Background and Purposes: Over the past three decades, numerous studies performed in Korea have reported that job stress is a determinant risk factor for chronic diseases and work disability. Every society has its own culture and occupational climate particular to their organizations, and hence experiences different occupational stress. An occupational stress measurement tool therefore needs to be developed to estimate it objectively. The purpose of this study is to develop and standardize the Korean Occupational Stress Scale (KOSS) which is considered to be unique and specific occupational stressors in Korean employees. Subjects and Methods: Data were obtained from the National Study for Development and Standardization of Occupational Stress (NSDSOS Project: 2002-2004). A total of 12,631 employees from a nationwide sample proportional to the Korean Standard Industrial Classification and the Korean Standard Occupational Classification were administered. The KOSS was developed for 2 years (2002-2004). In the first year, we collected 255 items from the most popular job stress measurement tools such as JCQ, ERI, NIOSH and OSI, and 44 items derived from the a qualitative study (depth interview). Forty-three items of KOSS, in the second year, were retained for use in the final version of the KOSS by using Delphi and factor analysis. Items were scored using conventional 1-2-3-4 Likert scores for the response categories. Results: We developed eight subscales by using factor analysis and validation process: physical environment (3 items), job demand (8 items), insufficient job control (5 items), interpersonal conflict (4 items), job insecurity (6 items), organizational system (7 items), lack of reward (6 items), and occupational climate (4 items). Together they explained 50.0% of total variance. Internal consistency alpha scores were ranged from 0.51 to 0.82. Twenty-four items of the short form of the KOSS (KOSS-SF) were also developed to estimate job stress in the work setting. Because the levels of the subscales of occupational stress were gender dependent, gender-specific standard norms for both the 43-item full version and the 24-item short form using a quartile for the subscales of KOSS were presented. Conclusion: The results of this study suggest that KOSS might be an appropriate measurement scale to estimate occupational stress of Korean employees. Further and more detailed study needs to be conducted to improve the validity of this scale.
Computer Graphics에 의한 3성분계 Wilson, NRTL 및 UNIQUAC 방식을 이용한 VLE 추정방식의 비교

The purpose of this study is to estimate VLE by means of the Wilson, NRTL, UNIQUAC equations and to compare with these three methods for two ternary systems: ethanol-benzene-cyclohexane, cyclohexanone-cyclohexanol-phenol, by the computer graphic technology. The temperatures and compositions of two systems were estimated by means of three TPTT equations. Isothermal lines of these two ternary systems were graphically illustrated from these estimated values and the reported experimental values. As a result of the analysis of the parameters and computer graphics, these parameter pairs changing with respect to the temperature and composition of the solutions. From the result of plotting the parameter pairs, A_12, vs, A_21, a locus contour between two parameters is observed, therefore numerous number of roots to satisfy these TPTT equations are found. These phenomena is nearly identical with the previously reported results that there are some correlation between these parameter pairs.

Objectives: This study was performed to find the association of cigarette smoking and blood lipid profiles in healthy young adults. Methods: The data was collected by a self-administered questionnaire and health examination in 489 professional soldiers from 6 to 12 May, 2004. The status of cigarette smoking was classified by non-smoking, ex-smoked, ≤4.9, 5-9.9, and ≥10 pack-years. The statistical analysis were used by chi-square test, analysis of variance and analysis of covariates. Results: In status of subjects cigarette smoking, current smoking, ex-smoked, and non-smoking were 71.8%, 11.2%, and 17.0%, respectively, In distribution by amount of smoking, non-smoking, ex-smoked, ≤4.9 pack-years, 5-9.9 pack-years, and ≥10 pack-years were 17.0%, 11.2%, 37.4%, 21.3%, and 13.1%, respectively. At other confounding variables were controlled, total cholesterol and LDL-cholesterol were increased significantly when amount of smoking was increased. However, triglyceride and HDL-cholesterol did not showed statistically significant differences, Conclusion: Smoking has relevant to total cholesterol and LDL-cholesterol and it is need to perform the further study for finding how blood lipid profiles are affected its level by cigarette smoking and to find an influence of cigarette smoking on cardiovascular disease.
Characterization of a stamen-specific cDNA encoding a novel plant defensin in Chinese cabbage

