      • RSV나 인플루엔자 A형 바이러스에 감염된 비천식 소아에게 비인두 흡인액의 Interleukin - 11 , Interferon - γ치와 eosinophil cationic protein 치의 비교

        N/A Background: Infection with respiratory virus has been shown to exacerbate asthma in humans. However, the role of a respiratory virus in the pathogenesis of chronic asthma and/or wheezing in young children has not been clearly defined. The objective of this study was to determine whether respiratory virus infections such as RSV, and influenza A virus are related to the productions of IL-11, IFN-γ, and ECP levels in nasopharyngeal secretions. Method: We compared IL-11, IFN-γ, and ECP levels in nasopharyngeal secretions from 38 non-asthmatic wheezing children with viral infections (RSV in 21 children, influenza A virus in 17 children), and 16 non-asthmatic healthy children who were included as the controls. IL-11, and IFN-y levels were analysed by ELISA. ECP concentrations were measured by monoclonal antibody-based fluorometric assay. Result. RSV infection in children induced a greater release of IL-11 in nasopharyngeal secretions than in influenza A virus infection, and in the controls. The release of IFN-γ levels in nasopharyngeal secretions from children with influenza A virus infection was significantly higher than in nasopharyngeal secretions from children with RSV. ECP levels of subjects with viral infection were significantly higher than in control children. Conclusion. This study demonstrated that RSV is a potent inducer of IL-11 elaboration in nasal epithelium and that IL-11 is an important mediator in the pathogenesis of RSV infection. Increased IFN-γ production in response to the influenza A virus infection may be related to effective Th1 responses.

      • RSV 와 인플루엔자 바이러스 A 형 감염에 의해 천명을 보이는 소아와 천식 소아에서 비인두 흡인액의 Interleukin-5 와 Interleukin-γ 치의 비교

        목 적 : 호흡기 바이러스 감염은 소아에서 천식을 악화시키는 것으로 잘 알려져 있다. 그러나 영유아기의 천식이나 천명발생에 있어서 호홉기 바이러스의 역할에 대해서는 명확하게 밝혀져 있지 않다. 소아기의 천명이 천식과 달리 하나의 독립된 질환인지, 아니면 같은 질환으로 달리 표현되는 것인지 논란이 되어왔다. 이에 본 저자들은 호흡기 바이러스 감염과 천명과의 상관관계와 기전을 이해하기 위하여 RSV 감염이나 influenza A 바이러스 감염에 의해 천명이 있는 소아와 바이러스가 증명되지 않고 천명이 있는 천식소아에서의 비인두 흡인액의 IL-5와 IFN-γ치를 측정하여 비교하였다. 방 법 : 호흡기 바이러스 감염군 38명(RSV감염군 21명, influenga A virus 감염군 17명), 바이러스가 증명되지 않은 천식환아군 12명, 정상 대조군 16명을 대상으로 double sandwich ELISA를 이용하여 비인두 흡인액에서 IL-5와 IFN-γ치를 측정하여 비교하였다. 결 과 : RSV 감영군에서 비인두 흡인액의 IL-5 평균치가 influenza A 바이러스군의 평균치보다 의미있게 높았으며, 천식군보다도 높은 양상을 보이나 의미있는 차이는 없었다 반면, influenza A 바이러스 감염군의 비인두 흡인액의 IFN-γ치가 RSV군에 비해 의미있게 높았으나 천식환아군이나 정상군에 비해 의미있게 높지는 않았다. 비인두 흡입액에서 IL-5와 IFN-γ치간의 상관관계는 없었다. 결 론 : RSV 감염군은 influenza A 바이러스 감염군에 비해 Th2 반응이 우세한 것으로 추정되며, 반면에 influenza A virus 감염군은 Th1 반응을 보이는 것을 알 수 있다. And it also has been debated whether virus-induced wheezing in young children is an entity different from allergic asthma, or just a different expression of the same disease. In this study, we attempted to evaluate the importance of eosinophilic inflammation, comparing IL-5 and IFN-γ levels in nasopharyngeal secretions in wheezing children with or without viral infection and the controls. Methods : We compared IL-5 and IFN-γ levels in nasopharyngeal secretions from 38 non-asthmatic wheezing children with viral infections (RSV in 21 children, influenza A virus in 17 children), 12 asthmatic children without viral infections and 16 children as the controls. Results : The present study reported that RSV infection in children induced more releasing of IL-5 in nasopharyngeal secretions than the influenza A virus infected ones and the controls. On the other hand, the releasing of IFN-γ levels in nasopharyngeal secretions from children with influenza A virus infection was significantly higher than those of the children with RSV infection or asthmatic children. Conclusion : RSV infection in children may play a role in the immune response toward a Th2 phenotype as increasing IL-5 secretion in nasopharyngeal secretion. Increased IFN-γ production in response to the influenza A virus infection may be related to the effective Th1 responses.

