        Expression of FMD Virus-like particles in yeast Hansenula polymorpha and immunogenicity of combine with CpG and Alumimun adjuvant

        Background: Inactivated vaccines are limited in preventing foot-and-mouth disease (FMD) due to safety problems. Recombinant virus-like particles (VLPs) are an excellent candidate for a novel vaccine for preventing FMD, given that VLPs have similar immunogenicity as natural viruses and are replication- and infection-incompetent. Objectives: The 3C protease and P1 polyprotein of type O FMD virus (FDMV) was expressed in yeast Hansenula polymorpha to generate self-resembling VLPs, and the potential of recombinant VLPs as an FMD vaccine was evaluated. Methods: BALB/c mice were immunized with recombinant purified VLPs using CpG oligodeoxynucleotide and aluminum hydroxide gel as an adjuvant. Cytokines and lymphocytes from serum and spleen were analyzed by enzyme-linked immunosorbent assay, enzyme-linked immunospot assay, and flow cytometry. Results: The VLPs of FMD were purified successfully from yeast protein with a diameter of approximately 25 nm. The immunization of mice showed that animals produced high levels of FMDV antibodies and a higher level of antibodies for a longer time. In addition, higher levels of interferon-γ and CD4+ T cells were observed in mice immunized with VLPs. Conclusions: The expression of VLPs of FMD in H. polymorpha provides a novel strategy for the generation of the FMDV vaccine.

        The Influence of Dietary Calcium and Phosphorus Imbalance on Intestinal NaPi-IIb and Calbindin mRNA Expression and Tibia Parameters of Broilers

        A $2{\times}2$ factorial experiment was conducted to study the effect of dietary calcium and non-phytate phosphorus (nPP) imbalance on calbindin and NaPi-IIb mRNA levels in the small intestine and tibia parameters of broiler chicks. One hundred and forty four 1-d-old Arbor Acres male broiler chicks were divided into four treatments consisted of six replicates with six chicks each. The two dietary calcium levels were 1.10% and 0.60%, and two dietary nPP levels were 0.50% and 0.27%. Results showed that a high Ca/nPP ratio diet (4.07:1) significantly depressed feed intake and weight gain of broilers (p<0.05), but a lower Ca:nPP ratio (1.2:1) had no influence (p>0.05). Low-Ca with low-P diet resulted in low tibia minerals and tibia breaking strength of broilers, and all the tibia parameters were further decreased when the dietary ratio of Ca to P was relative higher. Low dietary Ca or P up-regulated the calbindin and NaPi-IIb mRNA expression levels. Low Ca with normal P diet up-regulated duodenal calbindin mRNA expression level to the greatest extent. Low P with a normal Ca diet significantly enhanced NaPi-IIb mRNA expression level to the highest extent. These results suggest that the calbindin and NaPi-IIb mRNA expression were enhanced by the imbalance between dietary Ca and nPP, and their expression were not only influenced by Ca or nPP level, but also the ratio of Ca:nPP.

        Tunable stability of oil-containing foam systems with different concentrations of SDS and hydrophobic silica nanoparticles

        Experiment and molecular dynamics simulation were carried out to study the tunable stability of oilcontainingSDS-stabilized Nitrogen-in-water foam. The experimental results show that the foam stabilitycould be tuned by the concentrations of SDS and modified SiO2 nanoparticles. In the foam systems with alow SDS concentration (0.2 wt.%), the foams show poor stability and the foam stability was almost notaffected by the addition of modified SiO2 nanoparticles. The foam stability was greatly improved atmoderate SDS concentration (0.5 wt.%), it enhancedfirst and then weakened with the increase ofmodified SiO2 concentration, and the half-life time achieved a maximum value of 1292 s at 0.05 wt.%modified SiO2. However, at high SDS concentration (0.8 wt.%), the foam stability was pretty good exceptfor when the modified SiO2 concentration is too high (>0.2 wt.%). The microscopic mechanism wasobtained by investigating the structural and dynamic properties of the foamfilm. The simulation workshowed consistent results of foam stability with the experimental results. Moreover, it also revealed thatthe concentration and configuration of SDS will affect its interaction with SiO2 and oil molecule, which iscritical to foam stability.

