        LncRNA A2M-AS1 lessens the injury of cardiomyocytes caused by hypoxia and reoxygenation via regulating IL1R2

        Background: Myocardial ischemia and reperfusion injury (MI/RI) is a complex pathophysiological process, which can lead to severe myocardial injury. The long noncoding RNA alpha-2-macroglobulin antisense RNA 1 (A2M-AS1) has been revealed to be abnormally expressed in MI, However, its function in MI and the potential mechanism are still unclear. Objective: To evaluate the functional role of A2M-AS1 in hypoxia/reoxygenation (H/R)-induced neonatal cardiomyocytes and its potential molecular mechanism. Methods: Dataset GSE66360 was obtained from GEO database for analyzing the RNA expression of A2M-AS1 and interleukin 1 receptor type 2 (IL1R2). KEGG pathway enrichment analysis of the genes that co-expressed with A2M-AS1 was performed. Human neonatal cardiomyocytes were subjected to H/R to construct in vitro models. QRT-PCR and Western blot were adopted to test the levels of mRNA and protein. The viability and apoptosis of cardiomyocytes were tested by CCK-8 and flow cytometry assays, respectively. Results: The expression of A2M-AS1 was notably downregulated in H/R-treated cardiomyocytes. Overexpression of A2M-AS1 can notably enhance the cell viability of H/R-damaged cardiomyocytes, whereas knockdown of A2M-AS1 showed the opposite outcomes. Besides, a negative correlation was showed between A2M-AS1 and IL1R2 expression. In H/R-treated cardiomyocytes, overexpression of IL1R2 weakened the promoting proliferation and anti-apoptosis effects caused by overexpressing A2M-AS1, however, IL1R2-knockdown abolished the anti-proliferation and pro-apoptosis effects caused by silencing A2M-AS1. Conclusion: This study demonstrates the potential regulatory role of A2M-AS1/ IL1R2 axis in cardiomyocytes suffered from H/R, and provides insight into the protection of MI/RI.

        MicroRNAs (miRNAs) are endogenous non-coding genes that participate in post-transcription regulation by either degrading mRNA or blocking its translation. It is considered to be very important in regulating insect development and metamorphosis. Insects are the largest group of animals and are extremely valuable in biological and agriculture research. Insects are also important pests to human health and agriculture, and efforts are necessary protect both humans and plants from disease and damage. Despite their importance, insects lag behind mammals, nematodes, and plants in miRNA research. At present, only 279 insect miRNAs have been identified from Drosophila melanogaster, Anopheles gambiae, Apis mellifera, Bombyx mori, and D. pseudoobscura in miRBase, and most of these miRNAs were computationally predicted without experimental validation. Functional analysis of insect miRNAs has only been conducted in D. melanogaster.

        A Facile Method to Fabricate Bioenvironmentally Friendly Janus Nonwoven Medical Covers: Preparation and Property Evaluation

        Medical product contamination is a serious threat to patients’ health. However, designing medical covers thatensure Janus performance and outstanding abrasion resistance, mechanical performances has remained a great challenge. Inthis study, a novel Janus nonwoven fabric nonwoven fabric consists of a hydrophilic inner layer of polylactic acid (PLA)/lowmeltingpolylactic acid (LPLA) and a coated outer layer of hydrophobic thermoplastic polyurethane (TPU) is successfullyfabricated. The subsequent PLA/LPLA-TPU textiles exhibits excellent protective performance of inner absorption. Moreover, the coating improves tensile strength and increases abrasion resistance. A mechanism of mechanical failure isfurther studied. This kind of nonwoven fabric is suggested to be a promising candidate for medical covers and health supplies.

