      • 양배추 Peroxidase에 의한 효소적 포도당 분석에 미치는 환원제의 영향

        The enzymatic glucose assay using cabbage peroxidase was inhibited by various sulfhydryl reagents. Among test sulfhydryl reagents, reduced glutathione was most effecitive for the inhibition of enzymatic glucose assay. The sulfhydryl reagents were not decomposed at lower concentration of hydrogen peroxide, but it was decomposed partially by addition of higher concentration of hydrogen peroxide. Therefore, the sulfhydryl reagents seem to inhibit the glucose assay by inhibiting cabbage peroxidase used in the assay.

      • Administration of placenta-derived mesenchymal stem cells counteracts a delayed anergic state following a transient induction of endogenous neurogenesis activity after global cerebral ischemia

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P>Global cerebral ischemia (GCI) is a major obstacle for cardiac arrest survival. Recent studies have suggested the possibility of mesenchymal stem cell (MSC) as a novel therapeutic option for GCI, but these results were limited to the neuroprotective effects of MSCs. Therefore, we aimed to investigate specific characteristics of neurogenesis after transient GCI, and to assess the effect of MSC on these characteristics.</P> <P><B>Methods</B></P> <P>Adult male Sprague–Dawley rats were subjected to 7 min of transient GCI and randomized into 7 groups: baseline, MSC, and control administered groups, to be analyzed at 2, 3, and 4 weeks after GCI, respectively. The same interventions were repeated for sham operated animals. Rats were euthanized at the designated time after GCI.</P> <P><B>Results</B></P> <P>A comparison of GCI and sham groups without MSC treatment, showed that the counts of bromodeoxyuridine (BrdU)- and doublecortin (DCX)-positive cells were significantly increased in the GCI group at 1 week after insult, but the trend was reversed at 3 weeks after insult. The counts of BrdU-, Ki67- and DCX-positive cells and the intensity of zinc translocator 3 (ZnT3) were all significantly higher in the MSC-treated group than those in the control group at 3 weeks after GCI. The count of NeuN-positive cells in the hippocampus was significantly increased in the MSC group at 4 weeks after GCI.</P> <P><B>Conclusions</B></P> <P>GCI induces transient neurogenesis, followed by an anergic state. MSC may counteract this anergy of neurogenesis and result in an increase in intact neurons in later stages.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Neurogenesis increased after global cerebral ischemia but decreased soon. </LI> <LI> MSC administration counteracted the decrease of neurogenesis activity. </LI> <LI> Counteraction of MSC to anergy of neurogenesis resulted in more intact neurons. </LI> <LI> Effect of MSC on neurogenesis activity is consistent with ZnT3 expression. </LI> </UL> </P>

      • Surface plasmon resonance imaging-based protein arrays for high-throughput screening of protein-protein interaction inhibitors

        <P>The E7 protein produced by high-risk human papillomavirus (HPV) induces a degradation of the retinoblastoma tumor suppressor RB through direct interaction, which suggests that an inhibitor for the interaction can be a potential anticancer drug. A surface plasmon resonance (SPR) imaging-based protein array chip was developed for the high-throughput screening of inhibitor molecules targeting RB-E7 interaction. The glutathione S-transferase-fused E7 protein (GST-E7) was first layered onto a glutathionylated gold chip surface that had been designed to specifically bind to GST-fused proteins. Subsequently, a microarrayer was used to spot the hexa-histidine-tagged RB proteins (His<SUB>6</SUB>-RB) onto the GST-E7-layered gold chip surface, and the resulting SPR image was analyzed. Upon increased His<SUB>6</SUB>-RB concentration in the spotting solution, the SPR signal intensity increased proportionally, indicating that His<SUB>6</SUB>-RB bound to GST-E7 in a concentration-dependent manner. The His<SUB>6</SUB>-RB/GST-E7 interaction was challenged by spotting the His<SUB>6</SUB>-RB solution in the presence of a RB binding peptide (PepC) derived from a motif on E7. The SPR imaging data showed that PepC inhibited the His<SUB>6</SUB>-RB/GST-E7 interaction in a concentration-dependent manner. Our results show that the SPR imaging-based protein array chip can be applied to screen small molecule inhibitors that target protein-protein interaction.</P>

        한국산 무우 peroxidase 에 의한 포도당의 효소적 분석

        Korean-radish peroxidase can replace horseradish peroxidase for enzymatic determination of glucose. The glucose could be quantitatively assayed by using glucose oxidase and Korean-radish peroxidase. The optimum temperature and pH for glucose assay were of 40℃ and pH 5.5, respectively. Various cations and anions such as Ag^+, Cu^(++), Pb^(++), Fe^(++), N₃^-, S₂O₃^-, S₂O₄^- and NO₂^- inhibited the glucose assay completely. The relationship between absorbance and glucose concentration was linear up to 50μ㏖ of glucose under the optimum assay conditions.

