        Inhibition of Proinflammatory Cytokine Generation in Lung Inflammation by the Leaves of Perilla frutescens and Its Constituents

        This study was designed to find some potential natural products and/or constituents inhibiting proinflammatory cytokine generation in lung inflammation, since cytokines such as tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) are pivotal for provoking airway inflammation. In our preliminary screening procedure, the 70% ethanol extract of the leaves of Perilla frutescens (PFE) was found to clearly inhibit TNF-α production in the lung at 100 mg/kg, after intranasal lipopolysaccharide treatment of mice. Based on this result, ten constituents including phenylpropanoids (allyltetramethoxybenzene, caffeic acid, dillapiole, elemicin, myristicin, nothoapiole, rosmarinic acid methyl ester, rosmarinic acid) and monoterpenes (perilla aldehyde and perilla ketone) were successfully isolated from the extract. Among them, elemicin and myristicin were found for the first time to concentration-dependently inhibit IL-1β-treated IL-6 production from lung alveolar epithelial cells (A549) at concentrations of 10-100 μM. These findings suggest that the phenylpropanoids including elemicin and myristicin have the potential to be new inhibitory agents against lung inflammation and they may contribute, at least in part, to the inhibitory activity of PFE on the lung inflammatory response.

      • 광 산란 실험에 의한 pH 변화가 쪽거리 구조에 미치는 영향 연구

        광 산란 실험을 토대로 두가지 다른 염(NaCl)의 농도를 가지는 폴리스티렌(polystyrene) 수용액에서는 응집과정 동역학을 pH의 변화에 따라 조사하였다. 염의 농도가 3 mol/l인 폴리스티렌 수용액에서는 pH에 관계없이 쪽거리(fractal) 차원은 1.75로 주어지고 응집체의 성장형태는 확산 극한 무더기 무더기 응집(diffusion-limited cluster-cluster aggregation: DLCA)모델과 일치하지만, 염의 농도가 묽은 0.3 mol/l인 폴리스티렌 수용액에서는 응집체의 응집과정은 pH에 따라 느리게 또는 빠르게 진행된다. 산성인 pH 4와 pH 5인 경우에서는 쪽거리 차원은 1.81±0.05와 1.79±0.05로 각각 주어진다. 이런 경우에서는 응집과정이 빠르게 진행되고 DLCA 모델과 일치하는 결과를 주는 반면에, 중성인 pH 7과 염기성인 pH 11과 pH 12인 경우에서의 쪽거리 차원은 오차 범위내에서 2.05로 주어지고 응집과정은 느리게 진행된다. 이것은 반응 극한 무더기 무더기 응집(reaction-limited cluster-cluster aggregation)모델과 일치함을 알 수 있다. On the basis of light scattering experiments, the kinetics of the aggregation process in water solutions of polystyrene particles induced at two different salt concentrations (3 mol/liter and 0.3 mol/liter) was investigated for various pH values. for the water solution of polystyrene particles induced at the high salt concentration (3 mol/liter), the fractal dimension was given by 1.75, irrespective of the pH value, and the growth of clusters agreed with the diffusion-limited cluster-cluster aggregation (DLCA) model. However, for the case of the low salt concentration (0.3 mol/liter), aggregation of clusters took place either slowly or rapidly, depending on the pH value. The fractal dimensions for pHs 4 and 5 were given by 1.81±0.05, and 1.79±0.05, respectively. In this case, the kinetic growth process took place rapidly, which agreed with DLCA model. On the other hand, the fractal dimensions for pH 7 and pHs 11 and 12 were all given by 2.10 within the range of errors, and clusters grew slowly, which agreed with the reaction-limited cluster-cluster aggregation model.

