      • 腎皮質 S-Adenosyl-L-Methionine : Protein-Carboxyl O-Methyltransferase에 關한 硏究 Protein-Carboxyl O-Methyltransferase from Bovine Renal Cortex

        S-Adenosyl-L-methionine : protein-carboxyl 0-methyltransferase (EC 2.1. 1. 24, protein methylase II) was purified from bovine renal cortex approximately 6, 780-fold with a 9% yield by combination of ammonium sulfate precipitate, Sephadex G-75 chromatography, S-adenosyl-L-homocysteine-sepharose 4B chromatography and hydroxyapatite chromatography, and then its properties and physiological role were investigated. 1. The enzyme showed a pH optimum around 5.6-6. 0. It is heat labile, inactivated by heattreatment for 3 minutes at 60℃, and when stored at -20℃ in the presence of 10% glycerol, its activity has been almost stable in a month. 2. Copper ion (Cu^2+}) and zinc ion (Zn^2+) were a potent inhibitors, the activity being completely inhibited at 2 mM. The inhibition of copper and zinc ions were recovered 85%and 63% of its activity adding of 4 mM EDTA, respectively. The inhibition of copper ion was completely recovered by adding of 10 mM mercaptoethanol, but zinc ion did not almost affect. 3. The apparent Km value for S-adenosyl-L-methionine was 4 x 10^-6M. Kinetic analysis of the enzyme in the presence of 25 uM copper ion showed that the nature of the inhibition to the enzyme was noncompetitive, considering that Km was constant and Vmax was decreased, and 50uM zinc ion showed that it was uncompetitive, considering that both Km and Vmax were decreased. 4. Immunoglobulin A, histone II and histone H3 were relatively good substrates for the enzyme. 5. The enzyme activity was completely inhibited by 0.4 mM of p-hydroxymercuribenzoic acid, but all of the activity was recovered in adding 10 mM mercaptoethanol, and the molecular weight of the enzyme was 24, 000. 6. The carboxylmethylated fractions of the brush-border membrane of bovine renal cortex had molecular weight of 70,000, 67,000, 45,000, 40,000, 31,000 and 27,000 etc. (6 fractions), as determined by SDS/polyacrylamide gel electrophoresis. 7. The carboxylmethylation of the brush-border membrane increased with time, S-adenosylmethionine, substrate concentration, and methylated brush-border membrane was demethylated by 50% for 10 minutes at pH 7.4. 8. D-Glucose uptake was inhibited by carboxylmethylation of brush-border membrane. These results suggest that protein methylase II may play some roles in regulating physiological function of brush-border membrane.

        In this paper, reflex RGB data captured from WebCam is stored to HDD through USB port at every 5 seconds intervals. Also, if strangers are detected through Motion Detection routine, warning voice message is broadcasted and invasion message is transmitted by e-mail and transmit the e-mail title to mobile phone through WAP(Wireless Application Protocol) Push. The detected image is stored to hard-disk in 'month - day - hour - minute - second . jpg' data type. And the image data is transmitted to web server through FTP(File Transfer Protocol) because invader can deletes or destroys the image data on hard-disk. We implement a surveillance system which is able to utilize through internet regardless of time and space.

      • 精神分裂症의 生物學的 硏究 : 液體 크로마토그라프와 放財酵素法에 依한 Catecholamines 微量定量 A Rapid Microassay of Catecholamines by High Performance Liquid Chromatography-Radioenzymatic Method

