        LncRNA-IMAT1 Promotes Invasion of Meningiomas by Suppressing KLF4/hsa-miR22-3p/Snai1 Pathway

        Malignant meningiomas often show invasive growth that makes complete tumor resection challenging, and they are more prone to recur after radical resection. Invasive meningioma associated transcript 1 (IMAT1) is a long noncoding RNA located on Homo sapiens chromosome 17 that was identified by our team based on absolute expression differences in invasive and non-invasive meningiomas. Our studies indicated that IMAT1 was highly expressed in invasive meningiomas compared with non-invasive meningiomas. In vitro studies showed that IMAT1 promoted meningioma cell invasion through the inactivation of the Krüppel-like factor 4 (KLF4)/hsa-miR22-3p/Snai1 pathway by acting as a sponge for hsa-miR22-3p, and IMAT1 knockdown effectively restored the tumor suppressive properties of KLF4 by preserving its tumor suppressor pathway. In vivo experiments confirmed that IMAT1 silencing could significantly inhibit the growth of subcutaneous tumors and prolong the survival period of tumor-bearing mice. Our findings demonstrated that the high expression of IMAT1 is the inherent reason for the loss of the tumor suppressive properties of KLF4 during meningioma progression. Therefore, we believe that IMAT1 may be a potential biological marker and treatment target for meningiomas.

        서울과 상하이 사교육 비교분석

        [연구목적] 본 연구는 서울과 상하이의 사교육 수요실태, 사교육 공급실태, 사교육 정책실태에 대한 비교분석을 통하여 두 도시의 사교육 현상에 대한 이해를 제고시키고, 향후 사교육 정책적 시사점을 제공하는 데 그 목적이 있다. [연구방법] 이러한 목적을 달성하기 위해 국제비교자료(PISA), 정부의 국가·도시 대규모 통계 자료(서울 통계청, 상하이 CFPS, 녹색지표), 개인연구자, 기관보고서 등 자료를 활용하여 문헌분석방법과 비교연구방법을 병행하였다. [연구결과] 주요 연구결과는 첫째, 사교육 수요실태는 두 도시의 사교육 참여율과 비용이 높고, 참여시간이 길다는 점에서 사교육이 학생생활 및 가계경제에 영향을 미친다. 양극화(고액화·음성화) 현상이 발생하는 공통점이 있지만, 학교급별, 계열별, 유형별, 지출금액별 격차 등에서 다소 상의하다. 둘째, 사교육 공급실태는 그 유형이 두 도시 모두 개인과외로 시작하여 학원형태로 발전하였고, 최근에는 온라인 사교육으로 전개되었다. 사교육공급은 더욱 다양화, 대형화, 전문화, 세분화 되고, 사교육 상장기업들의 상장, 합병, 인수 등 자본화 활동이 활발하게 이루어지는 공통점이 있다. 하지만 학습지 사교육, 공교육 교사의 유상과외, 사교육 상장기업의 창립년도와 상장시기, 해외상장, 업무영역, 해외진출 등에서는 차이점이 있다. 셋째, 사교육 정책실태는 두 도시에서 사교육과 관련된 직접정책과 간접 정책이 수립되었지만, 대체적으로 사교육 문제를 해결하지 못한 공통점이 있다. [결론] 이에 사교육 현상에서 유사성을 유지하던 서울과 상하이는 사교육에 대한 인식과 정책 목적의 차이로 인해 향후 서로 다른 양상으로 전개될 가능성이 있다. [Purpose] The purpose of this study is to deepen the understanding of the private tutoring phenomenon in Seoul and Shanghai, and to provide future directional policy advice through a comparative analysis of Seoul and Shanghai’s private tutoring in demand conditions(participation rate, participation time, participation cost, participation motivation), supply conditions(supplier type, the status of private tutoring listed companies), and policy conditions(policy type). [Methods] This research simultaneously employed documentary analysis and comparative research methods. The research materials and data included international comparative data, state-led(official) largescale statistical data, and individual researchers’ data. [Results] The main findings are as follows: First, in the demand conditions, both Seoul and Shanghai have extremely high participation rates and participation cost of private tutoring, also both cities have characteristics of high expenditure and polarization in common. Second, in the supply conditions, concerning the types and characteristics of private tutoring providers, both cities recently through online forms have been diversifying and departmentalizing in common. Both of two cities’ private tutoring listed companies actively being listed, mergers, acquisitions, and other capital activities. Third, in the policy conditions, private tutoring related policies in Seoul and Shanghai are similar in type, institutionalization and diversification, but are different in purpose. [Conclusion] The development of private tutoring between Seoul and Shanghai could be different. Based on analysis results, discussion have been proposed.

