        Comprehensive genomic analyses with 115 plastomes from algae to seed plants: structure, gene contents, GC contents, and introns

        Background Chloroplasts are a common character in plants. The chloroplasts in each plant lineage have shaped their own genomes, plastomes, by structural changes and transferring many genes to nuclear genomes during plant evolution. Some plastid genes have introns that are mostly group II introns. Objective This study aimed to get genomic and evolutionary insights on the plastomes from green algae to flowering plants. Methods Plastomes of 115 species from green algae, bryophytes, pteridophytes (spore bearing vascular plants), gymnosperms, and angiosperms were mined from NCBI organelle genome database. Plastome structure, gene contents and GC contents were analyzed by the in-house developed Phyton code. Intronic features including presence/absence, length, intron phases were analyzed by manually in the annotated information in NCBI. Results The canonical quadripartite structures were retained in most plastomes except of a few plastomes that had lost an invert repeat (IR). Expansion or reduction or deletion of IRs resulted in the length variation of the plastomes. The number of protein coding genes ranged from 40 to 92 with an average 79.43 ± 5.84 per plastome and gene losses were apparent in specific lineages. The number of trn genes ranged from 13 to 33 with an average 21.19 ± 2.42 per plastome. Ribosomal RNA genes, rrn, were located in the IRs so that they were present in a duplicate except of the species that had lost one of the IR. GC contents were variable from 24.9 to 51.0% with an average 38.21 ± 3.27%, indicating bias to high AT contents. Plastid introns were present in 18 protein coding genes, six trn genes, and one rrn gene. Intron losses occurred among the orthologous genes in different plant lineages. The plastid introns were long compared with the nuclear introns, which might be related with the spliceosome nuclear introns and self-splicing group II plastid introns. The trnK-UUU intron contained the maturase encoding matK gene except in the chlorophyte algae and monilophyte ferns in which the trnK-UUU was lost, but matK retained. There were many annotation artefacts in the intron positions in the NCBI database. In the analysis of intron phases, phase 0 introns were more frequent than those of phase 2 and 3 introns. Phase polymorphism was observed in the introns of clpP which was derived from nucleotide insertion. Plastid trn introns were long compared to the archaeal or eukaryotic nuclear tRNA introns. Of the six plastid trn introns, one was at the D loop and other five were at the anticodon loop. The insertion sites were conserved among the trn genes in archaea, eukaryotic nuclear and plastid tRNA genes. Conclusions Current study refurbrished the previous findings of structural variations, gene contents, and GC contents of the chloroplast genomes from green algae to flowering plants. The study also included some noble findings and discussions on the plastome introns including their length variations and phase variation. We also presented and corrected some false annotations on the introns in protein coding and tRNA genes in the genome database, which might be confirmed by the chloroplast transcriptome analysis in the future.

        The effect of Carbontetrachloride administration on Lactatedehydrogenase isozyme patterns were analyzed by cellulose acetate electrophoresis for the brain, heart, kidney, lung, liver and mouse muscle. Each other tissue was found to have a characteristic distribution of these isozymes. At normal condition, aerobic tissue such as brain, heart, kidney, and lung were found to have all five isozymes from LDH_1 to LDH_5, while anaerobic tissues such as liver and muscle were found to have superiorly LDH_5 The toxicity of Carbontetrachloride administration was most prominence in the liver, and lung toxicity was occured also.

