        Evaluation of secondary cytoreduction surgery in platinum-resistant ovarian cancer patients within three-line recurrent: a multicenter, randomized controlled study

        Background: Epithelial ovarian cancer is the leading cause of death among gynecologicalmalignancies. Platinum resistance remains a dilemma and bottleneck in treatment, andsalvage chemotherapy has limited effectiveness. Recently, the role of secondar y cytoreductivesurger y (SCS) in patients with platinum-resistant recurrent ovarian cancer (ROC) has causedattention especially in patients with oligometastases. However, there is neither high-qualityevidence-based evidence nor standardized criteria for selecting SCS for patients withplatinum-resistant ROC until now. Methods: This multicenter, randomized, controlled clinical trial is to evaluate the valueof SCS and to clarif y reliable criteria of utilizing SCS in women with ROC, which is led byGynecologic Oncology Group, Women’s Hospital, Zhejiang University School of Medicine. Recruitment has started on Januar y 1st, 2023, and is scheduled to end in December 2026. One hundred and forty participants with platinum-resistant ROC who meet the “RSCScriteria” will be randomized assigned at a ratio of 1:1 to either the experimental arm or thestandard arm. Patients in the experimental arm will receive SCS followed by non-platinumsingle agent chemotherapy (paclitaxel, gemcitabine or liposomal adriamycin) for at least4 cycles while patients in the standard arm will be provided with only non-platinum singleagent chemotherapy. The primar y outcome is progression-free sur vival. The secondar youtcomes are overall sur vival, adverse events and health-related cancer-specific quality of life.         <P>Sneak path current is a severe hindrance for the application of high-density resistive random-access memory (RRAM) array designs. In this work, we demonstrate nonlinear (NL) resistive switching characteristics of a HfO<SUB> <I>x</I> </SUB>/SiO<SUB> <I>x</I> </SUB>-based stacking structure as a realization for selector-less RRAM devices. The NL characteristic was obtained and designed by optimizing the internal filament location with a low effective dielectric constant in the HfO<SUB> <I>x</I> </SUB>/SiO<SUB> <I>x</I> </SUB> structure. The stacking HfO<SUB> <I>x</I> </SUB>/SiO<SUB> <I>x</I> </SUB>-based RRAM device as the one-resistor-only memory cell is applicable without needing an additional selector device to solve the sneak path issue with a switching voltage of ~1 V, which is desirable for low-power operating in built-in nonlinearity crossbar array configurations.</P>

        For the promotion of community development, the first step is to investigate the specialty resources and human resources, to understand the process and the current appearance of community development, and to explore the future development vision or direction should be promoted. Secondly, the local smart people must have the abilities of cooperation, communication, cross-domain integration, and linking external support. It is one of the keypoints of adding-value workforce. Therefore, leaders of a community must find the local characteristics of the championship, and have the capacity to promote citizen participation and to integrate external support and cooperation, so that communities can acquire and integrate diverse and sufficient resources in order to succeed in community sustainability. Therefore, the main purpose of this paper is to establish a model of resource inventory and integration. Through the methods of literature analysis and focus group discussion, this paper explores the community cases of Guantu area in Taipei, Shennong Street and Jingliao community in Tainan how to inventory both natural and human resources, to seek community development and value-added directions, to promote civic participation, cooperation, and to link external groups, so as to achieve the success of internal and external resource integration.

        Combination of Tumor Volume and Epstein-Barr Virus DNA Improved Prognostic Stratification of Stage II Nasopharyngeal Carcinoma in the Intensity Modulated Radiotherapy Era: A Large-Scale Cohort Study

        Purpose Little is known about combination of the circulating Epstein-Barr viral (EBV) DNA and tumor volume in prognosis of stage II nasopharyngeal carcinoma (NPC) patients in the intensity modulated radiotherapy (IMRT) era. We conducted this cohort study to evaluate the prognostic values of combining these two factors. Materials and Methods By Kaplan-Meier, we compare the differences of survival curves between 385 patients with different EBV DNA or tumor volume levels, or with the combination of two biomarkers mentioned above. Results Gross tumor volume of cervical lymph nodes (GTVnd, p < 0.001) and total tumor volume (GTVtotal, p < 0.001) were both closely related to pretreatment EBV DNA, while gross tumor volume of nasopharynx (GTVnx, p=0.047) was weakly related to EBV DNA. EBV DNA was significantly correlated with progress-free survival (PFS, p=0.005), locoregional-free survival (LRFS, p=0.039), and distant metastasis-free survival (DMFS, p=0.017), while GTVtotal, regardless of GTVnx and GTVnd, had a significant correlation with PFS and LRFS. The p-values of GTVtotal for PFS and LRFS were 0.008 and 0.001, respectively. According to GTVtotal and pretreatment EBV DNA level, patients were divided into a low-risk group (EBV DNA 0 copy/mL, GTVtotal < 30 cm3; EBV DNA 0 copy/mL, GTVtotal  30 cm3; or EBV DNA > 0 copy/mL, GTVtotal < 30 cm3) and a high-risk group (EBV DNA > 0 copy/mL, GTVtotal  30 cm3). When patients in the low-risk group were compared with those in the high-risk group, 3-year PFS (p=0.003), LRFS (p=0.010), and DMFS (p=0.031) rates were statistically significant. Conclusion Pretreatment plasma EBV DNA and tumor volume were both closely correlated with prognosis of stage II NPC patients in the IMRT era. Combination of EBV DNA and tumor volume can refine prognosis and indicate for clinical therapy.

