        RhoGDI2 induced malignant phenotypes of pancreatic cancer cells via regulating Snail expression

        Background: Rho GDP dissociation inhibitor 2 (RhoGDI2) has been shown to contribute to the aggressive phenotypes of human cancers, such as tumor metastasis and chemoresistance. Objective: This study aimed to assess the effects of RhoGDI2 on tumor progression and chemoresistance in pancreatic cancer cells. Methods: The expression of RhoGDI2 in pancreatic cancer cells was detected by Western blot analysis. Gain-of-function and loss-of-function approaches were done to examine the malignant phenotypes of the RhoGDI2-expressing or RhoGDI2-depleting cells. The correlation between RhoGDI2 and Snail was also analyzed. Results: Differential expression of RhoGDI2 protein in pancreatic cancer cell lines was identified. Gain-of-function and loss-of-function experiments showed that RhoGDI2 induced the malignant phenotypes of pancreatic cancer cells, including proliferation, migration, invasion, and gemcitabine (GEM) chemoresistance. The upregulation of RhoGDI2 stimulated the expression of Snail, resulting in the altered expression of epithelial marker E-cadherin and mesenchymal marker Vimentin, which were characteristics of the tumorigenic activity of epithelial-mesenchymal transition. The expression of RhoGDI2 and Snail was upregulated in clinical tumor samples, and higher expression of RhoGDI2 or Snail was significantly associated with poor patient survival in pancreatic ductal adenocarcinoma (PDAC). Conclusion: The findings indicated that RhoGDI2 promoted GEM resistance and tumor progression in pancreatic cancer and that RhoGDI2 might be a potential therapeutic target in patients with PDAC.

        Study on Aging Characteristics and Chemical Composition of Hydrogenated Transformer Oil

        Under the condition of Baader aging, the chemical composition variation and the influence of transformer oil aging on electrical properties such as dielectric loss factor and physicchemical properties such as interfacial tension were studied in the aging precess. Moreover, the correlation between hydrogenated transformer oil electrical and physic-chemical properties and its chemical composition variation were also investigated. The results show that these parameters of physic-chemical and electrical properties of hydrogenated transformer oil relate to each other and have closed correlation with chemical composition.

        Impedance Modeling and Resonance Suppression Method for Current Source VSG Under Weak Grid Conditions

        A large proportion of renewable energy is integrated into power system, resulting in the gradual electrifi cation of the power system and appearing the characteristics of a weak power grid. Under weak grid, current source virtual synchronous generator (VSG) is prone to resonance. To ensure stable and uninterrupted operation of renewable energy through power electronic devices connected to grid, this paper proposes a composite control method based on grid voltage feedforward (GVF) and q-axis impedance reshaping (QAIR) for current source VSG converter resonance suppression. Firstly, sequence impedance modeling (SIM) and stability analysis are conducted on current source VSG converter. Under weak grid, the negative resistance eff ect caused by phase-locked loop (PLL) in the low frequency range makes system unstable in grid connection. To eliminate the negative resistance instability caused by PLL, a current source VSG GVF control resonance suppression method is established. SIM of this method and stability analysis are conducted. Secondly, a QAIR resonance suppression method for current source VSG converter is established. SIM of the method is conducted and stability analyzed. On the basis of above two methods, a composite resonance suppression method based on GVF and QAIR is established, and SIM and stability analysis are conducted. Simulation and experimental tests are conducted on the stability of GVF, QAIR, and GVF + QAIR composite resonance suppression current type VSG systems. The test results showed that the method based on GVF + QAIR resonance suppression is more eff ective, compared to GVF/QAIR resonance suppression method. Under weak grid, it has better ability to eliminate negative resistance eff ects and harmonic suppression caused by PLL, further improving grid-connected stability.

        Canonical correlation analysis based fault diagnosis method for structural monitoring sensor networks

        The health conditions of in-service civil infrastructures can be evaluated by employing structural health monitoring technology. A reliable health evaluation result depends heavily on the quality of the data collected from the structural monitoring sensor network. Hence, the problem of sensor fault diagnosis has gained considerable attention in recent years. In this paper, an innovative sensor fault diagnosis method that focuses on fault detection and isolation stages has been proposed. The dynamic or auto-regressive characteristic is firstly utilized to build a multivariable statistical model that measures the correlations of the currently collected structural responses and the future possible ones in combination with the canonical correlation analysis. Two different fault detection statistics are then defined based on the above multivariable statistical model for deciding whether a fault or failure occurred in the sensor network. After that, two corresponding fault isolation indices are deduced through the contribution analysis methodology to identify the faulty sensor. Case studies, using a benchmark structure developed for bridge health monitoring, are considered in the research and demonstrate the superiority of the new proposed sensor fault diagnosis method over the traditional principal component analysis-based and the dynamic principal component analysis-based methods.

        haracterization of a Recombinant Thermostable Xylanase from Hot Spring Thermophilic Geobacillus sp. TC-W7

        A xylanase-producing thermophilic strain, Geobacillus sp. TC-W7, was isolated from a hot spring in Yongtai (Fuzhou, China). Subsequently, the xylanase gene that encoded 407 amino acids was cloned and expressed. The recombinant xylanase was purified by GST affinity chromatography and exhibited maximum activity at 75℃ and a pH of 8.2. The enzyme was active up to 95℃ and showed activity over a wide pH range of 5.2 to 10.2. Additionally, the recombinant xylanase showed high thermostability and pH stability. More than 85% of the enzyme`s activity was retained after incubation at 70℃ for 90 min at a pH of 8.2. The activity of the recombinant xylanase was enhanced by treatment with 10 mM enzyme inhibitors (DDT, Tween-20, 2-Me, or TritonX-100) and was inhibited by EDTA or PMSF. Its functionality was stable in the presence of Li+, Na+, and K+, but inhibited by Hg2+, Ni2+, Co2+, Cu2+, Zn2+, Pb2+, Fe3+, and Al3+. The functionality of the crude xylanase had similar properties to the recombinant xylanase except for when it was treated with Al2+ or Fe2+. The enzyme might be a promising candidate for various industrial applications such as the biofuel, food, and paper and pulp industries.

