        한국인 A 형 간염 환자에서 A 형 간염 바이러스의 유전자형에 관한 연구

        $quot;목적: 한국에서 과거 A형 간염은 낙후된 개인위생 및 자연환경으로 유소아기에 불현성 감염으로 앓고 지나가게 되어 청소년 및 성인에서 HAV에 대한 항체 보유율이 매우 높고 성인에서의 급성 A형 간염은 매우 드물게 보고되었다. 그러나 고도의 경제성장과 더불어 식생활, 위생상태의 개선, 상하수도의 보급 등으로 유소아기의 A형 간염 이환율이 현격히 감소하면서 청소년 및 성인에서 HAV 항체가 없는 인구집단 즉 HAV에 감염되면 현증 간염이 발생할 수 있는 인구 집단이 증가되어 최근 수 년 사이에 현증 A형 간염이 폭발적으로 증가하게 되었다. 이에 저자는 HAV의 유전자형을 알아보고 이미 보고된 각국의 HAV와 연관성을 알아보고자 본 연구를 시행하였다. 대상과 방법: 1994년부터 1998년까지 한국에서 산발적으로 발생한 18예의 A형 간염 환자의 혈청에서 HAV RNA를 추출, 역전사-중화효소 연쇄반응법(RT-PCR)으로 증폭하여 직접 염기서열의 분석을 통해 국내 HAV 사이에 염기서열, 아미노산의 차이 등을 알아보고 세계 각국에서 보고되었던 HAV 분리주들과 비교하였다. 결과: HAV 게놈의 특정부위인 VP1/2A 연결부위 168 bp 길이에서 시행한 염기서열 분석 결과 한국에서 분리된 18예의 HA이러스는 최근 해외에서 유입된 바이러스일 가능성은 적으며 따라서 장기간 국내에서 토착화된 바이러스일 것으로 생각된다. $quot;Background/Aims: Studies of genotypes and changes in nucleotide and amino acid sequences of hepatitis A virus (HAV) may provide valuable information on the epidemiological aspects of a particular region. In Korea the prevalence of anti-HAV in the 1 - 20 year age group declined from 60% in 1980 to 9% in 1995. As a result this age group has a high risk of HAV infection. Actually over 1,500 cases of clinically overt hepatitis A occurred in 1998 while few cases of clinical hepatitis A had been reported until the early 1990s. The aims of this study are to determine the genotypes of HAV which have been circulating in Korea and to define the phylogenetic relationships of geographically defined isolates. Method: From 1994 - 1998 a total of 18 serum specimens was obtained from patients in Korea with sporadic form acute hepatitis A. The HAV nucleic acid from serum specimens was subjected to genomic sequence analysis following viral RNA extraction, reverse transcription and amplification of the cDNA by PCR. DNA sequencing was performed in both directions of each PCR product. Results: All isolates clustered within the subgenotype IA irrespective of the geographic locations and timing of the clinical hepatitis. Among 18 Korean isolates, 9 isolates had 2 amino acid sequence changes and 2 isolates had 1 amino acid sequence change. These changes in the amino acid sequences are unique and have never been reported in HAV subgenotype IA. Conclusions: All isolated HAV had genotype (IA). Eleven of 18 isolates had unique changes in amino acid sequences. These data indicate that the endemic HAV has been circulating in Korea over a long period of time.

        This study was intended to present the proper operational scheme and present the criterion for establishment of proper convenient facilities and its planning method and the proper method of laying all sorts of teaching aids out. For this purpose, it attempted to investigate the type of crippled disorder and the behavioral characteristics of schoolchildren with crippled disorder with a focus on the descriptive study on disabled schoolchildren receiving complete integrated education. And it sought to analyze the present situation of complete integrated education and the case of integrated education and dissect the realities of its operation and the realities of the establishment of convenient facilities for disabled schoolchildren with a focus on the survey research.

