        일본잎갈나무재의 수용성추출물 첨가가 표고버섯의 톱밥재배에 미치는 영향

        표고버섯(Lentinula edodes)의 생장이 매우 좋았던 일본잎갈나무재의 수용성 추출물(water-soluble fraction, WSF)을 분리하여 구성당을 분석한 결과, rhamnose 및 mannose는 거의 없고, glucose도 2.2%로 매우 낮았으며, arabinose가 18.3%, galactose가 75.3%로서 가장 많이 함유되어 있었다. 정제된 추출물의 arabinose와 galactose의 함량비는 1 : 3.4였다. 정제된 WSF를 완전메틸화·가수분해·가스크로마토그래피에 의하여 분석한 결과, β-1,3 결합한 D-galactopyranose가 주쇄를 이루며, 이에 galactose의 C6위에 측쇄로서 β-1,6 결합한 D-galactose와, β-1,6 결합한 L-arabinose가 존재하며, 전자는 2-3개의 D-galactopyranose가 결합을 하고 있고, 후자는 L-arabinofuranose가 1-2개 중간의 측쇄구조를 이루고, 말단은 L-arabinopyranose로 구성되어 있었다. PDA배지에 WSF첨가로 표고의 균사 생장이 매우 증진되었으며, 2-4% 첨가수준에서 균사 생장이 최대였고, 첨가량이 더 많으면 균사의 생장이 오히려 감소되었다. 톱밥배지에서도 PDA 배지에서의 결과와 마찬가지로 WSF첨가로 균사 생장이 증진되었으며, 산림 6호 및 Mok-H의 경우 모두 4% 첨가에서 균사의 생장이 최대로 나타났다. 톱밥배지에 WSF 첨가가 버섯생산량을 증가시켰는바, Mok-H의 경우 무첨가에 비해 약 1.3배, 산림 6호의경우 1.2배 증수되었다. 버섯균 세포벽의 주성분인 ergosterol 함량변화를 측정한 결과, WSF첨가로 미첨가에 비하여 ergosterol의 함량이 증가하였으며, 균사가 콜로니를 형성하는 초기단계에서는 낮은 ergosterol함량을 나타냈으나 자실체가 형성되는 시기에 급격히 높아지는 결과를 나타냈다. 따라서 배지내의 ergosterol함량이 자실체형성의 주요 예측수단으로 이용될 수 있을 것으로 판단된다. The water soluble fractions(WSF) from Japanese larch wood were isolated, purified by anion exchange resin and Sephadex gel filtration and identified its chemical structure by means of periodate oxidation and methylation reactions. Its major components are arabinose and galactose (1 : 3.4). Based on the results of periodate oxidation, methylation and gas chromatographic analysis of purified WSF, main chain is composed of β-1.3-glycosidic linkage among D-galactopyranoses, and two different side chains; β -1,6-glycosidic linkage among 2-3 units of D-galactopyranoses and β-1,6-glycosidic linkage between 1-2 units of Dgalactopyranose and L-arabinopyranose. Addition of WSF to culture media of oak mushroom (Lentinulu edodes) accelerated the mycelial growth. In the case of PDA cultures, 2 percent addition of WSF in Sanlim No. 6 strain and 4 percent of WSF in Mok-H strain mostly enhanced the mycelial growth of the mushroom. In the case of sawdust cultures, 4 percent addition of WSF in two strains showed the best mycelial growth. High percentages addition of WSF inhibited mycelial growth of the mushroom. Mushroom production was increased with addition of WSF. By the addition of WSF, ergosterol contents in the media were quite high at the colonized stage and rapidly increased at the fruiting stage. Therefore the ergosterol content could be utilized as an indicator to evaluate the culture maturity for the mushroom fruiting.

        The aging properties of cellular plastic insulations(EPS,PUF and UFF) were investigated by varying temperature, relative humidity and time. The time aging properties were changed with in 10 - 12% of initial properties at high temperature(above 318K) and relative humidity (above 90%). The amount of HCHO gas release from UFF was increased as temperature and relative humidity increased, and HCHO release rate was decreased exponentially with time.