        Mesenchymal Stem Cell Secreted-Extracellular Vesicles are Involved in Chondrocyte Production and Reduce Adipogenesis during Stem Cell Differentiation

        BACKGROUND: Extracellular vesicles (EVs) are derived from internal cellular compartments, and have potential as a diagnostic and therapeutic tool in degenerative disease associated with aging. Mesenchymal stem cells (MSCs) have become a promising tool for functional EVs production. This study investigated the efficacy of EVs and its effect on differentiation capacity. METHODS: The characteristics of MSCs were evaluated by flow cytometry and stem cell differentiation analysis, and a production mode of functional EVs was scaled from MSCs. The concentration and size of EVs were quantitated by Nanoparticle Tracking Analysis (NTA). Western blot analysis was used to assess the protein expression of exosomespecific markers. The effects of MSC-derived EVs were assessed by chondrogenic and adipogenic differentiation analyses and histological observation. RESULTS: The range of the particle size of adipose-derived stem cells (ADSCs)- and Wharton’s jelly -MSCs-derived EVs were from 130 to 150 nm as measured by NTA, which showed positive expression of exosomal markers. The chondrogenic induction ability was weakened in the absence of EVs in vitro. Interestingly, after EV administration, type II collagen, a major component in the cartilage extracellular matrix, was upregulated compared to the EV-free condition. Moreover, EVs decreased the lipid accumulation rate during adipogenic induction. CONCLUSION: The results indicated that the production model could facilitate production of effective EVs and further demonstrated the role of MSC-derived EVs in cell differentiation. MSC-derived EVs could be successfully used in cell-free therapy to guide chondrogenic differentiation of ADSC for future clinical applications in cartilage regeneration.

        Nomogram for predicting overall survival in children with neuroblastoma based on SEER database

        Purpose: This study was performed to establish and validate a nomogram for predicting the overall survival in children with neuroblastoma. Methods: The latest clinical data of neuroblastoma in Surveillance, Epidemiology, and End Results (SEER) database was extracted from 2000 to 2016. The cases included were randomly divided into training and validation cohorts. The survival curves were drawn with a Kaplan-Meier estimator to investigate the influences of certain single factors on overall survival. Also, least absolute shrinkage and selection operator regression was applied to further select the prognostic variables for neuroblastoma. Additionally, receiver operating characteristic (ROC) curves and calibration curves were used to evaluate the accuracy of the nomogram. Results: In total, 1,262 patients were collected and 8 independent prognostic factors were achieved, including patients’ age, sex, race, tumor grade, radiotherapy, chemotherapy, tumor site, and tumor size. Then we constructed a nomogram by using the data of the training cohort with 886 cases. Subsequently, the nomogram was validated internally and externally with 886 and 376 cases, respectively. The internal validation revealed that the area under the curves (AUC) of ROC curves of 1-, 3-, and 5-year overall survival were 0.69, 0.78, and 0.81, respectively. Accordingly, the external validation also showed that the AUC of 1-, 3-, and 5-year overall survival were all ≥0.69. Both methods of validation demonstrated that the predictive calibration curves were consistent with standard curves. Conclusion: The nomogram possess the potential to be a new tool in predicting the survival rate of neuroblastoma patients.

      • The Ca <sub>V</sub> 3.2 T-Type Ca <sup>2+</sup> Channel Is Required for Pressure Overload-Induced Cardiac Hypertrophy in Mice

