        Development of an ISSR-Derived SCAR Marker in Korean Ginseng Cultivars (Panax ginseng C. A. Meyer)

        Recently, new ginseng cultivars having superior agricultural traits have been developed in Korea. For newly developed plant cultivars, the identification of distinctiveness is very important factors not only in plant cultivar management but also in breeding programs. Thus, eighty-five inter simple sequence repeat (ISSR) primers were applied to detect polymorphisms among six major Korean ginseng cultivars and two foreign ginsengs. A total of 197 polymorphic bands with an average 5.8 polymorphic bands and 2.9 banding patterns per assay unit across six Korean ginseng cultivars and foreign ginsengs from 236 amplified ISSR loci with an average 6.9 loci per assay unit were generated by 34 out of 85 ISSR primers. Three species of Panax ginseng including the Korean ginseng cultivars, P. quinquefolius, and P. notoginseng, could be readily discriminated using most tested primers. UBC-821, UBC-868, and UBC-878 generated polymorphic bands among the six Korean ginseng cultivars, and could distinguish them from foreign ginsengs. Sequence characterized amplified region (SCAR) marker system was introduced in order to increase the reproducibility of the polymorphism. One SCAR marker, PgI821C650, was successfully converted from the randomly amplified polymorphism by UBC-821. It showed the expected dominant polymorphism among ginseng samples. In addition, the specific polymorphism for Sunwon was generated by treating Taq I restriction enzyme to polymerase chain reaction products of PgI821C650. These results will serve as useful DNA markers for identification of Korean ginseng, especially Sunwon cultivar, seed management, and molecular breeding program supplemented with marker-assisted selection.

        A Novel Method to Differentiate Tonsil-Derived Mesenchymal Stem Cells In Vitro into Estrogen-Secreting Cells

        BACKGROUND: The advantages of tonsil-derived mesenchymal stem cells (TMSCs) over other mesenchymal stem cells (MSCs) include higher proliferation rates, various differentiation potentials, efficient immune-modulating capacity, and ease of obtainment. Specifically, TMSCs have been shown to differentiate into the endodermal lineage. Estrogen deficiency is a major cause of postmenopausal osteoporosis and is associated with higher incidences of ischemic heart disease and cerebrovascular attacks during the postmenopausal period. Therefore, stem cell-derived, estrogen-secreting cells might be used for estrogen deficiency. METHODS: Here, we developed a novel method that utilizes retinoic acid, insulin-like growth factor-1, basic fibroblast growth factor, and dexamethasone to evaluate the differentiating potential of TMSCs into estrogen-secreting cells. The efficacy of the novel differentiating method for generation of estrogen-secreting cells was also evaluated with bone marrow- and adipose tissue-derived MSCs. RESULTS: Incubating TMSCs in differentiating media induced the gene expression of cytochrome P450 19A1 (CYP19A1), which plays a key role in estrogen biosynthesis, and increased 17β-estradiol secretion upon testosterone addition. Furthermore, CYP11A1, CYP17A1, and 3β-hydroxysteroid dehydrogenase type-1 gene expression levels were significantly increased in TMSCs. In bone marrow-derived and adipose tissue-derived MSCs, this differentiation method also induced the gene expression of CYP19A1, but not CYP17A1, suggesting TMSCs are a superior source for estrogen secretion. CONCLUSION: These results imply that TMSCs can differentiate into functional estrogen-secreting cells, thus providing a novel, alternative cell therapy for estrogen deficiency.         청소년의 여가활동 실태에 관한 연구

        In this study, the status of teenager's leisure activity has been understood, examined and analyzed. The purpose of this study is to examine the status of teenager leisure activity and to provide way to improve the leisure space. The study has been based on questionnaire on middle and high school student, both male and female, in Seoul. The analysis was done by SPSSWIN Program Ver 11.0. Frequency, percentage,-test were methods of analysis. The results are as follows. Teenager are wasting their leisure time because of insufficient facilities for productive activities. They use the time, when analyzed in one week, for visiting shops, playing in the ground and playing in the game rooms. And when analyzed in a month, they use time for meeting friends in coffee shops, visiting restaurants, playing in the game rooms and visiting sports facilities. Teenagers' leisure time were different according to sex and age. Students hoped the leisure spaces to be healthy and active leisure activity, and facilities they wanted, were sports facilities, game rooms and parks. It is necessary that teenager have opportunities to learn how to use their leisure time healthy in the leisure space. This study was invested the status of teenager leisure activities, that were only divided sex, age among various social and environmental fators. Consquently, questions on that and arguments of different visual angles should be remained the study subject, later. This study was only composed of language levels, self-recorded method questionaire research. But, to get a result exactly is required the observation and interview

