        Bubble breakup dynamics and flow behaviors of a surface-functionalized nanocellulose based nanofluid stabilized foam in constricted microfluidic devices

        Nanocellulose was surface-functionalized toward the applications in enhanced oil recovery (EOR) as a green alternative. The focus of this paper is on the effect of this material based nanofluid (NF) on foam lamella stabilization through studying its bubble breakup dynamics and flow behaviors in constricted mircofluidic devices. The NF stabilized foam produced an improved flow resistance across the capillary largely due to the capillary trapped bubbles at the contraction. The “snap-off” caused the NF stabilized foam to produce finer textured bubbles, which can migrate readily forward to the deep porous media, as revealed by the pressure profiles.

        Insufficient radiofrequency ablation-induced autophagy contributes to the rapid progression of residual hepatocellular carcinoma through the HIF-1α/BNIP3 signaling pathway

        Currently speaking, it is noted that radiofrequency ablation (RFA) has been the most widely used treatment for hepatocellular carcinoma (HCC) occurring in patients. However, accumulating evidence has demonstrated that the incidence of insufficient RFA (IRFA) may result in the identified rapid progression of residual HCC in the patient, which can greatly hinder the effectiveness and patient reported benefits of utilizing this technique. Although many efforts have been proposed, the underlying mechanisms triggering the rapid progression of residual HCC after IRFA have not yet been fully clarified through current research literature reviews. It was shown in this study that cell proliferation, migration and invasion of residual HepG2 and SMMC7721 cells were significantly increased after the IRFA was simulated in vitro. In other words, it is noted that IRFA could do this by enhancing the image of autophagy of the residual HCC cell via the HIF-1 α/BNIP3 pathway. Consequently, the down-regulation of BNIP3 may result in the inhibition of the residual HCC cell progression and autophagy after IRFA. Our present study results suggest that IRFA could promote residual HCC cell progression in vitro by enhancing autophagy via the HIF-1 α/BNIP3 pathway. For this reason, it is noted that the targeting of the BNIP3 may be useful in preventing the rapid growth and metastasis of residual HCC after IRFA. [BMB Reports 2019; 52(4): 277-282]

        Research Articles : Synchronized Expression of Two Bombyx mori Caspase Family Genes, ice-2 and ice-5 in Cells Induced by Ultraviolet Irradiation

        The caspase family proteins play an important role in programmed cell death (apoptosis). To date, the expression profiles of the caspase family genes in Bombyx mori (Bm) are poorly known. In this study, we examined the expression profiles of two novel Bm caspase family genes (ice-2 and ice-5), the potential change of the mitochondrial membrane and the morphology in Bm cells after stimulation of ultraviolet (UV) irradiation. The results showed the potential change of the mitochondrial membrane occurred at 5 hours after UV irradiation treatment. Analysis of fluorescent quantitative RT-PCR demonstrated that both the ice-2 and ice-5 might be involved in UV induced apoptosis in Bm cells. Notably, after UV irradiating, expression pattern of ice-2 and ice-5 were remarkably different. The ice-2 gene was highly expressed at two time points, 0.5 and 5 hours after UV stimulating, while the expression level of ice-5 only peaked at 5 hours after UV stimulating. It indicated that apoptosis induced by UV irradiation was involved in the mitochondrial pathway and the two isoforms of Bm ice may act but play different role during the apoptosis of Bm cells.

        Effects of Dietary Zinc Level and an Inflammatory Challenge on Performance and Immune Response of Weanling Pigs

        Two experiments were conducted to determine the effect of dietary zinc level on growth performance and immune function in normal (Experiment 1) and immunologically challenged (Experiment 2) weanling pigs. Treatments consisted of the following: i) a corn-soybean meal basal diet containing 36.75 mg/kg total Zn, ii) basal diet+60 mg/kg added Zn as $ZnSO_{4}$, iii) basal diet+120 mg/kg added Zn as $ZnSO_{4}$. Each diet was fed to six pens of four pigs per pen (Exp. 1) or six pens of three pigs per pen (Exp. 2). In Exp. 1, the dietary zinc level had no effect on average daily growth (ADG), average daily feed intake (ADFI), or feed conversion ratio (FCR). Concentrations of tissue and serum zinc were not affected. Peripheral blood lymphocyte proliferation (PBLP) was not affected by dietary treatments. Supplementation of 120 mg/kg Zn decreased (p<0.05) the antibody response to bovine serum albumin (BSA) on d 7 compared with pigs fed the basal diet, but not on d 14. In Exp. 2, LPS challenge had no effect on ADG, ADFI and FCR in the entire trial (from d 0 to 21). LPS challenge significantly decreased ADG and ADFI (p<0.01) from d 7 to 14, but FCR was not affected. LPS challenge increased PBLP (p<0.05) and serum concentration of interleukin-1 (IL-1) (p<0.01), whereas the antibody response to BSA and serum concentration of interleukin-2 (IL-2) were not affected. Supplementation of Zn did not affect ADFI and FCR from d 7 to 14, but there was a trend for ADG to be enhanced with Zn supplementation (p<0.10). Supplementation of Zn tended to increase PBLP (p<0.10). Dietary treatment had no effect on the antibody response to BSA or concentrations of serum IL-1 and IL-2. Results indicate that the level of Zn recommended by NRC (1998) for weanling pigs was sufficient for optimal growth performance and immune responses. Zn requirements may be higher for pigs experiencing an acute phase response than for healthy pigs.