        류마티스 관절염환자의 혈청 Interleukin-10

        Objective: To investigate whether the serum levels of IL-10 in patients with rheumatoid arthritis are different from those of normal controls and SLE patients and to find out any correlation with disease activity parameters of rheumatoid arthritis. Methods: Sera from 20 healthy normal persons, 16 rheumatoid arthritis patients and 16 patients with systemic lupus erythematosus were collected and measured for IL-10 and IL-6. Various disease activity parameters were measured in RA patients. Results: The serum level of IL-10 in RA patients was significantly elevated compared to normal controls but lower than those of SLE patients. In RA patients there was no definite correlation between the disease activity parameters and serum IL-10 levels. Despite significant improvements in terms of various disease activity parameters, there was no significant change of serum IL-10 levels after treatment in RA patients. In seropositive RA patients, positive correlation was found between serun IL-10 and rheumatoid factor levels. Conclusion: Our findings show that the serum IL-10 levels in patietns with RA are elevated compared to normal controls but lower than those of SLE patients. There was no correlation between serum IL-l0 levels and disease acivity parameters of RA.

        Interleukin-13 Increases Podocyte Apoptosis in Cultured Human Podocytes

        Purpose: Podocytes are important architectures that maintain the crucial roles of glomerular filtration barrier functions. Despite this structural importance, however, the mechanisms of the changes in podocytes that can be an important pathogenesis of minimal change nephrotic syndrome (MCNS) are not clear yet. The aim of this study was to investigate whether apoptosis is induced by interleukin (IL)-13 in cultured human podocytes. Methods: Human podocytes were treated with different IL-13 doses and apoptotic cells were analyzed using terminal deoxynucleotidyl transferase dUTP nick-end labeling (TUNEL assay) and fluorescence-activated cell sorting (FACS). Results: The IL-13 increased the number of TUNEL-positive cells in a dose-dependent manner at 6 and 18 hours (P<0.05 and P<0.05, respectively). The apoptosis rate was appeared to be increased slightly in the IL-13-stimulated podocytes (8.63%, 13.02%, and 14.46%; 3, 10 and 30 ng/mL, respectively) than in the control cells (7.66%) at 12 hours by FACS assay. Conclusion: Our study revealed that IL-13 expression may increase podocyte apoptosis. Blocking the IL-13 signal pathway can potentially play an important role in regulating the apoptosis of podocytes.

      • Fucoidan isolated from the popular edible brown seaweed Sargassum fusiforme suppresses lipopolysaccharide-induced inflammation by blocking NF-κB signal pathway

        The anti-inflammatory effect of a fucoidan with a molecular weight of 102.67 kDa isolated from an enzymatic digest of Sargassum fusiforme was investigated in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages and zebrafish. The results indicated that the fucoidan significantly and dose-dependently inhibited the production of inflammatory molecules, including tumor necrosis factor-alpha (TNF-α), nitric oxide (NO), prostaglandin E2 (PGE2), interleukin-1 beta (IL-1β), and interleukin-6 (IL-6), as well as improved the viability of LPS-stimulated RAW 264.7 cells. Moreover, the fucoidan suppressed the expression of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) by regulating the nuclear factor kappa B (NF-κB) pathway in LPS-stimulated RAW 264.7 cells. Furthermore, in vivo test results suggested that the fucoidan remarkably reduced reactive oxygen species (ROS), cell death, and NO levels in LPS-stimulated zebrafish in a dose-dependent manner. Taken together, these results demonstrated that the fucoidan isolated from S. fusiforme possesses strong anti-inflammatory activities in vitro and in vivo, and could prove as an important candidate to be used to develop anti-inflammatory agents in pharmaceutical and cosmeceutical industries.