        Expression of FMD Virus-like particles in yeast Hansenula polymorpha and immunogenicity of combine with CpG and Alumimun adjuvant

        Background: Inactivated vaccines are limited in preventing foot-and-mouth disease (FMD) due to safety problems. Recombinant virus-like particles (VLPs) are an excellent candidate for a novel vaccine for preventing FMD, given that VLPs have similar immunogenicity as natural viruses and are replication- and infection-incompetent. Objectives: The 3C protease and P1 polyprotein of type O FMD virus (FDMV) was expressed in yeast Hansenula polymorpha to generate self-resembling VLPs, and the potential of recombinant VLPs as an FMD vaccine was evaluated. Methods: BALB/c mice were immunized with recombinant purified VLPs using CpG oligodeoxynucleotide and aluminum hydroxide gel as an adjuvant. Cytokines and lymphocytes from serum and spleen were analyzed by enzyme-linked immunosorbent assay, enzyme-linked immunospot assay, and flow cytometry. Results: The VLPs of FMD were purified successfully from yeast protein with a diameter of approximately 25 nm. The immunization of mice showed that animals produced high levels of FMDV antibodies and a higher level of antibodies for a longer time. In addition, higher levels of interferon-γ and CD4+ T cells were observed in mice immunized with VLPs. Conclusions: The expression of VLPs of FMD in H. polymorpha provides a novel strategy for the generation of the FMDV vaccine.

        Hexadecylamine Modified Copper Nanowire Coated Superhydrophobic Cotton Fabric for Antifouling, Oil-water Separation, and Infrared Reflection Applications

        Superhydrophobic surfaces by creating surface roughness followed by coating with low-surface-energyfluorinated materials have been intensively developed on cotton fabrics. However, exploring superhydrophobic surfaces withnon-fluorine components and further improving their attachment with cotton fabrics is still challenging. Herein, wedeveloped a superhydrophobic surface on cotton fabric by dip coating with hexadecylamine (HDA) modified coppernanowires (Cu NWs) based on the coordination between Cu atoms and amine groups. The HDA modified Cu NWs not onlycreated surface roughness, but also decreased the surface energy, leading to a superhydrophobic cotton fabric with watercontact angle of 164 °±2.0 °. Furthermore, ultrasonic washing and tape peeling tests exhibited the Cu NWs were adheredtightly to the surface of cotton fibers, owing to the strong hydrogen bond between amine groups on HDA molecules andhydroxyl groups on cotton fibers. In addition, the superhydrophobic cotton fabric showed excellent antifouling, oil-waterseparation and infrared reflection properties. It is believed that this facile method provide promising industrial applicationsfor fabricating superhydrophobic surfaces on other substrates.