      • 순조대 효명세자 예제(睿製) 정재(呈才) - `예제`의 범주 및 정재 창작시기와의 관련성을 중심으로 -

        The aim of this study is to investigate the category that King Sunjo`s son prince Hyomyong 孝明世子(1809-1830) made Yeje Chongjae 睿製呈才 and relevance to create date. This study is based upon such source materials a government office document called Muza chinjak uigwe 『戊子進爵儀軌』(1828) and Kich`uk chinch`an uigwe 『己丑進饌儀軌』(1829). Generally it mean that yeje was the King`s son created verse. But created category of yeje was to adapt court dance known as chongjae 呈才, only apply to the words of a song known as ch`angsa 唱詞,not dance. The truth of this matter was convinced of Yeje 『睿製』(1829). A kind of King Sunjo`s son Hyomyong create yeje chongjae were twenty. An item were Mugo 舞鼓,Muae 無導,Abak 牙拍,Kainjonmoktan 佳人剪牧丹,Ch`oehwamu 催花舞,Posangmu 寶相舞,Kyo ngp`ungdo 慶豊圖, Mansumu 萬壽舞,Mangsonmun 望仙門,Musanhyang 舞山香,Pakchop 撲蝶, Sasonmu 四仙舞,Yonbaekpokchirnu 演百福之舞,Yongji 影池,Changsaeng poyonjimu 長生寶宴之舞,Chesuchu`cang 帝壽昌,Ch`opsung 疊勝,Ch`undae okch`ok 春臺玉燭,Ch`unaengjon 春鶯?,Honch`onhwa 獻天花. For the most part of these chongjae 呈才 are agreement that created date of chongjae and the words of a song known as ch`angsa 唱詞 But Mugo 舞鼓,Muae 無導, Abak 牙拍,Kainjonmoktan 佳人剪牧丹,Ch`oehwamu 催花舞,Posangmu are not agreement. So we are acknowledge the capability of disagreement between create date of yeje chongjae and create date of chongjae.

        A cose of Aspergillosis is reported in a 12-day-old chick. The chick died with clinical signs such as dyspnea, gasping, and accelerated breathing. At necropsy, small white caseous nodules (approximately 1 ㎜ in diameter) scattered throughout lung tissue. Upon H&E staining, multiple granulomas were observed in tertialy bronchi. Lesions consisted of central area of necrosis surrounded by macrophages, giand cells, lymphocytes, and some fibrous tissues. Hyphae of Aspergillus in the lesion branched into Y type, which was observed by PAS staining for fungus.

        Single-Port Transaxillary Robotic Thyroidectomy (START) for Benign Thyroid Tumors

        Purpose: Single-port transaxillary robotic thyroidectomy using the da Vinci SP® system with a two-step retraction method is known to be a practical surgical method that maximizes the cosmetic and functional benefits for patients and reduces the workload fatigue of surgeons by increasing robotic dependency. This study aimed to evaluate its technical feasibility in the treatment of benign tumors. Methods: We retrospectively analyzed the data of patients who were diagnosed with benign thyroid disease after undergoing single-port transaxillary robotic thyroidectomy with a two-step retraction method using the da Vinci SP® robotic system at Severance Hospital, Yonsei University College of Medicine, Seoul, Korea, between May 2019 and May 2022. Patients with Graves’ disease were excluded from this study. Results: Twenty-two patients were enrolled in this study, 21 females and one male, with a mean age of 32.7 years (range: 17–52 years). The mean body mass index was 22.5±3.1 kg/m2 (range: 18.0–29.7 kg/m2). The mean tumor size was 2.8±1.5 cm (range, 0.4–5.9 cm), and the mean operation time was 173.4±26.8 min (range, 128–226 min). There were no intraoperative complications. The mean estimated blood loss was 3.2±7.6 mL (range, 0–30 mL). No serious postoperative complications occurred, except one case of seroma. Conclusion: Single-port transaxillary robotic thyroidectomy’s utilization of a narrow working space is useful for benign thyroid tumors. It is feasible and safe for experienced robotic surgeons treating benign thyroid tumors.