        Appropriate Soil Heat Treatment Promotes Growth and Disease Suppression of Panax notoginseng by Interfering with the Bacterial Community

        In our greenhouse experiment, soil heat treatment groups (50, 80, and 121°C) significantly promoted growth and disease suppression of Panax notoginseng in consecutively cultivated soil (CCS) samples (p < 0.01), and 80°C worked better than 50°C and 121°C (p < 0.01). Furthermore, we found that heat treatment at 80°C changes the microbial diversity in CCS, and the inhibition ratios of culturable microorganisms, such as fungi and actinomycetes, were nearly 100%. However, the heat-tolerant bacterial community was preserved. The 16S rRNA gene and internal transcribed spacer (ITS) sequencing analyses indicated that the soil heat treatment had a greater effect on the Chao1 index and Shannon’s diversity index of bacteria than fungi, and the relative abundances of Firmicutes and Proteobacteria were significantly higher than without heating (80 and 121°C, p < 0.05). Soil probiotic bacteria, such as Bacillus (67%), Sporosarcina (9%), Paenibacillus (6%), Paenisporosarcina (6%), and Cohnella (4%), remained in the soil after the 80°C and 121°C heat treatments. Although steam increased the relative abundances of most of the heat-tolerant microbes before sowing, richness and diversity gradually recovered to the level of CCS, regardless of fungi or bacteria, after replanting. Thus, we added heat-tolerant microbes (such as Bacillus) after steaming, which reduced the relative abundance of pathogens, recruited antagonistic bacteria, and provided a long-term protective effect compared to the steaming and Bacillus alone (p < 0.05). Taken together, the current study provides novel insight into sustainable agriculture in a consecutively cultivated system.

        Development of a Serum-Free Culture Method for Endothelial Cells of the Stria Vascularis and Their Pro-inflammatory Secretome Changes Induced by Oxidative Stress

        Objectives. Reactive oxygen species in the stria vascularis (SV) of the cochlea may be involved in the pathogenesis of sen-sorineural hearing loss. However, the effects of oxidative stress on SV endothelial cells (SV-ECs) remain largely un-known, and no feasible in vitro cell culture model exists for the functional study of SV-ECs. Methods. We isolated primary SV-ECs from the SV of neonatal mice. The apoptosis-reducing effects of fibronectin in SV-ECs cultured with serum-free medium were determined using β-galactosidase staining and flow cytometry. SV-ECsincubated in serum-free medium were treated with various H2O2 concentrations to evaluate the effects of H2O2 ontheir viability. The secretome of SV-ECs treated with or without H2O2 (100 μM or 500 μM) was analyzed using high-resolution mass spectrometry. The function of the SV-EC secretome was evaluated by a macrophage assay. Results. We successfully isolated and characterized the SV-ECs. Treatment with H2O2 at concentrations up to 500 μM for2 hours and further incubation with serum-free medium in plates precoated with fibronectin showed no significanteffect on apoptosis. Compared to the control SV-ECs, the amount of differential proteins in the secretome of SV-ECsstimulated with 500 μM H2O2 was much higher than in those treated with 100 μM H2O2. Kyoto Encyclopedia ofGenes and Genomes and Gene Ontology analyses suggested that the proteins differentially expressed in SV-ECstreated with 500 μM H2O2 were involved in the regulation of multiple signaling pathways and cellular processes. Thesecretome of H2O2-stimulated SV-ECs exhibited significant pro-inflammatory effects on macrophages. Conclusion. We successfully established an in vitro serum-free culture method, identified the differential proteins releasedby oxidative stress-induced ECs and their functions, and revealed the pro-inflammatory effects of the secretome ofH2O2-stimulated SV-ECs. Therefore, SV-ECs might elicit immunoregulatory effects on bystander cells in the microen-vironment of oxidative stress-induced cochlea, especially cochlear macrophages.

        <P>Rational bottorritup construction of two-dimensional (2D) covalent or nonCovalent organic materials with precise structural control at the: atomic or molecular level remains a challenge. The design and synthesis of metal organic frameworks (MOFS) based on new building blocks is of great significance in achieving new types of 2D monolayer MOF films. Here, we demonstrate that a complexation between copper(II) ions and tri(beta-diketone) ligands yields a novel 2D MOF siyucture, either-inthe form of a powder.or.as a monolayer filth. It has been Characterized by Fourier transform infrared, Raman, ultraviolet-visible, X-ray photoelectron, and electron paramagnetic-resonance spectroscopies. Selected area electron diffraction and powder X-ray diffraction results show that the MOF is Crystalline and has a hexagonal structure. A MOF-based membrane has been prepared by vacuum filtration of an aqueous dispersion of the MOF powder onto a porous Anodisc filter having pore-size 0.02 mu m. The porous MOF membrane filters gold nanOparticies with a cutoff of similar to 2.4 nm.</P>