        Production and Characterization of Raw Starch Hydrolyzing Enzyme from Bacteria

        토양으로부터 생전분을 분해하는 세균을 분리하였으며, Bacillus sp.로 동정하였다. 분리한 세균으로부터 생전분 분해효소의 생성을 위한 최적조건을 검토하였다. 세균은 탄소원으로 wheat 및 soluble starch를, 질소원으로 polypeptone을 사용했을 때 최대 효소생성을 얻을 수 있었다. 그리고 탄소원을 0.5%, 질소원을 0.5% 수준으로 사용했을 때 효소 생성을 극대화하였다. 그리고 배지의 초기 pH를 6.5, 배양온도를 35℃로 유지했을 때 효소생성이 높았다. Sepharose CL-6B 셀 여과 및 DEAE-Sephacel 이온교환수지를 사용하여 부분정제한 효소활성의 최적조건은 pH6.5, 온도 70℃였다. 효소는 pH8.0에서 가장 안정하였으며, 60℃ 이상에서 불활성화하였다. Fe^++, Hg^++ 및 Ag^+는 세균으로부터 효소 생성을 억제하였으며, 아울러 효소활성도 완전히 저해하였다. 효소의 기질에 대한 K_m값은, 1.7%(corn starch), 1.4%(wheat starch), 2.5%(rice starch) 및 1.0%(potato starch)였다. A bacterium capable of hydrolyzing raw starch was isolated from soil, which was identified as a strain of Bacillus. The effects of culture conditions and medium compositions on the enzyme production were investigated. Among tested carbon sources, soluble starch and wheat starch were most effective for the production of the enzyme, and the level of concentration for the optimal enzyme production was 0.5%. For nitrogen sources, polypeptone was best for the enzyme production, with the level of 0.5%. The enzyme was maximally produced by cultivating the organism at medium of initial pH 6.5, and temperature of 35℃. The enzyme was partially purified by Sepharose CL-6B gel filtration and DEAE-Sephacel ion-exchange chromatography. The optimal pH and temperature for the enzyme reaction were 6.5 and 70℃, respectively. The enzyme most stable at pH 8.0, and temperature up to 60℃. In kinetic studies, the k_m values for corn, wheat, rice and potato starch sere 1.7, 1.4, 2.5 and 1.0%, respectively.

The relationship between absorbance and glucose concentration was linear up to $50{\mu}mol$ of glucose under the optimum assay conditions. 한국산 무우 peroxidase는 포도당의 효소적 분석에 주로 사용되는 horseradish peroxidase를 대체할 수 있었다. 포도당은 glucose oxidase- 한국산 무우 peroxidase에 의하여 정량적으로 측정되었다. 포도당 측정의 최적조건은 $40^{\circ}C$ 및 pH 5.5이었다. 양이온 중에서 $Ag^+$, $Cu^{++}$, $Pb^{++}$, $Fe^{++}$ 그리고 음이온 중에서는 ${N_3}^-$, ${SO_3}^=$, ${S_2O_3}^=$, ${S_2O_4}^=$, ${NO_2}^-$는 효소반응을 완전히 저하시킴으로서 한국산 무우 peroxidase를 사용하여 포도당을 측정하는 것을 완전히 방해하였다. 흡광도와 포도당 농도와의 상관관계는 최적조건에서 분석하고자 하는 포도당의 양이 $50{\mu}mol$ 이하에서는 직선관계를 나타내었다.

        Background: Hyperuricemia is common drug effect during treatment of tuberculosis consisting pyrazinamide (PZA). However, it is unclear the relationship between PZA-induced hyperuricemia and major adverse cardiovascular events (MACE). Aim: We evaluated the relationship between MACE and hyperuricemia in patients treated with PZA-based antituberculous drugs. Methods: We retrospectively reviewed the medical charts of patients who treated with tuberculosis at Chonnam National University Hospital between January 2010 and June 2017. Results: Total 1,674 patients were treated with tuberculosis. Among them, 229 patients were excluded due to lack of available serum uric acid levels because of early follow-up loss or local transfer. Mean age was 57.9 and 887 patients (61.4%) were male. Most patients (75.6%) who treated with tuberculosis were diagnosed pulmonary tuberculosis. The initial standard treatment regimens of isoniazid, rifampin, ethambutol, and PZA were introduced to most patients (92.9%). Among them, 61.9% patients at 2 weeks and 59.5% at 2 months form starting treatment had hyperuricemia. MACE were occured only 17 patients (1.2%) during treatment. There was no relationship between hyperuricemia and MACE in the univariate analysis (odds ratio, 0.72; 95% confidence interval, 0.18─2.90; P=0.648). Conclusion: Most patients who received PZA-based antituberculous drugs had hyperuricemia, however, it was not associated with development of MACE.

