and serration of the plants were distinctly different depending on the native region from which they were collected. Karyotypic analysis showed that the chromosome number was all diploid (2n=18), with one pair of submetacentric satellite chromosomes. The chromosome composition included 7 pairs of metacentric chromosomes and 2 pairs of submetacentric chromosomes in all plants. However, chromosome order and the ranges of the chromosome lengths were a little different from plant to plant according to their native growing regions. The plants from Geoje-Do especially showed large differences in the chromosome lengths between the longest and the shortest compared to the plants from other places. This results provide important data to support the classification of the species into several sub-species.

        라식 혹은 라섹 후 근시퇴행 시 안압하강제와 0.1% 플루오로메토론의 병합치료 효과

        목적: 라식 라섹 후 근시퇴행으로 인해 시력저하를 호소하는 환자에서 안압하강제와 0.1% 플루오로메토론을 함께 사용하여 약물치료를 시행하였을 때 시력과 굴절값에 미치는 효과를 알아보고자 하였다. 대상과 방법: 본원에서 라식 혹은 라섹 수술을 받은 환자 중 2015년 1월부터 2016년 1월까지 시력저하로 내원하여 근시퇴행으로 진단받고 약물치료를 받은 환자 155명(155안)을 대상으로 약물치료 전후의 시력 및 굴절이상을 비교하고, 라식 라섹에서 그 결과가 차이있는지 알아보았다. 결과: 약물치료 시작 시기는 수술 후 평균 64.1 ± 36.8개월 후였다. 약물치료 후 시력이 호전되고 근시량이 감소한 약물치료 반응군은 63명(41%)이었으며, 이들에서 시력 호전은 -0.21 ± 0.11 logMAR, 근시량의 감소는 0.56 ± 0.32디옵터였다. 약물치료 반응군 중 완전히 회복되는 경우는 24명(15%), 부분적으로 회복된 경우는 39명(26%)이었다. 약물치료에 시력이 호전되는 빈도는 라섹 후보다 라식 후에 더 높았으나 통계적으로 유의하지는 않았다. 결론: 라식 라섹 후 근시퇴행으로 인한 시력저하 시 안압 하강제와 0.1% 플루오로메토론을 함께 사용한 약물치료는 약 41% 환자에서 시력을 호전시킬 수 있는 방법이며, 라식, 라섹 후 모두에서 효과적인 방법이다. Purpose: To evaluate the effect of combined medical treatment with anti-glaucoma eyedrops and 0.1% fluorometholone on visual acuity and refractive errors in patients complaining of blurred vision due to myopic regression after laser-assisted in-situ keratomileusis(LASIK) or laser-assisted sub-epithelial keratectomy (LASEK). Methods: This study comprised 155 patients (155 eyes) who were diagnosed with myopic regression after LASIK or LASEK and received medical treatment from January 2015 to January 2016. The visual acuity and refractive errors were compared before and after medical treatment and evaluated to determine whether the results differ between LASIK and LASEK. Results: The mean time of medical treatment was 64.1 ± 36.8 months after surgery. The responder group whose vision was improved and whose myopic error was decreased after medical treatment was comprised of 63 patients (41%). Theirvisualacuity in this group improved -0.21 ± 0.11 logMAR, and the amount of myopic error decreased 0.56 ± 0.32 diopters. The full responder group was 24 patients (15%), and the partial responder group was 39 patients (26%). The frequency of response to medical treatment was higher after LASIK than after LASEK, but the difference was not statistically significant. Conclusions: The combined medical treatment with anti-glaucoma eyedrops and 0.1% fluorometholone was effective in 41% of patients with regard to visual acuity improvement when used for post-LASIK or post-LASEK myopic regression. The medical treatment was effective after both LASIK and LASEK.

        2,4-bis (p-hydroxyphenyl)-2-butenal (HPB242) induces apoptosis via modulating E7 expression and inhibition of PI3K/Akt pathway in SiHa human cervical cancer cells.