        새로운 장류제품으로서 보리등겨의 이용방안을 모색하기 위하여 간장을 만들어 연구하였다. 보리로 제조한 간장의 갈색화는 점차적으로 증가 하였으며 완만한 변화를 보였다. 향기성분으로는 4-vinyl-2-methoxy-phenol, benzeneacetaldehyde, palmitic acid, 2-furancatboxaldehyde, methyl-9, 12-octadecadienoate, di-(2-ethylhexyl)phthalate, diethyl phtalate, dibytyl-1,2-benzenedicatboxylate, 5-methyl-2-furancarboxaldehyde, 3,4-dimethyl-1h-pyrazole, phenylethyl alcohol, dioctyl-hexanedioate, dimethyl-1,2-benzenedicatboxylate, benzaldehyde, methional, 2-methoxy-phenol, n-furfurylidene-3-methylbutyl amine, 1-furfuryl-2-formyl pyrrole, tetradrcanoic acid, 5-methyl-pyrimidine, 4-methyl-5-hydroxymethyl-imidazole, maltol, 5-(5-methyl-2-furanyl)methyl-2-furancarboxaldehyde 순으로 높은 함량을 차지 하였다. For investigation of new utilization as jang-products, kanjang was prepared using barely bran. This study was conducted to investigate flavor components of kanjang during fermentation time. The optical density was gradually increased. Among the flavor components identified in kanjang made with barley bran, the contents of 4-viny1-2-methoxy-phenol was the most in quantity followed by benzeneacetaldehyde, palmitic acid, 2-furancarboxaldehyde, methyl-9,12-octadecadienoate, di-(2-ethylhexyl)phthalate, diethyl phtalate, dibutyl-1,2-benzendicarboxylate, 5-methyl-2-furancarboxaldehyde, 3,4-dimethyl-1h-pyrazole, phenylethyl alcohol, dioctyl-hexanedioate, dimethyl-1,2-benzenedicarboxylate, benzaldehyde, methional, 2-methoxy-phenol, n-furfurylidene-3-methylbutyl amine, 1-furfuryl-2-formyl pyrrole, tetradecanoic acid, 5-methyl-pyrimidine, 4-methyl-5-hydroxymethyl-imidazole, maltol and 5-(5-methyl-2-furanyl)methyl-2-furancarboxaldegyde.

        Purpose : The purpose of this study was to identify the physical health status and health behavior practice of in-home elderly in order to provide basic data for effective nursing intervention to promote health and quality of life. Method : The subjects for this study were 299 elderly persons in the D city. Data were collected by interview with questionnaires. Result : The ratio of chronic disease was higher for the elderly who live alone than the elderly who live with family. Arthritis was the most common chronic disease of the elderly in spite of the fact that either they live alone or live with family. The physical health status score of the elderly who live with family was statistically higher than the elderly who live alone. The health behavior score of the elderly who live with family was statistically higher than the elderly who live alone. Conclusion : To promote health behavior of the elderly, it is necessary to develop nursing intervention that considers sociocultural tradition and demographic character.

The approach is capable of systematically calculating a set of appropriate weighting matrices for the quadratic performance index, so that the resulting optimal control law would not only minimize the index, but also place the eigenspectrum of the closed-loop system at some predefined location.

        2004년 7월 봇돌바다의 영양염과 Chlorophyll-a의 단기 변동

        In order to study the temporal variations of nutrients and chlorophyll-a in the Bottol Bada, three field observations were carried out on 20, 23 and 26 July, 2004. The low N:P values exhibit nitrogen deficiency during the periods of observation. This result is not representative of typical summer environment in the southern coast of Korea. The possible mechanisms are as follows: 1) The freshwater inflow was not sufficient for the supply of nitrogen because the total precipitation was 11.9 mm in July, 2004. This amount is no more than 5% in normal precipitation in July. 2) There was an inflow of oceanic water under the subsurface into the Bottol Bada. Even though the oceanic water comprises more nutrients, it produces the stratification between the surface and the subsurface water and seems to prevent the supply of nutrinets to the surface layer. 3) The high chlorophyll-a concentration of 1.2 ㎍/L was shown near the narrow channel between Gae-do and Geumo-do. This seems to be resulted from the inflow of water from Gamak Bay.