        Construction of a resequencing-based high-density genetic map for grape using an interspecific population (Vitis amurensis × Vitis vinifera)

        A high-density genetic map is a prerequisite for marker-assisted selection of crop species. Here, we developed a high-density genetic map of Vitis using 92 F1 progeny derived from the cross between two wine grape cultivars, ‘Shuanghong’ and ‘Italian Riesling’. Whole genome resequencing (WGS) technology was used to generate SNP markers for the map. The map consisted of 1351 recombination bin markers (210,873 SNPs), distributed on 19 linkage groups (LGs), with an average distance of 0.65 cM between adjacent bin markers and a map distance of 860.46 cM. The high levels of uniform coverage and collinearity between the 19 LGs and the grape reference genome indicated that the map is of high quality and can serve as a powerful platform for evaluating grapevines and deciphering the genetic mechanism of important economic and agronomic traits. Our findings will facilitate the process of mapping quantitative trait loci (QTL) that control important horticultural and economic traits of grape.

        Development and Practice of Performance-Based Seismic Design of High-Rise Buildings in China

        Seismic performance-based design methods are widely used in the field of engineering. This paper introduces the current status of seismic performance-based design methods for high-rise buildings in China, and summarizes latest advancements in seismic performance-based design methods for high-rise buildings in China, with a focus on the design methods based on predetermined yield mode and the design methods based on member ductility requirements. Finally, the development direction of seismic performance-based design method for high-rise buildings is prospected.

        Neuroprotective Effects of Luteolin Against Spinal Cord Ischemia–Reperfusion Injury by Attenuation of Oxidative Stress, Inflammation, and Apoptosis

        Luteolin (LU) is a widely distributed flavonoid with multitarget effects. The objective of this study was to determine whether LU could reduce the ischemia–reperfusion injury of the spinal cord (SCII) in a rat model. Forty-eight rats were divided into four groups: sham, SCII, SCII+L-LU (50 mg/kg), and SCII+H-LU (100 mg/kg). Abdominal aortic occlusion was carried out for 40 min in all groups. Hindlimb motor functions were evaluated using the Tarlov scoring system. Nissl and terminal deoxynucleotidyl transferase-mediated dUTP biotin nick end labeling (TUNEL) staining were used to detect cell survival and apoptosis in the spinal cord. Spinal cord samples were taken for determination of malondialdehyde, xanthine oxidase, superoxide dismutase, and glutathione peroxidase activities. The levels of tumor necrosis factor-α, interleukin (IL)-1β, and IL-18 were assessed using ELISA kits to examine the inflammatory responses in the spinal cord. Western blot analysis was used to examine the expression of nuclear factor erythroid 2-related factor (Nrf2) and nod-like receptor pyrin domain-containing 3 protein (NLRP3) levels. We found that LU pretreatment significantly improved the locomotor function of rats after SCII, increased neuron survival, and inhibited apoptosis in the spinal cord. Furthermore, the oxidative stress and inflammatory response were significantly suppressed upon treatment with LU. Finally, LU upregulated Nrf2 levels and downregulated NLRP3 protein expression in SCII tissues. Thus, LU exhibited a neuroprotective effect following SCII by alleviating oxidative stress and inhibiting inflammatory responses and cell apoptosis. The possible mechanism may be related to the activation of Nrf2 and inhibition of NLRP3 inflammasome pathway.

