      • Genetic polymorphism of DOCK8, IL17RA and KLK12 in Koreans with atopic dermatitis and allergic march

        Background: Atopic dermatitis (AD) is one of the most common childhood diseases and has a complex etiology involving genetic and environmental factors. Thus, a broad understanding of genetic background is needed for early diagnosis of AD. Objectives: The purpose of this study was to identify of novel candidate functional genetic variants in Koreans with atopic dermatitis and allergic march. Methods: Whole-exome sequencing (WES) was performed in 20 AD and 20 AM Korean patients, and 40 Korean personal genome project (KPGP) controls. Sanger sequencing was carried out to validate found variants in 86 AD, 72 AM patients and 81 controls. Results: Three candidate variants of DOCK8 (rs529208), IL17RA (rs1248468), and KLK12 (rs3745540) were identified. rs529208 had no correlation with the development of AD but the AA genotype of DOCK8 had significantly increased total IgE level. rs1248468 had significantly increased risk of AD and the CA genotype of IL17RA had also increased total IgE level. rs3745540 was significantly associated with AD. Conclusion: DOCK8 (rs529208), IL17RA (rs1248468) and KLK12 (rs3745540) were deemed functionally interesting based on WES. Our case-control study suggests that the three variants detected in WES may enhance the risk of AD.

        IRAK4 as a Molecular Target in the Amelioration of Innate Immunity–Related Endotoxic Shock and Acute Liver Injury by Chlorogenic Acid

        <P>Mice lacking the IL-1R-associated kinase 4 (IRAK4) are completely resistant to LPS-induced endotoxic disorder or the TLR9 agonist CpG DNA plus D-galactosamine-induced acute liver injury (ALI), whereas wild-type strains succumb. However, translational drugs against sepsis or ALI remain elusive. Lonicerae flos extract is undergoing the clinical trial phase I in LPS-injected healthy human volunteers for sepsis treatment. In the current study, chlorogenic acid (CGA), a major anti-inflammatory constituent of lonicerae flos extract, rescued endotoxic mortality of LPS-intoxicated C57BL/6 mice, as well as ameliorated ALI of LPS/D-galactosamine-challenged C57BL/6 mice. As a mechanism, CGA inhibited various TLR agonist-, IL-1 alpha-, or high-mobility group box-1-stimulated autophosphorylation (activation) of IRAK4 in peritoneal macrophages from C57BL/6 or C3H/HeJ mice via directly affecting the kinase activity of IRAK4, a proximal signal transducer in the MyD88-mediated innate immunity that enhances transcriptional activity of NF-kappa B or AP-1. CGA consequently attenuated protein or mRNA levels of NF-kappa B/AP-1 target genes encoding TNF-alpha, IL-1 alpha, IL-6, and high-mobility group box-1 in vivo under endotoxemia or ALI. Finally, this study suggests IRAK4 as a molecular target of CGA in the treatment of innate immunity-related shock and organ dysfunction following insult of various TLR pathogens from bacteria and viruses.</P>

        인간유방암 MDA-MB-231세포에서 peptide H에 의한 TNFα 발현 억제

        청국장은 다양한 peptide류를 포함한다. 청국장 유래의 peptide H를 인간유방암 MDA-MB-231 세포에 처리했을 때, $TNF{\alpha}$ 발현은 뚜렷하게 억제되었다. $TNF{\alpha}$에 의해 유도되는 IL6 발현 역시, peptide H에 의해 감소될 수 있음을 시사해 준다. Peptide H구조는 glucocorticoid, dexamethasone과 전혀 유사하지 않아 그들과 다른 기작으로 $TNF{\alpha}$ 발현억제에 작용할 것을 시사해 준다. Peptide H는 $TNF{\alpha}$ 발현 억제 효과가 있으므로 보다 깊이 있는 연구를 바탕으로, 류마티스 관절염, 크론병 등의 치료제로 개발될 수 있기를 기대해 본다. Chungkookjang (fermented soybeans) contains diverse peptides. Human breast cancer MDA-MB-231 cells were treated with peptide H derived from Chungkookjang, and $TNF{\alpha}$ expression in the cells was conspicuously repressed, suggesting peptide H's regulation of IL6 since IL6 is induced by $TNF{\alpha}$. The structure of peptide H was different from those of glucocorticoid and dexamethasone, suggesting different mechanisms of $TNF{\alpha}$ expression suppression. Peptide H which reduces $TNF{\alpha}$ expression can be developed as drugs for cancer, rheumatoid arthritis and Crohn's disease, after more investigation.

