      • ANXA2 Regulates the Behavior of SGC-7901 Cells

        ANXA2, a member of the annexin family, is overexpressed and plays important roles in tumor development. However, the significance of ANXA2 expression in gastric carcinoma has not been clarified.To elucidate its roles in growth of gastric cancer, ANXA2 expression in SGC-7901 cells was inhibited with a designated siRNA, then cell proliferation, cell cycling, apoptosis and motility were determined by MTT assay, flow cytometry, Hoechst 33342 staining and wound healing assay, respectively. To further assess the behavior of ANXA2 deleted SGC-7901 cells, changes of microstructures were observed under fluorescence microscopy, laser scanning confocal microscopy and electron microscopy. We found that inhibition of ANXA2 expression caused cell proliferation to decrease significantly with G1 arrest, motility to be reduced with changes in pseudopodia/filopodia structure and F-actin and ${\beta}$-tubulin expression, and apoptosis to be enhanced albeit without significance. At the same time, ANXA2 deletion resulted in fewer pseudopodia/filopodia, non-stained areas were increased, contact inhibition among cells reappeared, and expression of F-actin and ${\beta}$-tubulin was decreased, with induction of polymerized disassembled forms. Taken together, these data suggest that ANXA2 overexpression is important to maintain the malignancy of cancer cells, and this member of the annexin family has potential to be considered as a target for the gene therapy of gastric carcinoma.

        Resonant Tank Design Considerations and Implementation of a LLC Resonant Converter with a Wide Battery Voltage Range

        This paper illustrates resonant tank design considerations and the implementation of a LLC resonant converter with a wide battery voltage range based on the fundamental harmonic approximation (FHA) analysis. Unlike the conventional design at zero load, the parameter K (the ratio of the transformer magnetizing inductor L<sub>m</sub> to the resonant inductor L<sub>r</sub>) of the LLC converter in this paper is designed with two charging points, (V<sub>o_min</sub>, I<sub>o_max1</sub>) and (V<sub>o_max</sub>, I<sub>o_max2</sub>), according to the battery charging strategy. A 2.9kW prototype with an output voltage range of 36V to 72V dc is built to verify the design. It achieves a peak efficiency of 96%.

        Analysis and Design of a Multi-resonant Converter with a Wide Output Voltage Range for EV Charger Applications

        This paper illustrates the analysis and design of a multi-resonant converter applied to an electric vehicle (EV) charger. Thanks to the notch resonant characteristic, the multi-resonant converter achieve soft switching and operate with a narrowed switching frequency range even with a wide output voltage range. These advantages make it suitable for battery charging applications. With two more resonant elements, the design of the chosen converter is more complex than the conventional LLC resonant converter. However, there is not a distinct design outline for the multi-resonant converters in existing articles. According to the analysis in this paper, the normalized notch frequency $f_{r2n}$ and the second series resonant frequency $f_{r3n}$ are more sensitive to the notch capacitor ratio q than the notch inductor ratio k. Then resonant capacitors should be well-designed before the other resonant elements. The peak gain of the converter depends mainly on the magnetizing inductor ratio $L_n$ and the normalized load Q. And it requires a smaller $L_n$ and Q to provide a sufficient voltage gain $M_{max}$ at ($V_{o\_max}$, $P_{o\_max}$). However, the primary current increases with $(L_nQ)^{-1}$, and results in a low efficiency. Then a detailed design procedure for the multi-resonant converter has been provided. A 3.3kW prototype with an output voltage range of 50V to 500V dc and a peak efficiency of 97.3 % is built to verify the design and effectiveness of the converter.