        Enhancing Astaxanthin Accumulation in Haematococcus pluvialis by Coupled Light Intensity and Nitrogen Starvation in Column Photobioreactors

        Natural astaxanthin mainly derives from a microalgae producer, Haematococcus pluvialis. The induction of nitrogen starvation and high light intensity is particularly significant for boosting astaxanthin production. However, the different responses to light intensity and nitrogen starvation needed to be analyzed for biomass growth and astaxanthin accumulation. The results showed that the highest level of astaxanthin production was achieved in nitrogen starvation, and was 1.64 times higher than the control group at 11 days. With regard to the optimization of light intensity utilization, it was at 200 μmo/m2/s under nitrogen starvation that the highest astaxanthin productivity per light intensity was achieved. In addition, both high light intensity and a nitrogen source had significant effects on multiple indicators. For example, high light intensity had a greater significant effect than a nitrogen source on biomass dry weight, astaxanthin yield and astaxanthin productivity; in contrast, nitrogen starvation was more beneficial for enhancing astaxanthin content per dry weight biomass. The data indicate that high light intensity synergizes with nitrogen starvation to stimulate the biosynthesis of astaxanthin.

        A Putative Histone Deacetylase Modulates the Biosynthesis of Pestalotiollide B and Conidiation in Pestalotiopsis microspora

        Fungi of the genus Pestalotiopsis have drawn attention for their capability to produce an array of bioactive secondary metabolites that have potential for drug development. Here, we report the determination of a polyketide derivative compound, pestalotiollide B, in the culture of the saprophytic fungus Pestalotiopsis microspora NK17. Structural information acquired by analyses with a set of spectroscopic and chromatographic techniques suggests that pestalotiollide B has the same skeleton as the penicillide derivatives, dibenzodioxocinones, which are inhibitors of cholesterol ester transfer protein (CETP), and as purpactins A and C’, inhibitors of acyl-CoA:cholesterol acyltransferase (ACAT). Strain NK17 can make a fairly high yield of pestalotiollide B (i.e., up to 7.22 mg/l) in a constitutive manner in liquid culture. Moreover, we found that a putative histone deacetylase gene, designated as hid1, played a role in the biosynthesis of pestalotiollide B. In the hid1 null mutant, the yield of pestalotiollide B increased approximately 2-fold to 15.90 mg/l. In contrast, deletion of gene hid1 led to a dramatic decrease of conidia production of the fungus. These results suggest that hid1 is a modulator, concerting secondary metabolism and development such as conidiation in P. microspora. Our work may help with the investigation into the biosynthesis of pestalotiollide B and the development for new CETP and ACAT inhibitors.

        The Effect of Transformation on the Virulence of Streptococcus pneumoniae

        Although pneumococcus is one of the most frequently encountered opportunistic pathogen in the world, the mechanisms responsible for its infectiveness have not yet been fully understood. In this paper, we have attempted to characterize the effects of pneumococcal transformation on the pathogenesis of the organism. We constructed three transformation-deficient pneumococcal strains, which were designated as Nos. 1d, 2d, and 22d. The construction of these altered strains was achieved via the insertion of the inactivated gene, comE, to strains 1, 2 and 22. We then conducted a comparison between the virulence of the transformation-deficient strains and that of the wild-type strains, via an evaluation of the ability of each strain to adhere to endothelial cells, and also assessed psaA mRNA expression, and the survival of hosts after bacterial challenge. Compared to what was observed with the wild-type strains, our results indicated that the ability of all of the transformation-deficient strains to adhere to the ECV304 cells had been significantly reduced (p < 0.05), the expression of psaA mRNA was reduced significantly (p < 0.05) in strains 2d and 22d, and the median survival time of mice infected with strains 1d and 2d was increased significantly after intraperitoneal bacterial challenge (p < 0.05). The results of our study also clearly indicated that transformation exerts significant effects on the virulence characteristics of S. pneumoniae, although the degree to which this effect is noted appears to depend primarily on the genetic background of the bacteria.