      • Association of Increased Pulmonary Interleukin-6 with the Priming Effect of Intra-Amniotic Lipopolysaccharide on Hyperoxic Lung Injury in a Rat Model of Bronchopulmonary Dysplasia

        <P><I>Background:</I> The authors previously demonstrated the priming effect of intra-amniotic lipopolysaccharide (LPS) on hyperoxic lung injury in a rat model of bronchopulmonary dysplasia (BPD). <I>Objectives:</I> To investigate the mechanism underlying this priming effect by determining biochemical profiles in a rat model of BPD. <I>Methods:</I> The rat model involved intra-amniotic LPS administration and postnatal hyperoxia (85%). The mRNA expressions of <I>interleukin-6 (IL-6), vascular endothelial growth factor (VEGF), VEGF receptor-2 (VEGFR-2), basic fibroblast growth factor (bFGF),</I> and <I>transforming growth factor </I>β<SUB><I>1</I></SUB><I> (TGF-</I>β<SUB><I>1</I></SUB><I>),</I> as well as the protein levels of IL-6, VEGF, and protein carbonyl in lung tissue were compared between the LPS plus hyperoxia, the LPS only, the hyperoxia only, and the control groups. <I>Results:</I> Morphometric analysis of lung tissues demonstrated that alveolarization was significantly inhibited only in the LPS plus hyperoxia group. IL-6 protein levels and its mRNA expression in the lungs were significantly increased only in the LPS plus hyperoxia group. Neither LPS nor hyperoxia increased IL-6 in the lungs independently. <I>bFGF</I> mRNA expression was significantly decreased in the LPS-treated groups. VEGF protein levels were significantly reduced by hyperoxia, whereas protein carbonyl levels were increased by intra-amniotic LPS or hyperoxia. No additional significant change to VEGF or protein carbonyl levels was produced by intra-amniotic LPS or hyperoxia. There were no significant differences in the mRNA expressions of <I>VEGF, VEGFR-2,</I> and <I>TGF-</I>β<SUB><I>1</I></SUB>. <I>Conclusions:</I> The priming effect of intra-amniotic LPS on hyperoxic lung injury may be associated with IL-6 elevation in the lungs.</P><P>Copyright © 2009 S.         The purpose of this paper is to verify the development and operation process of Information System will affect the system performance. SDLC(System Development Life Cycle) is divided into four steps, that is analysis, design, implementation and maintenance. The operation will be done after the SDLC. It has been claimed that user participation, executive support and user training are considered as important factors for success during the each step of SDLC. But few attempts have been made to prove it. So the field study was used for the research purpose. The conclusion to be drawn here are these: 1) each step of SDLC has a significant effect on the following step and system success, 2) documentation has a significant effect on the development and operation process, 3) user participation have a significant effect on the maintenance and system success, 4) executive support has a significant effect on the implementation process, 5) User training has a significant effect on the system success, 6) system development process has a significant effect on the operation.

        The purpose of this study is to confirm whether the fitness between the interdependence and the information characteristics makes any effects on the user satisfaction by the computer literacy. The information characteristics is divided into the degrree of aggregation, and scope. User satisfaction was used as a surrogate measure of the performance of information systems. The computer self-efficacy was used to control any confounding effects. Field research method using questionnaires was employed to empirically investigate the relationship among the variables. Findings of the study are summarized as follows the fitness between the interdependence information turned out to have significant effects on information system success. The implication of such results is that under the environment of highly interdependence, more-aggregated and broad-scope information is appropriate.

        PWM converter has better power factor and low harmonic components than conventional diode rectifiers and phase controlled rectifier. Also the DC link voltage can be regulated fast and stable against the variation in the load. In order to regulate for DC link voltage, rapid current control is need for source current control. This paper focuses on the improvement of quickness of current controller to know the transient state characteristics of regulated DC link voltage against the variation in the load. To do this, it is adopted to three phase voltage type PWM converter with space vector pulse width modulation(SVPWM) method and synchronously rotating d, q axis current. Each d, q axis current is controlled independently. To control d, q axis current, Current control methods to be applied in system are PI current control and Predictive current control respectively. As a result, Transient response characteristic of Predictive current controller is superior to PI current controller.

        Hemifacial microsomia(HFM) is a craniofacial disorder and complex malformation syndrome with a variable and asymmetric malformation involving first and second pharyngeal arch derivatives. Multiple etiological mechanisms for HFM have been considered. Mechanisms proposed include genetic factors, early embryogenesis insults involving abnormal neural crest development and migration, and stapedial artery hematoma. HFM is the second most common craniofacial malformation but has been rarely reported in korea, and we report five cases of HFM in temporal bone CT findings. The temporal bone CT findings of five patients aged 6 days and 19 years(mean, 9 years) with hemifacial microsomia were reviewed and evaluated. Particular attention was focused on auricular malformation, external auditory canal, volume of middle ear cavity, ossicular anomaly, facial nerve anomaly and inner ear anomaly. In all patients, temporal bone CT scans revealed the stenosis or atresia of external auditory canals, decreased middle ear cavities, matoid underdevelopment and ossicular anomalies including ossicular ankylosis, but only one patient showed facial nerve or inner ear anomaly. In conclusion, temporal bone CT scan is a useful diagnosis modality of hemifacial microsomia, which is especially more useful in preoperative evaulation and estimation of postoperative prognosis that demonstate middle and inner ear anomalies.