      • 순조대 효명세자 대리청정시 정재(呈才)의 계승과 변화

        순조대 대리청정기는 정재사에서 가장 큰 변화의 시기로, 본고의 목적은 1차 자료의 검토를 통해, 효명세자(孝明世子) 대리청정기의 정재의 구성요소인 가·무·악(歌舞樂)에 어떠한 계승과 변화가 존재했는가를 밝히는 것이다. 첫째, 정재 창사는 조선전기와 마찬가지로 성수무강(聖壽無疆)과 태평성대(太平聖代)를 비는 내용이 많았다. 이러한 계승적 측면 위에, 효명세자가 지은 창사의 내용은 조선 전기에 비해 훨씬 풍부하고 다채로운 내용을 수용하는 변화가 있었다. 둘째, 정재 춤구성을 계승과 변화의 측면으로 본다면, 두 측면이 공존한 시기라고 하겠다. 당악정재 포구락과 수연장의 경우, 1828년 진작에서 일시적으로 죽간자가 생략되는 예가 있었으나 바로 다음 해에 다시 회복되어 당악정재의 틀은 그대로 고수되고 죽간자의 구호나 치어도 변하지 않은 채 계승되었다. 기존 정재를 계승하면서 변화도 꾀하였다. 효명세자 대리청정기에 독무인 춘앵전(春鶯囀)과 무산향(舞山香)의 등장은 새로운 변화의 정점이며, 정재가 완숙한 경지에 이르렀다는 것의 반증이다. 셋째로, 정재 반주음악은 하나의 별칭에 다양한 악곡명이 붙는 것이 특징이다. 또한 어떤 정재에 반드시 어떤 악곡이라는 고정성보다는, 때에 따라 다양한 악곡을 선택할 수 있는 탄력성을 가졌다. 효명세자 대리청정기 정재의 새로운 측면과 변화의 측면을 중심으로 살펴보면, 새로운 변화조차도 계승을 바탕으로 하고 있다는 사실을 확인 할 수 있었다. 이 시기 정재의 연행양상을 한마디로 정리한다면 ‘다양함의 공존’이라고 할 수 있겠다. 옛 것과 새 것이 함께 존재하며, 옛 것을 밑바탕으로 하여 새 것을 만들기도 하였다. 동일한 정재라도 어느 때는 창사가 불려지고, 어느 때는 생략되어 연행시간에 탄력성을 부여했다. 또한 전승되어온 정재이든 새로 창작된 정재이든, 동일한 정재라도 연향에 따라 무원수가 다르게 구성되는 경우가 많았다. 이는 정재 춤사위 대형의 변화로 이어져, 연행공간에 탄력성을 부여했다. 반주음악도 동일한 정재에 다른 악곡을 연주할 수 있었다. 이 시기 정재 연행에 나타난 ‘다양함의 공존’은 문화적 성숙도를 보여준다. 서로 다름을 폭넓게 끌어들여 자양분으로 삼았기 때문에 나타난 현상이라 생각한다. 연향의 자리는 효명세자가 자식된 입장에서 어버이에게 효를 다하는 것이면서 다른 한편으로는 국정최고 운영자로서 실추된 왕실의 권위를 회복하려는 현실적 의지의 발현이었는데, 그 맥락에 정재가 놓여있는 것이다. The aim of this study is to investigate an aspect of chongjae 呈才, during the time when prince Hyomyong 孝明世子(1809-1830) was the right power of representation attorney in the reign of King Sunjo. History of chongjae 呈才 in the reign of King Sunjo, it is very important time. Type and contents is various appears in this time. 17 repertory were succeeded in this time, and 23 repertory were created. So it is a point when the succession and change coexist. The scope of this research is between 1827 and 1830, when Prince Hyomyong 孝明世子(1809-1830) is the right power of representation attorney. This study is based upon such source materials a government official document called Muja chinjak ŭigwe 『戊子進爵儀軌』 (1828) and Kich'uk chinch'an ŭigwe 『己丑進饌儀軌』(1829). Chongjae 呈才 is synthetic performing art, so it composed of a center element dance, the words of a song known as ch'angsa 唱詞, and incidental music it will together research. Chongjae was not only the court performence but also the local governmental performence. But this research is restricted a scope, only in the court performence. It is composed three section, it examined from the viewpoint of succession and change of chongjae integral part. In the first section was examined ch'angsa 唱詞. Succession position of chongjae ch'angsa, the political contents was accomplishing the center is the thing. Renovation position of chongjae ch'angsa, the political contents becomes internalized and the lyrical contents was introduced is the thing. Ch'angsa of mugo 舞鼓, abak 牙拍, Kainjon moktan 佳人剪牧丹, and Posangmu 寶相舞 are the evidences. In the second section examined was dance composition of chongjae. Succession position of dance composition, the frame of existing dance was maintained almost like that. There is temporary fluctuation where the chukkanja 竹竿子 disappears from Suyonjang 壽延長 and Pogurak 抛毬樂, but features of origin it returned again. From renovation position of dance composition, most will pay attention the fact that was appearance of solo dance, for example Ch'unaengjon 春鶯囀 and Musanhyang 舞山香. In the third section was examined incidental music of chongjae. There was an accompaniment name which is various in chongjae one. Until now from the viewpoint of succession and change, it tried to observe the aspect which chongjae is various. The feature of at this time variety chongjae is able to summarize, as coexistence.