        <P>Voltage-gated T-type Ca(2+) channels (T-channels) are normally expressed during embryonic development in ventricular myocytes but are undetectable in adult ventricular myocytes. Interestingly, T-channels are reexpressed in hypertrophied or failing hearts. It is unclear whether T-channels play a role in the pathogenesis of cardiomyopathy and what the mechanism might be. Here we show that the alpha(1H) voltage-gated T-type Ca(2+) channel (Ca(v)3.2) is involved in the pathogenesis of cardiac hypertrophy via the activation of calcineurin/nuclear factor of activated T cells (NFAT) pathway. Specifically, pressure overload-induced hypertrophy was severely suppressed in mice deficient for Ca(v)3.2 (Ca(v)3.2(-/-)) but not in mice deficient for Ca(v)3.1 (Ca(v)3.1(-/-)). Angiotensin II-induced cardiac hypertrophy was also suppressed in Ca(v)3.2(-/-) mice. Consistent with these findings, cultured neonatal myocytes isolated from Ca(v)3.2(-/-) mice fail to respond hypertrophic stimulation by treatment with angiotensin II. Together, these results demonstrate the importance of Ca(v)3.2 in the development of cardiac hypertrophy both in vitro and in vivo. To test whether Ca(v)3.2 mediates the hypertrophic response through the calcineurin/NFAT pathway, we generated Ca(v)3.2(-/-), NFAT-luciferase reporter mice and showed that NFAT-luciferase reporter activity failed to increase after pressure overload in the Ca(v)3.2(-/-)/NFAT-Luc mice. Our results provide strong genetic evidence that Ca(v)3.2 indeed plays a pivotal role in the induction of calcineurin/NFAT hypertrophic signaling and is crucial for the activation of pathological cardiac hypertrophy.</P>

        Construction of DOPA-SAM multilayers with corrosion resistance via controlled molecular self-assembly

        Although nano “green” coatings with excellent corrosion resistance have attracted great attention, the inhibition efficiency is still limited due to the lack of knowledge about the correlation between molecular structure and anticorrosion performance. Here, we fabricated a series of 3,4-dihydroxy-l-phenylalanine adlayers on self-assembled monolayers (SAMs) with varying end groups. We found that both NH2 and CF3SAMs were more conducive to the adsorption of DOPA and a flat adsorption conformation was preferentially adopted, with the plane of the phenylene ring parallel to the surface via cation-π interactions or hydrophobic interactions, leading to a compact and dense adlayer. Such DOPA-SAM multilayers can effectively protect the substrate from corrosion by suppressing the diffusion of aggressive water and acid molecules as well as the electrodissolution of metals. The lowest corrosion current of adlayers reaches 6.96 μA cm−2 which is much lower than that of bare substrate and other anticorrosion surfaces reported previously. The results provide guidance on the design of green anticorrosion materials via selecting SAMs that bridge organic and metal interface.

        FePO4-coated Li5Cr7Ti6O25 nanocomposites as anode materials for high-performance lithium-ion batteries

        FePO4 coated Li5Cr7Ti6O25 nanocomposites are synthesized according to a simple sol–gel method followingby a calcination procedure. XRD and Rietveld refinement result indicate that FePO4 decorating doesnot change the crystal structure and lattice parameters of Li5Cr7Ti6O25. SEM and TEM prove that the particlesize of all sample are evenly distributed in the 50–100 nm range. Both energy dispersion spectroscopymapping and HRTEM indicate the existence of FePO4 on the surface of Li5Cr7Ti6O25, whichprovides a good conductive contact. CV and EIS exhibit that FePO4 (3 wt%) coated Li5Cr7Ti6O25 materialhas lower polarization, larger Li+ diffusion coefficient and higher conductivity than other pureLi5Cr7Ti6O25 and other FePO4 coated Li5Cr7Ti6O25 composites. Therefore, FePO4 (3 wt%) coatedLi5Cr7Ti6O25 anode material displays the highest charge and discharge capacity at each rate. Theenhanced electrochemical performance of FePO4-coated Li5Cr7Ti6O25 result from the enhanced lithiumion and electron transfer kinetics. The pristine Li5Cr7Ti6O25 only shows a specific capacity of197 mAh g 1 at 500 mA g 1 after 100 cycles, but FePO4 (3 wt%)-coated Li5Cr7Ti6O25 achieves a highcapacity of 237.9 mAh g 1. Therefore, the FePO4 coating can be regarded as a promising strategy toimprove the electrochemical properties of Li5Cr7Ti6O25.

        Candida albicans ENO1 Null Mutants Exhibit Altered Drug Susceptibility, Hyphal Formation, and Virulence

        We previously showed that the expression of ENO1 (enolase) in the fungal pathogen Candida albicans is critical for cell growth. In this study, we investigate the contribution of the ENO1 gene to virulence. We conducted our functional study of ENO1 in C. albicans by constructing an eno1/eno1 null mutant strain in which both ENO1 alleles in the genome were knockouted with the SAT1 flipper cassette that contains the nourseothricin-resistance marker. Although the null mutant failed to grow on synthetic media containing glucose, it was capable of growth on media containing yeast extract, peptone, and non-fermentable carbon sources. The null mutant was more susceptible to certain antifungal drugs. It also exhibited defective hyphal formation, and was avirulent in BALB/c mice.