        Luteolin Modulates the NF-E2-Related Factor 2/Glutamate–Cysteine Ligase Pathway in Rats with Spinal Cord Injury

        Spinal cord ischemia–reperfusion injury (SCII) easily causes unalterable neurological deficits. We previously demonstrated that the flavonoid luteolin (LU) has strong antioxidant, anti-inflammatory, and other neuroprotective efficacies against SCII. In our current study, we examined the contributions of the NF-E2-related factor 2 (Nrf2)/glutamate–cysteine ligase (GCL) pathway to LU-mediated neuroprotection in the transient abdominal aorta occlusion rat model of SCII. Rats were divided into four groups: Sham surgery, SCII alone, SCII plus LU pretreatment (SCII + LU), and SCII plus cotreatment with LU and the Nrf2 inhibitor ML385 (SCII + LU + ML385). The Basso–Beattie–Bresnahan (BBB) scale was used to assess neurological function, hematoxylin and eosin staining to evaluate pathological change to the spinal cord, and enzyme-linked immunosorbent assay to measure tissue markers of oxidative stress and inflammation induced by SCII. Mitochondrial injury and apoptosis were examined by flow cytometry and expression levels of Nrf2, GCL catalytic subunit (GCLc), and GCL modifier subunit (GCLm) by real-time quantitative polymerase chain reaction. LU pretreatment significantly enhanced recovery of motor function as evidenced by the BBB score and attenuated the pathological damage. Furthermore, LU effectively enhanced the antioxidative activity, alleviated mitochondrial swelling, decreased the expression levels of several proinflammatory cytokines after SCII, and significantly upregulated Nrf2, GCLc, and GCLm expression levels. Cotreatment with ML385 reversed all these protective effects of LU except the anti-inflammatory response. Collectively, these findings indicate that the neuroprotective efficacy of LU depends on suppression of oxidative stress and preservation of mitochondrial function through signaling pathways involving Nrf2 activation and downstream gene expression.

        The impacts of LPCVD wrap-around on the performance of n-type tunnel oxide passivated contact c-Si solar cell

        In this paper, Tunnel Oxide Passivated Contact (TOPCon) silicon solar cells with the industrial area (244.32 cm2) are fabricated on N-type silicon substrates. Both the ultra-thin tunnel oxide layer and phosphorus doped polycrystalline silicon (polysilicon) thin film are prepared by the LPCVD system. The wrap-around of polysilicon is observed on the surface of borosilicate glass (BSG). The polysilicon wrap-around can form a leakage current path, thus degrades the shunt resistance of solar cells, and leads to the degradation of solar cell efficiency. Different methods are adopted to treat the polysilicon wrap-around and improve shunt resistance of solar cells. The experimental results indicate that a chemical etching method can effectively solve the problem of polysilicon wrap-around and improve the performance of solar cells. Finally, a conversion efficiency of 22.81% has been achieved by our bifacial TOPCon solar cells, with Voc of 702.6 mV, Jsc of 39.78 mA/cm2 and FF of 81.62%.

        Macroscopic Zn-doped a-Fe2O3/graphene aerogel mediated persulfate activation for heterogeneous catalytic degradation of sulfamonomethoxine wastewater

        In order to obtain a robust, durable and efficient heterogeneous catalyst, macroscopic monolithic Zndopeda-Fe2O3/graphene aerogel (GA) hybrid architecture with integrated morphology and hierarchicallyporous structure were controllably synthesized via a facile in-situ hydrothermal method and then used aspersulfate (PS) activator for sulfamonomethoxine (SMM) wastewater purification. Several key reactionparameters including the initial SMM concentration, reaction temperature, coexisting inorganic anionsand SMM in real natural water samples had different influence on the SMM removal efficiency. The catalyticefficiency of Zn-doped a-Fe2O3/GA with the molar ratio of Fe/Zn = 2:1.5 was about 66%, 62%, 66%and 11%33% higher than that of GA, a-Fe2O3/GA, Zn/GA and other Fe/Zn molar ratio. The improvedactivity of Fe/Zn = 2:1.5 benefits from the synergistic effects of the sp2 hybridized carbon and porousframework, as well as the surface oxygenic functional groups, which accelerate the pollutant/oxidant dispersionand electron transfer. Electron paramagnetic resonance results indicate that OH, 1O2 and SO4radicals account for the catalytic degradation of SMM and the activation of PS in present system is differentfrom conventional homogeneous systems, and speculate mechanism was proposed based on theobtained data.
