      • 枳實에 의한 아나필락시의 抑制效果

        The effect of water extract of Poncirus trifoliata fruit (ATFE) on compound 48/80 induced anaphylaxis was studied in the rat. The following results have obtained. 1. Optimal lethal dose of compound 48/80 was 8ug/gm B.W. 2. PTFE was found to exhibit an inhibitory activity on the anaphylaxis induced by compound 48/80. 3. Administration of PTFE 60 min before or 5 min after had a better result than 10 min after anaphylaxis induced by compound 48/80. 4. PTFE also inhibited significantly the serum histamine release in anaphylaxis induced by compound 48/80 in the rat. 5. Administration of PIFE before 30 min decreased PCA reaction induced by anti DNP-IgE and DNP-HSA. This result indicates that poncirus trifoliata suppresses the anaphylaxis induced by compound 48/80 with decreasing the serum histamine release.

        직업군인의 일-가정 양립 지원을 위한 정책방안 연구

        This study has researched concept of work-family compatibility and support policies synthetically in order to develop a policy for work family compatibility, to prepare institutional base to improve working condition for professional soldiers, and to make a strategy to expand system. Vision of work-family compatibility policy of professional soldiers was chosen to "balanced life of work and family of professional soldiers". Purpose of the policy is (1) "to expand parenting and educational support for professional soldiers", (2) "to build family-friendly circumstance to build a healthy family of professional soldiers", (3) "to improve understanding of army members and family for professional soldiers' work and family compatibility, (4) "to vitalize support policies for work and family compatibility of professional soldiers, and 3 sections of policy and the detailed plans has been made. The first section is "support for birth/parenting and the detailed plan consists of (1) to offer prior opportunity to enter day care center, (2) to build more day care centers in army, (3) emergency support in parenting service, (4) support for going see a obstetrician regularly. The second section is "family support" and the detailed plans are (1) offer mentoring for soldiers' children, (2) provide information for parenting and education, (3) consult soldiers' children, (4) offer family harmony program. The third section is "organizational culture and environmental support", and (1) support Return-to-Work program for users of Parental Leave, (2) offer a program to improve understanding of work and family compatibility for Military Commanders, (3) to make good examples of work and family compatibility and its promotion, (4) DB management and monitoring circumstance of work and family compatibility of professional soldiers, (5) to create manual of work and family compatibility and its distribution, (6) to operate a support system for work and family compatibility have been proposed. 이 연구는 직업군인의 일-가정 양립 지원을 위한 정책방안을 수립하기 위해 일반적인 일-가정 양립의 개념 및 직업군인의 일-가정 양립 지원의 의의·지원정책 현황·지원요구 등을 분석하여 직업군인의 일-가정 양립 지원 방안을 도출하였다. 직업군인의 일-가정 양립 지원을 위한 정책방안의 비전으로는「직업군인의 일과 가정의 조화로운 삶」으로 선정되었다. 정책방안의 목표로는 ①「직업군인의 자녀 양육 및 교육 지원 확대」, ②「직업군인의 건강한 가족 형성을 위한 가족친화 여건 조성」, ③「직업군인의 일-가정 양립을 위한 군 구성원 및 군인가족의 인식 제고」, ④「직업군인의 일-가정 양립을 위한 지원 정책 활성화」가 도출되었고 이에 따라 다음과 같은 세 가지 정책영역과 영역별 세부 추진방안이 도출되었다. 첫째 영역은 「출산/육아·보육 지원」이며, 세부 추진 방안으로 ① 보육시설 입소 우선권 제공, ② 군내 보육시설 운영 확대, ③ 긴급 보육서비스 지원, ④ 부인과 통원진료 지원으로 구성되어 있다. 둘째 영역은 「가족 지원」이며, 세부 추진 방안으로 ① 군 자녀 멘토링 프로그램 제공, ② 자녀양육 및 교육정보 제공, ③ 군 자녀 상담제공, ④ 가족화합 프로그램 제공을 제시하였다. 셋째 영역은 「조직문화 및 환경지원」이며, ① 육아휴직자 복귀지원 프로그램 운영, ② 상급지휘관 일-가정 양립 인식 개선 프로그램 제공, ③ 일-가정 양립 우수사례 발굴 및 홍보, ④ 직업군인의 일-가정 양립 현황 DB 관리 및 모니터링, ⑤ 일-가정 양립제도 사용 매뉴얼 개발 및 배포, ⑥ 일-가정 양립 지원 시스템 운영을 새로운 정책으로 제시하였다.

        지실(枳實)의 항알러지 작용에 대한 연구

        The unripe fruit of Poncirus trifoliata Raf has been used for the treatment of allergic disease. Recently it was reported that the fruit inhibits passive cutaneous anaphylaxis and histamine release at mast cell. Type I immediate hypersensitivity of anaphylactic type is caused by released mediate chemical at mast cell. Histamine is also known as one of potent mediate chemical. Also release of mediate chemical is affected by specific stimulation of IgE combined with mast cell. Activation of mast cell is known to be stimulated by compound 48/80 and inhibited by increase of cAMP. In this experiment, the effect of water extract of Poncirus trifoliata Raf fruit (PT) on a histamine release, cAMP concentration and IgE production was measured. Compound 48/80 was administrated to the mouse peritoneal cell which was pretreated with PT. PT dosedependently inhibited histamine release at peritoneal mast cell and the serum level of histamine induced by compound 48/80. PT also instantly increased cAMP level of peritoneal mast cell right after it was added and the level gradually decreased. Production of IgE induced by antigens at mouse peritoneal cell was inhibited by PT. The IgE synthesis is induced by IL-4 and it is known that lipopolysaccharide(LPS) plus IL-4 cause an increase in IgE secretion by murine B cells. The effects of PT inhibited the production of IgE activated by LPS plus IL-4 at human U266B1 cells. These results indicate that PT has antiallergic activity by Inhibition of IgE production from B cells and histamine release by increase of cAMP.

