        DEPDC1 is a novel cell cycle related gene that regulates mitotic progression

        DEPDC1 is a recently identified novel tumor-related gene that is upregulated in several types of cancer and contributes to tumorigenesis. In this study, we have investigated the expression pattern and functional implications of DEPDC1 during cell cycle progression. Expression studies using synchronized cells demonstrated that DEPDC1 is highly expressed in the mitotic phase of the cell cycle. Immunofluorescence assays showed that DEPDC1 is predominantly localized in the nucleus during interphase and is redistributed into the whole cell upon nuclear membrane breakdown in metaphase. Subsequently, siRNA-mediated knockdown of DEPDC1 caused a significant mitotic arrest. Moreover, knockdown of DEPDC1 resulted in remarkable mitotic defects such as abnormal multiple nuclei and multipolar spindle structures accompanied by the upregulation of the A20 gene as well as several cell cycle-related genes such as CCNB1 and CCNB2. Taken together, our current observations strongly suggest that this novel cancerous gene, DEPDC1, plays a pivotal role in the regulation of proper mitotic progression.         We report on a patient with a partial deletion on the short arm of chromosome 18 (del 18p), who presented with dysmorphic features and delayed developmental milestones as well as with a patent ductus arteriosus (PDA) and pulmonary valve stenosis (PS). Several forms of congenital heart disease (CHD) are found in about 10% of patients with del (18p), but coexisting PDA and PS have not been reported. Del (18p) must be considered in patients with characteristic phenotypic abnormalities and congenital heart disease, including a combination of PDA and PS.

        Control Strategy and Characteristic Analysis of Hybrid Active Power Filters with the Resonant Impedance Principle

        A new kind of resonant impedance type hybrid active filter (RITHAF) is proposed for dynamic harmonic current suppression and high capacity reactive compensation in medium and high voltage systems. This paper analyzed the different performance of the RITHAF when the active part of the RITHAF is controlled as a current source and as a voltage source, respectively. The harmonic suppression function is defined in this paper. The influences of the changes caused by the grid impedance and the detuning of the passive power filter on the compensating characteristics of the RITHAF are studied by analyzing the suppression function. Simulation and industrial application results show that the RITHAF has excellent performances in harmonic suppression and reactive compensation, which is suitable for medium and high voltage systems.

        Butyrolactones Derivatives from the Fermentation Products of an Endophytic Fungus Aspergillus versicolor

        Two new butyrolactones, asperphenol A (1) and B (2), together with four known butyrolactones (3-6) were isolated from the fermentation products of an endophytic fungus Aspergillus versicolor. Their structures were elucidated by spectroscopic methods including extensive 1D- and 2D-NMR techniques. Compounds 1-6 were also tested for their anti-tobacco mosaic virus (anti-TMV) activities. The results showed that compound 2 exhibited high anti-TMV activity with inhibition rate of 46.7%. The other compounds also exhibited potential anti-TMV activities with inhibition rates in the range of 21.8-28.4%.

        High Infectivity of an Endoparasitic Fungus Strain, Esteya vermicola, against Nematodes

        Esteya vermicola, as the first recorded endoparasitic fungus of pinewood nematodes, exhibits great potential as a biological agent against nematodes. However, only two strains of this species have been described so far. In this study, we identified a novel endoparasitic fungal strain, CNU 120806, isolated from infected nematodes in forest soil samples during a survey of nematophagous fungi in Korea. This strain showed similar morphological characteristics and infection mode with the two previously described strains of E. vermicola. All strains are characterized by the ability to produce two types of conidiogenous cells and conidia, and to parasitize nematodes with lunate adhesive conidia. Moreover, the CNU 120806 strain showed 100% identity with E. vermicola CBS 115803 when their partial sequences of 28S rRNA gene were compared. Molecular phylogenetic analysis further identified CNU 120806 as a strain of E. vermicola, by clustering CNU 120806 and E. vermicola CBS 115803 into a single subclade. Culture medium influenced the proportion of dimorphic CNU 120806 conidia, and further changed the adhesive and mortality rates of nematodes. The CNU 120806 strain exhibits high infection activity against nematodes on nutrient-rich PDA medium. Almost all tested nematodes were killed within 8~10 days after inoculation. This study provides justification for further research of E. vermicola, and the application and formulation of this fungus as a bio-control agent against nematodes.