        A direct rapid radioenzymatic assay in combination with high performance liquid chromatography (HPLC) for measuring simultaneously picogram quantities of nore pinephrine, epinephrine and dopamine has been developed, and the relationship of changes in cerebrospinal fluid (CSF) norepinephrine concentration during haloperidol treatment to changes in psychosis rating of twelve schizophrenic patients has been investigated. The three catecholamines in 20?l of CSF are converted to their 0-methylated ana-logues by catechol-0-methyltransferase in the presence of S-adenosyl-L-1{methyl-3H} methionine. After the addition of tetraphenylborate, these derivatives are extracted. with diethylether at pH 8.2 followed by re-extraction with 0.1N HCI and then isol-ated by HPLC using, ?Bondapak C18column and 0.17 M acetic acid-60% methanol- 0.02 M Na2SO4 as solvent. Manual collection of each fraction is started two seconds after in appeared mark. The HPLC fraction of norepinephrine and epinephrine deri-vatives are oxidized to vanillin followed by extracting the vanillin with toluene, and dopamine derivatives fraction is extracted with toluene/isoamylalcohol and then re-extracted with 0.1N HCl. Internal standard curve shows a good linearity from 5 to 30 picogram per samples, and incorporated radioactivity of norepinephrine, epinephrine and dopamine derivatives are 163±11.4, 125±10.5 and 112±3.9cpm per picogram and calculated sensitivities are 0.4, 0.3 and 1 picogram, respectively. 3H-Normetanephrine, 3H-metanephrine and 3H-methoxytytamine are well separated by HPLC, and each retention times are 3.52, 4.08 and 5.62 minutes and yields are 75.6?0.9, 76.1?0.8 and 70.1?0.4%. Separation of the methylated derivatives by HLPC are performed within 8 minutes, and thus enabled us to complete the differential analysis of 15 samples (30 tubes) in a day. CSF norepinephrine concentration in schizophrenic patients show rostral-caudal gradient and haloperidol induced decreases of norepinephrine and the decreases correlated significantly with psychosis rating, From the above rsults, it is suggested that this method is more excellent than any others in sensitivity, accuracy, reproducibility and specificity of the differential assay, and CSF norepinephrine can be used for evaluation of neuroleptics treatment of schizophrenia by this method.

      • Polyvinylfluorocinnamate 광배향막 표면에서의 액정배향에 관한 연구

        광배향에 의하여 액정의 방향자가 유도되는 메카니즘을 이해하기 위하여 광반응성 고분자인 polyvinylfluorocinnamate의 UV 조사에 따른 구조변화를 FTIR과 UV/Vis spectroscopy를 이용하여 조사하였다. 선편광이 된 UV를 등방성의 고분자 필름에 조사하여 필름 내에 광학적 anisotropy가 형성되는 것을 확인하였고 이러한 배향막의 anisotropy에 의하여 액정의 방향자가 결정되는 것을 알 수 있었다. 광조사에 의하여 UV의 편광방향과 수직으로 늘어서게 된 분자 사슬내의 group들이 분산력에 의하여 액정분자의 배향을 조사된 UV의 편광방향과 수직인 방향으로 유도하는 것이다. In order to understand the induction mechanism of the liquid crystal director, structural changes of photosensitive polyvinylfluorocinnamate with UV irradiation were investigated using FTIR and UV/Vis spectroscopy. It was found that optical anisotropy was induced in the film by irradiation of linearly polarized UV light onto the isotropic polymer film, and such anisotropy in the film determined the direction of the liquid crystal director. The groups in the polymer, which are preferentially aligned perpendicular to the polarization direction after the UV exposure, induce liquid crystal alignment perpendicular to the polarization direction of UV via dispersive intermolecular interactions.

        There are many theoretical indices for chemical carcinogenicity. Among the various indices, the LUMO electron density and approximate superdelocalizability have taken to compared and discussed for the carcinogenicity of PAHs, aromatic amines and Azo-compounds. The sum of LUMO electron density of the trans-butadiene type 1, 2, 4 positions in a molecule has been known to be in more agreement with the observed values than those of approximate superdelocalizability. It is suggested that any trans-butadiene type 1, 2, 4 positions play an importmant role in the elucidation of mechanism of chemical carcinogenesis.

      • 發癌物質과 非發癌物質의 DNA-Modifying Activity에 대한 量子生物學的 解釋

        LCAOMO calculation has been carried out for thirty chemical carcinogens and nine metabolites of them. The LUMO electron density which is a measure of ability electron to be accepted and observed DNA-Modifying activity which is a measure of mutation relationship was considered. It was found that the sum of LUMO electron density of 1, 2, 4-positions of a relevant trans-butadiene type (F??) in the compound was correlative to DNA-Modifying activity. This means that carcinogenesis closely relate to mutagensis, since the LUMO electron density is a measure of carcinogenic activity of carcinogens as shown earlier. It was also found that LUMO electron density of 1, 2, 4-positions in compounds increase with their metabolic activation. This is in agreement with the experimental result that carcinogen is more electrophilic as metabolic activation. In conclusion, it may be possible to give a theoretical interpretation on the mutation of chemical carcinogens and explain theoretically the fact that the compound have a strong elestrophilicity on metabolic activation.