        <P>The Maillard reaction is a chemical reaction occurring between an amino acid and a reducing sugar, usually requiring thermal processing. Maillard reaction products (MRPs) have antioxidant, antimutagenic, and antibacterial effects, and although 2,4-bis (p-hydroxyphenyl)-2-butenal (HPB242), a fructose-tyrosine MRP, appears to inhibit proliferation of cancer cells, its mechanism of action has not been studied in detail. We found that HPB242 treatment modulated expression of cyclins and tumor suppressor genes in SiHa human cervical cancer cell lines: cyclins and phospho-pRB were downregulated, whereas the expression of CDK inhibitors and p53 was enhanced. HPB242 induced apoptosis dose-dependently by suppressing E7 expression and leading to sub-G1 cell-cycle arrest in SiHa cell lines; treatment also led to the proteolytic cleavage of caspase-3, -9, and poly (ADP-ribose) polymerase. Moreover, HPB242 upregulated Fas expression, altered expressions of pro- and antiapoptotic factors, and also inhibited nuclear translocation of nuclear factor κB and phosphorylation of IκB. HPB242 treatment decreased phosphatidyl inositol-3 kinase and p-Akt expression levels, demonstrating that this survival pathway may also be inhibited by HPB242. Cumulatively, HPB242 promotes apoptosis by influencing E7 expression, inducing cell-cycle arrest at sub-G1 phase, and promoting both intrinsic (mitochondrial) and extrinsic (Fas-dependent) apoptosis in SiHa human cervical cancer cells.</P>

        바른체형운동 지도가 정신장애인의 우울감 및 폐활량에 미치는 효과

        본 연구는 바른체형운동 프로그램이 정신장애인의 폐활량 및 우울감에 미치는 영향을 알아보고자 경북 P시에 거주하고 정신장애 판정을 받았으며 정신재활프로그램에 참여하는 정신장애인을 대상으로 실시하였다. 참여한 피험자에게 연구목적과 내용을 설명하였고, 연구 참가동의서를 받은 19명을 최종 선정하여 10명은 운동군, 9명은 통제군으로 분류하여 12주간 바른체형운동 프로그램을 실시하였고, 연구결과는 다음과 같다. 우선 폐활량의 변화를 살펴본 결과 PEF (최대호기속도)와 FEV1/FVC (1초간 노력성 호기량의 노력성 폐활량에 대한 비)가 바른체형운동 적용 후에 증가하여 호기기능이 유의하게 향상됨을 확인하였다. 또한, 바른체형운동 프로그램 참여에 따른 운동군의 우울감의 변화는 통계적으로 유의한 차이는 나타나지 않았으나 우울정도의 감소폭이 통제군보다 운동군에서 훨씬 크게 나타나 바른체형운동이 정신장애인의 우울감 정도의 감소에 긍정적인 변화를 나타냈다고 할 수 있다. 그리고 본 연구에서는 프로그램 참가자들의 특수성으로 인한 표본 수의 한계로 인해 통계적 검증의 한계가 명확히 존재함을 고려하여 참여자들의 주관적 경험을 심층적으로 이해하는 것을 목적으로 한 질적분석(qualitative research)을 병행하여 실시하였다. 프로그램 참가자 중 동의하는 사람에 한해 심층적 면담을 실시하였으며 이를 통해 참여자들이 표현하는 내용의 의미단위를 검토하여 분류하고 세분화하는 작업을 통해 주제분석(thematic analysis)을 실시한 결과, 전반적으로 바른체형운동 프로그램에 참여한 정신장애인들의 우울감 정도가 줄어든 것을 알 수 있었다. 우울감의 변화과정은 ‘변화의 시작’, ‘프로그램에 대한 흥미’, ‘신체적 건강의 필요성에 대한 인식’, ’신체적 건강증진’, ’신체적 기능의 회복’, ’부정적인 습관(자세)의 변화’, ’우울감의 감소’, ’할 수 있다는 자신감’, ’미래에 대한 희망’과 같은 3개의 주제와 6개의 하위 주제로 나타났다. 따라서 바른체형운동이 정신장애인들의 신체적 건강증진을 위해 심폐지구력을 높여 신체적 건강증진을 도모할 뿐 아니라 더 나아가 정신적 우울감의 감소에도 효과성을 높여 정신적 건강증진에도 도움을 줄 수 있는 프로그램으로 정신장애인의 삶의 질 증진을 위해서는 보급이 반드시 필요할 것으로 사료된다. The purpose of this study was to investigate the effects of the regular Upright Body Type Excercise on posture, muscle strength of leg, lung capacity and depression of people with mental illness. The subjects of this study were recruited from B mental health center (5 years and more mental illness patients, n=19) located in P city. For the exercise group, They were carried out the Upright Body Type Excercise Program during 8 weeks (60 min/time, 3 times/week). They were randomly divided into two groups. Exercise group (n=10) and Control group (n=9). And also, they were agreed with consent forms before the experiments. Research results were as follows. Through the upper body type exercise, there was significantly difference in PEF and FEV1/FVC in the trained group. And also, there was much improved in depression level in the trained group. The change of melancholy feeling before and after the program was not statistically significant. However, because of limitations of sampling size due to the peculiarities of the program participants, in consideration of the limit of statistical validation exists clearly, intended to better understand the subjective experience of attendees, qualitative analyzing(qualitative research) was carried out in parallel. It was conducted a deep interview only person accepted among program participants, thematic analysis, subject analysis tasks to be subdivided by classifying by considering the semantic units of what participants expressed, was thus carried out. It found that the degree of melancholy feeling of mental disorders who participated in the Upright Body Typed Exercise Program was reduced. The course of the experience of change in depression appered the three subjects and six sub-themes such as “the start of the change”, “interest of the program”, “recognition of the need of the body`s health”, “physical health promotion”, “recovery of physical function”, “negative change of habits (attitude)”, “reduction of sense of depression”, “confidence that it is possible to”, “hope for the future”. Therefore, upper body type exercise is much helpful in lung capacity and mental health of people with mental illness. So, this type of exercise mostly needed in the people with mental illness group than the other group for the quality of life.