        Antioxidant Enzymes and Photosynthetic Responses to Drought Stress of Three Canna edulis Cultivars

        Edible canna is a productive starch source in some tropical and semitropical regions. In these regions, water deficit stress is one of factors that limit the crop yield. In the present study, we investigated seven physiological indexes and photosynthetic responses of three edible canna (Canna edulis Ker.) cultivars ('PLRF', 'Xingyu-1', and 'Xingyu-2') under 35 days drought stress. Our results indicated that drought treatment caused visible wilting symptoms in all cultivars, especially in 'Xingyu-1'. Coupled with the increase of wilting symptoms, relative water content (RWC) and chlorophyll content decreased progressively, malondialdehyde (MDA) content gradually increased, and key antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) activities increased first and then decreased in all three cultivars. The effect of water stress was more pronounced in 'Xingyu-1' than in 'PLRF' and 'Xingyu-2', and in lower leaves than in upper leaves. In addition, 35 days drought stress also significantly reduced the photosynthetic capacity. Consistent with antioxidant parameters, photosynthetic changes of 'Xingyu-2' were less than those of the other cultivars under water deficit stress. Drought stress caused a significant increase of water use efficiency (WUE) in 'Xingyu-2', but little in 'PLRF', and obvious decrease in 'Xingyu-1'. These results indicated that 'Xingyu-2' was more tolerant to drought stress than 'PLRF' and 'Xingyu-1' by maintaining lower lipid peroxidation and higher antioxidant enzyme activities.

      • Lack of Influence of an XRCC3 Gene Polymorphism on Oral Cancer Susceptibility: Meta-analysis

        Background: To systematically summarize the association between the X-ray repair cross complementing 3 (XRCC3) gene polymorphism and oral cancer susceptibility by meta-analysis. Materials and Methods: Databases including PubMed, EMbase, CNKI, VIP and WanFang Data were searched to identify case-control studies concerning the association between an XRCC3 gene polymorphism and the risk of oral cancer from the inception to June 2014. Two reviewers independently screened the literature according to the criteria, extracted the data and assessed the quality. Then meta-analysis was performed using Stata 11.0 software. Results: Seven published case-control studies including 775 patients with oral cancer and 1922 controls were selected. Associations between the rs861539 polymorphism and overall oral cancer risk were not statistically significant in all kinds of comparison models (CT vs CC: OR=0.94, 95%CI=0.74-1.18; TT vs CC: OR=0.94, 95%CI=0.64-1.38; dominant model: OR=0.95, 95%CI=0.76-1.18; recessive model: OR=0.94, 95%CI=0.69-1.29; allele T vs C: OR=0.97, 95%CI=0.84-1.11). In the stratified analysis by ethnicity, no significant associations were found among Asians and Caucasians. On stratification by tumor type, no significant associations were found for cancer and oral premalignant lesions. Conclusions: The XRCC3 gene polymorphism was not found to be associated with the risk of oral cancer. Considering the limited quality of the included case-control studies, more high quality studies with large sample size are needed to verify the above conclusion.

        High Efficiency Onboard Charger Based on Two-Stage Circuit

        On-board charger (OBC) is key part of electric vehicles. Limited to space and weight, design objectives of OBC are high power-density and high effi ciency. Two-stage circuit is commonly used for 3.3 kW OBC, interleaved power factor correction (ILPFC) is utilized for power factor correction and DC bus voltage regulation, LLC resonant converter is utilized for voltage and power regulations. In this paper, the relationships between the internal parameters and effi ciency of ILPFC are studied by discrete iterative method, and the internal parameters are optimized to improve ILPFC`s effi ciency. Meanwhile, the relationships between the resonant parameters and effi ciency of LLC converter are also studied by fundamental harmonic approximation method to optimize the effi ciency in wide charging voltage. A 3.3 kW OBC prototype is developed to verify the eff ectiveness and correctness of the optimal method, the power factor and total harmonic distortion at full-load state are about 99.99% and 2.98% with the charging voltage ranging from 230 to 430 V, respectively
