Sustained-release praziquantel tablet : pharmacokinetics and the treatment of clonorchiasis in beagle dogs

Praziquantel is rapidly absorbed and secreted; and thus fractional doses are recommended for the treatment of cestode and trematode infections. In the present study, we developed a new praziquantel tablet formula allowing sustained-release (SRP). In vitro dissolution of SRP tablets showed that praziquantel at 300 ㎎/tablet combined with hydroxypropyl methylcellulose dissolved completely at a constant rate over 10 h, whereas the conventional praziquantel tablet (PZQ) was only 40% dissolved. Pharmacokinetic studies in dogs confirmed that SRP was absorbed more slowly than PZQ. The mean value of the area under the concentration/time curve from 0 h to the final observation time, the maximum concentration in serum, and the time of maximum concentration in serum for SRP were 3.471.500 ng/min for 0.25 ㎖, 10,300 ng for 0.25 ㎖, and 192 min. while the values for PZQ were 688,600 ng/min for 0.25㎖, 2,500 ng for 0.25㎖, and 135 min. The cure rate in dogs with a heavy infection (500 metacercariae) treated with a single dose of SRP (150 ㎎/tablet) at 50 ㎎/㎏ was 80%, while in dogs treated with a single dose of SRP (300 ㎎/tablet) at 30 mg/kg it was 60%, and the cure rate with PZQ was 20%. In each case. the egg reduction rate was similar (over 90%). No abnormal liver functions or hepatic or renal pathologies were observed in dogs administered with SRP at 30 ㎎/㎏. The SRP tablet showed sustained release and slow absorption; and it had an improved anthelmintic efficacy against Clonorchis sinensis in experimental dogs. compared with conventional praziquantel.

흰쥐에서 Cumambrin A의 대동맥 이완작용

We previously reported that the exogenous administration of cumambrin A, a sesquiterpene lactone from the dried flowers of Chrysanthemum boreale Makino has a pharmacological effect on normalization of blood pressure in the spontaneously hypertensive rats (SHR). In the present study, we further investigated the effect of cumambrin A on the relaxation of phenylephrine-induced precontracted rat aortic artery rings. The potency of cumambrin A was then compared to verapamil, a well known Ca^(2+)-channel blocker. The results demonstrate that the isolated rat aortic arteries are relaxed to basal tension at a concentration of 5X10^(-5)M cumambrin A treatment. The results also show that the phenylephrine-induced contraction is inhibited by a pretreatment of cumambrin A. Co-treatment of cumambrin A and verapamil showed a strong synergetic effect on the relaxation of rat aortic artery rings. Thus, these data demonstrate that cumambrin A is a potent relaxant of rat aortic smooth muscle and suggest that cumambrin A modulates intracellular or extracellular Ca^(2+) mobilization.

19世紀 前半基의 社會變動과 勞動思想 : Louis Blanc를 中心으로

European society of the former half of the nineteenth century was heavily influenced by the Dual Revolution, that is, the French Revolution and the Industrial Revoltion. The Industrial Revolution produced the working class and the problems of labour, though it accomplished the development of modern capitalism. In the nineteenth century the so-called utopian socialists-Fourier, Owen, Saint-Simon-attempted to solve these problems in their own ways. They tried 'to discover a new and more perfect system of social order and to impose this upon society from without by propaganda, and, wherever it was possible, by the example of model experiments. These new social systems were foredoomed as utopians' (Engels). Louis Blanc(1811∼82), born later than these utopians and lived through almost the whole century, showed a more practical and concrete way to socialism. He vehemently condemned the limitless competition of capitalist society and deeply sympathised with the poverty-ridden workers. In order to eliminate such social wrongs he proposed to create 'the social workshop', a kind of producer's corporation. Initially established under state sponsorship, these workships should be run by the workers themselves. Blanc believed that his social workshops would prevail over the existing factories based on competition because of their increased productivity based on fraternal solidarity. In the same way, he expected a fraternal absorption of the bourgeoisie rather than a violent revolutionary triumph over it by the proletariat. Blanc firmly believed that a democratically elected republic would willingly initiate the social revolution without violence and terror. So there would be no dictatorial institutions, for even a brief time. Blan's ideas gained wide popularity among workers during 1840s and had a precarious chance to be realized in the spring of 1848, but failed. Blanc's socialism stood between the utopian socialism and Marxism, chronologically and ideologically. His belief in peaceful transition to socialism may have some connection with modern French humanistic socialism(socialisme humaniste).

Alcohol dehydrogenase from Thermomicrobium roseum was purified to electrophoretic homogeneity approximately 77-fold with a 38% activity yield by adenosine 5'-monophosphate and Mono Q column chromatography. The molecular weight of the enzyme was determined to be approximately 43,000 by SDS-polyacrylamide gel electrophoresis and 88,000 by gel chromatography, indicating a homodimeric structure. The Km and Vmax values of the enzyme for ethanol were 24.2 mM and 12.8 ㎛ol/min/mg, respectively. Pyrazole notably inhibited the enzymatic activity. Ca2+ ion activated the enzyme activity, but Cu2+ inhibited. The activity of the enzyme was optimal at pH 10.0 and 60℃.