        전위된 관절내 원위 요골의 관절경을 이용한 고정술

        목적: 전위된 관절내 원위 요골 골절에 관절경을 이용한 정복술과 경피적 K 강선 및 외고정기기를 이용한 수술법의 결과를 알아보고자 함이다. 대상 및 방법: 2001년 1월부터 8월까지 수술한 11명 12예를 대상으로 하였으며, 평균 추시 기간은 14개월 이었다. 골절의 형태는 AO 분류에 의하면 1예는 B3형, 4예는 C2형, 7예는 C3형 골절이었다. 전예에서 2.7㎜ 관절경을 이용하였으며 추가적인 관혈적 정복은 시행하지 않았다. 경피적 K 강선 고정술 후 외고정 기기를 사용하였다. 관절내 골연골 골편을 제거하였으며 추가적인 수근 골간 인대와 삼각 섬유 연골 손상의 적극적인 처치는 없었다. 고정물 제거 시기는 경피적 K 강선은 술후 4주, 외고정 기기는 7주에 시행하였다. 결과: 평균 수근 관절의 신전 및 굴곡은 정상의 72%였다. 평균 요측 길이는 12.3 ㎜, 전방경사는 5.7°, 척측 경사는 22.4°였다. 평균 관절면의 불일치는 1㎜였다. 술후 구획 증후군의 발생은 없었으며 치료 중 골절 함몰 소견은 없었다. 임상적으로 Gartland와 Werley의 평가에 의하면 8예에서 우수, 3예에서 양호, 1예에서 보통의 결과를 얻었다. 결론: 관절내 분쇄 골절의 치료에서 관절경을 이용한 관절면 정복술은 수술적 연부 조직을 최소화하면서 관절면을 비교적 정확하게 정복함으로 관절면의 불일치를 감소시켜 손상 후 좋은 결과를 얻을 수 있는 효과적인 방법으로 사료된다. Purpose: To investigate the result of arthroscopically assisted reduction of intraarticular fracture of distal radius with percutaneous K-wires and external fixation. Materials and Methods: We reviewed 12 intraarticular distal radial fractures of 11 patients treated arthroscopically assisted reduction in Orthopedic Department of Sung-Ae hospital between January, 2001 and August, 2001. The mean length of follow-up was 14 months. Analysis of them revealed that B3 was 1 case, C2 4 cases and C3 7 cases according to the AO classification. All cases were treated by 2.7 ㎜ arthroscopic devices and percutaneous K-wires pining with external fixationg, but additionally invasive reductional technique was not used. We removed the osteochondral flap but not treated. The K-wires were removed at 4 weeks, external fixation was 7 weeks, respectivley. Result: The mean active range of movement in the affected wrist was total arc of the flextion-extension 72% of the opposite side. Radiographically mean volar tilt, radial inclination and radial length compartment syndrome and fractures collapse were not occured during follow-up period. According to the Gartland and Werley demerit-point system, eleven cases were a excellent or good one was a fair result. Conclusion: Arthroscopically assisted fixation is a useful method for reducing the soft tissue injuies and preventing the articular surface incogruency by anatomically reduction un cases of intraarticular comminution.

      • ICR계 마우스에서 홍화씨 에탄올 추출물의 급성독성시험

        The present study was carried out to investigate the potential acute toxicity of safflower seed by a single oral dose in ICR mice. Twenty animals of each sex were randomly assigned to four groups of 5 rats each and were administered singly by gavage at dose levels of 0, and 2,500 mg/kg bodyweight. Mortalities, clinical findings, and body weight changes were found and complete gross postmortem examinations were performed. Throughout the study period, no treatment-related deaths were observed. There were no adverse effects in clinical signs, body weight, and gross findings at any dose tested. The results showed that the single oral administration of safflower seed did not cause any toxic effect at the dose levels of 2,500 mg/kg in rats and the minimal lethal dose was considered to be over 2,500 mg/kg body weight for both sexes.
