Background: Panax ginseng Meyer (P. ginseng), a herb distributed in Korea, China and Japan, exerts benefits on diverse inflammatory conditions. However, the underlying mechanism and active ingredients remains largely unclear. Herein, we aimed to explore the active ingredients of P. ginseng against inflammation and elucidate underlying mechanisms. Methods: Inflammation model was constructed by lipopolysaccharide (LPS) in C57BL/6 mice and RAW264.7 macrophages. Molecular docking, molecular dynamics, surface plasmon resonance imaging (SPRi) and immunofluorescence were utilized to predict active component. Results: P. ginseng significantly inhibited LPS-induced lung injury and the expression of proinflammatory factors, including TNF-a, IL-6 and IL-1b. Additionally, P. ginseng blocked fluorescence-labeled LPS (LPS488) binding to the membranes of RAW264.7 macrophages, the phosphorylation of nuclear factor-kB (NF-kB) and mitogen-activated protein kinases (MAPKs). Furthermore, molecular docking demonstrated that ginsenoside Ro (GRo) docked into the LPS binding site of toll like receptor 4 (TLR4)/myeloid differentiation factor 2 (MD2) complex. Molecular dynamic simulations showed that the MD2-GRo binding conformation was stable. SPRi demonstrated an excellent interaction between TLR4/MD2 complex and GRo (KD value of 1.16 × 10<SUP>-9</SUP> M). GRo significantly inhibited LPS488 binding to cell membranes. Further studies showed that GRo markedly suppressed LPS-triggered lung injury, the transcription and secretion levels of TNF-α, IL-6 and IL-1β. Moreover, the phosphorylation of NF-kB and MAPKs as well as the p65 subunit nuclear translocation were inhibited by GRo dose-dependently. Conclusion: Our results suggest that GRo exerts anti-inflammation actions by direct inhibition of TLR4 signaling pathway.
Functional mechanisms for diabetic nephropathy-associated genetic variants

Diabetic nephropathy (DN) is one of the major complications of diabetes. A tremendous amount of genetic variations have been identified to be associated with DN. However, most of them only generate from statistical associations at the DNA level, generally without direct functional evidence regarding their association mechanisms underlying DN. Based on the publicly available datasets and resources, this study performed integrative analyses (expression quantitative trait loci analysis, differential gene expression analysis and functional prediction analysis) to detect the molecular functional mechanisms underlying the associations for DN. Among 150 selected (P\E-4) genetic associations that were archived in the public databases, two single nucleotide polymorphisms (SNPs) (rs3135377 and rs9469220) have been found to act as cis-effect regulators of the ‘‘identified’’ gene (HLADRA and HLA-DRB1). These eQTL genes have differential expression signals in the DN-associated cell groups. These SNPs were predicted as regulatory sites by utilizing online prediction tools. Our data suggest potential mechanistic links underlying the association between DN and two identified SNPs. These results could help us to have a deeper understanding of the functional relevance of genetic variants with susceptibility to DN, which is useful for pursuit of in-depth validation studies to dissect their involvements and molecular functional mechanisms in DN.
Numerical Simulation of Spray Flow in Injector For the Steamjet Engine

In this paper, according to the work mechanism of SteamJet, With the certain small scaled turbojet for an influence, along flying trajectory calculating mass of import air and injection water, establishing the spray physics model/ mathematics model. Based on the equation of three-dimension Renault average N-S, air to adopt the limited physical volume method, the liquid adopts to the Lagrangian orbit model, with straight injecting the current method ,Simulate the field of air/ liquid two phase flow field to distribute of inlet to carry on mutually. The results indicate that spray have an effect on flow field configuration. It is concluded that the numerical model and CFD method for modeling spray flow processes in gas/liquid injector will be satisfactory. Similar to the SteamJet the United States puts forward, adopting water injection can satisfy to import the conditional request for pressure and temperature of compressor.

Purpose: Human Papillomavirus (HPV) vaccination is readily available, but immunization rate is minimal. Lack of knowledge concerning the HPV infection and vaccine is expected as the primary reason. The correlation between knowledge levels and vaccination will provide applicable guidelines for successful interventions. Method: 309 female university students have completed surveys September 2016, reporting of HPV infection knowledge, HPV vaccine knowledge, source of knowledge, and attitudes concerning vaccination. Result: HPV infection and vaccine knowledge levels were both suboptimal(9.57±1.84, 7.59±1.58), and the Internet(16.2%) was recognized as the most generally used means of obtaining information on the HPV vaccine. However, vaccination rates were reported to be low(28.2%), with lack of information(46.6%) hindering immunization. Correlations were found between HPV infection knowledge and cervical cancer insight, but the correlations between knowledge levels and vaccination rates were insignificant. Conclusion: Prior information on cervical cancer had an effect on the knowledge levels of HPV infection, but other factors were insignificant. Lack of information on the HPV vaccine was speculated, thus programs should focus on accessibility and effectiveness.
Effects of SRBSDV-infected rice plants on the fitness of vector and non-vector rice planthoppers

