        Effects of Rehmanniae Radix and Radix Aconiti Water Extracts on Blood Pressure in Hypertensive Rats

        한의학에 있어서 음양의 조절은 매우 중요하다. 숙지황은 음허를 치료하는 주약이며, 부자는 양허를 치료하는 주약이다. 한의학에서 신성고혈압의 발생은 음허와 관련이 있다고 하였다. 본 연구는 실험적 신성고혈압과 자연발증고혈압 백서에 숙지황과 부자 전탕액을 투여 후 혈압의 변동과 혈장 renin 활성도, aldosterone 및 심방이뇨호르몬 (atrial natriuretic hormone)의 변동을 관찰하였다. 숙지황 투여후 실험적 신성고혈압 백서의 혈압은 감소하였다. 부자 전탕액 투여 후 자연발증고혈압 백서의 혈압 상승이 억제되었다. 숙지황 및 부자 전탕액 투여 후 자연발증고혈압 백서의 혈장 renin 활성도는 감소하였다. 부자 전탕액 투여 후 신성고혈압 백서의 혈장 ANP 농도는 감소하였다. 부자 전탕액의 혈압강하 작용은 ANP 및 aldosterone 농도의 변동과 관련이 있다고 사료된다.

        아토피 피부염 치료 처방에 대한 방제학적(方劑學的) 고찰(考察)

        In Oriental Medicine, Atopic dermatitis(AD) belongs to the category of the Naseun, Taesun(胎癬), Taeryumchang(胎斂瘡), Eczema(濕疹), Seupchang(濕瘡), Samanpung(四彎風), Chimumchang(浸淫瘡). The Basic Prescriptions which have been used for treatment of Atopic dermatitis are saengryusamultang(生料四物湯), Onchungeum(溫淸飮), Seungmagalgeuntang(升麻葛根湯),Hoichunryangkyuksan(回春凉膈散), Doghengsan(導赤散), Pyungweesan(平胃散), Heungbangpaedoksan(荊防敗毒散), Goomigangharltang(九味羌活湯), Baekhotang(白虎湯), Gaegytang(桂枝湯), Yukmigihyuangtang(六味地黃湯). AD can divide three groups, Acute-Type, Semiacute-Type, Chronic-Type, according to it's condition. This study shows that it can be applicate Seungmagalgeuntang(升麻葛根湯), Hoichunryangkyuksan(回春凉膈散), Doghengsan(導赤散), Heungbangpaedoksan(荊防敗毒散), Goomigangharltang(九味羌活湯), Baekhotang(白虎湯), Gaegytang(桂枝湯) for treatment of Acute-Type, Pyungweesan(平胃散) for treatment of Semiacute-Type, and saengryusamultang(生料四物湯), Oncungeum(溫淸飮) for treatment fo Chronic-Type.

        동의보감(東醫寶鑑) 중(中) 박하(薄荷)가 주약(主藥)으로 배오(配伍)된 방제(方劑)의 활용(活用)에 대한 고찰(考察)

        Objectives : This report describes 48 prescriptions related to the use of Herba Menthae main blended from Dongeuibogam. The following conclusions were drawn through investigations on the prescriptions that use Herba Menthae as a key component. Prescriptions that Herba Menthae was taken as a monarch drug are utilized for 15 therapeutic purposes, for example, wind stroke, throat disease, nose disease, head disease and tooth disease. In particular, 12.5% of prescriptions appear in the chapter of each wind stroke, throat disease, nose disease. Methods : Prescriptions that utilize Herba Menthae as the main ingredient are used in the treatment of urticaria, hemoptysis-hematemesis, loss of consciousness due to wind stroke and they are also used for treating 41 different types of disease. Herba Menthae is used for pathogenic factors such as wind and heat. Results : The dosage of Herba Menthae is 0.15pun(about 0.06g) to 4don(about 14g), however 5pun(about 1.88g) has been taken the most for clinical application. Conclusions : Gamgiltang, Piryongbanggamgiltang and Hoichunyanggyeoksan are the most useful base prescriptions which use the Herba Menthae as the main ingredient.