        한반도 서·남해 주요 갯벌 퇴적물의 시·공간적 변화: 입도, 유기물, 중금속

        국가해양생태계종합조사의 일환으로 2015년부터 2017년까지 3년간 강화남단, 가로림만, 증도, 순천만 등 한반도 4개 대표 갯벌에서 시계열 변화에 따른 퇴적환경 및 유 무기 오염현황을 조사하였다. 조사시기 및 지역별 퇴적물의 평균입도는 강화남단 $5.0-5.3{\varnothing}$, 가로림만 $4.5-4.8{\varnothing}$, 증도 $6.1-6.5{\varnothing}$, 순천만 $8.6-8.7{\varnothing}$이었다. 전반적으로 강화남단에서 순천만으로 남하함에 따라 입도가 조립해지는 경향을 나타내었고, 순천만에서 가장 세립한 경향을 보였다. 조사시기별로 강화남단과 증도의 경우 2015년보다 2017년에 평균입도가 보다 세립화되는 경향을 나타내었다. 강열감량은 2015년 순천만에서 15.5%로 다른 지역에 비해 상대적으로 높았으나 2016년 8.3%, 2017년 7.0%로 시간의 경과에 따라 점차 감소되었다. 미량금속 농도는 증도와 순천만의 일부정점에서 Zn와 As가 주의기준(TEL)을 초과하였으나 기타 금속의 경우 오염농도 이하의 범위로 분석되어 오염의 개연성이 나타나지 않았다. 연구지역내 퇴적물중 평균입도와 미량금속 농도 사이의 관계를 살펴본 결과, 증도의 경우 수은을 제외한 모든 미량금속과 강열감량이 평균입도와 정의 상관성(r=0.40-0.88, P < 0.05)을 보였다. 반면 순천만의 경우, 입도와 미량금속은 유의한 상관성이 없는 것으로 파악되었으며, 강열감량과 미량금속 간에 음의 상관성(p < 0.05)을 나타내었다. 퇴적물의 미량금속 오염을 평가하기 위하여 농집지수 $I_{geo}$를 계산한 결과, Cu, Zn, Pb, Cd, Hg에 대해서는 1보다 작은 범위로 오염되지 않았고, As의 경우 순천만과 증도의 일부정점에서 약간 오염된 수준에 해당되었다. As a part of the national marine ecosystem monitoring program, the temporal and spatial variation of sedimentary environment and pollution of organic matters and trace metals from four major tidal flats, i.e., Ganghwa Is., Garolim bay, Jeung Is., Suncheon bay, was investigated for 3 yerars from 2015 to 2017. The mean grain size of the sediment was $5.0-5.3{\varnothing}$ at Ganghwa Is, $4.5-4.8{\varnothing}$ at Garolim bay, $6.1-6.5{\varnothing}$ at Jeung Is, and $8.6-8.7{\varnothing}$ at Suncheon bay. The mean grain size (Mz) tended to decrease from the north (Ganghwa Is.) to the south (Suncheon bay). The ignition loss (IL) was 15.5% in Suncheon bay in 2015, which was relatively high compared to other sites, but gradually decreased over time from 8.3% in 2016 to 7.0% in 2017. In Jeung Is. and Suncheon bay, the concentration of Zn and As exceeded the threshold effect level (TEL) at some stations, but the range of trace metals in the other sites was below the level. In Jeung Is., the Mz and concentration of trace metals except Hg was positively correlated (r= 0.40-0.88, P<0.05). On the other hand, Mz was negatively correlated with trace metals (P<0.05) in Suncheon bay. The geoaccumulation index ($I_{geo}$) to evaluate contamination status of sediments for trace metal was less than 1(not contaminated) for Cu, Zn, Pb, Cd and Hg, and 2-3 (moderately to strongly polluted) for As at several stations in Suncheon bay and Jeung Is.