        당일 라식, 라섹 수술과 기존 수술법의 수술 후 결과 비교

        Purpose: To evaluate the postoperative results of one day laser-assisted in-situ keratomileusis (LASIK) or laser-assisted sub-epithelial keratectomy (LASEK) procedures, which were performed on the same day as preoperative examinations, including fundus examinations after dilating the pupil. Methods: This study included 226 LASIK patients (226 eyes) and 201 LASEK patients (201 eyes) who underwent surgery with Visumax and EX500 from January to December in 2016. We divided the patients into two groups. The one-day surgery group (one-day group) underwent surgery on the same day as preoperative examinations, including dilated fundus examinations. The scheduled surgery group (scheduled group) underwent surgery on the scheduled day after the preoperative examinations. In the one-day group, the surgery was usually performed 2–5 hours after instillation of the pupil dilating eye drops. Results: Among LASIK patients, the one-day group included 109 patients and the scheduled group included 117 patients. The postoperative myopic errors were 0.06 ± 0.37 diopters (D) and 0.07 ± 0.36 D, respectively (p = 0.91). The postoperative astigmatism was -0.38 ± 0.24 D and -0.37 ± 0.24 D, respectively (p = 0.77). The postoperative uncorrected visual acuity was -0.05 ± 0.03 logMAR and -0.06 ± 0.03 logMAR, respectively (p = 0.13). Among LASEK patients, the one-day group included 107 patients and the scheduled group included 94 patients. The postoperative myopic error was 0.18 ± 0.52 D and 0.22 ± 0.54 D, respectively (p = 0.95). The postoperative astigmatism was -0.48 ± 0.30 D and -0.46 ± 0.29 D, respectively (p = 0.14). The postoperative uncorrected visual acuity was -0.05 ± 0.03 logMAR and -0.06 ± 0.03 logMAR, respectively (p = 0.33). Conclusions: The postoperative results of the one-day LASIK and LASEK patients, whose surgery was performed on the same day as the preoperative examinations, were not significantly different from those using the conventional method.         The Biflavonoid Amentoflavone Induces Apoptosis via Suppressing E7 Expression, Cell Cycle Arrest at Sub-G1 Phase, and Mitochondria-Emanated Intrinsic Pathways in Human Cervical Cancer Cells

        Amentoflavone, a biflavonoid from Selaginella tamariscina, is known to possess several bioactivities such as antitumor, anti-inflammatory, and antifungal effects. However, the mechanism of the anticancer effects of amentoflavone on human cervical cancer cells has not been studied in detail. In this study, we demonstrated that amentoflavone induces apoptosis in SiHa and CaSki cervical cancer cells by suppressing human papillomavirus protein E7 expression. The cyclins and tumor suppressors were modulated by amentoflavone in SiHa and CaSki human cervical cancer cells: cyclin and hyperphosphorylated retinoblastoma (p-pRb) were down-regulated, whereas cyclin-dependent kinase inhibitors and p53 were enhanced. Amentoflavone up-regulated peroxisome proliferator-activated receptor γ (PPARγ) and phosphatase and tensin homolog deleted on chromosome 10 (PTEN) expression levels while inhibiting E7-mediated cyclooxygenase-2 (COX-2)/interleukin-32 (IL-32) expressions were downregulated, and Akt phosphorlylation was decreased in an amentoflavone-induced apoptotic process, suggesting that amentoflavone may be a PPARγ activator. Additionally, the expression of the anti-apoptotic factor Bcl-2 was decreased, whereas that of the well-known apoptotic factor Bax was increased, thereby releasing cytochrome c into cytosol in amentoflavone-treated cervical cancer cells. Furthermore, amentoflavone treatment led to the activation of caspase-3 and -9 and proteolytic cleavage of poly(ADP-ribose) polymerase. The expression level of the extrinsic death receptor Fas (CD95) was not altered by amentoflavone treatment. When these findings are taken together, the biflavonoid amentoflavone activates PPARγ/PTEN expressions and induces apoptosis via suppressing E7 expression, cell cycle arrest at sub-G1 phase, and mitochondria-emanated intrinsic pathways in SiHa and CaSki human cervical cancer cells. These findings suggest that amentoflavone has potential for development as a therapeutic agent for human cervical cancer.

