Suicide cancer gene therapy using pore-forming toxin, streptolysin O.

<P>We cloned the streptolysin O gene from the Streptococcus pyogenes genome and tested the possibility of using it as an anticancer reagent. Transient transfection of the streptolysin O gene efficiently killed 293T cells after 12 hours of transfection as determined by lactate dehydrogenase release and propidium iodide uptake. No caspase activity was observed and necrosis was prominent during streptolysin O-induced cell death. Biochemical analysis of streptolysin O protein revealed that the deletion of only 5 amino acids from the COOH-terminal region of streptolysin O, which is essential for cholesterol binding activity, abolished its cell-killing activity, whereas the NH2-terminal region was more resilient, i.e., up to 115 amino acids could be deleted without changing its cell-killing activity. We generated a streptolysin O-expressing adenovirus and injected it into human cervical cancer cell-derived tumors grown in a nude mouse model. Twenty-one days postinjection, the average size of tumors in the streptolysin O adenovirus-injected group was 29.3% of that of the control PBS-treated group. Our results show that the genes of pore-forming toxins, like streptolysin O protein, have the potential to establish a novel class of suicide gene therapeutic reagents.</P>

Background : Despite recent advances in antifungal chemotherapy, invasive aspergillosis remains an important cause of morbidity and mortality in immunocompromised patients. Interleukin-15 (IL-15) is a cytokine that is known to enhance antifungal activities of monocytes against Candida albicans. Objective: The purpose of this study is to investigate the potentials of IL-15 to enhance antifungal activities of monocytes against Aspergillus fumigatus. Methods: Peripheral blood monocytes from healthy adults were incubated with 0,1,10,100 ng/ml of IL-15 for 1, 2, and 4 days. Then, the ability of IL-15 to elicit the production of superoxide anion, the damage of hyphae by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyletrazolium bromide assay, and the killing ability of Aspergillus fumigatus conidia was investigated. Results: Incubation of peripheral blood monocytes with 100 ng/ml of IL-15 enhanced hyphal damage after 2 days (p<0.05), condicidal activity from the first day (p<0.05), and increased the production of superoxide anion (O2-) in response to phorbol myristate acetate. Conclusion: Our results indicate that IL-15 augments the microbicidal activity of human monocytes against Aspergillus fumigatus .[Kor J Med Mycol 6(2): 64-69]

Hepatic yersiniosis caused by Yersinia enterocolitica 4:O3 in an adult dog.

<P>A 2-year-old Pekinese dog was diagnosed with hepatic yersiniosis. Grossly, white-to-yellow nodules consisting of degenerated inflammatory cells, cell debris, and bacterial clumps were scattered throughout the liver. Histopathologically, suppurative and necrotizing hepatitis was apparent. Yersinia enterocolitica biotype 4, serotype O3 (4:O3) was identified and confirmed in the liver immunohistochemically, using a monoclonal antibody. The virulence genes ystA and ail were detected, but the isolate was negative for autoagglutination and calcium-dependent growth. To confirm systemic yersiniosis in animals, it is imperative that the organism(s) be identified because the hepatic lesions are similar to those of Y. pseudotuberculosis and other diseases, including plague, which is also a zoonotic pathogen.</P>

This study aims to explore whether students' mother tongue (Korean) or the target language (English) is used as teacher/student communication channel has any relationship with students' English speaking achievements. To that end, the study employs two native speakers of English as the teachers, one with only target language in comparison group and the other with fluent Korean in experimental group. The study has used oral tests for measuring the students' English achievements, and questionnaires for understanding their perceptions about their classes. After six months' experiment duration, the study has concluded that the two groups do not demonstrate significant differences with each other, even though both of the groups have shown significant improvements in six months.

Resilon을 이용한 근관충전 시 수종의 치면처리제에 따른 미세누출 평가

이 실험의 목적은 Resilon으로 근관충전된 치아에서 자가산부식전처리제에 따른 치근단 미세누출을 평가하고자 함이다. 70개의 단근치를 사응하였고 .04 Taper ProFile을 사용하여 ISO #40까지 근관성형 하였다. 치아는 근관충전 재료와 자가산부식 전처리제에 따라서 15개씩 4개의 실험군과 5개씩 2개의 대조군으로 나누었다. 실험 치아는 치아 장축에 평행하게 잘라내어 염색액의 최대 침투 길이를 측정하였다. 통계분석은 One-way ANOVA test로 분석하였으며 Scheffe test로 사후검정 하였다. 실험결과 자가산부식전처리제를 사용한 2, 3, 4군 사이에서는 통계적으로 유의한 차이를 나타내지 않았다. 그러나 1군과 4군 사이에서는 통계적으로 유의한 차이가 나타났으며 (p < 0.05), 1군에서 가장 적은 치근단 미세누출을 나타냈다. 이 실험 결과에 따르면 Resilon으로 근관충전을 한 치아는 gutta percha와 AH26^(ⓡ)으로 근관충전할 치아와 비교했을 때 더 좋은 치근단 밀폐를 나타내지 않는다는 결론을 얻었으며, Resilon을 사용하여 근관충전을 한 경우에 자가산부식 전처리제의 차이에 따른 치근단 밀폐효과의 차이를 보이지 않는다는 결론을 얻었다. The purpose of this study was to compare the apical microleakage in root canal filled with Resilon by several self-etching primers and methacrylate-based root canal sealer. Seventy single-rooted human teeth were used in this study. The canals were instrumented by a crown-down manner with Gate-Glidden drills and .04 Taper Profile to ISO #40. The teeth were randomly divided into four experimental groups of 15 teeth each according to root canal filling material and self-etching primers and two control groups (positive and negative) of 5 teeth each as follows: group 1 - gutta percha and AH26^(ⓡ) sealer; group 2 - Resilon, RealSeal™ primer and RealSeal™ sealer; group 3 - Resilon, Clearfil SE Bond^(ⓡ) primer and RealSeal™ sealer group 4 - Resilon, AdheSe^(ⓡ) primer and RealSeal™ sealer. Apical leakage was measured by a maximum length of linear dye penetration of roots sectioned longitudinally by diamond disk. Statistical analysis was performed using the One-way ANOVA followed by Scheffe' s test. There were no statistical differences in the mean apical dye penetration among the groups 2, 3 and 4 of self-etching primers. And group 1, 2 and 3 had also no statistical difference in apical dye penetration. But, there was statistical difference between group 1 and 4 (p < 0.05). The group 1 showed the least dye penetration. According to the results of this study, Resilon with self-etching primer was not sealed root canal better than gutta precha with AH26^(ⓡ) at sealing root canals. And there was no significant difference in apical leakage among the three self-etching primers.

