        DBA1J 마우스에서 가감보중익기탕이 콜라젠 유도 관절염 억제에 미치는 효과

        N/A Objectives : The present study has been undertaken to investigate the effect of Kagam-Bojungikgitang (KBT) decoction on development of collagen-induced arthrtis (CIA) in DBAl J mice. Methods : CIA was induced in UBAJ1 mice by immunization with bovine type II collagen (CIU and treatment with lipopolysacchride (LPS). KBT is the water extracts prepared from Ginseng Radix, Astragali Radix, Angelicae gigantis Radix, Astractylodis Rhizoma alba, Aurantii nobilis Pericarpium, Glycyrrhizae Radix, Artemisiae iwayomogii Herba, Scutellariae Radx. This is a modifitd prescription of Bojungikgitang, which has been used for the treatment of indigestion, and immunological disease in oriental countries. KBT was orally administered at difference doses. The incidence of arthritis, arthritis index, levels of tumor necrosis factor -alpha (TNF-a). interferon-gama (IFN-V), interleukin lbeta (IL-lP), IL - 3 . IL--10, II- 13. and prostaglandin E? (PGE.1 and cyclooxygenase~-2 (COX21 activity were investigated. Results : KBT dose- dependently suppressed the release of T h l cytokines (TNF-a, IFN-.v, and IL-1 8 ) and inflammatory mediators (PGE? and COX--2 caused by immunization of CII and stimulation of 1,PS. KUT without cytotoxic effext. Kagam-bojunyikgitang`s Inhibitory effects were better than Ehjungikgitang in PGEz production and COX-2 expression. However, KBT increased markedly the production of Th:! cytokines (IL-4, IL 10, and IL -13). Therefor , KBT suppressed markedly the incidence of arthritis and arthritis index caused by immunization of CII and stimulation of LPS. Conclusion : These results suggest that KBT suppress inflammatory mediators and regulates Th l and Th2 cytokmes. And these properties may contribute to the strong anti-arthritis of KBT.

        Activation of Transient Receptor Potential Melastatin Family Member 8 (TRPM8) Receptors Induces Proinflammatory Cytokine Expressions in Bronchial Epithelial Cells

        Purpose: Cold air is a major environmental factor that exacerbates asthma. Transient receptor potential melastatin family member 8 (TRPM8) is a cold-sensing channel expressed in the airway epithelium. However, its role in airway inflammation remains unknown. We investigated the role of TRPM8 in innate immune responses in bronchial epithelial cells and asthmatic subjects. Methods: The TRPM8 mRNA and protein expression on BEAS2B human bronchial epithelial cells was examined by real-time polymerase chain reaction (PCR), immunofluorescence staining and western blotting. Additionally, interleukin (IL)-4, IL-6, IL-8, IL-13, IL-25 and thymic stromal lymphopoietin (TSLP) levels before and after menthol, dexamethasone and N-(4-tert-butylphenyl)-4-(3-chloropyridin-2-yl) piperazine-1-carboxamide (BCTC) treatments were measured via real-time PCR. TRPM8 protein levels in the supernatants of induced sputum from asthmatic subjects and normal control subjects were measured using enzyme-linked immunosorbent assay, and mRNA levels in sputum cell lysates were measured using real-time PCR. Results: Treatment with up to 2 mM menthol dose-dependently increased TRPM8 mRNA and protein in BEAS2B cells compared to untreated cells (P < 0.001) and concomitantly increased IL-25 and TSLP mRNA (P < 0.05), but not IL-33 mRNA. BCTC (10 μM) significantly abolished menthol-induced up-regulation of TRPM8 mRNA and protein and IL-25 and TSLP mRNA (P < 0.01). TRPM8 protein levels were higher in the supernatants of induced sputum from asthmatic subjects (n = 107) than in those from healthy controls (n = 19) (P < 0.001), and IL-25, TSLP and IL-33 mRNA levels were concomitantly increased (P < 0.001). Additionally, TRPM8 mRNA levels correlated strongly with those of IL-25 and TSLP (P < 0.001), and TRPM8 protein levels were significantly higher in bronchodilator-responsive asthmatic subjects than in nonresponders. Conclusions: TRPM8 may be involved in the airway epithelial cell innate immune response and a molecular target for the treatment of asthma.

