        RESEARCH ARTICLE: Grafting-Induced Gene Expression Change in Brassica rapa Leaves is Different from Fruit Trees

        Grafting has widely used in the agriculture of fruit-bearing crops and trees because rootstocks have a profound influence on many aspects of scion development and scion responses to the environmental changes including biotic and abiotic stress tolerance. These effects appear to result from the change of gene expression on scion, but only limited numbers of papers have been published demonstrating it. To identify altered expression of genes in Chinese cabbage, Chiifu(Brassica rapa ssp. pekinensis, inbred line) shoot was grafted on three Brassica rootstocks: mustard, turnip and broccoli. After head formation, Br300K microarray experiment was conducted using total RNAs from scion leaves collected in two different seasons, spring (June) and fall (October). A large number of differentially expressed genes (DEGs) were identified both in two seasonal samples, but DEGs were more notable in June sample than in October sample. However, the number of DEGs by three rootstocks were high in October with respect to up-regulation, but high in June for down-regulation. Categories of DEGs included metal ion binding, response to hormonal stimuli, response to endogenous stimuli, regulation of transcription, oxidation reduction and response to stress. Up-regulated genes in both June and October samples were similar in mustard and turnip rootstocks, but different in broccoli rootstock. Two genes were found to respond to all experimental conditions: Brapa_ESTC049008 (hypothetical protein) as an up-regulated gene and Brapa_ESTC016027 (CNGC12) as a down-regulated gene. Together with the previous reports, these results suggest that grafting-induced gene expression depends on the species involved.

      • Gene Identification and Molecular Marker Development Related to High-Temperature Tolerance in Cabbage

        To select genes associated with the high-temperature tolerance from Brassica, two transcriptomic analyses have been used: microarray and RNA Seq. Using two contrasting inbred lines of B. rapa, Chiifu and Kenshin, version 3 microarray (135 K microarray) was conducted to RNA samples extracted from series of 45℃-treated leaves and 29 genes were selected for genomic DNA cloning of cabbage. Of 29 genes, 8 genes contain 40 SNPs, 11 SSRs and 23 In-Del markers that distinguish high-temperature tolerant and susceptible cabbages, BN1 and BN2. These 8 genes include a unknown gene, AP2, SMP, FBD, SKP2B, IAA16, HSP21 and OLI2-2. We also selected 16 cabbage genes from RNA Seq analysis using two inbred lines, BN1 and BN2: 5 genes for BN1-high expression, 5 genes for BN1-specific expression, 5 genes for BN2-specific expression, and BoCaMB. Using RNA sequences, genomic DNAs corresponding to 16 genes have been clones and analyzed to find out molecular markers. Markers were further transformed into PCR-based marker and confirmed with additional cabbage genetic lines. We are currently transforming PCR-makers into SNP markers. To examine function of high-temperature tolerant genes, we also transformed 5 genes into Arabidopsis plants. We will describe detailed methods and results in a poster.         Organic Nutrition and Gene Expression in Different Tissues of Chinese Cabbage

        To evaluate the nutritional value of Chinese cabbage, carbohydrate, vitamin C, carotenoid and free amino acid levels were measured in six different tissues (leaf and midrib of outer leaves, leaf and midrib of middle leaves, and leaf and midrib of inner leaves) of fully-headed Chinese cabbage. Gene expression associated with the biosynthesis of these compounds was also examined. High levels of starch and vitamin C were detected in the leaves while midrib parts contained low levels. The outer leaf parts contain the lowest level of glucose and sucrose, in contrast to high levels observed in inner parts. However, the outer leaves contained the highest content of carotenoids. While Chinese cabbage contains high levels of Glu, Ala, Asp, and Arg, the levels of most essential amino acids are very low. Transcript levels tested in this study did not correspond to nutrient levels, implying biosynthesis of organic nutrients lacks regulation at the transcriptional level. These results collectively show that although the outer leaves of Chinese cabbage contain more health compounds such as vitamins and carotenoid, the inner leaves are more nutritious.