        3D Electromagnetic-Temperature Field Close-Coupling Calculation of Losses and Heat in the Damper Winding of a Large Tubular Hydro-Generator

        To obtain more precise and rational calculation details of the loss and heat of a damper winding in a tubular hydro-generator, this study develops a three-dimensional, fi nite-element, electromagnetic-temperature fi eld close-coupling model of the damper winding. On the basis of multi-physical fi eld coupled theory, the model fully considers the temperature eff ects of the damper winding resistivities and heat conductivities, and the eddy current loss in the end region of the damper winding. The model was verifi ed by direct tests of the damper winding temperature. Unlike the conventional weak-coupling model, the proposed close-coupling model fully captures the interaction between the electromagnetic and temperature fi elds. Therefore, of the model more accurately and reasonably calculates the loss and heat of the damper winding than the conventional model. The proposed calculation model can properly assess the loss and heat of damper windings in large hydro-generators, which is helpful for improving the design standards of hydro-generators.

      • Risk Factors for Surgical Site Infections after Liver Resec-tion (A Multivariate Analysis of 6,132 Patients)

        Aims: Surgical site infection (SSI) is a common postoperative complication and associated with an increased morbidity, hospital stay, and overall cost. The aim of the present study was to identify risk factors for SSIs after hepatic resection based on a large single-center cohort. Methods: A retrospective study was conducted of 6,132 patients who underwent liver resection without concomitant biliary reconstruction or gastrointestinal procedures between 2014 and 2016 at the largest hepatic center in China. The occurrences of SSI, classified as incisional SSI and organ/space SSI within 30 days after operation were investigated. Patient- and surgical-related risk variables were collected using standardized data collection form. A likelihood ratio forward regression model was used to assess the independent association of risk factors with SSI. Results: SSI developed in 587 patients (9.6%), including superficial/deep incisional SSI in 357 patients (5.8 %), and organ/ space SSI in 304 patients (5.0 %). Multivariate logistic regression analysis showed that obesity, diabetes mellitus, ASA score ≥ 2, liver cirrhosis, re-hepatectomy, hepatoliathiasis, and intraoperative blood transfusion were independent risk factors of overall SSI. However, incisional and organ/space SSI differed from each other with respect to risk factors. Among a variety of risk factors, hepatolithiasis, liver cirrhosis, and intraoperative blood transfusion were consistently associated with both incisional and organ/space SSI. Conclusions: SSI is a common complication after liver resection, and more caution should be taken in patients with hepatolithiasis or liver cirrhosis. Prevention strategies focusing on factors associated with SSI is necessary in order to reduce SSI after liver resection.

      • Large-Scale Genome-Wide Association Study of East Asians Identifies Loci Associated With Risk for Colorectal Cancer

        <P><B>Background & Aims</B></P> <P>Genome-wide association studies (GWASs) have associated approximately 50 loci with risk of colorectal cancer (CRC)—nearly one third of these loci were initially associated with CRC in studies conducted in East Asian populations. We conducted a GWAS of East Asians to identify CRC risk loci and evaluate the generalizability of findings from GWASs of European populations to Asian populations.</P> <P><B>Methods</B></P> <P>We analyzed genetic data from 22,775 patients with CRC (cases) and 47,731 individuals without cancer (controls) from 14 studies in the Asia Colorectal Cancer Consortium. First, we performed a meta-analysis of 7 GWASs (10,625 cases and 34,595 controls) and identified 46,554 promising risk variants for replication by adding them to the Multi-Ethnic Global Array (MEGA) for genotype analysis in 6445 cases and 7175 controls. These data were analyzed, along with data from an additional 5705 cases and 5961 controls genotyped using the OncoArray. We also obtained data from 57,976 cases and 67,242 controls of European descent. Variants at identified risk loci were functionally annotated and evaluated in correlation with gene expression levels.</P> <P><B>Results</B></P> <P>A meta-analyses of all samples from people of Asian descent identified 13 loci and 1 new variant at a known locus (10q24.2) associated with risk of CRC at the genome-wide significance level of <I>P</I> < 5 × 10<SUP>–8</SUP>. We did not perform experiments to replicate these associations in additional individuals of Asian ancestry. However, the lead risk variant in 6 of these loci was also significantly associated with risk of CRC in European descendants. A strong association (44%–75% increase in risk per allele) was found for 2 low-frequency variants: rs201395236 at 1q44 (minor allele frequency, 1.34%) and rs77969132 at 12p11.21 (minor allele frequency, 1.53%). For 8 of the 13 associated loci, the variants with the highest levels of significant association were located inside or near the protein-coding genes <I>L1TD1</I>, <I>EFCAB2</I>, <I>PPP1R21</I>, <I>SLCO2A1</I>, <I>HLA-G</I>, <I>NOTCH4</I>, <I>DENND5B</I>, and <I>GNAS</I>. For other intergenic loci, we provided evidence for the possible involvement of the genes <I>ALDH7A1</I>, <I>PRICKLE1</I>, <I>KLF5</I>, <I>WWOX</I>, and <I>GLP2R</I>. We replicated findings for 41 of 52 previously reported risk loci.</P> <P><B>Conclusions</B></P> <P>We showed that most of the risk loci previously associated with CRC risk in individuals of European descent were also associated with CRC risk in East Asians. Furthermore, we identified 13 loci significantly associated with risk for CRC in Asians. Many of these loci contained genes that regulate the immune response, Wnt signaling to β-catenin, prostaglandin E2 catabolism, and cell pluripotency and proliferation. Further analyses of these genes and their variants is warranted, particularly for the 8 loci for which the lead CRC risk variants were not replicated in persons of European descent.</P>