        고온 집진용 Fail Safety 소결 필터의 통기도와 분진제거 특성

        The fail safety filter is an assistant filter element to be mounted in order to intercept the particles leaked when the main filter elements are broken. So it should have two contrary functions of being plugged easily to meet the purpose of dust sealing and a high permeability to save the space. The permeability of the metal filter elements were effectively controlled by the following factor: powder size(53-840 ${\mu}m$) and applied pressure(1000-2000 $kgf/cm^2$), and then the compact were sintered for 1 hour at $1200^{\circ}C$ in vacuum sintering furnace. The sintered metal filters was evaluated for the function of the fail safety filter in an experimental unit. The maximum allowable particle size was 420-840 ${\mu}m$, when a CIP pressure of 1500 $kgf/cm^2$ was applied reveals a permeability of about $1.2{\times}10^{10}m^2$ and pore size of about 60 ${\mu}m$. The metal filter produced with stainless steel powder of 480-840 ${\mu}m$ size, which presented excellent permeability than commercial ceramic filter element and plugged with in 3 minutes with the leak of the maximum particle size less than 3 ${\mu}m$.

        실크단백질의 식이 공급이 아토피 피부염 동물 모델 NC/Nga Mice 피부의 세라마이드 함량 및 관련인자 발현에 미치는 영향

        본 연구에서는 아토피 피부염 동물 모델인 NC/Nga mice에 실크단백질 sericin과 fibroin을 식이 공급 후 피부의 세라마이드 함량 및 관련인자 발현 변화를 정상대조군인 BALB/c mice 및 아토피 피부염 대조군과 비교 분석하였다. 세라마이드 함량은 아토피 대조군인 CA군이 정상대조군 C군보다 현저히 낮았으나, 정제된 건조 분말을 별다른 처리 없이 그대로 실크단백질 sericin을 식이공급한 S군은 정상대조군인 C군 이상의 수준으로 높였으며, 실크단백질 fibroin(F군)은 세라마이드 함량을 정상대조군 수준이상으로 올려주지는 못하였다. 세라마이드 합성효소인 SPT의 mRNA 및 protein 발현은 아토피 대조군인 CA군은 정상대조군인 C군보다 높았으나, 실크단백질 공급군인 S군 및 F군은 모두 정상대조군 C군보다 현저히 낮았다. 반면, 세라마이드 분해효소인 ceramidase의 mRNA 및 protein 발현은 아토피 대조군인 CA군은 정상대조군인 C군보다 높았으나, 실크단백질을 식이섭취한 S군 및 F군에서는 정상대조군인 C군과 유사한 수준으로 CA군에 비해서는 유의적으로 낮았다. 결론적으로 실크단백질 sericin의 10주간 식이섭취는 아토피 피부염 동물모델 NC/Nga mice 표피의 세라마이드 함량을 정상대조군 수준 이상으로 증가시켰으며, 궁극적으로 표피의 장벽기능을 정상대조군의 수준으로 변화시켰다. 이는 serine이 다량 함유되어있는 실크단백질 sericin을 공급에 의해 증가되었는데, serine은 그 자체로서 보습기능을 하여 표피의 장벽기능을 보완하였기 때문에 SPT에 의한 생합성은 불필요하였으며 또한, ceramidase에 의한 세라마이드 분해를 정상수준이하로 억제하여 세라마이드의 생성보다는 분해억제에 의한 것으로 여겨진다. Ceramide rich intercellular lipid lamellae are thought to be particularly important in maintaining the structural integrity of epidermal barrier. Ceramide is synthesized de novo by serine palmitoyltransferase (SPT) phospholipid intermediates, serine and palmitic acid persist within the stratum corneum. The ceramide which is synthesized is degraded with fatty acid and sphingosine by degradative enzyme ceramidase. The depletion of ceramide in stratum corneum was reported in the atopic dermatitis. As an effort to search for the dietary source for improving the level of ceramide in epidermis, the dietary effects of various-typed silk protein were compared. Seventy male NC/Nga mice, an animal model of atopic dermatitis, were divided into seven groups: group CA as an atopic control with control diet, group S: 1% crude sericin diet, group F: 1% crude fibroin diet, group PS : peptide pattern of sericin(Mw 5000), group PF: peptide pattern of fibroin (Mw 1500), group AS: manufactured the same as amino acid profile of sericin and group AF: manufactured the same as amino acid profile of fibroin. Ten male BALB/c mice were served as group C (control group) control diet. All mice were fed on diet and water ad libitum for 10 weeks. Dry skin condition was established in group CA as ceramide content was decreased. Despite a marked decrease of mRNA and prorein expression of SPT, enzyme do novo synthesis, ceramide content of group S was dramatically increased by inhibiting the mRNA and protein expression of degradative enzyme ceramidase. However, dietary supplementation of crude silk fibroin protein (group F) and in other groups that were supplemented with either amino acid or peptide type of sericin or fibroin did not increase the level of ceramide. Together, our data demonstrate that dietary supplementation of crude sericin is more effective at improving ceramide level in epidemis of NC/Nga mice.