        K-562 백혈병 세포주에서 저근백피와 Gleevec을 처리에 의한 유전자 발현 비교 분석

        In this study, we investigated gene expression patterns induced by Ailanthus altissima extract and compared it with Gleevec, a well-known anti-leukemia drug, in K562 chromic leukemia cells. Ailanthus altissima extract(100 ug/ml) and Gleevec(50 ug/ml) were treated to cells for 1h, 2h, 4h, and 16h and total RNA was extracted. Gene expressions were evaluated using cDMA microarray, in which 24,000 genes were spotted. Hierarchical clustering analysis showed that expression of genes included in two clusters were increased or decreased time dependently by both Ailanthus altissima extract and Gleevec. Genes included in another cluster were induced by Ailanthus altissima extract but not by Gleevec. In biological process analysis, expression of genes involved in apoptosis, growth arrest and DNA-damage were increased, but genes stimulating cell cycle were decreased. This study provides comprehensive comparison of the patterns of gene expression changes induced by Ailanthus altissima extract and Gleevec in K-562 leukemia cells.

        마우스에서 황금 유래 Wogonin의 Th1과 Th2 사이토가인 조절 효과

        In the present study, wogonin, a major flavone isolated from Scutellaria Radix were examined for its imunosuppressive activity on the 2,4-dinitro-fluorobenzene (DNFB)-induced delayed type hypersensitivity (DTH) in C3H/HeN mice. This compound inhibited selectively $TNF-\alpha$ and IL-4, but not IL-6, IL-10 and $IFN-\gamma$ on DNFB-induced Th1 and Th2 cytokines in a concentration-dependent manner. Interestingly, this compound increased the expression of heme oxygenase-1 (HO-1) in a concentration-dependent manner The above results reveal that wogonin possesses anti-inflammatory, humoral and cellular immunomodulatory, and stress reducing activities on the DNFB-induced DTH in mice and these properties may contribute to the anti-atopic dermatitis activity of Scutellaria Radix.

        숙지황, 부자 전탕액이 고혈압 백서의 혈압에 미치는 영향

        $CD4^+$ Jurkat T 세포주에서 Th1과 Th2 사이토가인 조절에 미치는 황금 유래 Baicalin, Baicalein 및 Wogonin의 효과

        In the present study, baicalin, baicalein, and wogonin, a major flavone isolated from Scutellaria Radix were examined for their effects on PMA-induced Interlukin-6 (IL-6), $interferon-\gamma(IFN-\gamma)$, tumor necrosis factor $(TNF)-\alpha$, IL-4, IL-10, and IL-13 productions in the PMA-stimulated $CD4^+$ Jurkat T cells. These three compounds inhibited PMA-induced Th1 cytokine $(IL-6,\;IFN-\gamma,\;TNF-\alpha)$ and Th2 cytokine (IL-4 and IL-13) productions in a concentration-dependent manner. But wogonin, but not baicalin baicalein, increased PMA-induced IL-10 production. These results suggest that baicalin, baicalein, and wogonin, a major flavone modulate Th1 and Th2 cytokine productions in $CD4^+$ Jurkat T cells and these properties may contribute to the anti-atopic dermatitis activity of Scutellaria Radix.