        $CD4^+$ Jurkat T 세포주에서 Th1과 Th2 사이토가인 조절에 미치는 황금 유래 Baicalin, Baicalein 및 Wogonin의 효과

        In the present study, baicalin, baicalein, and wogonin, a major flavone isolated from Scutellaria Radix were examined for their effects on PMA-induced Interlukin-6 (IL-6), $interferon-\gamma(IFN-\gamma)$, tumor necrosis factor $(TNF)-\alpha$, IL-4, IL-10, and IL-13 productions in the PMA-stimulated $CD4^+$ Jurkat T cells. These three compounds inhibited PMA-induced Th1 cytokine $(IL-6,\;IFN-\gamma,\;TNF-\alpha)$ and Th2 cytokine (IL-4 and IL-13) productions in a concentration-dependent manner. But wogonin, but not baicalin baicalein, increased PMA-induced IL-10 production. These results suggest that baicalin, baicalein, and wogonin, a major flavone modulate Th1 and Th2 cytokine productions in $CD4^+$ Jurkat T cells and these properties may contribute to the anti-atopic dermatitis activity of Scutellaria Radix.

        Suppression of lipopolysaccharide-induced expression of inflammatory indicators in RAW 264.7 macrophage cells by extract prepared from <i>Ginkgo biloba</i> cambial meristematic cells

        <P><I>Context</I>: <I>Ginkgo biloba</I> L. (Ginkgoaceae) leaves have been used as an herbal medicine that has a complex range of biological activities. However, when we consider that biological activity of plant extracts is highly variable according to the source, location, and harvest season, technology to obtain the natural products with homogeneity is extremely important.</P><P><I>Objective</I>: We established the technology to obtain the cambial meristematic cells (CMCs) of <I>Ginkgo biloba</I>, which were expanded <I>in vitro</I> with homogeneity through a suspension culture and then determined the anti-inflammatory activity of fractionated samples prepared from the ethanol extract of CMCs.</P><P><I>Materials and methods</I>: We determined the anti-inflammatory activity of samples using lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophage cells. Especially, influence of sample treatment on the expression of various indicators, such as nitric oxide (NO), inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2, mitogen-activated protein (MAP) kinases, transcription factor, and cytokines, involved in inflammatory activity was assessed.</P><P><I>Results</I>: A fractionated sample demonstrated 53.4% inhibition of LPS-induced NO production from the cells. Additionally, when fractionated samples were treated, iNOS and COX-2 expressions were almost completely suppressed. Fractionated samples also inhibited the phosphorylation of LPS-induced extracellular signal-regulated (ERK) and p38 MAP kinases more than 60%. IκB phosphorylation and subsequent nuclear factor (NF)-κB activation were also suppressed by fractionated samples. The expression of pro-inflammatory cytokines, IL-6 and tumor necrosis factor (TNF)-α, was significantly inhibited by the sample treatment.</P><P><I>Discussion and conclusion</I>: Fractionated samples from the ethanol extract of <I>Ginkgo biloba</I> CMCs could potentially be the source of a powerful anti-inflammatory substance.</P>

        금은화(金銀花)와 황금(黃芩)이 배오(配伍)된 처방제(處方劑)의 항염증(抗炎症) 효과(效果) 연구(硏究)