      • 충청지방에서 발생한 한국형출혈열 54예의 임상적 고찰

        In Dae Jeon Eul-Ji General Hospital, activated since 1581, 54 patients with Korean Hemorrhagic Fever Who were all positive for Korea antigen were observed from January, 1981 to December, 1984. Epidemiology, clinical features, laboratory findings, complications and clinical course in Korean Hemorrhagic Fever were analyzed. The results were as follows: 1. The most prevalent area was OK-Chun Kun. 2. The peak incidence of age was fifth decade and male to female ratio was 2:1. 3. The peak incidence was November to December. 4. The most frequent symptomes were epigastric pain, nausea and vomiting and followed by hemsturia. 5. Conservative treatment was done and hemodialysis was performed in 19 patients without death.

        A new device, muscle clamping system was developed to facilitate exact quatifying technique A strabismus surgery, and reduce the risk of complications. The device is composed of a lower fixing body with three jaws and an upper supporting body. They are used to clip an extraocular muscle and fix it to the sclera with a single bite. Superior rectus recession on 16 rabbit eyes were performed with this new device. Conjunctival injection, muscle adhesion strength, and light microscopic findings were examined at 1, 2, 4 and 8 weeks postoperatively. The Conjunctival injection were minimal, adhesion power ranged from 420 to 600 gram gravity, which is sufficient in withstand the normal pull of human extraocular muscle. In microscopic exam, some Inflammatory cells and fibrosis were found. The new device was technically easy, fast, and accurate, so it may be useful in stabismus surgery.

        Biodegradable retinal fixation devices obtain mechnical fixation of the retina with desirable chorioretinal scarring and with the potential for local, sustained release of antimetabolites and steroids to inhibit proliferative vitreoretinopathy. We manufactured a biodegradable retinal tack with barb that was designed in order to prevent intrusion from implantation of retinal tacks. This study was carried to evaluate the efficacy for retinal fixation and the capability for sustained release of drugs with a newly designed biodegradable retinal tack Biodegradable retinal tacks were made of polymers of glycolic acids and were designed with barbs in a shape to prevent the disinsertion. Biodegradale retinal tacks are divided into 3 parts, a conical portion that is inserted into the sclera, a cylinder portion that remains in the vitreous, and a neck portion between the pin and the cylinder. The tapered conical end was manufactured to allow easy insertion through the retina and choroid into the sclera. A cylinder portion was manufactured with a tapered angle that fixes firmly into the orifice of 19 gauge spinal needle. A neck portion, 0.4 mm in diameter, was designed to prevent disinsertion from following implantation of retinal tack. The applicator was a 19 gauge spinal needle and its orifice was prepared to 15°angle to accept the tapered cylinder portion of the retinal tack. The retinal tacks, secured in the needles, were passed through the formed vitreous and inserted into the retina, choroid, and sclera and were released by pushing the internal needle, usually within 2-3mm of the medullary ray of the posterior rabbit retina A retinal tack was placed in each of 8 pigmented rabbit eyes. Slit-lamp biomicroscopy, indirect ophthalmoscopy and fundus photography were performed periodically from 1 day to 8 weeks after surgery. Eight eyes were enucleated and studied by light microscopy at 8 weeks. Biomicroscopic evaluation of the animals revealed edemas adjacent to the retinal surfaces immediately after insertion of the biodegradable retinal tacks in all the animals. These edemas disappeared after 1 week. The first noticeable change in the size of retinal tacks was shown after 2weeks. The size of the retinal tacks gradually got smaller, decreasing to about one-half at 4 weeks and about one-third at 8 weeks. All retinal tacks remained in inserted places without any movement for an 8 week period. On light microscopy, epiretinal proliferations were seen to extend into the vitreous cavity. Cellular capsules that lined the inner aspect of the scleral defect caused by tack insertion were found. However the adjacent retina had a normal cytologic appearance and architecture in all specimens. We manufactured a biodegradable retinal tack that is designed to prevent intrusion from implantation of retinal tacks. All biodegradable retinal tacks reduce in size with time, but no retinal tacks extruded from the inserted place. The newly designed biodegradable retinal tack can be used for retinal fixation and may be used as a vehicle for the introduction of pharmacologic agents to prevent the cellular events that promote proliferative vitreoretinopathy.