        솔수염하늘소(Monochamus alternatus)에 대한 수종 살충제의 살충효과

        To develop efficient control program against Japanese pine sawyer (JPS). Monochamus alternatus Hope, laboratory bioassay using 7 insecticides, and the residual effects of fog-machine treatment and ultra-low-volume spray by helicopter of fenitrothion induced 100% mortality 4 days after exposure (DAE), and exposure for 4 minutes did that 2 DAE. Even the periods when they were alive, they did not feed on treated P. densiflora shoots. All the topical applications of the fenitrothion on elytra, head, mouth, tarsus, and ventral parts show 100% mortality. The residual effect of fog-machine treatment of fenitrothion was very effective against JPS adults. When the adults were inoculated on the treated shoots at the 1st day of treatment with 50,000 and 5,000 ppm solution of fenitrothion, the LT_(100) was only 1 day; even at the 11th day it was only 3 days. The residual efficacy of ultra-low-volume spray with helicopter lasted longer than that of fog-machine treatment. When the JPS adults were inoculated on the sprayed shoots even at the 10th day of the spray with 60,240, 5,000 and 1,000 ppm, the LT_(100) was only a day.

      • 요와 농 검체에서의 Vancomycin 내성 장구균 분리 2예

        65세 여자 환자의 요 검체에서 vancomycin 내성 E. faecalis를, 욕창의 농에서 vancomycin 내성 E. faecium을 분리하였는데, 환자의 병력으로 미루어 미국에서 정착된 세균에 의하여 감염되었을 것으로 생각되었다. 분리 균주에 대한 vancomycin의 MIC는 각각 258과 32㎍/mL, teicopoanin은 0.25, 0.5㎍/mL이어서 VanB 표현형으로 추측되었다. 우리 나라에서의 VRE 확산을 예방하기 위한 조치의 일환으로서 의사를 포함한 병원 직원에게 VRE 감염증의 심각성을 홍보하여야 하며, 미생물검사실에서는 임상검체에서 분리된 장구균의 vancomycin 내성을 시험하고 장내 VRE 보균자의 주기적 검색이 필요하다고 사료되었다. Vancomycin-resistant Enterococcus faecalis and E. faecium were isolated from urine and decubitus ulcer discharge, respectively, from a 65-year-old woman. Proir to the transfer to this hospital in December, 1995, she had been admitted to the Chicago Rehabilitation Center in the U.S. for the treatment of olivopontocerebellar atrophy, quadriplegia, and neurogenic bladder. She was also treated with multiple antimicrobials including vancomycin for pneumonia at intensive care unit, which suggested that she acquired the organisms in the U.S. The minimal inhibitory concentrations of vancomycin and teicoplanin against the isolates were: E. faecalis 256 ㎍/mLand 0.5㎍/mL; E. faecium 32 ㎍/mL and 0.5 ㎍/mL, respectively. The susceptibility patterns suggest the isolates are vanB type. Although other species of vancomycin-resistant Enterococcus spp. were reported previously in Korea, these isolates may be the first vancomycin-resistant E. faecalis and E. faecium from clinical specimens.