        일반계 고등학교 교사의 혁신 교육 참여경험

        이 연구는 일반계 고등학교의 혁신 교육 현장을 이해하기 위한 것이다. 이를 위해 연구진은 대규모 혁신 고등학교 방문, 전문적 학습공동체와 진로 탐색 프로그램 관찰, 혁신학교 참여 교사 10명과의 개별 면접을 진행하였다. 그 결과 교사들의 교육과정 경험은 학생주도권 강화 노력 및 자원 확보와 근무여건 악화에 대한 고민으로 구성되었다. 수업경험은 학력 격차 해소 가능성 발견 및 재미와 학력 성장이 함께 이루어지는 수업의 가능성 발견이었다. 평가 경험은 과정 중심 수행평가 시행의 고달픔과 공정성 확보의 어려움 및 교육적 효과 확인 등으로 구성되었다. 학교 풍토는 다양한 시도에 대한 적극적 지원과 반론이 허용되는 관계로 경험되었다. 혁신학교 운영을 통한 교사의 성장과 좌절 경험은 교사로서의 자존감 회복, 변화에의 용기, 전문성 강화와 혁신 교육의 가치에 대한 재다짐 등을 포함하였다. 이 연구는 일반계 고등학교 혁신 교육 참여 교사의 이야기를 정리하였다는 점에서 의의가 있다. This study is to understand innovative education in general high schools. To this end, researchers visited the school, observed professional learning communities and career programs, and interviewed with ten teachers participating in innovative education. The curriculum experiences of the teachers consisted of efforts to strengthen student initiative, worries about securing resources and worsening working conditions. The class experience was to discover the possibility of overcoming the academic gap between students, classes with fun and academic growth. The evaluation experience consisted of the difficulty of implementing a process-based performance evaluation, securing fairness, and verifying educational effects. The school climate was experienced as active support for various attempts and being allowed for a counterargument. Finally, the teacher s growth and frustration experiences included recovering self-esteem, strengthening expertise, courage for change, and reaffirming innovative education’s value. This study is meaningful because it summarizes the stories of teachers participating in innovative education in general high school.

        Factors to Assess for Re-breeding after Parturition in Milking Cows

        The purpose of this study was to assess for re-breeding concentrate period in postpartum in milking cows. The 48 cows aged 3.5~5.5 years and of 400~600 kg body weight were examined every 3rd day from 15 to 36 day postpartum. Blood samples for progesterone and estradiol 17 β hormone analyses were withdrawn from the coccygeal vein every third day until the end of the experiment. The ovarian follicular numbers were verified and measured using a multi frequency probe. The least squares means are presented for each day by GLM of SAS. The results showed that ovary lengths (right ovary; 1.64±0.62 cm, left ovary; 1.44±0.46 cm) were similar in right and left ovary activity level during estrous cycle of postpartum cows. We were judged completed uterus on day at 2.31±0.17 cm level of cervix diameter. And we were monitoring started at 6.44±2.03 cm from day 15 after postpartum. The results showed that mean plasma concentration of progesterone (3.28 ng/ml) in large follicle gradually increased days 30 in postpartum. And, monitoring of estradiol 17β (22.18 pg/ml) hormone during postpartum period would be useful to predict the ovarian and uterus activity for re-breeding in postpartum milking cows. From these results, we conclude that cervix diameter (mean: 2.31 cm) was very important for reproductive organ recovery standard level of postpartum milking cows, hormone secretion level (P4: 3.28 ng/ml, E2: 22.18 pg/ml) and body condition score (2.5~2.75) level about 30 days in postpartum period.

        한국(韓國)의 강릉단오제(江陵端午祭)와 중국(中國)의 반한(反韓) 정서(情緖)

        By the modern and present the Korean-Chinese relations which were severed show activity as never before since the establishment of diplomatic relations in 1992. Close relationship between Korea and China are reaping a great success in many fields such as not only economy but also cultural and private exchanges. However, there are a lot of dissonance in the rear of the friendly relationship between two countries. The historical disputes between Korean and Chinese, which has become an issue after ‘Gangneung Dano Festival’ of South Korea was designated as world Cultural Heritage in 2005, were provoked into anti-korean sentiment in China. Even now many Chinese people consider that Korea plunders Chinese culture. Thus, the paper examines ‘Korean Genesis Theory’ which mainly disclosing chinese anti-korean sentiment in Korean-Chinese historical disputes. Even though most of the claims have been turned out fabricated falsehood by certain Chinese, these claims have already spreaded throughout many Chinese, and distorting the image of Korea. Next, it analyzes the phenomenon of anti-Korean sentiment, and tries to find the cause and solution. Through the past history, it also looks for the origin of the historical disputes between the two countries. Finally, it suggests several proposals for resolving Korea-China historical disputes and conflicts.

