A new method for quantitative determination of Hyoscyamine and Scopolamine in Datura stramonium L. by High Performance Liquid Chromatography(HPLC) was established. A reversed-phase system with a μ Bondapak C18 column using methanol in 0.2% aqueous phosphoric acid(40:60) with PIC A as a mobile phase was developed. Hyoscyamine and Scopolamine were detected at 254nm and the analysis was successfully carried out within 20min.

Thirteen compounds were synthesized by condensing the N-heterocyclic amines (piperidine, pyrrolidine, morpholine) and secondary aliphatic amines (dimethylamine, diethylamine) with 3,4-methylenedioxyphenylalkenoic acid chlorides for developing CNS depressants. Among them, N,N-diethyl-3,4-methylenedioxycinnamamide (IX) and N,N-dimethyl-5-(3,4-methylenedioxyphenyl)-2,4-pentadienoic acid amide (XII) exhibited strong activity in antagonism against pentylenetetrazole-induced convulsion, strychnine-induced convulsion and maximal electroshock seizure. N,N-Dimethyl-3,4-methylenedioxycinnamide (VIII) showed more potent activity in antagonism against strychnine-induced convulsion and maximal electroshock seizure and in the prolongation of hexobarbital sleeping time.

In studies of the aminopyrine-alcohol-buffer solution on the occult blood test using the filter paper method, it is found that the experimental optimum conditions are 4% aminopyrine. 10% of H_2O_2, pH5 of the Walpole buffe、r solution Toyo filter paper No.2, 1 min of standing time, and the 7 : 3 ratio of aminopyrine to H_2O_2 (V/V) As the sensitivity of this method agrees to that of Hematest, it is considered that this method can be used as a clinical test in occult blood.
사티오시아노코발트[Ⅱ]산 이온을 이용한 植物性鹽基成分의 確認 및 定量

A Coloring and spectrophotometric method using tetrathiocyanatocobaltate(Ⅱ) ion as a complexation reagent was developed for the identification of Coptidis Rhizoma and Phellodendri Cortex in herb medicine. Identification tests could be carried out coloring in test tube and ultraviolet-visible spectrophotometry. Coptidis Rhizoma and Phellodendri Cortex: with a solution of 1,2-dichloroethane in thst tube - yellowish green. The ultraviolet - visible spectrum of base components in 1,2-dichloroethane exgibits maxima at 270, 340, 442 and 624nm and was developed for the determination of berberine or palmatine in Coptidis Rhizoma and Phellodendri Cortex. Optimum conditions for good determination was determined: Maximum wavelength 442nm; Determination of berberine was possible to the range of 0.01mg/ml-0.10mg/ml in 1,2-dichloroethane. Calibration curve showed that the coefficient of variation was 0.999% for berberine. And determination of palmatine was possible to the range of 0.10mg/ml - 0.60mg/ml in 1,dichloroethane. The method was rapid, accurate. It was considered that spectrophotometry could be greatly applied to the analysis of base components in Coptidis Rhizoma and Phellodendri Cortex containing relatively small amount of gerb medicine extract.

Evodiamine and evocarpine are basic components to contain in Evodiae Fructus. After Evodiamine and evocarpine heve been isolated from the Evodiae fructus as the precipitate, they could be identifified and separated by coloured test, UV/Vis. absorption wavelength or TLC The presence of evodiamine and evocarpine are confirmed by its reaction with the TCR ion, [Co(SCN)_4]^2-, to produce the blue or yellowish blue precipitate, It is based on the formation of complex compounds by evodiamine and evocarpine with TCR reagent. The method has been found to be simple, convenient and suitable for routine identification of evodiamine and evocarpine, related basic constituents, the crude drug "Evodiae fructus" prepared from the Evodia officinalis Dode or Evodia rutaecarpa Bentham(Rutaceae).

This study was undertaken to develope a method for the determination of aconitine alkaloids from Aconitum ciliare Tuber by spectrophotometry. The complex of cobalt(Ⅱ)ion and thiocyanate ion forms tetrathiocyanatocobaltate(Ⅱ)ion in aqueous solution containing an excess of thiocyanate ion. The tetrathiocyanatocobaltate(Ⅱ)ion produces the complex with aconitine alkaloids. The complex are extracted from aqueous solution to 1. 2-dichlorethane. A spectrophotometry for determination of aconitine alkaloids with tetrathiocyanatocobaltate(Ⅱ)ion were, established by 1, 2-dichlorethane extraction. Determination of aconitine standard was possible to the rage of 0.4-0.6㎎/㎖ in 1.2-dichlorethane. To examine the determination of aconitine alkaloids in Aconitum ciliare Tuber were determined by using acid-alkali titration, spectrophotometry from ether extracts. The average recovery of aconitine standard sloution was 97.9% and standard deviation 2.97%. As this method is accurate and precise, is applicable to the determination of aconitine alkaloids in Aconitum ciliare Tuber.