        4 Dipyridamole (99m)Tc-MIBI SPECT의 육안 분석과 정량적 분석

        연구배경 : 관상동맥 질환을 진단하고 병소의 위치를 결정하거나 그 기능적 의의를 평가하는 비관혈적인 검사법으로 심근 SPECT의 유용성이 널리 인정되고 있다. 저자들은 심근 SPECT의 육안 분석과 정량적 분석방법의 진단적 우위를 비교하기 위하여 본 연구를 시행하였다. 방법 : 허혈성 심장 질환을 의심하여 심근 SPECT와 관상동맥 조영술을 모두 실시한 126명의 환자를 연구 대상으로 하였다. Dipyrdamole 0.56mg/kg을 정주하고 ^(99m)Tc-MIBI 15mCi를 투여한 후 rotating gamma camera를 이용하여 부하 스캔을 구하였고, 4시간 후 ^(99m)Tc-MIBI 25mCi를 다시 투여한 후 재분포 스캔을 시행하였다. 단층 영상을 3방향으로 재배치하여 육안적으로 분석하였다. Technetium 흡수의 분포도로부터 전체 좌심실을 대표하는 2차원의 극지도를 그리고, CSMC에 의해 개발된 정상치와 비교함으로써 최종적인 병변 극지도를 구하여 정량적으로 분석하였다. 관류 결손의 재관류 여부를 육안분석으로 완전 재관류, 부분 재관류, 무 재관류군으로 구분하였고, 각 군에서의 정량적 관류 정도를 비교하였다. 결과 : 관상동맥 질환 진단의 전체적인 민감도는 육안 분석으로 91%, 정량적 분석으로는 94%로서 유의한 차이가 없었으며, 특이도는 각각 76% 및 71%로 역시 유의한 차이가 없었다. 개개의 관상동맥 분지의 병변을 육안 및 정량적 분석으로 발견할 수 있는 민감도는 좌전하행지가 각각 89% 및 92%, 우관상 동맥이 84% 및 93%, 좌회선지가 71% 및 77%로 모든 혈관에서 육안 분석보다 정량적 분석이 높았으나 유의한 차이는 없었다. 육안 및 정량적 분석의 특이도는 좌전하행지가 각각 79% 및 70%, 우관상 동맥이 77% 및 82%, 좌회선지가 96% 및 92%로 유의한 차이는 없었다. 육안 분석에 관류 결손의 완전 재관류는 협심증군에서 심근경색증군보다 유의하게 높았다. 육안분석에 의하여 분류된 각 군의 정량적 분포 정도는 완전 재관류군과 무재관류군 사이나, 부분재관류군과 무재관류군 사이에서는 유의한 차이가 있었으나, 완전 재관류군과 부분재관류군 사이에서는 유의한 차이가 없었다. 결론 : Dipyridamole 정주 후 ^(99m)Tc-MIBI SPECT검사의 정량적 분석이 육안 분석보다 더욱 정확한 분석 방법은 아닌 것으로 생각되며, 재관류 평가의 임상적 중요성에 비추어 볼 때 병변의 심한 정도와 재관류의 정도를 정량화 하는 방법에 대한 연구가 더욱 진행되어야 할 것으로 사료된다. Background: The usefulness of myocardial SPECT in diagnosing coronary artery disease, & assessing the functional significance of the coronary stenosis is confirmed by many investigators. To determine the superiority of the quantitative analysis method over the visual analysis method in assessing myocardial SPECT this study was performed. Methods: One hundred & twenty six patients with suspicious coronary artery disease had both intravenous dipyridamole ^(99m)Tc-MIBI SPECT and coronary angiougraphy within an interval of 5 days. The scintigraphic findings were assessed visually and quantitatively using two dimensional polar map. The results of visual and quantitative assessments were compared to the coronary angiographic findings. In 85 patients who showed perfusion defect on stress scintigraphy, the reversibility of the defect was evaluated by visual analysis. Results: For detection of coronary artery disease, the overall sensitivity of the visual and quantitative analyses was 91% & 94% respectively (p>0.05). The overall specificity was 76% by visual and 71% by quantitative analyses(p>0.05). For detection of individual coronary branch diseases, the quantitative analysis was higher than visual analysis in the sensitivity(LAD: 89% vs. 92%, RCAl 84% vs.93%, LCx: 71% vs. 77%), but the differences were not statistically significant. The specificities of both visual and quantitative analyses were similary high(LAD: 79% vs. 70%, RCA: 77% vs. 82%, LCx: 96% vs. 92%), but there were no significant differences also. The reversibility of perfusion defect was assessed by visual analysis. Of the 144 abnormal vascular territories in 85 patients, 49 vascular territories showed complete redistribution and 30 territories showed partial redistribution. The patients with angina had significantly higher percentage of complete redistribution (angina: 50% vs. MI: 15%, P<0.05). Conclusion: Our results showed that quantitative analysis of ^(99m)Tc-MIBI tomography may not be more accurate technique than visual analysis for detection of coronary artery disease and for localization of individual coronary stenoses.

        자가면역성 만성 췌장염 17예의 임상상

        Background/Aims: Autoimmune pancreatitis is a distinctive type of chronic pancreatitis with reversibility. We analyzed the largest number of patients with autoimmune pancreatitis among the studies carried out at a single institution in Korea. Methods: We retrospectively analyzed the clinical, radiologic, laboratory and histologic features in 17 patients who were diagnosed as autoimmune pancreatitis. Results: The patients were predominately elderly men who presented with jaundice or nonspecific gastrointestinal symptoms. In most of the patients, serum pancreatic enzymes were normal or mildly elevated with an accompanying cholestatic biochemical profile. Elevated IgG levels and detection of autoantibodies were observed in 47% (8/17) and 35% (6/17), respectively. Thirteen patients showed histologic findings of lymphoplasmacytic infiltration and fibrosis, and one patient showed predominant infiltration of eosinophils. Fifteen patients revealed radiologic images of diffuse swelling of pancreas and the sclerosing pattern on direct pancreatogram. All of the features improved in response to oral steroid therapy. Two cases showed focal narrowing with upstream duct dilatation and thus, they were diagnosed as focal type. Diabetes mellitus concurred in 13 cases and primary sclerosing cholangitis and Sjoren`s syndrome were present in 1 case, respectively. Conclusions: The recognition of autoimmune pancreatitis allows patients to avoid unnecessary surgery and recover with the administration of oral steroid. (Korean J Gastroenterol 2004; 43:112-119)