        간호실무에서 신봉이론(Espoused theories)에 대한 탐색적 연구

        As a nursing practice involves nurses' actions in a specific context of health care, this study has focused on exploring the espoused theories in nursing practice within the action science perspectives, Espoused theories are the belief, principles, and rationale expressed by the practitioner as guiding her/his actions in a situation of practice. The data were analysed qualitatively and 25 elements of espoused theories of nursing action were identified and clustered into 6 categories. The 25 elements of espoused theories are as follows: The clinical nurse worked in wholistic and individual nursing, focussed on the patient's needed, comfort and supportive nursing (5 theories of nursing goal) ; excellent skills, knowledge based, assessment and data collection, explaining, educating or a scientific basis(6 theories of nursing intervention): advocacy, value oriented, treatment, account- ability and commitment(4 theories of nursing ethics) ; human respect. partnership, trust(3 theories of patient-nurse relationship) : knowledgable, accumulated clinical experiences and personally lived experiences. positive perspectives(4 theories of nurse), role of intervention. rewarding peer relationship(3 theories of situations) The above mentioned espoused theories are similar to that of nursing textbooks which students learned through basic nursing education and almost the same as the Acts ofa Nurse in Korean. However, we are doubtful whether nurses actually do as they think. Therefore. it is recommended to review the theories-in-use in order to and any discrepancies between the espoused theories and the reality of nursing actions

        Biotransformation of Pueraria lobata Extract with Lactobacillus rhamnosus vitaP1 Enhances Anti-Melanogenic Activity

        Isoflavone itself is less available in the body without the aid of intestinal bacteria. In this study, we searched for isoflavone-transforming bacteria from human fecal specimens (n = 14) using differential selection media. Isoflavone-transforming activity as the production of dihydrogenistein and dihydrodaidzein was assessed by high-performance liquid chromatography and we found Lactobacillus rhamnosus, named L. rhamnosus vitaP1, through 16S rDNA sequence analysis. Extract from Pueraria lobata (EPL) and soy hypocotyl extract were fermented with L. rhamnosus vitaP1 for 24 and 48 h at 37℃. Fermented EPL (FEPL) showed enhanced anti-tyrosinase activity and antioxidant capacities, important suppressors of the pigmentation process, compared with that of EPL (p < 0.05). At up to 500 μg/ml of FEPL, there were no significant cell cytotoxicity and proliferation on B16-F10 melanoma cells. FEPL (100 μg/ml) could highly suppress the content of melanin and melanosome formation in B16-F10 cells. In summary, Lactobacillus rhamnosus vitaP1 was found to be able to biotransform isoflavones in EPL. FEPL showed augmented anti-melanogenic potential.

        경기 화성지역 중학생의 환경호르몬에 대한 지식수준과 식품포장재의 소비행동

        Purpose: This study investigated the consumption behavior (positive and negative) of food packing materials and the knowledge level (low, medium and high groups) of endocrine disruptors, among middle school students (n=240) in the Hwaseong region. Methods: This research was conducted between 18 and 22 May, 2015, and was achieved using the questionnaire method. Data were analyzed by applying the chi-square test, t-test and one way ANOVA test. Results: Considering the gender ratio, higer knowledge level was obtained in girls than in boys (p<0.05). Difference of knowledge score by gender was also higher in girls in 4 of the factors evalulated: harmfulness (p<0.05), size of organism and accumulation (p<0.01), excessive using of pesticide in fruit (p<0.05), putting hot foods in plastic materials (p<0.05). Assessment of positive consumption behaviors revealed that girls (3.20 score) showed a higher score in caution of plastics or vinyls when putting hot food than boys (2.89 score, p<0.05). Conversely, evaluation of negative consumption behaviors revealed that boys showed a higher score in eating all the soup of cup ramen (boys 3.07 score, girls 2.54 score, p<0.01) and microwave cooking of cup ramen made of styrofoam (boys 2.19 score, girls 1.91 score, p<0.05). In the comparison of groups for positive consumption behavior by knowledge level, a higher score (3.33) was obtained by the high group in following recipe when using a microwave oven, than the low (2.70 score) and medium (3.17 score) groups (p<0.05). Moreover, the negative consumption behavior of reusing polyethylene terephthalate (PET) bottle to put hot water was higher in the low group (2.15 score) as compared to the medium (1.93 score) and high (1.63 score) groups (p<0.05). Conclusion: Taken together, our results indicated that to induce good consumption behavior of food packing materials, considering the gender need to take precedence when imparting knowledge improvement related to endocrine disruptors.