      • Effects of Moxibustion to Zusanli(ST36) on Alteration of Natural Killer Cell Activity in Rats

        Moxibustion is one of the major healing techniques in Oriental medicine. It has been widely used in many diseases such as rheumatoid arthritis, Hashimoto disease, breech presentation, etc. However, till now, effects of moxibustion on natural killer (NK) cell activity and relations between sympathetic nerve system (SNS) and the immune alteration induced by moxibustion wert not well studied. This study was designed to evaluate effects of moxibustion on NK cell activity and the Intervention of SNS in the alteration of NK cell activity induced by moxibustion. Splenic NK cell cytotoxicity was measured in a standard 4-hour ^(51)Cr release assay. We measured the NK cell cytotoxicity after moxibustion stimulation for 1, 3, 5 and 7 days, and also measured the NK cell cytotoxicity after 3 and 7 days burn stimulation with similar temperature. Interleukin (IL)-2, -4 and interferon (INF)-γ in serum were measured by rat IL-2, -4 and INF-γ ELISA test kit. To evaluate the effects of sympathectomy on alteration of NK cell cytotoxicity, 6-hydroxydopamine (6-OHDA: 50 mg/kg) was used. We showed that NK cell activity of moxibustion stimulation group incrcased at the 3rd day, and declined at the 7th day in comparison with that of the control group. In thc moxibustion stimulation group, NK cell activity was significantly higher than the sham group at the 3rd day. On the contrary, in the burn stimulation group, NK cell activity was significantly higher than that of the sham groups at 3rd and 7th days. INF-γ level after 3 days in the moxibustion stimulation group was significantly higher than that of the sham group. IL-2 Ievel among groups were not different. IL-4 was not detected in serum with this method. Sympathectomy abolished the NK cell activity alteration induced by moxibustion. The results suggest that moxibustion modulates NK cell activity, along with INF-γ, and SNS is mediating these effects.

      • 六味知黃湯 投與가 高壓環境에 露出된 흰쥐의 血液造成과 臟器造織에 미치는 影響

        This study was purposed to investigate the effect of yukmijihwangtang on rat blood components and organ histopathologic changes in the different environments of hyperbaric ambient air. The pharmacological effects of yukmijiwhangtang in oriental medicine have been reported to improve general condition and treat mental and physical weakness due to overload. 84 Male Sparague-Dawely rats, 200-250 g weight, were housed two per cage, fed Standard Rodent Diet with water ad libitum. The animals were assigned to 3 main groups for the blood components and gas analysis : normobaric control group (NC ; N=4), hyperbaric control group (HC ; N=8), hyperbaric medication group (HM ; N=8). HC and HM group were devided into 2 subgroups and kept on different situation of the hyperbaric ambient air (2ATA or 3ATA) for 2 hrs. HM group was administered 10cc extract of boiled yukmijiwhangtang before exposure. For histopathological analysis, hyperbaric groups(HC and HM : N=80) was further devided into 2 subgroups according to atmosphere absolute and exposure period : 2ATA (acute, 1, 2, 3, 4 wks), 3ATA (acute, 1, 2, 3, 4 wks). There were 20 groups in total, with 4 animals in each group. Experimental rats of HM group were administered 10cc extract of boiled yukmijiwhangtang 2 times per day and kept on situation of the hyperbaric ambient air (2ATA and 3ATA) 2 hrs per day for four weeks. In order to record the possible effects of yukmijiwhangtang in hyperbaric condition on the blood components, the rats were killed by decapitation directly after the hyperbaric exposure for 2 hrs. But the administered rats were killed on the morning following the last hyperbaric exposure during 4 wks. The samples were taken 2 hrs, 1, 2, 3, 4 wks after the begining of this study. Blood was collected from posterior abdominal vena cava immediately after decapitation. Organs for histopathological assays in hyperbaric groups (HC and HM) were also removed 2 hrs, 1, 2, 3, 4 wks and fixed at 10% formalin solution. The whole blood of 1㎖ was used for the analysis of pH, PO₂, PCO₂, HCO₃, BE, O₂CT, O₂SAT level by Blood Gas Autoanalyzer (Coning 175, U.S.A). And The rest whole blood of 1㎖ was utilized for the measurement of WBC (white blood cell), RBC (red blood cell), Hb (hemoglobin), Hct (hematocrit), MCV (mean corpuscular volume), MCH (mean corpuscular hemoglobin) level by Microcell Counter (Model; CC-170-TDA Co., Japan). Hematological and blood gas analysis and organ histopathological changes after exposure to hyperbaric ambient air were as follow; (1) Hematological parameters were lower in the hyperbaric medication group than all the control groups without WBC, MCV (2ATA) and MCV, MCH (3ATA). But a statistical significance among the groups was not shown. (2) The hyperbaric medication group was lower than another control groups on all parameters of blood gas analysis after 2 hours exposure to hyperbaric ambient air (2ATA and 3ATA) without O₂SAT. But a statistical significance among the groups was not shown. (3) On observing the histopathological system of the lung with microscope, the congestion, edema and hemorrhage on the alveolar sacs and alveolar wall damage in the hyperbaric control group were heavier than in the hyperbaric medication group.