        사관생도의 체육활동을 통한 사회성발달요인이 신체적 자기효능감에 미치는 영향

        The purpose of this study of cadets social development through physical activity factor out the impact of physical self-efficacy are seeing. To this end, on the basis of previous research and theory, cadets according to the individual characteristics of participants in physical activity factors, social development, and physical effects on self-efficacy was determined empirically. The subjects of this study are based in KAFA cadets to participate in physical activity among the 200 selected patients, 169 were used in the final part were used. By the way, more studies were summarized as follows. First, the social development of the sub-factors of the social factors, governance, stability, no statistically significant differences between men and women in the show, and overall social development factors in men than in women was higher. Physical self-efficacy subscale of physical competence and physical attractiveness in the sense of gender showed statistically significant differences, and about the physical self-efficacy was higher in women than men. Second, the social development of the factors involved in the stability of the sub-factors, depending on stocks showed a statistically significant difference, and the overall social development factors were the highest in the group event. Self-efficacy of physical competence subscale of the body involved in the events showed significant differences according to the tatistical and physical competence, the individual events and the most underestimated, interpersonal and organization in order. In other words, the overall physical self-efficacy and the lowest individual events, personal events, corporate events in order, respectively. Third, social development, physical factors impact on self-efficacy to evaluate the results with social dominance physical competence, physical self-expression, sense of physical attractiveness showed a positive correlation, and perceived physical ability and a negative correlation with has shown. Stability in the perceived physical ability, has shown a positive correlation.

        This paper discusses about the implementation of a simulator for the Navigation Control System(NCS) of bimodal tram. To operate a NCS of bimodal tram, values of all sorts of sensors installed in bimodal tram should be transmitted to the NCS, and the NCS calculates the measured sensor values to determine traveling direction, traveling speed, current position etc. The implementation of the simulator consists of a device applying driver’s input transaction function & virtual sensor program and a sub-rack device that controls communication with the NCS to evaluate navigation control function. The virtual sensor program can create routes (map), traveling profiles & seat information et cetera in order to transmit to the NCS, analyzes driver's input values and NCS output values to create virtual sensor values. The sub-rack device takes charge of communication with the NCS using CAN-OPEN, CAN-J1939, MVP protocols. This paper discusses about the implementation of the simulator and afterwards analyzes and evaluates the NCS simulation results.

      • 한국과 C.I.S 우수 레슬링 선수들의 엉치걸이 동작에 대한 운동학적 요인 분석

        This study investigated kinemtic factors of hip throw as a representative skill to make a comparative study between Korean and C. I. S wrestlers Four elite wrestlers were filmed using high - speed cinematography. Kinematic data for the each phase were derived from the digitized film. Analyzing variables included center of gravity(CG) displacement and velocity, hip joint, angular displacement and angular velocity The results were as followers ; 1. Elapse time of CG at conducting hip throw were showed similar result between two nations. 2. C. I. S wrestlers showed more displacement than Korean wrestlers in the harizontal phase (P<0.001) but, Korean wrestlers showed large movemnet, in the vertical phase(P <0.001). 3. In average velocity at whole phase, Korean wrestlers showed rapid velocity in the vertical phase(P<0.001) but, C. I. S wrestlers at conducting throw partner down showed rapid velocity in the horizontal phase(P<0.05). 4. C. I. S wrestlers showed large angular displacement in the hip joint at Ph. I. (P <0.001). 5. C. I. S wrestlers showed rapid angular velocity in the hip joint at Ph. I (P<0.001).
