Expression of Transforming Growth Factor-β Receptors inFood Protein-Induced Enterocolitis Syndrome in InfancyHai Lee Chung, M.D., Sun Mi Chung, M.D., Gyung Ah Ha, M.D., Jeong Jin Lee, M.D.Eun Jin Choi, M.D., Jin Gyung Kim, M.D., Woo Taek Kim, M.D.Un Seok Nho, M.D., Jin Bok Hwang, M.D. and Jeong Ja Park, M.D.* 목 적 : IgE와 관련되지 않은 위장관 과민 반응의 한 형태인 food protein-induced enterocolitis syndrome(FPIES)은 구토와 설사를 주소로 주로 영아기에 관찰되나 그 병리학적 기전은 아직 자세히 알려져 있지 않다. 저자들은 FPIES으로 진단되었던 환아들의 소장 점막에서 transforming growth factor(TGF)-β의 수용체들의 표현 정도를 조사하고 장점막 손상 정도와의 상관 관계도 함께 조사하여 FPIES를 비롯한 식품 단백 특이 IgE와 관련되지 않은 위장관 과민 반응의 기전을 이해하는데 도움을 주고자 하였다.방 법 : 구토와 설사를 주소로 소아과에 입원하여 임상적 기준 및 유발 시험에 의해 FPIES으로 진단되었던 평균 연령 49일의 환아 28명을 대상으로 하였으며 내시경 검사를 실시하여 얻은 소장의 조직에 대해 type 1과 type 2 TGF-β 수용체들에 대한 면역 조직 화학 검사를 실시하였다.결 과 : TGF-β 수용체들은 융모와 음와의 상피 세포 모두에 표현되는 것이 관찰되었고 type 1 수용체는 융모의 위축을 동반하는 경우에서 그렇지 않은 경우에 비해 의미 있게 낮은 표현 정도를 나타내었으며(P<0.001) 융모의 위축 정도와 역상관관계를 나타내었다(r=-0.59, P<0.001). 두가지 수용체 모두 고유판의 세포들에는 거의 표현되지 않는 양상을 보였다. 대조군에서는 두 수용체 모두 중등도 이상의 표현 정도를 나타내었다.

BACKGROUND: We first determined the efficacy of lesional injection of tonsil-derived MSCs (mesenchymal stem cells) for the treatment of 5-fluorouracil induced oral mucositis. METHODS: Oral mucositis was induced in hamsters by administration of 5-fluorouracil (day 0, 2, 4) followed by mechanical trauma (day 1, 2, 4). The experimental groups included MT (mechanical trauma only), 5-FU ? MT (mechanical trauma with 5-fluorouracil administration), TMSC (mechanical trauma with 5-fluorouracil administration, tonsilderived mesenchymal stem cells injection), DEXA (mechanical trauma with 5-fluorouracil administration, dexamethasone injection), and saline (mechanical trauma with 5-fluorouracil administration, saline injection). RESULTS: On day 10, gross and histologic analyses showed that nearly complete healing and epithelialization of the cheek mucosa of the TMSC group, whereas the other groups showed definite ulcerative lesions. Compared with the MT and DEXA groups, CD31 expression was greater in the TMSC group on days 10 and 14. Tendency towards a decrease in MMP2 expression with the time in the TMSC group was observed. In addition, the TMSC group showed higher expression of TGF-b, and NOX4 on day 10 compared with the other groups. Scratch assay demonstrated that the conditioned media harvested from tonsil-derived MSCs significantly increased migratory efficacy of NIH3T3 cells. Transwell assay showed that the preferential migration of tonsil-derived MSCs to the wound area. CONCLUSION: Intralesional administration of tonsil-derived MSCs may accelerate wound healing of 5-fluorouracil induced oral mucositis by upregulating neovascularization and effective wound contraction. In addition, tonsil-derived MSCs might contribute to oral ulcer regeneration via the stimulation of fibroblast proliferation and migration.

Marker-assisted backcrossing is a powerful method for developing new cultivars. To develop genomic-wide markers, genotyping-by-sequencing(GBS) can be an efficient method. However, unrefined low-quality markers and missing data between markers can contribute to hamperingthe marker selection process, particularly in multi-way crosses. In this study, we aimed to calculate the recovery rate of offspring individualsand minimize errors that occur among a large number of markers. Initially, missing data were imputed by comparing samples using the k-nearestneighbor (k-NN) algorithm. Thereafter, low-quality single-nucleotide polymorphisms (SNPs) were corrected by applying the graphical representationmethod based on the k-NN algorithm in order of the SNPs in a chromosome designed for a multi-parental population. Four-way cross anddouble-backcrossed tomato BC1F1 (230 lines) and BC2F1 (96 lines) populations were genotyped by GBS. The genotype of samples of theBC1F1 and BC2F1 populations was determined based on the parental haplotype. Thus, the method of visualizing the genotype of offspringindividuals, generated via crosses of multiple parents, not only improves estimation of the recovery rate but also facilitates easier selectionin breeding programs.