        Linoleic acid 함량과 ω6/ω3 비율이 침수속박 스트레스에 의한 위궤양의 발병에 미치는 영향

        This study was designed to determine the effects of linoleic acid contents and ω6/ω3 ratios on the induction of gastric ulcer by water immersion and restraint stress. Sprague-Dawley rats were fed 5 diets containing 7% fat (w/w) for 6 weeks. These diet groups were Lh, Mh, Hh, Mm, Ml, :3 different linoleic acid levels(0.3% of energy(L), 3.5(M), 10(H)) and 3 different ω6/ω3 ratios(11(l), 33(m), 100(h)) with beef tallow, sunflower or fish oil. The Lh group showed a significantly higher ulcer index (UI) than the Mh and Hh groups(p<0.05). At the same linoleic levels, the UI had no significant difference within the ω6/ω3 ratios. The Mh group showed significantly higher(p<0.05) PGE₂ and TBX₂ content than any other group. Pearson's correlation coefficients between UI and PGE₂ and TBX₂ had a negatively significant correlation(p<0.05). Linoleic acid of gastric mucosal phospholipids was reflected by the diet, but was not significantly different. The most significant finding of this study is that not only the absolute amount of linoleic acid, but also the ω6/ω3 ratios are important factors for the prevention of gastric ulcer.

      • 졸-겔공정을 이용한 초고용량 캐패시터용 Nickel oxide 나노입자의 합성

        The Ni (OH)₂ nanoparticles with average diameter of 50 nm were synthesized by hydrolyzing nickel acetate with water, and were dispersed in the distilled water to prepare the coating solution. The electrodes for supercapacitor were prepared by dip-coating the coating solution on the Ni-foam. The electrodes heat-treated at several temperatures showed the highest specific capacitance, 160 F/g, at 250℃. It can be explained that the Ni(OH)₂ particles heat-treated at 100 and 200℃ on the electrodes contained the impurities such as acetate groups in the pores, and resulted in low specific surface areas. However, as the calcination temperature increases. the impurities incorporated in the pores are removed at 250℃, and thus, the particles on the electrodes show the highest specific surface area (177 ㎡/g).

        This study was formed the basis of propelling for being closely connecting between School PE(Physical Education) and Local Society Exercise for regarding the cooperation possibility depending on the actual situation. From this, it is helpful for evaluating the total understanding of the present condition form School PE and Local Society Exercise and grasping the realities concretely. To gain the above result, the subject of study is on 48 agencies among 50 agencies that include 30 elementary and middle schools, 15 government offices, 5 YMCA offices. After investigating the plan of providing physical activity program except result PE class and the connecting program with School PE and its activation, it comes to the conclusion below this. There is the period of possible time for propelling the cooperating model at School and Local Society PE which is special activity class after school, club activity class, discretional activity class and the vacation, weekends. And also there are many participating agencies that are school, local society, public sports center and community, the youth group, local autonomous entity PE post, prfessional physical association and community, city and countries Education Office and the sports-lover's group etc. So far, the connecting program is operated at 24 agencies which are 3 elementary school(30%), 5 middle school(50%), 4 high school(40%)m 7 government office(70%), 5 YMCA office(80%). As the reason of non-operating it, the first is the lack of the charger, spare time and considering teacher's advantage when time is arranged at the teacher and leader's part. The second is the falling behind facility and the lack of place and appliance for practicing. The third is the insufficient of connecting program at program part and increasingly number of students who are individual participating at the side of local society physical agency at school. The forth is the lack of finances for running at economy part and overpaying of using fee. The fifth is the lack of the activity time at the elementary school and putting first in importance the exam for university enterance at middle and high school. The sixth is the parents and principals and school committee have little understanding of the physical educating in common at recognition part. The seventh is the cooperating company's preparation is not enough and incomplete relation each other. Looking into the opinion for activating plan of connecting program which is classified into common contents, the result is presented the same reaction both of them that the professional leader arrangement is needed at teacher and leader's part. At the facility expansion and open program part is required for a various program. In the case of school at economy condition part, more cheaper than expenses for facilities is needed and also the renovation of the education system And at the recognition part, the positive attitude of school is needed. At the safety part, the solution of safety problem for responsibility is needed. At the administration part, the support for professional agency and the change of systemic role for connecting agency.