        턱끝절골술 : 189명에 대한 고찰

        The chin plays a decisive role in the contour of the lower face. And harmonious jaw will correspond to good facial profile and positive image. Osseous genioplasty is a widely used method to correct the 3-dimensional(sagittal, vertical, transverse) deformity of the chin with minimal complications. During an 8 year period from 1988, the authors at the In-Je University Seoul Paik Hospital experienced 198 patients of osseous genioplasty. Results of clinical analysis are as follows: 1)Mail to female ratio was 1 : 3 with female being predoniment. The average age was 24.9 years. 2) Horizontal advancement genioplasty in 169 cases was the most common precedure with 7.0mm of average advancement. There was 4 cases of horizontal retrusion with average retrusion of 4.3mm. 52 cases had vertical augmentation with average of 7.5mm increase. 9 cases had vertical reduction with average redution of 4.9mm. 3) Although malocclusion is not necessarilly contraindication to genioplasty alone, genioplasty following surgical with or without orthodontical correction of malocclusion is the way to get better result with patient satisfaction. 4) 77% of the patients had adjunctive procedures simultaneously to acheive a better contour and profile 5) Complications such as hematoma, infection, fracture, nerve damage were minimal.

        Oxalate와 Enrofloxacin투여한 랫트신장에 대한 병리조직학적관찰

        To investigate the renal effects of enrofloxacin administration on rats induced with dehydration or hyperoxaluria, male rats were treated with enrofloxacin of 50 mg to 500 mg/kg b.w.. The microscopical observations of kidney and urine sediment were carried out in the experimental groups. The result obtained were as follows; The male rats deprived of water for 72 hours and administered with enrofloxacin. As enrofloxacin administration dose was increased, clinical signs such as loss of appetite, depression, weakness, and loss of urine output became more severe. In the histopathological findings, there were hyperemia and hemorrhage in renal cortex, vacuolation and necrosis of renal tubular epithelia, proteinous casts within renal tubules. The male rats were orally administered with sodium oxalate and injected with enrofloxacin for 7days. As enrofloxacin administration dose was increased, clinical signs such as the loss of appetite and water consumption, and weakness became more severe. In the histopathological findings, there are hemorrhage of glomeruli and cortical hyperemia, vacuolation and necrosis of tubular epithelia, proteinous casts in renal tubules. In the microscopical findings of urine sediment, there are calcium oxalate crystal (diamond-like type) and magnesium ammonium phosphate crystals (rhomboid). The male rats were intraperitoneally injected with sodium oxalate and administered with enrofloxacin for 7days. As enrofloxacin administration dose was increased, clinical signs such as the loss of appetite and water consumption, weakness were more severe. In the histopathological findings, there were hyperemia and hemorrhage in both glomeruli and renal cortex. Severe necrosis of renal tubular epithelia, bluish materials within renal tubules were also found. In the microscopical findings of urine sediment, there were many calcium oxalate crystals. The present results suggest that enrofloxacin has some injurious effects in rats having dehydration or hyperoxaluria, and clinically, we should consider these renal injury effects when we use enrofloxacin in patients accompanied renal disease, dehydration and hyperoxaluria conditions.

      • 알코올 섭취 후 운동이 시간 경과에 따른 혈중 알코올 농도와 간 기능 효소에 미치는 영향

        This study was done in order to investigate the effects of performing the light exercise right after alcohol drinking on the changes of blood alcohol and ALT, AST concentration, All seven healthy collegiate students volunteered to two sections of experiment, Each subject drank one bottle of Soju(360ml, 75.6g ethanol) For forty minutes at the same speed of drinking with the same kind and volume of carbohydrate snacks. Each section of the study was conducted by a two-cross-over design with four days wash-out period, All volunteers performed brisk walking exercise corresponding to about 50-60%HRmax of exercise intensity For 30minutes immediately after alcohol drinking. Blood sampling was conducted by drawing about 10ml venous blood from antecubital vein at pre -alcohol drinking, and post- 1, 2, 4 hour alcohol drinking. Blood alcohol concentration reduced significantly at all time course in exercise group compared to Control group. However, the concentration of serum aspartate aminotransferase and alanine aminotransferase, the indicator of liver cell damage, showed no significant difference between two groups. In conclusion, performing light exercise such as brisk walking after alcohol drinking may somewhat help to reduce the concentration of blood alcohol Faster by influencing the alcohol metabolism.