Minimization of the stress concentration is very important in the life cycle extension of structures and machine parts, which have cracks or notches. Industrial field, stop-hole is drilled on the structures and machine parts to prevent the origination and propagation of cracks and to reduce the stress concentration. But, there is no a study on the optimal dimension of stop-hole for the various loading and geometrical conditions. Therefore, in this study, a study on the optimal dimension of stop-hole with various conditions is done by photoelastic experimental method.

A number of contemporary critics have indicted Joyce as a chauvinist author singularly devoted to projects of male linguistic mastery and to a celebration of the Freudian phallus. In Joyce's canon, woman is both desirable object and subject of desire-the incomparable Other who remains mysterious and enigmatic. His female characters frequently embody that imaginary maternal presence that the unconscious constructs as a corollary to leasure principle of infantile need. Woman emerges in Joyce's work as virgin or whore, madonna or temptress, but usually within a problematic context of maternity. A "woman's love," Joyce declared, "is always maternal and egoistic. (Letters Ⅱ, 912) Even the temptress is implicitly portrayed as a potential mother in training. From her Oedipal role as a"Papa's little bed-pal Lump of love" (U 3:83), the adolescent girl learns to weave a web of romantic entrapment. Thoughout Ulysses, Joyce deliberately problematizes gender and interrogates sexual practice. The novel unfolds as a radical exploration of different forms of psychic mobility. Sexual identity is at once ambiguous and polymorphous, a state to be achieved through the textual exploration of a series of ostensibly perverse psychological drives. Characters are radically decentered by erotic fantasy and attempt to uncover new identities latent in residual needs that overflows the boundaries of sex-role expectations. In the "Penelope" episode, Molly Bloom emerges as the clue of Joyce's novel(LettersI,17O)-a clue to the male-dominated, antiheroic modern epic. She proves, in her own right, to be a poet of the imagination whose final soliloquy elevates Ulysses to the heights of lyrical discourse. by virtue of her capacious monologue, Molly can be envisaged as both goddess and whore, Dublin housewife and archetypal precursor, the Great mother/lover of Ulysses.

Oscar Wilde's aesthetics is reflected in his works, The Picture of Dorian Gray. In this novel he reveals the puritan hypocrisy and materialism of Victorian society. He is against Victorian vulgar materialims, moralism and philistinism. His The Picture of Dorian Gary is the most representative homosexual novel among his works. Wilde confesses his double life, homosexual temperament and beauty for beauty's sake through Dorian's life in this novel. In view of Wilde's aesthetics, art is prior to reality. Wilde's understanding with an assertion that "LIfe imitates Art far more than Art imitates Life," presents his attitude about art and life. At the beginning of the novel, Dorian is described as an image of good but gradually becomes an image of evil. Dorian has come to realize that good and evil coexist in himself. Dorian, a good looking young man, stays young because he sold his soul. As time goes on, Dorian becomes a murderer and a liar. When Dorian looks at a portrait of himself, because it is grotesque to him, he stabs himself with a knife causing his own death. Although Wilde concludes the novel with Dorian's death, he achieves his aim which is to reveal Victorian hypocrisy and its corrupted mores. Wilde is interested in describing the follies of the Victorian morality and satirizing the hypocrisy of the Victorian upper classes at the end of the nineteenth century. The modern notion of homosexuality is revealed by Basil. Basil's effusive longings for Dorian mean not only artistic desire but also homosexual desire. From this point of view, Basil who believes in Christianity represents Victorian hypocrisy. While depicts his real life in this novel. Wilde himself was arrested for homosexual offenses under the Criminal Law Amendment Act of 1895. In the problem of homosexuality it must be pointed out that in spite of his crime, Wilde as an artist plays a role of martyr in Victorian society. There is no important act or immoral act in Wilde's aesthetics. The most important thing is that art is superior to ethics.