We isolated a stamen-specific cDNA, BSD1(Brassica stamen specific plant defensin 1)that encodes a novel plant defensin peptide in Chinese Chinese cabbage (Brassica campestris L. ssp. pekinensis). Plant defensins are antimicrobial peptides containing eight highly conserved cysteine residues linked by disulfide bridges. In BSD1, the eight cysteine residues and a glutamate residue at position 29 are conserved whereas other amino acid residues of the plant defensins consensus sequence are substituted. BSD1 transcripts accumulate specifically in the stamen of developing flowers and its level drops as the flowers mature. The recombinant BSD1 produced in Escherichia coli showed antifungal activity against several phytopathogenic fungi. Furthermore, constitutive over-expression of the BSD1 gene under the control of the cauliflower mosaic virus (CaMV) 35S promoter conferred enhanced tolerance against the Phytophthora parasitica in the transgenic tobacco plants.

Objective: To differentiate the human embryonic stem cells (hESCs) into the retinal pigment epithelium (RPE) in the defined culture condition and determine its therapeutic potential for the treatment of retinal degenerative diseases. Methods: The embryoid bodies were formed from hESCs and attached on the matrigel coated culture dishes. The neural structures consisting neural precursors were selected and expanded to form rosette structures. The mechanically isolated neural rosettes were differentiated into pigmented cells in the media comprised of N2 and B27. Expression profiles of markers related to RPE development were analyzed by reverse transcription-polymerase chain reaction and immunostaining. Dissociated putative RPE cells ($10^5$ cells/5 ${\mu}L$) were transplanted into the subretinal space of rat retinal degeneration model induced by intravenous sodium iodate injection. Animals were sacrificed at 1, 2, and 4 weeks after transplantation, and immnohistochemistry study was performed to verify the survival of the transplanted cells. Results: The putative RPE cells derived from hESC showed characteristics of the human RPE cells morphologically and expressed molecular markers and associated with RPE fate. Grafted RPE cells were found to survive in the subretinal space up to 4 weeks after transplantation, and the expression of RPE markers was confirmed with immunohistochemistry. Conclusion: Transplanted RPE cells derived from hESC in the defined culture condition successfully survived and migrated within subretinal space of rat retinal degeneration model. These results support the feasibility of the hESC derived RPE cells for cell-based therapies for retinal degenerative disease.

The Ca^(2+)-binding protein calmodulin mediates cellular Ca^(2+) signals in response to a wide array of stimuli in higher eukaryotes. Plants express numerous CaM isoforms. Transcription of one soybean (Glycine max) CaM isoform, SCaM-4, is dramatically induced within 30 min of pathogen or NaCl stresses. To characterize the cis-acting element(s) of this gene, we isolated an approximately 2-kb promoter sequence of the gene. Deletion analysis of the promoter revealed that a 130-bp region located between nucleotide positions -858 and -728 is required for the stressors to induce expression of SCaM-4. A hexameric DNA sequence within this region, GAAAAA (GT-1 cis-element), was identified as a core cis-acting element for the induction of the SCaM-4 gene. The GT-1 cis-element interacts with an Arabidopsis GT-1-like transcription factor, AtGT-3b, in vitro and in a yeast selection system. Transcription of AtGT-3b is also rapidly induced within 30 min after pathogen and NaCl treatment. These results suggest that an interaction between? GT-1 cis-element and a GT-1-like transcription factor plays a role in pathogen- and salt-induced SCaM-4 gene expression in both soybean and Arabidopsis.

Plants produce numerous calmodulin isoforms that exhibit differential gene expression patterns and sense different Ca^(2+)signals. This diversity results in different physiological responses to particular stimuli. GmCaM-4 and -5 are two divergent calmodulin isoforms from the soybean (Glycine max) that have been reported to be involved in plant disease resistance. However, little is known about the pathway by which these specific isoforms transduce the defense signal and up-regulate pathogenesis-related (PR) genes. Here we report that overexpression of GmCaM-4/-5 induces constitutive PR gene expression and enhances disease resistance in wild-type Arabidopsis, but not in the nim1 mutant of Arabidopsis. GmCaM-4/-5 also appear to activate trans-acting elements that bind to cis-acting elements in the Arabidopsis PR-1 promoter. Thus up-regulation of PR genes by these GmCaM isoforms is dependent on NIM1 (Non immunity 1) and unknown transcription factors.