        Association of the Single-Nucleotide Polymorphism and Haplotype of the Complement Receptor 1 Gene with Malaria

        Purpose: Although the polymorphisms of erythrocyte complement receptor type 1 (CR1) in patients with malaria have been extensively studied, a question of whether the polymorphisms of CR1 are associated with severe malaria remains controversial. Furthermore, no study has examined the association of CR1 polymorphisms with malaria in Chinese population. Therefore, we investigated the relationship of CR1 gene polymorphism and malaria in Chinese population. Materials and Methods:We analyzed polymorphisms of CR1 gene rs2274567 G/A, rs4844600 G/A, and rs2296160 C/T in 509 patients with malaria and 503 controls, using the Taqmangenotyping assay and PCR-direct sequencing. Results: There were no significantdifferences in the genotype, allele and haplotype frequencies of CR1 gene rs2274567 G/A, rs4844600 G/A, and rs2296160 C/T polymorphisms between patientswith malaria and controls. Furthermore, there was no association of polymorphismsin the CR1 gene with the severity of malaria in Chinese population. Conclusion:These findings suggest that CR1 gene rs2274567 G/A, rs4844600 G/A, and rs2296160 C/T polymorphisms may not be involved in susceptibility to malariain Chinese population.

        Traditional Chinese medicine for COVID-19 pandemic and emerging challenges: An online cross-sectional survey in China

        Background We aimed to investigate use of infection control behaviours, preventative and therapeutic interventions, and outcomes among respondents to an online survey during the COVID-19 pandemic in China. Methods The survey was designed by an international team, translated and adapted to simplified Chinese, including 132 kinds of traditional Chinese medicine (TCM) preparation recommended by guidelines. It was distributed and collected from February to May 2021, with data analysed by WPS spreadsheet and wjx.cn. Descriptive statistics were used to describe demographics and clinical characteristics, diagnosis, treatments, preventative behaviours and interventions, and their associated outcomes. Results The survey was accessed 503 times with 341 (67.8%) completions covering 23 provinces and four municipalities in China. Most (282/341, 82.7%) respondents reported no symptoms during the pandemic and the majority (290/341, 85.0%) reported having a SARS-CoV-2 PCR test at some point. Forty-five (13.2%) reported having a respiratory infection, among which 19 (42.2%) took one or more categories of modern medicine, e.g. painkillers, antibiotics; 16 (35.6%) used TCM interventions(s); while seven respondents combined TCM with modern medicine. All respondents reported using at least one behavioural or medical approach to prevention, with 22.3% taking TCM and 5.3% taking modern medicines. No respondents reported having a critical condition related to COVID-19. Conclusion We found evidence of widespread use of infection control behaviours, modern medicines and TCM for treatment and prevention of COVID-19 and other respiratory symptoms. Larger scale studies are warranted, including a more representative sample exploring TCM preparations recommended in clinical guidelines.         Influence of Pinewood Nematode, Bursaphelenchus xylophilus, on the Growth of Endoparasitic Fungus Esteya vermicola