        Modeling and Analysis of Leakage Currents in PWM-VSI-Fed PMSM Drives for Air-Conditioners with High Accuracy and within a Wide Frequency Range

        Leakage currents occur in pulse-width-modulated voltage source inverter (PWM-VSI)-fed permanent magnet synchronous motor (PMSM) drives for air-conditioners, which seriously affect system safety and operation performance. High accuracy modeling and prediction of leakage currents are key issues for the design and implementation of air-conditioning products. In this study, the generation mechanism of leakage currents is discussed. A systematic modeling approach of leakage currents is proposed, including the modeling of leakage current sources and leakage current paths. By using the proposed approach, the complete model of leakage currents in PWM-VSI-fed PMSM drives for air-conditioners has been developed based on the extraction of all parameters. A comparison between the simulated leakage currents based on the developed model and measured leakage currents in the outdoor unit of an air-conditioning product is conducted. The comparison verifies the effectiveness of the proposed modeling approach, and the developed model exhibits high accuracy within a wide frequency range.

        Isolation and Molecular Characterization of a New CRT Binding Factor Gene from Capsella bursa-pastoris

        A new CRT binding factor (CBF) gene designated Cbcbf25 was cloned from Capsella bursa pastoris, a wild grass, by the rapid amplification of cDNA ends (RACE). The full-length cDNA of Cbcbf25 was 898 bp with a 669 bp open reading frame (ORF) encoding a putative DRE/CRT (LTRE)-binding protein of 223 amino acids. The predicted CbCBF25 protein contained a potential nuclear localization signal (NLS) in its N-terminal region followed by an AP2 DNA-binding motif and a possible acidic activation domain in the C-terminal region. Bioinformatic analysis revealed that Cbcbf25 has a high level of similarity with other CBF genes like cbfl, cbf2, and cbf3 from Arabidopsis thaliana, and Bncbf5, Bncbf7, Bncbfl6, and Bncbfl7 from Brassica napus. A cold acclimation assay showed that Cbcbf25 was expressed immediately after cold triggering, but this expression was transient, suggesting that it concerns cold acclimation. Our study implies that Cbcbf25 is an analogue of other CBF genes and may participate in cold-response, by for example, controlling the expression of cold-regulated genes or increasing the freezing tolerance of plants.

      • AZD1480 Can Inhibit the Biological Behavior of Ovarian Cancer SKOV3 Cells in vitro

        Objective: To study the mechanism of effects of AZD1480 on the SKOV3 ovarian cancer cell line. Methods: The MTT method was used to assess cellular proliferation, flow cytometry for cellular apoptosis, the scratch test to determine migration, transwell chamber assays to detect cellular invasion, plate clone experiments to detect the clone forming ability and Western blotting to determine p-STAT3 protein levels. Results: The proliferation rate, migration ability, invasiveness and the clone forming ability of SKOV3 cells were reduced after treatment with AZD1480, while apoptosis rate and chemotherapeutic susceptibility were increased. After treatment with AZD1480 plus cisplatin, the apoptosis rate increased significantly while the expression level of p-STAT3 protein was decreased. Conclusion: AZD1480 can inhibit the proliferation, invasion, metastasis and clone formation of SKOV3 cells, induce cellulsar apoptosis, increase the chemotherapeutic sensitivity and reduce the expression level of p-STAT3 protein.

        Microstructures and Mechanical Properties of Equiatomic NiTi Shape Memory Alloy Undergoing Local Canning Compression and Subsequent Annealing

        Local canning compression is imposed on equiatomic NiTi shape memory alloy (SMA) with complete B19′ martensite. Afraction of retained nanocrytalline grains are embedded in the dominant amorphous phase. The compressed NiTi samples areannealed for 2 h at various temperatures, including 300, 450 and 600 °C. Grain size increases with increasing crystallizationtemperature during heat treatment. Under annealing at 300 °C, nanocrystalline grains are dominant in NiTi SMA, where thereexists local amorphous zone. Under annealing at 450 °C, almost complete nanocrystalline can be obtained. Under annealingat 600 °C, grain size increases substantially, but there still exists a small amount of nanocrystalline grains. Under annealingat 300 °C, NiTi SMA exhibits extremely high elastic limit, but its plasticity is poor. Under annealing at 450 °C, NiTi SMAexhibits very high yield stress and it simultaneously keeps high plasticity. Under annealing at 600 °C, NiTi SMA exhibitsrelatively low yield stress, but its yield stress is still higher than that of as-received NiTi SMA and its plasticity is also high.