      • 天然 및 養殖産 뱀장어의 蛋白質 및 아미노酸 組成比較

        우리나라 전역에 걸쳐서 分布하고 있는 뱀장어의 營養學的 基礎資料를 提示하기 위하여 天然産과 養殖産으로 分類하여 蛋白質의 組成, 蛋白質의 아미노산酸 組成을 比較, 分析하였다. 그리고 天然 및 養殖産 뱀장어 肉蛋白質의 組成을 엄밀히 檢討하기 위해 肉蛋白質中 比較的 많은 量을 차지하는 筋形質蛋白質과 筋原纖維蛋白質을 SDS-PAG電氣泳動 分析하여 subunit의 差를 比較하였다. 또한 사료蛋白質의 아미노酸 組成을 分析함으로서 肉蛋白質의 아미노酸 組成에 미치는 영향도 아울러 比較, 檢討하였다. 天然 및 養殖産 뱀장어의 粗蛋白質 合量은 各各 14.3%와 15.4%이었으며, 粗脂肪은 25.0%와 29.7%로서 養殖産 뱀장어의 脂肪 含量이 天然産에 比하여 높은 편이있다. 肉蛋白質을 構成하는 蛋白質 組成은 天然産의 경우 筋形質蛋白質이 30.78%, 筋原纖維蛋白質이 59.02%, 細胞內殘渣蛋白質이 9.73%, 基質蛋白質이 2.47%이었으며, 養殖産은 筋形質蛋白質이 31.81%, 筋原纖維蛋白質이 58.37%, 細胞內殘渣蛋白質이 8.16%, 基質蛋白質이 1.80%를 차지하여 天然産과 養殖産뱀장어의 肉蛋白質 組成은 비교적 유사함을 알 수 있었다. 筋形質蛋白質과 筋原纖維蛋白質 分劃의 一部에 대하여 SDS-PAG電氣泳動 分析을 실시한 結果, 天然産 뱀장어의 筋形質蛋白質은 16개의 subunit로 構成되어 있었고, 養殖産은 分子量이 36,500, 46,000, 58,500, 75,000, 170,000, 235,000에 해당하는 4개의 subunit가 더 많은 22개의 subunit로 構成되어 있었다. 한편 筋原纖維蛋白質은 天然産이 14개의 subunit이었으며, 養殖産은 分子量 45,000에 해당하는 未知의 subunit가 하나 더 많은 15개로 이루어져 있었다. 肉蛋白質의 蛾眉노산 組成은 天然産이 養殖産에 比하여 glycine含量이 다소 높은 것을 제외하고는 대체로 비슷하였으며, lysine, aspartic acid, glutamic acid, glycine이 전체아미노酸의 45% 가량을 점하고 있었다. 사료蛋白質의 아미노酸 組成은 뱀장어의 필수아미노산 要求量을 만족시킬 만큼 양호한 組成을 보여주었으며, aspartic acid와 glutamic acid를 제외하고는 魚肉의 축적단백질로서 魚肉蛋白質의 아미노 組成과 비례관계를 갖는 것으로 판단되었다. The muscles of wild and cultured eel, Anguilla japonica, were analyzed for the protein composition and amino acid profile. The differences of the subunit distribution for the sarcoplasmic and myofibrillar proteins were discussed with sodium dodecylsulfate(SDS) polyacryamide gel electrophoresis. The muscle protein in wild eel was composed 30.78% of sarcoplasmic, 59.02% of myofibrillar, 9.73% of residual intracellular and 2.47% of stroma fraction. That in cultured eel was composed of 31. 81%, 58.37%, 8.16% and 1.80%, respectively. The sarcoplasmic and myofibrillar proteins were composed of 16 and 14 subunits in wild eel, and 22 and 15 subunts in cultured eel. The sarcoplasmic protein between wild and cultured muscles showed a similar trend in the subunits, except a few subunits such as 36,500, 46,000, 58,500, 75,000, 170,000 and 235,000 daltons in cultured eel. Only the existence of 45,000 dalton subunit was the difference between wild and cultured eel in myofibrillar protein. The distribution patterns of total amino acid in muscles of wild and cultured eel were found to be very similar trend, although glycine content in wild eel was slightly higher than that in cultured one.