        High dose Methotrexate 의 Pharmacokinetics 와 진행된 High dose Methotrexate , 5 - Fluorouracil 및 Adriamycin 복합화학요법

        N/A The folic acid antagonist, methotrexate (MTX), was shown to have antitumor activity due to inhibition of the enzyme dihydrofolate reductase. In the last two decades, the drug has been administered in conventional doses of 30 ㎎/㎡, however, during the last 10 years, very high doses of methotrexate in the range of 1-10 g/㎡, coupled with citrovorum factor rescue have been found to be strikingly more effective for chemotherapy of osteogenic sarcoma, malighant lymphoma, and squamous carcinoma of the head and neck. Klein (1983) reported a high response rate (63%) by MTX and 5-fluorouracil (5-FU) combination chemotherapy in cases with advanced stomach cancer. In Korea, the incidence of stomach cancer is higher than other tumors, and the usual chemotherapy program for advanced stomach cancer is a 5-fluorouracil, adriamycin and mitomycin-C combination regimen (FAM); but there has been no pharmacokinetic study or clinical application of MTX. This paper presents a pharmacokinetic and clinical study of 18 cases with advanced stomach cancer, admitted from January 1986 to December 1986. The results were as follows: 1) After intravenous infusion of MTX 1.5 ㎎/㎡, the plasma MTX levels were recorded as follows: At 30 min after discontinuation of MTX infusion, the plasma MTX level was 7.17×10-5±3.63×10-5 M, At 6 hours, it was 1.38×10-5±0.75×10-5 M; at 12 hours, 3.71×10-6±2.23×10-6 M; at 24 hours, 9.30×10-7±1.02±10-7 M; at 48 hours, 1.99×10-7±1.70×10-7 M; and at 72 hours, 1.27×10-7±1.25±10-7 M. Forty-eight hours after discontinuing the drug, the plasma level of MTX fell below 1×10-7 M in half of the cases. After 72 hours, 66.6% of the cases fell below the 1×10-7 M level. 2) The response rate for the HDMTX, 5-FU and ADR combination chemotherapy was 11.2% in all cases. One case (9.1%) out of 11 cases with previous chemotherapy, showed a partial response; also out of 7 case without previous chemotherapy, one case (14.2%) showed a partial response. 3) Toxicities with high dose MTX were: pancytopenia (1 case), nausea and vomiting (14 cases), stomatitis (8 cases), skin rash (2 cases) and diarrhea (2 cases). In our study the patient with pancytopenia died due to sepsis. There was only one case with an elevated creatinine level. In summary, the results of this study for pharmacokinetic changes of that MTX were similar to other reports. However, over 72 hours after discontinuation of high dose MTX and 5-FU combination chemotherapy, the risk of toxicity is reduced by the administration of CF. The response rates of HDMTX, 5-FU and ADR combination chemotherapy were low; therefore further studies should be done to elucidate a more exact response rate.

      • Medicinal Chemistry : Research Article ; Methyl Protodioscin from the Roots of Asparagus cochinchinensis Attenuates Airway Inflammation by Inhibiting Cytokine Production

        The present study was designed to find pharmacologically active compund against ariway in flammation from the roots of Asparagus cochinchinensis. The 70% ethanol exreact of the roots of A. cochinchinsis (ACE)was found to inhibit IL 6 production from IL_1B treated lung epithelial cells(A549)and the major constiuent, methyl protodiscin (MP), also stronglay inhibited the production dif IL-6,IL-8, and tumor neceosis factir (TNF) a from A549cellsat10-100uM. Thisdownregulating effect of proimflammatory cytikine pridyction was found to be mediated, at least in part,via inhibition of c-Jun N-terminal kinase(JNK) and C-Jun activatuib pathway. When examined in an in vivo mofrl of airway inflammation in mice, lipopolysaccharide-(LPS) induced acute lung injury, ACE, and MP significantly inhibited cell infiltration in the bronchialveolar lavage fluid by the oral treatment at doses fo 100-400mg/kg and 30-60mg/kg, respectively.MP also inhibited the production of Proinflammatory cytikines such as 1L-6, TNF-a, and 1L-1B in l ung tissue. All of these findings provide scientific evjdence supporting the role of A. cochinchinisi as a herbal remedy in treating ariway intlammation and also suggest a therapeutic balue of MP on airway inflammatory disriders.