        Inflammatory reaction is characterized by over-production of inflammatory mediators due to an up-regulation of inflammatory pathways, which produce pro-inflammatory mediators, such as interleukin-1beta (IL-$1{\beta}$), IL-6, tumour necrosis factor alpha (TNF-$\alpha$), prostaglantin $E_2$ ($PGE_2$), and nitric oxide (NO) in U937 cells. We investigate the anti-inflammatory effects of water extracts from Lonicerae Flos and Scutellariae Radix in lipopolysaccharide (LPS)-stimulated U937 cells. Each extract suppressed the production of inflammatory mediators (NO, IL-$1{\beta}$, TNF-$\alpha$, and $PGE_2$) and the expression of inducible NO synthase and cyclooxygenase-2 in LPS- stimulated U937 cells in a dose-dependent manner. These suppressive effects were synergistically increased by their combination. Their combination extract also inhibited NF-${\kappa}B$-DNA complex of NF-${\kappa}B$ binding activity and translocation of NF-${\kappa}B$ from cytosol to nucleus. These results suggest that the combination of water-extractable components of Lonicerae Flos and Scutellariae Radix may be useful for therapeutic drugs against inflammatory immune diseases, probably by suppressing the production of inflammatory mediators.

        MicroRNA-155 as a proinflammatory regulator via SHIP-1 down-regulation in acute gouty arthritis

        <P><B>Introduction</B></P><P>Gout is characterized by episodes of intense joint inflammation in response to intra-articular monosodium urate monohydrate (MSU) crystals. miR-155 is crucial for the proinflammatory activation of human myeloid cells and antigen-driven inflammatory arthritis. The functional role of miR-155 in acute gouty arthritis has not been defined. Therefore, the aim of this study was to examine the role of miR-155 in pathogenesis of acute gouty arthritis.</P><P><B>Methods</B></P><P>Samples from 14 patients with acute gouty arthritis and 10 healthy controls (HCs) were obtained. Peripheral blood mononuclear cells (PBMCs) and synovial fluid mononuclear cells (SFMCs) were cultured <I>in vitro</I> with MSU crystals, and gene expression (human miR-155 and SHIP-1) were assessed by real-time PCR. THP-1 cells were stimulated by MSU crystals and/or miR-155 transfection and then subjected to Western blot analysis. Levels of human tumor necrosis factor-alpha (TNF-α) and interleukin (IL)-1β in cell culture supernatants were measured by Luminex. Immunohistochemistry was performed on formalin-fixed gout tissues with anti–SHIP-1 antibody. A C57BL/6 J male mouse model of gout was used to analyze the expressions of miR-155, SHIP-1, and inflammatory cytokines.</P><P><B>Results</B></P><P>The samples from gouty arthritis were highly enriched in miR-155, with levels of expression being higher than those found in PBMC from HC. Treatment of the cells with MSU crystals strongly induced miR-155. In addition, overexpression of miR-155 in the cells decreased levels of SHIP-1 and promoted production of MSU-induced proinflammatory cytokines, such as TNF-α and IL-1β. Consistent with <I>in vitro</I> observations, miR-155 expression was elevated in the mouse model of gout. The production of inflammatory cytokines was markedly increased in MSU crystal induced peritonitis mice.</P><P><B>Conclusions</B></P><P>Overexpression of miR-155 in the gouty SFMC leads to suppress SHIP-1 levels and enhance proinflammatory cytokines.</P>

        마우스에서 황금 유래 Wogonin의 Th1과 Th2 사이토가인 조절 효과

        In the present study, wogonin, a major flavone isolated from Scutellaria Radix were examined for its imunosuppressive activity on the 2,4-dinitro-fluorobenzene (DNFB)-induced delayed type hypersensitivity (DTH) in C3H/HeN mice. This compound inhibited selectively $TNF-\alpha$ and IL-4, but not IL-6, IL-10 and $IFN-\gamma$ on DNFB-induced Th1 and Th2 cytokines in a concentration-dependent manner. Interestingly, this compound increased the expression of heme oxygenase-1 (HO-1) in a concentration-dependent manner The above results reveal that wogonin possesses anti-inflammatory, humoral and cellular immunomodulatory, and stress reducing activities on the DNFB-induced DTH in mice and these properties may contribute to the anti-atopic dermatitis activity of Scutellaria Radix.