        정상인의 체위 변화시 Bioimpedance 측정법을 이용한 혈역학적 변화의 관찰

        Introduction : The purpose of positioning is to allow the procedure to be performed as simply as possible and to achieve the best possible results free of complications. In a normal person, many investigators have researched the hemodynamic changes during positional adjustments. But the delicate qualitative hemodynamic changes in various positions has not been recorded very much. So we studied the qualitative hemodynamic changes in various positions with a noninvasive ioimpedance method. Method : Healthy adult males were studied (n=25). The protocol was that the position changed from supine to head-up, head-down, lithotomy, sitting, right-lateral, prone and Jackknife (J-K) position. Each position was sustained for three minutes. The mean arterial pressure (MAP), heart rates (HR), left cardiac work index (LCWI), systemic vascular resistance index (SVRI), cardiac index (CI), stroke index (SI) and end diastolic index (EDI) were measured with the bioimpedance method and an automated blood pressure device. Result : The contractility of heart expressed by LCWI, CI and SI decreased in head-down, sitting, right-lateral, prone and J-K positions. The preload expressed by EDI decreased in right-lateral and J-K positions. The afterload expressed by SVRI increased in sitting, prone and J-K positions, and decreased in right-lateral position. Conclusions : These results may be used as reference for anesthesiologist managing many patients of various physical status. (Korean J Anesthesiol 1998; 35: 710∼715)

        Reactive oxygen species-dependent activation of Bax and poly(ADP-ribose) polymerase-1 is required for mitochondrial cell death induced by triterpenoid pristimerin in human cervical cancer cells.

        <P>Naturally occurring triterpenoid compounds have long been used as anti-inflammatory, antimalarial, and insecticidal agents. It has become evident that some of the natural or synthetic triterpenoids have promising clinical potential as both a therapeutic and chemopreventive agent for cancer. However, the molecular basis for the antitumor activity of triterpenoid has yet to be defined. In this study, we show that pristimerin, a natural triterpenoid, induces mitochondrial cell death in human cervical cancer cells and that reactive oxygen species (ROS)-dependent activation of both Bax and poly(ADP-ribose) polymerase-1 (PARP-1) is critically required for the mitochondrial dysfunction. We also showed that c-Jun N-terminal kinase (JNK) is involved in ROS-dependent Bax activation. Treatment of pristimerin induced an increase in intracellular ROS, JNK activation, conformational change, and mitochondrial redistribution of Bax, mitochondrial membrane potential loss, and cell death. The PARP-1 was also found to be activated by pristimerin treatment. An antioxidant, N-acetyl-l-cysteine (NAC), inhibited pristimerin-induced JNK activation, Bax relocalization, and PARP-1 activation, as well as mitochondrial cell death. Moreover, inhibition of JNK clearly suppressed conformational change and mitochondrial translocation of Bax and subsequent mitochondrial cell death but did not affect PARP-1 activation. Inhibition of PARP-1 with 1,5-dihydroxyisoquinoline (DIQ) or with small interfering RNA of PARP-1 significantly attenuated pristimerin-induced mitochondrial membrane potential loss and cell death but did not affect JNK activation and Bax relocalization. These results indicate that the natural triterpenoid pristimerin induces mitochondrial cell death through ROS-dependent activation of both Bax and PARP-1 in human cervical cancer cells and that JNK is involved in ROS-dependent Bax activation.</P>