        동의보감(東醫寶鑑) 허로문(虛勞門) 처방(處方)의 방제(方劑) 분석(分析)에 대한 연구

        Objectives : Consumptive is an unhealthy condition that are caused by lack of blood and essence, and that means also some stages of chronic diseases. The purpose of this study is to analysis 108 Prescriptions of Consumptive part in Dongui Bogam. Methods : The 108 Prescriptions of Consumptive part in Dongui Bogam analysed frequency of basic prescriptions, symptoms of prescriptions and the pathology. Results : Ssangbohwan, Yiuihwan, Gamrihwan were used for tonifying were mostly used as basic prescriptions in whole Consumptive part respectively. There are common symptoms in consumptive part in Dongui Bogam. That symptoms are "tidal fever, night sweating, nocturnal emission, cough, sputum, skinny body, weak pulse, spontaneous sweating, deafness, dim vision and tuberculosis". Qi blood(yin yang) pathologies in prescriptions on consumptive part are "yin deficiency, yin deficiency with effulgent fire, yang qi deficiency, dual damage of qi and blood, non-interaction between fire and water, collapse of yang and exhaustion of yin, less blood". viscera and bowels pathologies in prescriptions on consumptive part are "heart and kidney deficiency, spleen-stomach weakness, spleen and kidney great deficiency, weakness of kidney qi, meridian waste in heart, spleen and kidney, damage in heart and lung". Conclusions : As a result of Study on Analysis all Prescriptions of Consumptive part in Dongui Bogam, We can understand more about basic prescriptions, symptoms of prescriptions and the pathology that are using for curing consumptive. We expected that this study will can help to give rationale for future study of consumptive caring.

        금은화(金銀花)의 항(抗)류마티즘 효능(效能)에 대한 연구(硏究)

        Rheumatoid arthritis is characterized by the focal loss of cartilage due to an up-regulation of inflammatory pathways, which produce inflammatory mediators, such as interleukin-1beta(IL-$1{\beta}$), IL-6, tumour necrosis factor alpha(TNF-$\alpha$), prostaglandin, and nitric oxide(NO). We investigated the anti-arthritic effects of water extract from FLOS LONICERAE(FLWE) in vitro and in vivo. Extract inhibited the production of inflammatory mediators(NO, IL-$1{\beta}$, TNF-$\alpha$, and prostaglandin $E_2$) and the expression of inducible NO synthase(iNOS) and cyclooxygenase-2(COX-2) in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages in a dose-dependent manner. FLWE also inhibited TNF-$\alpha$, IL-$1{\beta}$, IL-6, and $PGE_2$ production as well as COX activity in collagen-induced mouse arthritis. Moreover, FLWE significantly suppressed collagen-induced mouse arthritis. These results suggest that FLOS LONICERAE may be useful for therapy against inflammatory immune diseases and rheumatoid arthritis, probably by suppressing the production of inflammatory mediators.

        DBA1J 마우스에서 가감보중익기탕이 콜라젠 유도 관절염 억제에 미치는 효과

        N/A Objectives : The present study has been undertaken to investigate the effect of Kagam-Bojungikgitang (KBT) decoction on development of collagen-induced arthrtis (CIA) in DBAl J mice. Methods : CIA was induced in UBAJ1 mice by immunization with bovine type II collagen (CIU and treatment with lipopolysacchride (LPS). KBT is the water extracts prepared from Ginseng Radix, Astragali Radix, Angelicae gigantis Radix, Astractylodis Rhizoma alba, Aurantii nobilis Pericarpium, Glycyrrhizae Radix, Artemisiae iwayomogii Herba, Scutellariae Radx. This is a modifitd prescription of Bojungikgitang, which has been used for the treatment of indigestion, and immunological disease in oriental countries. KBT was orally administered at difference doses. The incidence of arthritis, arthritis index, levels of tumor necrosis factor -alpha (TNF-a). interferon-gama (IFN-V), interleukin lbeta (IL-lP), IL - 3 . IL--10, II- 13. and prostaglandin E? (PGE.1 and cyclooxygenase~-2 (COX21 activity were investigated. Results : KBT dose- dependently suppressed the release of T h l cytokines (TNF-a, IFN-.v, and IL-1 8 ) and inflammatory mediators (PGE? and COX--2 caused by immunization of CII and stimulation of 1,PS. KUT without cytotoxic effext. Kagam-bojunyikgitang`s Inhibitory effects were better than Ehjungikgitang in PGEz production and COX-2 expression. However, KBT increased markedly the production of Th:! cytokines (IL-4, IL 10, and IL -13). Therefor , KBT suppressed markedly the incidence of arthritis and arthritis index caused by immunization of CII and stimulation of LPS. Conclusion : These results suggest that KBT suppress inflammatory mediators and regulates Th l and Th2 cytokmes. And these properties may contribute to the strong anti-arthritis of KBT.