        Fabrication of polyacrylic acid-based composite binders with strong binding forces on copper foils for silicon anodes in lithium-ion batteries

        The commercial application of silicon (Si) anodes with high theoretical capacity is hampered by the poorcyclic stability because the huge volume change of Si during discharge/charge processes results in thepulverization of electrode materials and the electric contact loss of electrode materials with copper(Cu) foils. Binders play an important role for adhering active materials and conductive additives togetheronto Cu foils. Herein, we design and develop a three-dimensional networked composite binder (PAA-co-SN) via an amidation reaction between polyacrylic acid (PAA) and thiourea (SN). The granular anchors(CuxS) are constructed at the interface between PAA-co-SN binders and Cu foils, resulting from the reactionof S and Cu during the drying processes of Si electrodes. When used as binders for Si nanoparticles,they exhibit stable cyclic performance (1580 mAh g1 after 500 cycles). The anchoring mechanismbetween composite binders and Cu foils provides a strategy for the improvement of electrochemicalproperties of Si-based anodes for lithium-ion batteries.

      • Expression of Fas/FasL in CD8<sup>+</sup> T and CD3<sup>+</sup> Foxp3<sup>+</sup> Treg Cells - Relationship with Apoptosis of Circulating CD8<sup>+</sup> T Cells in Hepatocellular Carcinoma Patients

        Aims: Dysfunction of the host immune system in cancer patients can be due to a number of factors, including lymphocyte apoptosis. Several studies showed that $Foxp3^+T$ cells take part in inducing this process by expressing FasL in tumor patients. However, the relationship between apoptosis, $CD8^+T$ cells and $Foxp3^+T$ cells in HCC patients is still unclear. The present study was designed to investigate the correlation between apoptosis levels and Fas/FasL expression in $CD8^+T$ lymphocytes and $Foxp3^+T$ cells in patients with HCC. Methods: $CD8^+T$ cells and $CD3^+Foxp3^+T$ cells were tested from peripheral blood of HCC patients and normal controls and subjected to multicolor flow cytometry. The expression of an apoptosis marker (annexin V) and the death receptor Fas in $CD8^+T$ cells and FasL in $CD3^+Foxp3^+T$ cells were evaluated. Serum TGF-${\beta}1$ levels in patients with HCC were measured by enzyme-linked immunosorbent assay. The relationship between apoptosis and Fas expression, as well as FasL expression in $CD3^+Foxp3^+T$ cells was then evaluated. Results: The frequency of $CD8^+T$ cells binding annexin V and Fas expression in $CD8^+T$ cells, were all higher in HCC patients than normal controls and the proportion of apoptotic $CD8^+T$ cells correlated with their Fas expression. Serum TGF-${\beta}1$ levels correlated inversely with $CD3^+Foxp3^+T$ cells. Conclusions: Fas/FasL interactions might lead to excessive turnover of $CD8^+T$ cells and reduce anti-tumor immune responses in patients with HCC. Further investigations of apoptosis induction in $Fas^+CD8^+T$ cells in vitro are required.

        A Facile Method to Fabricate Bioenvironmentally Friendly Janus Nonwoven Medical Covers: Preparation and Property Evaluation

        Medical product contamination is a serious threat to patients’ health. However, designing medical covers thatensure Janus performance and outstanding abrasion resistance, mechanical performances has remained a great challenge. Inthis study, a novel Janus nonwoven fabric nonwoven fabric consists of a hydrophilic inner layer of polylactic acid (PLA)/lowmeltingpolylactic acid (LPLA) and a coated outer layer of hydrophobic thermoplastic polyurethane (TPU) is successfullyfabricated. The subsequent PLA/LPLA-TPU textiles exhibits excellent protective performance of inner absorption. Moreover, the coating improves tensile strength and increases abrasion resistance. A mechanism of mechanical failure isfurther studied. This kind of nonwoven fabric is suggested to be a promising candidate for medical covers and health supplies.