        본 논문은 선충 포식성 곰팡이에 대한 선충의 영향을 조사한 연구로, 소나무 기생곰팡이인 E. vermicola에 미치는 소나무 선충의 영향을 체계적으로 분석하였다. 선충 대사물과 선충 균질액 모두 E. vermicola의 성장을 빠르게 촉진하였으나, 선충 대사물이 선충 균질액보다 약간 높은 효과를 보였다. 또한 소나무 선충이 생성하는 휘발성 물질이 E. vermicola 성장에 미치는 영향을 분석하는 새로운 실험방법을 개발하였다. 본 연구결과에 따르면, 선충의 휘발성 물질은 E. vermicola에 아무런 영향을 끼치지 않는다는 재현성 있는 결과를 도출하였다. 본 연구는 소나무 선충의 생물학적인 조절인자로서 E. vermicola을 적용할 수 있는 정보를 제공할 수 있을 것이다. The influence of nematodes on nematophagous fungi has seldom been investigated. In the present study, the influence of pinewood nematode on its endoparasitic fungus, Esteya vermicola, was investigated systemically. Although both nematodal metabolite and nematodal homogenate could stimulate and speed up the growth of E. vermicola, the impact of nematodal metabolite was slightly higher than that of nematodal homogenate. In addition, a method was developed to investigate the influence of volatiles, discharged by pinewood nematodes in their metabolic process, on the growth of E. vermicola. Reproductive results were given and confirmed that nematodal volatiles have no influence on the cell growth of E. vermicola. This study may provide information for the application of E. vermicola as biological control agent of pinewood nematode.

        Excitation frequency, fastener stiffness and damping, and speed of the moving harmonic load on the dynamic response of the track structure

        The dynamic response of a track structure under a uniform-speed moving harmonic load is researched according to dynamic response characteristics of a periodic structure under moving harmonic load in the frequency domain. The track was assumed as a simple Euler beam model periodically supported by continuous discrete point, and mathematical model of the dynamic differential equation of vertical vibration for the track structure is built. Then, the analytical equation for the dynamic response of any point on the track structure is concluded in the frequency domain for the following research. The dynamic responses of the track structure under the uniform-speed moving harmonic load are investigated using the theory of infinite periodic structure. Finally, the effects of excitation frequency, fastener stiffness, fastener damping, and speed of the moving harmonic load on the dynamic response of the track structure are studied. Results indicate that the response peaks of the rail under moving harmonic load occur near the excitation frequency, and the dynamic response decreases rapidly in the area far from the excitation frequency. The response peaks of the rail will move slightly toward a high frequency with the increase in the excitation frequency. The increase in the fastener stiffness will lead to improvement of the dynamic response of the rail in the nonresonant region at a high frequency, equivalent to the high rigidity of the rail fastener and intense vibration of the rail. The changes in fastener damping exert no significant effect on the resonant peak and peak bandwidth of the system. The fastener damping plays a significant role in restraining the vibration at a high frequency. The strong vibration of the track structure can be effectively controlled by an increase in the damping.

      • Clinicopathological and Prognostic Characteristics of Triple-Negative Breast Cancer (TNBC) in Chinese Patients: A Retrospective Study

        Aims: To determine the clinical, pathological and prognostic features associated with triple-negative breast cancer (TNBC). Methods: Clinical and histologic data of 21,749 breast cancer patients who were treated at Tianjin Medical University Cancer Institute and Hospital between July 2002 and December 2011 were collected. Patients were divided into two groups: those with TNBC and those with other types of breast cancer. Patients and tumor characteristics were compared between the two groups using the Chi-square test. The prognostic results of 9,823 patients in the study population were also analyzed to determine long-term survival rates in the two groups of breast cancer patients. Results: Among the breast cancer patients treated in our hospital between 2003 and 2011, 10.4%-13.5% of them had triple-negative breast cancers. Data analyses revealed significant differences in disease onset age, family history of breast cancer, tumor size, tumor histologic grade, lymph note positivity and metastatic status between TNBC and non-TNBC patients. There were also significant differences in 5-year, 7-year and 9-year disease-free and 7-year and 9-year overall survival probability between the groups. Conclusions:TNBC are associated with younger disease onset age, larger tumor size, higher rate of axillary lymph node positivity, and higher tumor histologic grade. TNBC is also related to family history of breast cancer, increased metastatic risk and poor prognosis.