        의료보험 구강요양급여비에 관한 조사연구

        The government of our country has started Medical insurance policy, one of the social security systems. However, statistical date of medical insurance benefits is not much. Especially dental data is insufficient and irrational. In order to get reliable data which offer a valuable suggestion to direction of health care insurance system, the author studied the amount of total care benefits at 1998 which revealed by the survey and analysis of medical statistic index. And it divided into medical insurance, dental insurance, korean-medicine insurance benefits. The dental insurance benefits divided into dental survey and dental disease care benefits again. The dental disease care expenses is composed of dental caries and its advanced disease care expenses, periodontal disease care expenses, other dental disease care expenses. Its date is revealed by the survey and investigation of the demands of 12 dental clinic in 1998. The major findings of the analysis are summarized as follows; 1. In Medical insurance system, the total insurance benefits(include pharmaceutical expenses) was ₩10,115,324,330,000 and the total medical insurance benefits was ₩9,730,793,383,000. The Total dental insurance benefits was ₩647,033,656,600. The ratio of total medical insurance benefits to the total insurance benefit was the 96.20%, and the ratio of dental insurance benefits to the total medical insurance benefits was the 6.65% 2. The individual dental insurance benefits was ₩13,935.7. Among the individual dental insurance benefits, the proportion of the individual dental surveying expenses was 34.62% and individual dental disease care expenses was 46.04% 3. The most part of the dental insurance benefits was dental caries and its advanced disease care expenses and periodontal disease care expenses. 4. Among the dental caries its advanced disease care benefits, the percentage of tertiary preventive expenses was 40.52%. 5. To reduce the expenditure of finance of the health care insurance system and to poromote national oral health, it is necessary to increase the primary and the secondary preventive expenses. 6. Oral diagnostic expenses, oral health care planning expenses and preventive expenses for oral disease, prosthetic expenses in medical insurance benefits didn's supplied. 7. It should be make the legal basis of supply for oral diagnostic expenses and oral health care expenses. 8. Statistic data of the dental insurance should be more rationalize.

        An experimented study was carried out for the estimate of the bond strength property of underwater non-segregation concrete. The action of bond force and slip are affected by various factors. Bond is made up of three components of chemical adhesion and friction, mechanical interaction between concrete and steel. But properties of underwater non-segregation concrete is different from other type of concrete. This study addresses the comparison of physical properties of concrete according to the use of different cement types. It is also recommended to select a proper cement type depending on structural characteristics. Rational analytic formula for the modulus of rupture and bond stress are to be predicted from compressive strength of concrete.

        Characteristics of Korean ginseng varieties of Gumpoong, Sunun, Sunpoong, Sunone, Cheongsun, and Sunhyang

        Background: Ginseng (Panax ginseng Meyer) is an important medicinal herbs in Asia. However, ginsengvarieties are less developed. Method: To developed ginseng varieties, a pure line selection method was applied in this study. Results: Gumpoong was testing of 4-yr-old specimens in 2002, the proportions of the below-groundroots that were rusty colored for Gumpoong was 1.29 in Daejeon and 1.45 in Eumseong, whereas theproportions for its yellow berry variant were 2.60 and 2.45 in the two regions, respectively. Thus theGumpoong was resistant to root rust. Sunpoong has a high yielding property. Its average root weight is70.6 g for 6-yr-old roots. Its yield is 2.9 kg/1.62m2 and the rate of heaven- and earth-grade product is20.9%, which is very high compared to 9.4% for Yunpoong. Sunone is resistance to root rot and thesurvival rate of 4-yr-old roots was 44.4% in 1997, whereas that of the violet-stem variant landrace was21.7%. Sunhyang has content of arginyl-fructosyl-glucose (AFG), which produces the unique scent of redginseng, is 95.1 mmol/g and greater than the 30.8 mmol/g of Chunpoong in 6-yr-old plants. Sunun andCheongsun are being nurtured to protect genetic resources. Conclusion: Developed ginsneg varieties will be used as the basis for the protection of genetic resourcesand breeding. Jang-Ho Lee