사람 위선암에서 cathepsin L의 발현증가

cathepsin L은 lysosomal cysteine 단백분해효소로서 기저막(basement membrane)과 세포외기질(extracellular matrix)을 파괴하여 암세포의 침윤과 전이에 중요한 역할을 하는 물질로 알려져 있다. 이러한 cathepsin L에 대한 mRNA 발현도를 5개의 사람 위선암(gastric adenocarcinoma) 세포주와 5명의 위선암 환자에서 채취한 조직에서 방사능으로 표지된 cathepsin L특이 cDNA를 사용한 Northern blot법으로 측정하였다. 위암의 전이병소에서 수립한 세포주인 SNU-5, SNU-16, MKN-45와 Kato Ⅲ에서는 cathepsin L mRNA가 발현되었으나 원발병소에서 수립한 AGS 세포주에서는 mRNA의 발현이 관찰되지 않았다. 5명의 위암 환자에서는 원발병소, 전이가 확인된 임파절 및 암 근처 정상 위점막에서 각각 조직을 채취하여 cathepsin L mRNA의 발현을 측정하였다. 원발병소와 전이병소에서는 모두 cathepsin L mRNA가 발현되었으나 정상 위점막조직에서는 전예에서 mRNA 발현이 관찰되지 않았다. 한편 mRNA의 발현도는 1예에서는 전이병소가 원발병소에 비해 높았으나, 2예에서는 전이병소에서 발현도가 낮았으며, 나머지 2예에서는 원발병소와 전이병소 사이에 차이가 없어, 원발병소와 전이병소 사이에 mRNA의 발현도의 일관성 있는 경향은 관찰되지 않았다. 이상의 결과는 cathepsin L은 위암의 발생과 진행에 있어 암세포의 침윤과 전이를 촉진하는 것 이외에 또 다른 역할을 할 가능성을 시사하고 있다고 사료된다. Cathepsin L, a lysosomal cysteine protease, is known to play an important role in cancer invasion and metastasis by degrading the components of basement membrane and extracellular matrix. The mRNA expression of cathepsin L was determined by Northern blot analysis using a radiolabeled cDNA specific for cathepsin L in five human gastric adenocarcinoma cell lines and five surgical specimens of primary gastric adenocarcinomas, their metastatic lymph nodes and matched adjacent normal mucosae. The mRNA of cathepsin L was expressed in all of the four cell lines established from the metastatic sites, SNU-5, SNU-16, MKN-45 and Kato Ⅲ, while not detected in one cell line established from the primary site, AGS. The mRNA was expressed in all of the five primary and five metastatic cancer specimens tested, while it was not detected in all matched normal mucosae. The intensities of the mRNA expressions, however, did not show the consistent pattern between primary sites and metastatic lymph nodes. These results suggest that cathepsin L may have the other function in addition to facilitation of the invasion and metastasis during the development and progression of stomach cancer.

Dynamic Programming을 이용한 상호인증 경로기반 인증서 검증 기술 개발

본 논문은 국가간 상호인증 경로 기반 인증서 검증에서 인증 정책의 연결 상태를 효율적으로 검증하는 소프트웨어 개발에 대해 논하였다. 본 연구에서 개발된 소프트웨어의 핵심모듈인 "valid policy set"은 ITU-T및 IETF 표준을 준수한다. 본 연구에서 제안된 "동적배열"은 자원 관리 및 계산량에 있어서 보다 효율적인 정책 연결 과정을 제공한다. 그러므로, 제안된 동적 배열 모듈을 이용함으로써 이질적인 파라미터 값을 갖는 정책의 연결 관리를 고속으로 구현할 수 있다. This paper presents a software development which efficiently verifies policy mappings based on the mutual path validation among different CAs. The developed core module,"valid policy set" observes the ITU-T and IETF path validation standards. A policy mapping sequence based on "the dynamic array" we have proposed provides the efficiency in the viewpoints of computational cost and resource management. Therefore,the proposed dynamic array accelerates administrating a valid policy set combined with heterogeneous policy parameters.

유산양에서 발정동기화 유도와 인공수정 효율

This study was conducted to evaluate the response of CIDR-S on estrus synchronization of dairy goats and the effect of artificial insemination on conception rate. Estrus synchronization of sexually matured Saanen goats was induced with insertion of CIDR-S into vagina for 15 days and injection of 500 IU PMSG before removal of CIDR-S. Artificial insemination was conducted using liquid or frozen semen by intra-cervical insemination with synchronized does. Estrus synchronization was 90% using CIDR-S insertion for 15 days. Conception rate of inseminated does was 20% - 25%, while that of natural mated does was 100%. These results suggested that synchronization of dairy goats can be successfully induced and artificial insemination method has to be improved for the practical use in dairy goats.

암세포의 증식과정에서 20 kDa 단백질의 arginine 메칠화 반응에 관한 연구

Enzymatic methylation of endogenous protein (s) by protein arginine N-methyl- transferase(EC. 2.1.1.23) in several cancer cell lines was investigated in an attempt to understand possible regulatory role of the modification in relation to cellular proliferation. When several cancer cells (HeLa, HCT-48, A549, HepG2) were incubated with S-adenosyl-L-〔rnethyl-³H〕methionine, the endogenous 20- kDa species in the extracts were most intensely 〔methyl-³H 〕-labeled compared to that in normal colon tissues. Whether the 〔methyl-³H 〕-labeld 20-kDa is due to carboxyl-O-methylation, the alkali-lability of the purified and methylated 20- kDa species were examined at pH 9-11, at 37℃ for 60 min. The results indicated that 〔methyl-³H 〕-label on the 20-kDa species of HCT-48 cells did not decrease after alkali treatment quantified by Fuji BAS 2500 Bio-Image analyzer. The methylation of 20-kDa did not increase in the presence of 4 mM GTPγS, further indicating the methylation is not due to C-terminal farnesylated cysteine carboxyl methylation. Also, none of purified histones showed the same mobility on SDS-PAGE as the 20-kDa endogenoiis substrate. In conclusion, in the present study, a new novel endogenous 20-kDa methyl accepting protein for protein arginine methyltransferase was found which is closely correlated with cell proliferation suggesting a possible role of this post-translational modification during cellular proliferation.