        Improvement of FK506 Production by Selection of 4-Aminobutyric Acid-tolerant Mutant and Optimization of its Fermentation Using Response Surface Methodology

        Tacrolimus (FK506), a 23-membered polyketidemacrolide with immunosuppressant activity, can be produced byStreptomyces tsukubaensis. We studied a variety of mutant strainsof S. tsukubaensis for the microbial production of FK506. Thebest strain (CZ-19) was obtained from the parent strain LLZ-1 bynitrosoguanidine mutation and 4-Aminobutyric acid (FK506precursor structure analogs) adaption. In the shake-flask experiments,titer of FK506 by CZ-19 was 532.44 mg/L, increased by 65.13%compared to that of the parent strain. Through single factorexperiments and response surface methodology, we furtheroptimized the medium for improved FK506 production by CZ-19in shake flask culture. The optimal medium for enhanced FK506production was as follows: 17.19 g/L corn starch, 21.78 g/Lglucose, 8.06 g/L peptone, and 18.98 mg/L 4-aminobutyric acid. The predicted FK506 titer was 906.49 mg/L, and the experimentaldata confirmed the validity of the model. The present studydemonstrates that S. tsukubaensis CZ-19 is a promising strain forindustrial production of FK506.

        The expression of Rab5 and its effect on invasion, migration and exosome secretion in triple negative breast cancer

        Triple-negative breast cancer (TNBC) is the most aggressive subtype of breast cancer and current therapeutic strategies are limited in their effectiveness. The expressions of Rab5 and the M2 tumor-associated macrophage marker CD163 in tissues were detected by Western blot. The migration and invasion of cells were determined using a Transwell assay. The expressions of the exosome markers were evaluated by Western blot. The polarization of human macrophages (THP-1) was determined by incubation of THP-1 cells with conditioned medium or exosomes collected from MDA-MB-231 cells with indicated transfections or by a coculture system of THP-1 and MDA-MB-231 cells. The M1 and M2 macrophage markers were evaluated by qRT-PCR. The expression of Rab5 in TNBC was significantly higher than that in normal breast tissue. Rab5 expressions in triple-negative and luminal A breast cancer were higher than those in other molecular subtypes. Higher CD163 expression was observed in triple-negative breast cancer and in triple-negative and luminal B subtypes. Rab5 knockdown suppressed but Rab5 overexpression promoted the migration and invasion capacity of MDA-MB-231 cells. The levels of CD63 and CD9 in the medium of Rab5 knockdown cells were lower than those in control cells, whereas higher levels of CD63 and CD9 were observed in Rab5 overexpression cells. Rab5 knockdown decreased the excretion but did not alter the diameter of the exosomes. Knockdown of Rab5 facilitated the anti-tumor polarization of macrophages, which was partially reversed by Rab5 overexpression. Therefore, Rab5 is expected to be a potential therapeutic target for triple-negative breast cancer.

        Ganoderic acid A attenuates high‑fat‑diet‑induced liver injury in rats by regulating the lipid oxidation and liver inflammation

        Ganoderic Acid A (GA) has many pharmacologicaleffects such as anti-tumor, antibacterial, anti-inflammatory,and immunosuppressive effects. However, the protectiveeffect of GA on liver injury has not been reported. Thisstudy aimed to investigate the action of GA on insufficientmethionine and choline combined with high-fat diet (HFD)-induced non-alcoholic fatty liver disease (NAFLD) in rats. NAFLD model was established by insufficient methionineand choline combined with high fat feeding to rats. The levelsof Acetyl-CoA carboxylase, fatty acid synthase, sterolregulatory element binding protein, liver X receptors, AMPactivatedprotein kinase, peroxisome proliferator-activatedreceptor α, PPARg coactivator 1α and NF-κB pathway inthe liver were detected by western blot. The results of thisstudy demonstrated that the expression of GA can not onlysignificantly decrease the live weight and liver weight perbody weight of HFD mice, but also restore the alanine aminotransferase,aspartate aminotransferase, total bilirubinlevels, triglyceride and cholesterol in serum. In addition,the expression of GA increased the levels of high-densitylipoprotein cholesterol in serum, ameliorated pathologicalchanges and decreased NAS score of mice’s liver. In conclusion,the treatment with GA could improve NAFLD in ratsby regulating the levels of signaling events involved in freefatty acid production, lipid oxidation and liver inflammation.