Southern rice black-streaked dwarf virus (SRBSDV) is a new member of the genus Fijivirus and transmitted by whitebacked planthopper (WBPH), Sogatella furcifera (Horváth). The rice area affected by SRBSDV and the subsequent damages are increasing rapidly. In current study, we evaluated the effects of SRBSDV-infected plants on the ecological fitness and the tolerance to adverse environmental factors of vector WBPH and non-vector brown planthopper (BPH), Nilaparvata lugens (Stål). Our study revealed that the fitness of WBPH and BPH were both influenced by feeding on SRBSDV-infected rice. Fecundity ofWBPH feeding on the SRBSDV-infected rice plants were higher than those on healthy plants, nymphal duration of male WBPH prolonged markedly by 0.77 d (P b 0.05), while the weight of brachypterous female adults significantly decreased (P b 0.05), and the average survival time shortened both at 26 °C and 31 °C, respectively. Median lethal time of WBPH on SRBSDV-infected and healthy rice plants were 80 h and 112 h at 26 °C, 64 h and 88 h at 31 °C, respectively. As for non-vector BPH, nymph survival rate decreased significantly by 11.22% (P b 0.05) and the longevity of female adults also shortened (P b 0.05), while hatchability of BPH eggs increased (P b 0.05). Survival time of BPH feeding on healthy rice at 26 °Cwas significant longer than those feeding on healthy and infected rice at 31 °C. The above results imply that the vectorWBPH suffers much stronger effects than non-vector BPH fromfeeding on SRBSDVinfected rice plants.

Poly(ε-L-lysine) (ε-PL) is a novel bioactive polymer secreted by filamentous bacteria. Owing to lack of a genetic system for most ε-PL-producing strains, very little research on enhancing ε-PL biosynthesis by genetic manipulation has been reported. In this study, an effective genetic system was established via intergeneric conjugal transfer for Streptomyces albulus PD-1, a famous ε-PL-producing strain. Using the established genetic system, the Vitreoscilla hemoglobin (VHb) gene was integrated into the chromosome of S. albulus PD-1 to alleviate oxygen limitation and to enhance the biosynthesis of ε-PL in submerged fermentation. Ultimately, the production of ε-PL increased from 22.7 g/l to 34.2 g/l after fed-batch culture in a 5 L bioreactor. Determination of the oxygen uptake rate, transcriptional level of ε-PL synthetase gene, and ATP level unveiled that the expression of VHb in S. albulus PD-1 enhanced ε-PL biosynthesis by improving respiration and ATP supply. To the best of our knowledge, this is the first report on enhancing ε-PL production by chromosomal integration of the VHb gene in an ε-PL-producing strain, and it will open a new avenue for ε-PL production.
Study on the space frame structures incorporated with magnetorheological dampers

Magnetorheological damper has received significant attention in recent years due to the reason that it can offer adaptability of active control devices without requiring the associated large power sources. In this paper, performance tests on a MR damper are carried out under different currents, excitation amplitudes and frequencies, the damping characteristics and energy dissipation capacity of the MR damper are analyzed. Elasto-plastic dynamic analysis on a space frame structure incorporated with MR dampers is conducted, and numerical analysis results show that MR dampers can significantly mitigate the structural vibration responses. Finally, the genetic algorithm with the improved binary crossover and mutation technique is adopted to optimize the arrangement of MR dampers. Numerical results show that dynamic responses of the optimal controlled structure are mitigated more effectively.

Herein, a series of lasiokaurin derivatives were designed and synthesized. All the derivatives together with lasiokaurin and oridonin were tested for their antimicrobial and antiproliferative activity. Compound 16 showed the most promising antimicrobial activity with MICs of 2.0 and 1.0 lg/mL against Gram-Positive bacteria S. aureus and B. subtilis, respectively. All the synthetic lasiokaurin derivatives showed better antiproliferative activity than parent compound lasiokaurin 1. Compound 10 exhibited the strongest cytotoxicity with IC50 values of 0.47 and 0.20 lM against MGC-803 and CaEs-17 cells, accordingly. Moreover, it was shown to have potent antitumor activity in vivo in a murine model of MGC-803 gastric cancer. Preliminary SARs were also concluded based on obtained data. The apoptosis-inducing effects of 10 were further investigated using CaEs-17 cells. The results showed that lasiokaurin derivative 10 could induce apoptosis via mitochondria related pathway and arrest CaEs-17 cell cycle at S phase. Compound 10 could also affect apoptosisrelated proteins that was up-regulation of CDK2 and downregulation of ATM and cyclin A1.