        In recent years, gonorrhea has been pandemic and remains one of the most common STDs in the world, especially in developing countries. For the detection of a more effective therapeutic regimen and assessing the prevalence of Penicillinase Producing Neisseria gonorrhoeae(PPNG), we have been trying to study the patients who have visited the Venereal Disease Clinic of Choong-Ku Public Health Center in Seoul since 1980 by menas of the chromogenic cephalosporin method. In 1998, 93 strians of N. genorrhoeae were isolated, among which 60(64.5%) were PPNG. The prevalence of PPNG in Seoul, which had been decreased to 39% in 1996 after a peak of 74.3% in 1993, is increased to 64.5% in 1998.

        Antioxidant properties and antitumor activity of Coprinus comatus

        In this study, we investigated the antioxidant activity and anticancer effect of Coprinus comatus (C. comatus) extracts. Free radical scavenging capacity of the C. comatus extracts was evaluated in vitro by the DPPH assay. C. comatus exhibited the strong DPPH radical scavenging activity. In vitro tests were also performed to examine the anticancer activity of C. comatus against 293 human normal epithelial kidney cells and various human cancer cell lines including AGS, Hela and HCT-15. C. comatus extracts showed the potent cancer cell-selective growth inhibitory activity against AGS, Hela, and HCT-15 cells, but slightly inhibited 293 normal cell growth. We examined several biological activities such as antitumor effects of C. comatus extracts in vivo. Solid tumors were induced by Sarcoma-180 inoculation in the left groin of BALB/c mice and then C. comatus extracts were oral administered. After 5 weeks, all mice were sacrificed and their tumor weights were measured. C. comatus extracts exhibited the most effective antitumor activity, and the tumor growth rate was inhibited by 55% in comparison with control group. Taken together, these results indicate that C. comatus extracts showed high antioxidant activity and possessed powerful antitumor activity.

        Rotenoisin A is a novel anti-adipogenic compound

        <P><B>Abstract</B></P> <P>The purpose of this study was to investigate the mechanisms underlying the inhibitory effects of rotenoisin A on adipogenesis in 3T3-L1 preadipocytes. 3T3-L1 cells were treated with rotenoisin A for 8 days after the induction of differentiation. Oil-red O staining showed that rotenoisin A significantly inhibited DMI-induced lipid accumulation and adipocyte differentiation. We found that rotenoisin A treatment of 3T3-L1 preadipocytes significantly reduced the mRNA and protein levels of the key adipocyte-specific transcription factors C/EBPβ, C/EBPα, and PPARγ and markedly inhibited the expression of fatty acid-binding protein (aP2), fatty acid synthase (FAS), and lipoprotein lipase (LPL). Furthermore, we observed that rotenoisin A substantially increased the phosphorylation of AMP-activated protein kinase (AMPK) and its downstream target phosphorylated acetyl CoA carboxylase (ACC). However, co-treatment with Compound C, an AMPK inhibitor, reversed the rotenoisin A-induced inhibition of the expression of the adipogenic transcription factors C/EBPα and PPARγ and decreased the levels of phosphorylated AMPK in differentiated 3T3-L1 cells. These results demonstrated that the anti-adipogenesis mechanism involves the down-regulation of critical adipogenic transcription factors, including C/EBPβ, C/EBPα, and PPARγ, through activation of the AMPK signaling pathway by rotenoisin A.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We investigated the anti-obesity effect of rotenoisin A in 3T3-Ll adipocytes. </LI> <LI> Rotenoisin A reduced the expression of C/EBPα and PPARγ in 3T3-L1 adipocytes. </LI> <LI> Rotenoisin A increased AMPK and ACC phosphorylation in 3T3-L1 adipocytes. </LI> <LI> Rotenoisin A inhibited adipocyte differentiation and adipogenesis in 3T3-L1 cells. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>
